| 2 | Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. | 3 | Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. | 4 | All-fiber passively mode-locked laser using nonlinear multimode interference of step-index multimode fiber显示文摘We experimentally demonstrate for the first time, to the best of our knowledge, an all-fiber passively mode-locked laser operation based on the nonlinear multimode interference of step-index multimode fiber. Such a structure couples the light in and out of the multimode fiber via single-mode fibers, and its physical mechanisms for saturable absorption have been analyzed theoretically based on the third-order nonlinear Kerr effect of multimode fiber. Using the nonlinear multimode interference structure with 48.8 mm length step-index multimode fiber, the modulation depth has been measured to be ~5%. The passively mode-locked laser output pulses have a central wavelength of 1596.66 nm, bandwidth of 2.18 nm, pulsewidth of ~625 fs, and fundamental repetition rate of8.726 MHz. Since virtualized desktop system is based on multiple virtual machines (VM), it is necessary to design a distributed storage system to manage the VM images. In this paper, we design a distributed storage system, VMStore, by taking into account three important characteristics: high performance VM snapshot, booting optimization from multiple images and redundancy removal of images data. We adopt a direct index structure of blocks for VM snapshots to speed up VM booting performance significantly; provide a distribute storage structure with good bandwidth scalability by dynamically changing the number of storage nodes; and propose a data preprocessing strategy with intelligent object partitioning techniques, which would eliminate duplication more effectively. Performance analysis for VMStore focuses on two metrics: the speedup of VM booting and the overhead of de-duplication. The results show that a group of strong aftershocks which occurred at the end of July to early August in 2008 at the north of Wenchuan were obviously triggered by earth tide, the same conclusion is drawn by Schusters smooth test of the tidal force, tidal stress and tidal coulomb failure stress. In addition, the Wenchuan aftershock activity is obviously triggered by fortnight tide. | 14 | Characteristics of the May 12,2008 Wenchuan M_S8.0 Earthquake Sequence and Discussion on Relevant Problems显示文摘The aftershock activity of the May 12,2008 Wenchuan M_S8.0 Earthquake Sequence shows an obvious segmented feature.Most of the large aftershocks were distributed in the north and south parts of the aftershock zone.Thrusting was dominant with a small amount of strike-slip component in the south part.The aftershock activity decayed gradually,presenting the sequence features of a mainshock-aftershock pattern.The north part was the ending area of the mainshock fracture where strike-slipping was dominant,showing an obvious swarm feature.Therefore it became the major area for large aftershocks.The modulation of the earth tide on aftershock activity is remarkable;most large aftershocks occur during the period of flood and neap tide.The time period around 16:00 was the dominant occurring time for large aftershocks.The p-value,a parameter of modified Omori formula,increases gradually with time,and reaches about 1 at the end.Based on previous study,the sequence patterns,magnitude of maximum aftershock,as well as the duration of aftershock activity has been discussed.The primary results also show that the magnitude difference between the mainshock and the maximum aftershock is proportional to the rupture size of the mainshock for huge earthquakes of about M_S8.0.This means that when the magnitudes of the earthquakes are nearly the same,large rupture size corresponds to sufficient energy release. | Jiang Haikun Li Mingxiao Wu Qiong Song Jin | 2009 | Earthquake Research in China2009,23,1: | 1 |
| 15 | Porosity, permeability and rock mechanics of Lower Silurian Longmaxi Formation deep shale under temperature-pressure coupling in the Sichuan Basin, SW China显示文摘To investigate the porosity, permeability and rock mechanics of deep shale under temperature-pressure coupling, we selected the core samples of deep shale from the Lower Silurian Longmaxi Formation in the Weirong and Yongchuan areas of the Sichuan Basin for porosity and permeability experiments and a triaxial compression and sound wave integration experiment at the maximum temperature and pressure of 120 ℃ and 70 MPa. The results show that the microscopic porosity and permeability change and the macroscopic rock deformation are mutually constrained, both showing the trend of steep and then gentle variation. At the maximum temperature and pressure, the porosity reduces by 34%–71%, and the permeability decreases by 85%–97%. With the rising temperature and pressure, deep shale undergoes plastic deformation in which organic pores and clay mineral pores are compressed and microfractures are closed, and elastic deformation in which brittle mineral pores and rock skeleton particles are compacted. Compared with previous experiments under high confining pressure and normal temperature,the experiment under high temperature and high pressure coupling reveals the effect of high temperature on stress sensitivity of porosity and permeability. High temperature can increase the plasticity of the rock, intensify the compression of pores due to high confining pressure, and induce thermal stress between the rock skeleton particles, allowing the reopening of shale bedding or the creation of new fractures along weak planes such as bedding, which inhibits the decrease of permeability with the increase of temperature and confining pressure. Compared with the triaxial mechanical experiment at normal temperature, the triaxial compression experiment at high temperature and high pressure demonstrates that the compressive strength and peak strain of deep shale increase significantly due to the coupling of temperature and pressure. The compressive strength is up to 435 MPa and the peak strain exceeds 2%, indicating that high temperature is not conducive to fracture initiation and expansion by increasing rock plasticity. Lithofacies and mineral composition have great impacts on the porosity, permeability and rock mechanics of deep shale. Shales with different lithologies are different in the difficulty and extent of brittle failure. | 19 | Preventive effect of kiwi berry(Actinidia arguta)on loperamide-induced constipation显示文摘Constipation is a common intestinal disease.Kiwi berries can effectively prevent constipation.However,studies have yet to be done to determine how kiwi berries prevent constipation.For two weeks,mice in this study were continually orally gavaged with kiwi berry,loperamide,or a combination of the 2.This study found that the kiwi group's feces had more water than the constipated mice.In addition,kiwi berries can speed up gastrointestinal transit(GI),shorten the time it takes to pass the first dark stool,and dramatically enhance body weight gain.In the interstitial cells of Caj al(ICC)cells and colon tissues,alterations in the protein expression of vasoactive intestinal peptide(VIP),cyclic adenosine monophosphate(cAMP),protein kinase A(PKA),and aquapcrin-3(AQP3)were found.At 3,6,and 12 h of ICC cells and mouse colon,the kiwi group's VIP,cAMP,PKA,and AQP3 protein expression levels were lower than those of the constipated mice.The kiwi berry can decrease the Firmicutes to Bacteroidetes ratio and boost the diversity and quantity of gut microbiota.By influencing the gut microbiota and VIP-cAMP-PKA-AQP3 signaling pathway,kiwi berries prevent constipation. | Jiyue Zhang Bin Li Ningxuan Gao Haikun Li Xingyue Cui Hanqian Jiang Siyi Tang Chenyu Jin Jinlong Tian | 2024 | Food Science and Human Wellness2024,13,3: | 0 |
| 20 | NUIST ESM v3 Data Submission to CMIP6显示文摘This paper introduces the experimental designs and outputs of the Diagnostic,Evaluation and Characterization of Klima(DECK),historical,Scenario Model Intercomparison Project(MIP),and Paleoclimate MIP(PMIP)experiments from the Nanjing University of Information Science and Technology Earth System Model version 3(NESM3).Results show that NESM3 reasonably simulates the modern climate and the major internal modes of climate variability.In the Scenario MIP experiment,changes in the projected surface air temperature(SAT)show robust“Northern Hemisphere(NH)warmer than Southern Hemisphere(SH)”and“land warmer than ocean”patterns,as well as an El Niño-like warming over the tropical Pacific.Changes in the projected precipitation exhibit“NH wetter than SH”and“eastern hemisphere gets wetter and western hemisphere gets drier”patterns over the tropics.These precipitation patterns are driven by circulation changes owing to the inhomogeneous warming patterns.Two PMIP experiments show enlarged seasonal cycles of SAT and precipitation over the NH due to the seasonal redistribution of solar radiation.Changes in the climatological mean SAT,precipitation,and ENSO amplitudes are consistent with the results from PMIP4 models.The NESM3 outputs are available on the Earth System Grid Federation nodes for data users. | Jian CAO Libin MA Fei LIU Jing CHAI Haikun ZHAO Qiong HE Bo WANG Yan BAO Juan LI Young-min YANG Hua DENG Bin WANG | 2021 | Advances in Atmospheric Sciences2021,38,2: | 0 |
