| 1 | Complement C3 activation regulates the production of tRNA-derived fragments Gly-tRFs and promotes alcohol-induced liver injury and steatosis显示文摘Complement is known to play a role in alcoholic fatty liver disease (AFLD), but the underlying mechanisms are poorly understood, thereby constraining the development of a rational approach for therapeutic intervention in the complement system. C3 deficiency has been shown to impart protective effects against ethanol-induced hepatic steatosis and inflammation. Here we demonstrate a protection effect in wild-type mice by treatment with CR2-Crry, a specific inhibitor of C3 activation. The expression of glycine transfer (t) RNA-derived fragments (Gly-tRFs) is upregulated in ethanol-fed mice and inhibition of Gly-tRFs in vivo decreases chronic ethanol feeding-induced hepatosteatosis without affecting inflammation. The expression of Gly-tRF was downregulated in C3-deficient or CR2-Crry-treated mice, but not in C5-deficient mice;Gly-tRF expression was restored by the C3 activation products C3a or Asp (C3a-des-Arg) via the regulation of CYP2E1. Transcriptome profiling of hepatic tissues showed that Gly-tRF inhibitors upregulate the expression of sirtuin1 (Sirt1) and subsequently affect downstream lipogenesis and β-oxidation pathways. Mechanistically, Gly-tRF interacts with AGO3 to downregulate Sirt1 expression via sequence complementarity in the 3′ UTR. Notably, the expression levels of C3d, CYP2E1 and Gly-tRF are upregulated, whereas Sirt1 is decreased in AFLD patients compared to healthy controls. Collectively, our findings suggest that C3 activation products contribute to hepatosteatosis by regulating the expression of Gly-tRF. | 3 | A Genome Sequence of Novel SARS-CoV Isolates: the Genotype, GD-Ins29, Leads to a Hypothesis of Viral Transmission in South China显示文摘We report a complete genomic sequence of rare isolates (minor genotype) of theSARS-CoV from SARS patients in Guangdong, China, where the first few casesemerged. The most striking discovery from the isolate is an extra 29-nucleotidesequence located at the nucleotide positions between 27,863 and 27,864 (referredto the complete sequence of B J01) within an overlapped region composed of BGI-PUP5 (BGI-postulated uncharacterized protein 5) and BGI-PUP6 upstream ofthe N (nucleocapsid) protein. The discovery of this minor genotype, GD-Ins29,suggests a significant genetic event and differentiates it from the previously re-ported genotype, the dominant form among all sequenced SARS-CoV isolates. A17-nt segment of this extra sequence is identical to a segment of the same size intwo human mRNA sequences that may interfere with viral genome replication andtranscription in the cytosol of the infected cells. | 4 | Gene expression profiles of hepatocytes treated with La(NO_3)_3 of rare earth in rats显示文摘AIM: To compare the gene expression between La(NO3)3-exposed and control rats in vivo. METHODS: Rats were fed La(NO3)3 once daily at a dose of 20 mg/kg for one month by gavage. Gene expression of hepatocytes was detected using mRNA differential display (DD) technique and cDNA microarray and compared between treated and control groups. RESULTS: Six differentially expressed sequence tags were cloned by DD, of which five were up regulated and one was down regulated in treated rats. Two sequences were determined. One band was novel. The other shared 100% sequence homology with AU080263 Sugano mouse brain mncb Mus musculus cDNA clone MNCb-5435 5'. With DNA microarray, 136 differentially expressed genes were identified including 131 over-expressed genes and 5 under-expressed genes. Most of these differentially expressed genes were cell signal and transmission genes, genes associated with metabolism, protein translation and synthesis. CONCLUSION: La(NO3)3 could change the expression levels of some kinds of genes. | 5 | Fabrication of ZnO films by radio frequency magnetron sputtering and annealing显示文摘ZnO thin films were deposited on Si(l11) substrates through a radio frequency (rf) magnetron sputtering system.Then the samples were annealed at different temperatures in air ambience and ammonia ambience respectively. The struc-ture and composition of the ZnO films were studied by X-ray diffraction (XRD) and X-ray photoelectron spectroscopy(XPS). The morphology of the samples was studied by scanning electron microscopy (SEM). Measured results show thatZnO films with hexagonal wurtzite structure were grown on Si(111) substrates when annealed in the two ambiences. | 6 | The in vitro reconstitution of nucleosome and its binding patterns with HMG1/2 and HMG14/17 proteins显示文摘Using atomic force microscopy (AFM), the dynamic process of the in vitro nucleosome reconstitution followed by slow dilution from high salt to low salt was visualized. Data showed that the histone octamers were dissociated from DNA at 1M NaCl. When the salt concentration was slowly reduced to 650 mMand 300 mM, the core histones bound to the naked DNA gradually. Once the salt concentration was reduced to 50 mM the classic 'beads-on-a-string' structure was clearly visualized. Furthermore, using the technique of the in vitro reconstitution ofnucleosome,the mono- and di- nucleosomes were assembled in vitro with both HS2core (-10681 to -10970 bp) and NCR2 (-372to -194 bp) DNA sequences in the 5'flanking sequence of human b-globin gene. Data revealed that HMG 1/2 and HMG 14/17 proteins binding to both DNA sequences are changeable following the assembly and disassembly of nucleosomes. | 7 | Fabrication and photoluminescence of GaN nanowires prepared by ammoniating GaeO3/BN films on Si substrate显示文摘GaN nanowires were successfully pre-pared on Si(111) substrate through ammoniating Ga2O3/BN films deposited by radio frequency mag-netron sputtering system. The synthesized nanowires were confirmed as hexagonal wurtzite GaN by X-ray diffraction, selected-area electron diffraction and Fourier transform infrared spectra. Scanning electron microscopy and transmission electron microscopy revealed that the grown GaN nanowires have a smooth and clean surface with diameters ranging from 40 to 160 nm and lengths typically up to several tens of micrometers. The representative photolumi-nescence spectrum at room temperature exhibited a strong UV light emission band centered at 363 nm and a relative weak purple light emission peak at 422 nm. | 8 | Effect of substrate temperature on microstructural and optical properties of ZnO films grown by pulsed laser deposition显示文摘ZnO thin films were deposited on n-Si (111) at various substrate temperatures by pulsed laser deposition (PLD). X-ray diffraction (XRD), photoluminescence (PL), Fourier transform infrared spectrophotometer (FTIR), and scanning electron microscopy (SEM) were used to analyze the structure, morphology, and optical property of the ZnO thin films. An optimal crystallized ZnO thin film was obtained at the substrate temperature of 600°C. A blue shift was found in PL spectra due to size confinement effect as the grain sizes decreased. | 11 | An experimental study of acoustic scattering from thermocline in shallow sea显示文摘This work was done in Summer of 1988, 1989 and 1992, at three different stations (34, 39 and 44 m under sea level), in the south Yellow Sea using narrow band acoustic pulses with center frequencies of 120, 160 and 200 kHz.The pulses were echoed back from three different depths. We used CTD to record the temperature and salinity of different depth day and night, and the plankton samples at different layers were hauled up for comparison. We applied the numerical method introduced by R. Kind to calculate reflected plane wave spectrum, measured temperature and salinity in vertical direction of non-uniform layer distribution and obtained the numerical simulation signals of backward scattering sound waves from thermocline layers. We then compared them with the experimental data. Through the experiments of the same periords of three years we have confimed that the measured time that layer-scattering-waves to reach our recorders agreed well with that obtained by temperature sectional simulated data. On the other hand, there is no apparent effects of plankton distribution.
