1Three-dimensional-printed individualized porous implants:A new“implant-bone”interface fusion concept for large bone defect treatment显示文摘Bone defect repairs are based on bone graft fusion or replacement.Current large bone defect treatments are inadequate and lack of reliable technology.Therefore,we aimed to investigate a simple technique using three-dimensional(3D)-printed individualized porous implants without any bone grafts,osteoinductive agents,or surface biofunctionalization to treat large bone defects,and systematically study its long-term therapeutic effects and osseointegration characteristics.Twenty-six patients with large bone defects caused by tumor,infection,or trauma received treatment with individualized porous implants;among them,three typical cases underwent a detailed study.Additionally,a large segmental femur defect sheep model was used to study the osseointegration characteristics.Immediate and long-term biomechanical stability was achieved,and the animal study revealed that the bone grew into the pores with gradual remodeling,resulting in a long-term mechanically stable implant-bone complex.Advantages of 3D-printed microporous implants for the repair of bone defects included 1)that the stabilization devices were immediately designed and constructed to achieve early postoperative mobility,and 2)that osseointegration between the host bone and implants was achieved without bone grafting.Our osseointegration method,in which the“implant-bone”interface fusion concept was used instead of“bone-bone”fusion,subverts the traditional idea of osseointegration.Teng Zhang Qingguang Wei Hua Zhou Zehao Jing Xiaoguang Liu Yufeng Zheng Hong Cai Feng Wei Liang Jiang Miao Yu Yan Cheng Daoyang Fan Wenhao Zhou Xinhong Lin Huijie Leng Jian Li Xinyu Li Caimei Wang Yun Tian Zhongjun Liu 2021Bioactive Materials2021,6,11:9

7Microtubule-bundling protein Spef1 enables mammalian ciliary central apparatus formation显示文摘Cilia are cellular protrusions containing nine microtubule(MT)doublets and function to propel cell movement or extracellular liquid flow through beating or sense environmental stimuli through signal transductions.Cilia require the central pair(CP)apparatus,consisting of two CP MTs covered with projections of CP proteins,for planar strokes.How the CP MTs of such‘9+2’cilia are constructed,however,remains unknown.Here we identify Spef1,an evolutionarily conserved microtubule-bundling protein,as a core CP MT regulator in mammalian cilia.Spef1 was selectively expressed in mammalian cells with 9+2 cilia and specifically localized along the CP.Its depletion in multiciliated mouse ependymal cells by RNAi completely abolished the CP MTs and markedly attenuated ciliary localizations of CP proteins such as Hydin and Spag6,resulting in rotational beat of the ependymal cilia.Spef1,which binds to MTs through its N-terminal calponin.homologous domain,formed homodimers through its C-terminal coiled coil region to bundle and stabilize MTs.Disruption of either the MT-binding or the dimerization activity abolished the ability of exogenous Spef1 to restore the structure and functions of the CP apparatus.We propose that Spefl bundles and stabilizes central MTs to enable the assembly and functions of the CP apparatus.Jianqun Zheng Hao Liu Lei Zhu Yawen Chen Huijie Zhao Wei Zhang Fan Li Lele Xie Xiumin Yan Xueliang Zhu 2019Journal of Molecular Cell Biology2019,11,1:1
8Robust boron nanoplatform provokes potent tumoricidal activities via inhibiting heat shock protein显示文摘Near-infrared(NIR)-light-triggered photothermal therapy(PTT)is a promising treatment for breast cancer.However,its therapeutic efficiency is often compromised due to the heatinduced up-regulation of heat shock proteins,which confer photothermal resistance.To solve this urgent problem,PEGylated two-dimensional boron nanosheets(B-PEG)-which allow both multimodal imaging and photothermal conversion-were loaded with gambogic acid(GA),which can inhibit heat shock protein 90(Hsp90).Experimental findings indicated that this combination of B-PEG and GA could serve as an integrated drug delivery system for cancer diagnosis and treatment.It could be used to administer mild PTT as well as chemotherapy for breast cancer,provide improved anti-tumor effects,and reduce the toxicity of PTT,all while inhibiting breast cancer growth.This drug delivery system could offer a novel tool for administering chemotherapy combined with PTT while avoiding the adverse effects of traditional PTT.Yuying Zhao Ning Liu Piaoxue Liu Taojian Fan Rui Ma Huijie Hong Xiaojia Chen Zhongjian Xie Han Zhang Qi Wang Tongkai Chen 2022Asian Journal of Pharmaceutical Sciences2022,17,5:0

10Integrative multiomics analysis identifies a metastasis-related gene signature and the potential oncogenic role of EZR in breast cancer显示文摘Distant metastasis is a major cause of increased mortality in breast cancer patients,but the mechanisms underlying breast cancer metastasis remain poorly understood.In this study,we aimed to identify a metastasis-related gene(MRG)signature for predicting progression in breast cancer.By screening using three regression analysis methods,a 9-gene signature(NOTCH1,PTP4A3,MMP13,MACC1,EZR,NEDD9,PIK3CA,F2RL1 and CCR7)was constructed based on an MRG set in the BRCA cohort from TCGA.This signature exhibited strong robustness,and its generalizability was verified in the Metabric and GEO cohorts.Of the nine MRGs,EZR is an oncogenic gene with a well-documented role in cell adhesion and cell migration,but it has rarely been investigated in breast cancer.Based on a search of different databases,EZR was found to be significantly more highly expressed in both breast cancer cells and breast cancer tissue.EZR knockdown significantly inhibited cell proliferation,invasion,chemoresistance and EMT in breast cancer.Mechanistically,RhoA activation assays confirmed that EZR knockdown inhibited the activity of RhoA,Rac1 and Cdc42.In summary,we identified a nine-MRG signature that can be used as an efficient prognostic indicator for breast cancer patients,and owing to its involvement in regulating breast cancer metastasis,EZR might serve as a therapeutic target.GUODONG XIAO FENG CHENG JING YUAN WEIPING LU PEILI WANG HUIJIE FAN 2022Oncology Research2022,30,1:0

12Application of multidisciplinary in situ simulation training in the treatment of acute ischemic stroke: a quality improvement project显示文摘BACKGROUND:Ischemic stroke refers to a disorder in the blood supply to a local area of brain tissue for various reasons and is characterized by high morbidity,mortality,and disability.Early reperfusion of brain tissue at risk of injury is crucial for the treatment of acute ischemic stroke.The purpose of this study was to evaluate comfort levels in managing acute stroke patients with hypoxemia who required endotracheal intubation after multidisciplinary in situ simulation training and to shorten the door-to-image time.METHODS:This quality improvement project utilized a comprehensive multidisciplinary in situ simulation exercise.A total of 53 participants completed the two-day in situ simulation training.The main outcome was the self-reported comfort levels of participants in managing acute stroke patients with hypoxemia requiring endotracheal intubation before and after simulation training.A 5-point Likert scale was used to measure participant comfort.A paired-sample t-test was used to compare the mean self-reported comfort scores of participants,as well as the endotracheal intubation time and door-to-image time on the fi rst and second days of in situ simulation training.The door-to-image time before and after the training was also recorded.RESULTS:The findings indicated that in situ simulation training could enhance participant comfort when managing acute stroke patients with hypoxemia who required endotracheal intubation and shorten door-to-image time.For the emergency management of hypoxemia or tracheal intubation,the mean post-training self-reported comfort score was signifi cantly higher than the mean pre-training comfort score(hypoxemia:4.53±0.64 vs.3.62±0.69,t=-11.046,P<0.001;tracheal intubation:3.98±0.72 vs.3.43±0.72,t=-6.940,P<0.001).We also observed a decrease in the tracheal intubation and door-to-image time and a decreasing trend in the door-to-image time,which continued after the training.CONCLUSION:Our study demonstrates that the implementation of in situ simulation training in a clinical environment with a multidisciplinary approach may improve the ability and confi dence of stroke team members,optimize the fi rst-aid process,and eff ectively shorten the door-to-image time of stroke patients with emergency complications.Ganying Huang Huijie Yang Huan Yao Xinxin Fan Wenqin Xia Yuansheng Xu Xiaoling Shen Xue Zhao 2024World Journal of Emergency Medicine2024,15,1:0
