| 1 | In vivo theranostics with near-infraredemitting carbon dots—highly efficient photothermal therapy based on passive targeting after intravenous administration显示文摘Carbon dots that exhibit near-infrared fluorescence(NIR CDs)are considered emerging nanomaterials for advanced biomedical applications with low toxicity and superior photostability and targeting compared to currently used photoluminescence agents.Despite progress in the synthesis of NIR CDs,there remains a key obstacle to using them as an in vivo theranostic agent.This work demonstrates that the newly developed sulfur and nitrogen codoped NIR CDs are highly efficient in photothermal therapy(PTT)in mouse models(conversion efficiency of 59%)and can be readily visualized by photoluminescence and photoacoustic imaging.The real theranostic potential of NIR CDs is enhanced by their unique biodistribution and targeting.Contrary to all other nanomaterials that have been tested in biomedicine,they are excreted through the body’s renal filtration system.Moreover,after intravenous injection,NIR CDs are accumulated in tumor tissue via passive targeting,without any active species such as antibodies.Due to their accumulation in tumor tissue without the need for intratumor injection,high photothermal conversion,excellent optical and photoacoustic imaging performance,and renal excretion,the developed CDs are suitable for transfer to clinical biomedical practice. | Xin Bao Ye Yuan Jingqin Chen Bohan Zhang Di Li Ding Zhou Pengtao Jing Guiying Xu Yingli Wang Kateřina Holá Dezhen Shen Changfeng Wu Liang Song Chengbo Liu Radek Zbořil Songnan Qu | 2018 | Light(Science & Applications)2018,7,1: | 13 |

| 5 | Cryo-EM structures of PAC1 receptor reveal ligand binding mechanism显示文摘The pituitary adenylate cyclase-activating polypeptide type I receptor(PAC1R)belongs to the secretin receptor family and is widely distributed in the central neural system and peripheral organs.Abnormal activation of the receptor mediates trigeminovascular activation and sensitization,which is highly related to migraine,making PAC1R a potential therapeutic target.Elucidation of PAC1R activation mechanism would benefit discovery of therapeutic drugs for neuronal disorders.PAC1R activity is governed by pituitary adenylate cyclase-activating polypeptide(PACAP),known as a major vasodilator neuropeptide,and maxadilan,a native peptide from the sand fly,which is also capable of activating the receptor with similar potency.These peptide ligands have divergent sequences yet initiate convergent PAC1R activity.It is of interest to understand the mechanism of PAC1R ligand recognition and receptor activity regulation through structural biology.Here we report two near-atomic resolution cryo-EM structures of PAC1R activated by PACAP38 or maxadilan,providing structural insights into two distinct ligand binding modes.The structures illustrate flexibility of the extracellular domain(ECD)for ligands with distinct conformations,where ECD accommodates ligands in different orientations while extracellular loop 1(ECU)protrudes to further anchor the ligand bound in the orthosteric site.By structureguided molecular modeling and mutagenesis,we tested residues in the ligand-binding pockets and identified clusters of residues that are critical for receptor activity.The structures reported here for the first time elucidate the mechanism of specificity and flexibility of ligand recognition and binding for PACT R,and provide insights toward the design of therapeutic molecules targeting PAC1R. | Jia Wang Xianqiang Song Dandan Zhang Xiaoqing Chen Xun Li Yaping Sun Cui Li Yunpeng Song Yao Ding Ruobing Ren Essa Hu Harrington Liaoyuan AHu Wenge Zhong Cen Xu Xin Huang Hong-Wei Wang Yingli Ma | 2020 | Cell Research2020,30,5: | 1 |

| 8 | Optimising multi-vent module-based adaptive ventilation using a novel parameter for improved indoor air quality and health protection显示文摘As infectious respiratory diseases are highly transmissible through the air,researchers have improved traditional total volume air distribution systems to reduce infection risk.Multi-vent module-based adaptive ventilation(MAV)is a novel ventilation type that facilitates the switching of inlets and outlets to suit different indoor scenarios without changing ductwork layout.However,little research has evaluated MAV module sizing and air velocity selection,both related to MAV system efficiency in removing contaminants and the corresponding level of protection for occupants in the ventilated room.Therefore,the module-source offset ratio(MSOR)is proposed,based on the MAV module size and its distance from an infected occupant,to inform selection of optimal MAV module parameters.Computational fluid dynamics simulations illustrated contaminant distribution in a two-person MAV equipped office.Discrete phase particles modelled respiratory contaminants from the infected occupant,and contaminant concentration distributions were compared under four MAV air distribution layouts,three air velocities,and three module sizes considered using the MsOR.Results indicate that lower air velocities favour rising contaminant levels,provided the ventilation rate is met.Optimal contaminant discharge can be achieved when the line of outlets is located directly above the infected occupant.Using this parameter to guide MAV system design,85.7% of contaminants may be rendered harmless to the human body within 120 s using the default air vent layout.A more appropriate supply air velocity and air vent layout increases this value to 91.4%.These results are expected to inform the deployment of MAV systems to reduce airborne infection risk. | Haotian Zhang Xiaoxiao Ding Weirong Zhang Weijia Zhang Yingli Xuan | 2024 | Building Simulation2024,17,1: | 0 |
