2Cardiotoxicity of 5-fluorouracil and capecitabine in Chinese patients: a prospective study显示文摘Background:5-Fluorouracil(5-FU)and capecitabine-associated cardiotoxicity ranging from asymptomatic electro-cardiography(ECG)abnormalities to severe myocardial infarction has been reported in a number of studies,but such cardiotoxicity in Chinese patients with malignant diseases has not been investigated to date.In the present study,we aimed to prospectively evaluate the incidence rates and clinical manifestations of 5-FU-and capecitabine-associated cardiotoxicity in cancer patients recruited from multiple centers in China.Methods:Among the 527 patients who completed the study,196 received 5-FU-based chemotherapy and 331 received capecitabine-based chemotherapy as either first-line or adjuvant therapy.Adverse events were reported during the treatment and up to 28 days of follow-up.Outcome measures included ECG,myocardial enzymes,cardiac troponin,brain natriuretic peptide and echocardiography.Univariate analysis and logistic regression were performed for subgroup analysis and identification of significant independent variables that are associated with cardiotoxicity of both agents.Results:In total,161 of 527 patients(30.6%)experienced cardiotoxicity.The incidence rate of cardiotoxicity was 33.8%(112/331)in the capecitabine group,which was significantly higher than the rate of 25%(49/196)in the 5-FU group(P=0.0042).110/527 patients(20.9%)suffered arrhythmia,105/527(19.9%)developed ischemic changes,while only 20/527 patients(3.8%)presented heart failure and 6/527 patients(1.1%)had myocardial infarction.Pre-existing cardiac disease,hypertension,capecitabine-based chemotherapy and duration of treatment were identified as sig-nificant risk factors associated with cardiotoxicity.The odds ratio were 15.7(prior history of cardiac disease versus no history),1.86(capecitabine versus 5-FU),1.06(5-8 versus 1-4 chemotherapy cycles)and 1.58(hypertension versus no hypertension),respectively.Conclusions:Cardiotoxicity induced by fluoropyrimidines in the Chinese population may be underestimated in clini-cal practice.Close monitoring of patients is recommended,especially for those patients at high risk for cardiotoxicity.Possible risk factors are duration of treatment,capecitabine-based chemotherapy,pre-existing cardiac diseases and hypertension.Trial registration This study was initiated on January 22,2014 and has been retrospectively registered with the registra-tion number ChiCTR1800015434 .Jianjun Peng Chao Dong Chang Wang Weihua Li Hao Yu Min Zhang Qun Zhao Bo Zhu Jun Zhang Wenliang Li Fenghua Wang Qiong Wu Wenhao Zhou Ying Yuan Meng Qiu Gong Chen 2018Cancer Communications2018,38,1:23
3Three-dimensional bioprinting of multicell-laden scaffolds containing bone morphogenic protein-4 for promoting M2 macrophage polarization and accelerating bone defect repair in diabetes mellitus显示文摘Critical-sized bone defect repair in patients with diabetes mellitus remains a challenge in clinical treatment because of dysfunction of macrophage polarization and the inflammatory microenvironment in the bone defect region.Three-dimensional(3D)bioprinted scaffolds loaded with live cells and bioactive factors can improve cell viability and the inflammatory microenvironment and further accelerating bone repair.Here,we used modified bioinks comprising gelatin,gelatin methacryloyl(GelMA),and 4-arm poly(ethylene glycol)acrylate(PEG)to fabricate 3D bioprinted scaffolds containing BMSCs,RAW264.7 macrophages,and BMP-4-loaded mesoporous silica nanoparticles(MSNs).Addition of MSNs effectively improved the mechanical strength of GelMA/gelatin/PEG scaffolds.Moreover,MSNs sustainably released BMP-4 for long-term effectiveness.In 3D bioprinted scaffolds,BMP-4 promoted the polarization of RAW264.7 to M2 macrophages,which secrete anti-inflammatory factors and thereby reduce the levels of pro-inflammatory factors.BMP-4 released from MSNs and BMP-2 secreted from M2 macrophages collectively stimulated the osteogenic differentiation of BMSCs in the 3D bioprinted scaffolds.Furthermore,in calvarial critical-size defect models of diabetic rats,3D bioprinted scaffolds loaded with MSNs/BMP-4 induced M2 macrophage polarization and improved the inflammatory microenvironment.And 3D bioprinted scaffolds with MSNs/BMP-4,BMSCs,and RAW264.7 cells significantly accelerated bone repair.In conclusion,our results indicated that implanting 3D bioprinted scaffolds containing MSNs/BMP-4,BMSCs,and RAW264.7 cells in bone defects may be an effective method for improving diabetic bone repair,owing to the direct effects of BMP-4 on promoting osteogenesis of BMSCs and regulating M2 type macrophage polarization to improve the inflammatory microenvironment and secrete BMP-2.Xin Sun Zhenjiang Ma Xue Zhao Wenjie Jin Chenyu Zhang Jie Ma Lei Qiang Wenhao Wang Qian Deng Han Yang Jinzhong Zhao Qianqian Liang Xiaojun Zhou Tao Li Jinwu Wang 2021Bioactive Materials2021,6,3:12
4Three-dimensional-printed individualized porous implants:A new“implant-bone”interface fusion concept for large bone defect treatment显示文摘Bone defect repairs are based on bone graft fusion or replacement.Current large bone defect treatments are inadequate and lack of reliable technology.Therefore,we aimed to investigate a simple technique using three-dimensional(3D)-printed individualized porous implants without any bone grafts,osteoinductive agents,or surface biofunctionalization to treat large bone defects,and systematically study its long-term therapeutic effects and osseointegration characteristics.Twenty-six patients with large bone defects caused by tumor,infection,or trauma received treatment with individualized porous implants;among them,three typical cases underwent a detailed study.Additionally,a large segmental femur defect sheep model was used to study the osseointegration characteristics.Immediate and long-term biomechanical stability was achieved,and the animal study revealed that the bone grew into the pores with gradual remodeling,resulting in a long-term mechanically stable implant-bone complex.Advantages of 3D-printed microporous implants for the repair of bone defects included 1)that the stabilization devices were immediately designed and constructed to achieve early postoperative mobility,and 2)that osseointegration between the host bone and implants was achieved without bone grafting.Our osseointegration method,in which the“implant-bone”interface fusion concept was used instead of“bone-bone”fusion,subverts the traditional idea of osseointegration.Teng Zhang Qingguang Wei Hua Zhou Zehao Jing Xiaoguang Liu Yufeng Zheng Hong Cai Feng Wei Liang Jiang Miao Yu Yan Cheng Daoyang Fan Wenhao Zhou Xinhong Lin Huijie Leng Jian Li Xinyu Li Caimei Wang Yun Tian Zhongjun Liu 2021Bioactive Materials2021,6,11:9
5In vitro degradation and biocompatibility evaluation of typical biodegradable metals (Mg/Zn/Fe) for the application of tracheobronchial stenosis显示文摘Tracheobronchial obstruction in children due to benign stenosis or tracheobronchomalacia still remains a challenging matter of concern.Currently,there is 10%–20%complication rate in clinical treatment.The nonbiodegradable property of silicone stents and nickel-titanium memory alloy stents take the primary responsibility for drawbacks including stimulating local granulation tissue proliferation,displacement,and stent-related infections.Permanent tracheobronchial stent will be a persistent foreign object for a long time,causing excessive secretion of tracheal mucosa,ulceration and even perforation,which is particularly unsuitable for young children with persistent tracheal growth.In this study,the degradation and biocompatibility performance of three typical biodegradable metals were investigated as potential tracheobronchial stent materials.The results exhibited that these materials showed different degradation behaviors in the simulating respiratory fluid environment compared with SBF.Except for pure iron group,high purity magnesium and zinc showed favorable cell adhesion and proliferation in three culture methodologies(direct culture,indirect culture and extraction culture).The proper corrosion rate and good biocompatibility indicated that high purity magnesium and zinc may be good candidates as tracheobronchial stent materials.Yangyang Li Jianglong Yan Wenhao Zhou Pan Xiong Pei Wang Wei Yuan Yufeng Zheng Yan Cheng 2019Bioactive Materials2019,4,1:8
6Genome-wide CRISPR screen identifies synthetic lethality between DOCK1 inhibition and metformin in liver cancer显示文摘Metformin is currently a strong candidate anti-tumor agent in multiple cancers.However,its anti-tumor effectiveness varies among different cancers or sub-populations,potentially due to tumor heterogeneity.It thus remains unclear which hepatocellular carcinoma(HCC)patient subpopulation(s)can benefit from met-formin treatment.Here,through a genome-wide CRISPR-Cas9-based knockout screen,we find that DOCK1 levels determine the anti-tumor effects of met-formin and that DOCK1 is a synthetic lethal target of metformin in HCC.Mechanistically,metformin promotes DOCK1 phosphorylation,which activates RAC1 to facilitate cell survival,leading to metformin resistance.The DOCK1-selective inhibitor,TBOPP,potentiates anti-tumor activity by metformin in vitro in liver cancer cell lines and patient-derived HCC organoids,and in vivo in xenografted liver cancer cells and immunocompetent mouse liver cancer models.Notably,metformin improves overall survival of HCC patients with low DOCK1 levels but not among patients with high DOCK1 expression.This study shows that metformin effective-ness depends on DOCK1 levels and that combining metformin with DOCK1 inhibition may provide a promising personalized therapeutic strategy for met-formin-resistant HCC patients.Junru Feng Hui Lu Wenhao Ma Wenjing Tian Zhuan Lu Hongying Yang Yongping Cai Pengfei Cai Yuchen Sun Zilong Zhou Jiaqian Feng Jiazhong Deng Ying Shu Kun Qu Weidong Jia Ping Gao Huafeng Zhang 2022Protein & Cell2022,13,11:5
7Analysis of grinding mechanics and improved grinding force model based on randomized grain geometric characteristics显示文摘Too high grinding force will lead to a large increase in specific grinding energy, resulting in high temperature in grinding zone, especially for the aerospace difficult cutting metal materials,seriously affecting the surface quality and accuracy. At present, the theoretical models of grinding force are mostly based on the assumption of uniform or simplified morphological characteristics of grains, which is inconsistent with the actual grains. Especially for non-engineering grinding wheel,most geometric characteristics of grains are ignored, resulting in the calculation accuracy that cannot guide practical production. Based on this, an improved grinding force model based on random grain geometric characteristics is proposed in this paper. Firstly, the surface topography model of CBN grinding wheel is established, and the effective grain determination mechanism in grinding zone is revealed. Based on the known grinding force model and mechanical behavior of interaction between grains and workpiece in different stages, the concept of grain effective action area is proposed. The variation mechanism of effective action area under the influence of grain geometric and spatial characteristics is deeply analyzed, and the calculation method under random combination of five influencing parameters is obtained. The numerical simulation is carried out to reveal the dynamic variation process of grinding force in grinding zone. In order to verify the theoretical model, the experiments of dry grinding Ti-6Al-4 V are designed. The experimental results show that under different machining parameters, the results of numerical calculation and experimental measurement are in good agreement, and the minimum error value is only 2.1 %, which indicates that the calculation accuracy of grinding force model meets the requirements and is feasible. 8Electrostatic atomization minimum quantity lubrication machining:from mechanism to application显示文摘Metal cutting fluids(MCFs)under flood conditions do not meet the urgent needs of reducing carbon emission.Biolubricant-based minimum quantity lubrication(MQL)is an effective alternative to flood lubrication.However,pneumatic atomization MQL has poor atomization properties,which is detrimental to occupational health.Therefore,electrostatic atomization MQL requires preliminary exploratory studies.However,systematic reviews are lacking in terms of capturing the current research status and development direction of this technology.This study aims to provide a comprehensive review and critical assessment of the existing understanding of electrostatic atomization MQL.This research can be used by scientists to gain insights into the action mechanism,theoretical basis,machining performance,and development direction of this technology.First,the critical equipment,eco-friendly atomization media(biolubricants),and empowering mechanisms of electrostatic atomization MQL are presented.Second,the advanced lubrication and heat transfer mechanisms of biolubricants are revealed by quantitatively comparing MQL with MCF-based wet machining.Third,the distinctive wetting and infiltration mechanisms of electrostatic atomization MQL,combined with its unique empowering mechanism and atomization method,are compared with those of pneumatic atomization MQL.Previous experiments have shown that electrostatic atomization MQL can reduce tool wear by 42.4%in metal cutting and improve the machined surface Ra by 47%compared with pneumatic atomization MQL.Finally,future development directions,including the improvement of the coordination parameters and equipment integration aspects,are proposed.Wenhao Xu Changhe Li Yanbin Zhang Hafiz Muhammad Ali Shubham Sharma Runze Li Min Yang Teng Gao Mingzheng Liu Xiaoming Wang Zafar Said Xin Liu Zongming Zhou 2022International Journal of Extreme Manufacturing2022,4,4:4

10De Novo and Inherited SETD1A Variants in Early-onset Epilepsy显示文摘Early-onset epilepsy is a neurological abnormality in childhood, and it is especially common in the first2 years after birth. Seizures in early life mostly result from structural or metabolic disorders in the brain, and the genetic causes of idiopathic seizures have been extensively investigated. In this study, we identified four missense mutations in the SETD1 A gene(SET domain-containing 1 A, histone lysine methyltransferase): three de novo mutations in three individuals and one inherited mutation in a four-generation family. Whole-exome sequencing indicated that all four of these mutations were responsible for the seizures. Mutations of SETD1 A have been implicated in schizophrenia and developmental disorders, so we examined the role of the four mutations(R913 C, Q269 R, G1369 R, and R1392 H) in neural development. We found that their expression in mouse primary cortical neurons affected excitatory synapse development. Moreover, expression of the R913 C mutation also affected the migration of cortical neurons in the mouse brain.We further identified two common genes(Neurl4 and Usp39) affected by mutations of SETD1 A. 11The Epigenetic Switches for Neural Development and Psychiatric Disorders显示文摘The most remarkable feature of the nervous system is that the development and functions of the brain are largely reshaped by postnatal experiences,in joint with genetic landscapes.The nature vs.nurture argument reminds us that both genetic and epigenetic information is indispensable for the normal function of the brain.The epigenetic regulatory mechanisms in the central nervous system have been revealed over last a decade.Moreover,the mutations of epigenetic modulator genes have been shown to be implicated in neuropsychiatric disorders,such as autism spectrum disorders.The epigenetic study has initiated in the neuroscience field for a relative short period of time. 13Influences of retrogressive erosion of reservoir on sedimentation of its downstream river channel-A case study on Sanmenxia Reservoir and the Lower Yellow River显示文摘Retrogressive erosion, a widespread phenomenon of sediment transport in reservoirs, often impacts on both the reservoir capacity and the sedimentation in the downstream river channel. Based on field data from the Sanmenxia Reservoir and the Lower Yellow River over the past decades, three courses of retrogressive erosion with distinctive features were analyzed. The results indicate that retrogressive erosion,especially caused by rapid reduction in the water level till the reservoir is empty, often results in the serious siltation of the lower Yellow River and threatens the safety of the flood control in the Lower Yellow River. Unreasonable operation of the reservoir and incoming hyperconcentrated floods accompanied by retrogressive erosion also aggravate the siltation of the main channel of the river. However, a reasonable operation mode of the reservoir so named 'storing the clear(low sediment concentration)water in the non-flood season, and sluicing the muddy(high sediment concentration) water in the flood season' was found, which might mitigate the deposition in both the reservoir and the Lower Yellow River. 14Genomic landscapes of Chinese sporadic autism spectrum disordersrevealed by whole-genome sequencing显示文摘Autism spectrum disorder (ASD) is a neurodevelopmental disorder with considerable clinical and genetic heterogeneity.In this study,we identified all classes of genomic variants from whole-genome sequencing (WGS) dataset of 32 Chinese trios with ASD,including de novo mutations,inherited variants,copy number variants (CNVs) and genomic structural variants.A higher mutation rate (Poisson test,P<2.2×10^(-16)) in exonic (1.37×10^(-8)) and 3'-UTR regions (1.42×10^(-8)) was revealed in comparison with that of whole genome (1.05×10^(-8)).Using an integrated model,we identified 87 potentially risk genes (P<0.01) from 4832 genes harboring various rare deleterious variants,including CHD8 and NRXN2,implying that the disorders may be in favor to multiple-hit.In particular,frequent rare inherited mutations of several microcephaly-associated genes (ASPM,WDR62,and ZNF335)were found in ASD.In chromosomal structure analyses,we found four de novo CNVs and one de novo chromosomal rearrangement event,including a de novo duplication of UBE3A-containing region at 15q11.2-q13.1,which causes Angelman syndrome and microcephaly,and a disrupted TNR due to de novo chromosomal translocation t (1;5) (q25.1;q33.2).Taken together,our results suggest that abnormalities of centrosomal function and chromatin remodeling of the microcephaly-associated genes may be implicated in pathogenesis of ASD.Adoption of WGS as a new yet efficient technique to illustrate the full genetic spectrum in complex disorders,such as ASD,could provide novel insights into pathogenesis,diagnosis and treatment.Jinyu Wu Ping Yu Xin Jin Xiu Xu Jinchen Li Zhongshan Li Mingbang Wang Tao Wang Xueli Wu Yi Jiang Wanshi Cai Junpu Mei Qingjie Min Qiong Xu Bingrui Zhou Hui Guo Ping Wang Wenhao Zhou Zhengmao Hu Yingrui Li Tao Cai Yi Wang Kun Xia Yong-Hui Jiang Zhong Sheng Sun 2018Journal of Genetics and Genomics2018,45,10:3
155-氟尿嘧啶和卡培他滨对中国患者的心脏毒性:一项前瞻性研究显示文摘背景与目的已有许多研究报道5-氟尿嘧啶(5-Fluorouracil,5-FU)和卡培他滨相关的心脏毒性,严重程度从无症状心电图(electrocardiography,ECG)异常到严重心肌梗塞而不同。目前为止,对中国恶性肿瘤患者的这种心脏毒性尚无研究。本研究旨在前瞻性地对多个中心招募的癌症患者进行5-FU和卡培他滨相关心脏毒性的发生率和临床表现进行评估。方法在完成研究的527例患者中,196例接受基于5-FU的化疗,331例接受基于卡培他滨的化疗并以其为一线或辅助治疗。在治疗期间和长达28 d的随访期间对所发生的不良反应进行记录。疗效测量指标包括心电图、心肌酶、心肌肌钙蛋白、脑钠肽和超声心动图。使用单因素分析和logistic回归方法进行亚组分析和鉴定与两种药物的心脏毒性相关的显著性独立变量。结果 527例患者中有161例(30.6%)出现心脏毒性。卡培他滨组的心脏毒性发生率为33.8%(112/331),显著高于5-FU组25%(49/196)的发生率(P=0.0042)。110/527患者(20.9%)发生心律失常,105/527(19.9%)发生缺血性改变,而只有20/527患者(3.8%)出现心力衰竭,6/527患者(1.1%)出现心肌梗塞。既往的心脏病、高血压、卡培他滨为基础的化疗和治疗时间被确定为与心脏毒性相关的显著风险因素。优势比分别为15.7(心脏病史vs.无病史)、1.86(卡培他滨vs. 5-FU)、1.06(5–8个化疗周期vs. 1–4个化疗周期)和1.58(高血压vs.无高血压)。结论氟嘧啶类药物在中国人群中引起的心脏毒性可能在临床实践中被低估。建议密切监测患者,尤其是对心脏毒性高风险的患者。可能的风险因素包括治疗持续时间、基于卡培他滨的化疗、既往心脏病史和高血压史。Jianjun Peng Chao Dong Chang Wang Weihua Li Hao Yu Min Zhang Qun Zhao Bo Zhu Jun Zhang Wenliang Li Fenghua Wang Qiong Wu Wenhao Zhou Ying Yuan Meng Qiu Gong Chen 2018癌症2018,37,10:3
16High-risk phenotypes of genetic disease in a Neonatal Intensive Care Unit population显示文摘To the Editor: Genetic diseases contribute to 35% of deaths during the first year of life and are a significant cause of intensive care.[1] A previous study based on the China Neonatal Genomes Project investigated the genetic causes of early infant deaths and found that >25% of deceased neonates with genetic diagnoses can be cured if diagnosed in time.[2] Therefore, it is crucial to target and diagnose neonates with genetic diseases as early as possible. According to our experience, the typical phenotypes, such as special facial features or multiple congenital anomalies (MCAs), indicate a high risk of genetic disease and lead physicians to perform genetic testing in neonates as early as possible. However, in practice, infants without typical phenotypes typically undergo a long and costly diagnostic process before genetic diagnoses are confirmed. Moreover, a recent survey by the American College of Medical Genetics and Genomics (ACMG) and other national professional organizations indicated that there are insufficient numbers of qualified geneticists to fulfil genetic service needs.[3] The ACMG published the general clinical features for genetic testing indications. For example, patients with phenotypes or family history data that strongly implicate a genetic cause may undergo genetic testing.[1] However, the study indicated that many genetic conditions arise de novo or are inherited with no family history.[1] A previous study attempted to apply the non-phenotype-driven panel approach in neonates admitted to the neonate intensive care unit (NICU).[4] However, at present, the diagnostic yield is only 3.45% (1/29).[4] In addition, the economic and ethical issues associated with genomic screening remain challenging. 19Influence of Sr on microstructure and mechanical properties of ZL114 cast alloy显示文摘In the present work, the effects of trace element Sr on the microstructure, tensile properties, fractography and morphology of the eutectic Si of ZL114A (Al-7Si) cast alloy were investigated. The cast ZL114A aluminum alloy was prepared using a metal mold, and then heat treated with the T6 technique. The test results of mechanical properties show that modification element Sr can improve significantly the ultimate tensile strength, elongation and hardness of the ZL114A-T6 aluminum alloy. By adding 0.04%Sr, the values of the tensile strength Rm, elongation A and hardness HB increase from 230 MPa, 1% and 65 to the maximum of 305 MPa, 8%, and 100, respectively. SEM analysis indicates that the near tensile fracture surface on longitudinal section of the ZL114A-T6 alloy reveals a transgranular fracture mode without Sr modification, while the tensile fracture is an intergranular fracture mode after Sr modification. 20Roof filling control technology and application to mine roadway damage in small pit goaf显示文摘To recover coal resources that have been damaged by traditional mining methods and ensure stability of the lower roadway in a small pit goaf,the goaf area must be filled and reinforced.In this research,the 1202 working face of the Hanzui mine is considered as an example for classifying the roof of the mining tunnel under the small kiln destruction zone,the effect of the goaf on the roadway is determined based on the radio tunnel penetration method,a mechanical model to determine the roof filling control mechanism was established,and the duct foaming system and roof filling process were designed.The results show that the scope and degree of influence of the goaf on the mining lane are large,but safe tunneling can be ensured through the use of a steel shed and advanced grouting techniques.When the roof conditions are not similar,materials with different filling heights and filling strengths can be used to control the roof filling of the roadway.By combining field experience and laboratory tests,it was determined that a high-foaming material with a water-cement ratio of 1:0.6,a suitable high-foaming additive,and a water volume ratio of 1:30 is cost-efficient for filling and meets the filling strength requirements.Finally,the reliability of the proposed technology was verified by field experiments,which provide a reference for filling operations in similar mines.Weiyi Cai Zechao Chang Dongsheng Zhang Xufeng Wang Wenhao Cao Yazhou Zhou 2019International Journal of Mining Science and Technology2019,29,3:2
