2GABAergic Abnormalities Associated with Sensorimotor Corticostriatal Community Structural Deficits in ErbB4 Knockout Mice and First-Episode Treatment-Naive Patients with Schizophrenia显示文摘The current study was designed to explore how disruption of specific molecular circuits in the cerebral cortex may cause sensorimotor cortico-striatal community structure deficits in both a mouse model and patients with schizophrenia.We used prepulse inhibition(PPI)and brain structural and diffusion MRI scans in 23 mice with conditional ErbB4 knockout in parvalbumin interneurons and 27 matched controls.Quantitative real-time PCR was used to assess the differential levels of GABA-related transcripts in brain regions.Concurrently,we measured structural and diffusion MRI and the cumulative contribution of risk alleles in the GABA pathway genes in firstepisode treatment-naı¨ve schizophrenic patients(n=117)and in age-and sex-matched healthy controls(n=86).We present the first evidence of gray and white matter impairment of right sensorimotor cortico-striatal networks and reproduced the sensorimotor gating deficit in a mouse model of schizophrenia.Significant correlations between gray matter volumes(GMVs)in the somatosensory cortex and PPI as well as glutamate decarboxylase 1 mRNA expression were found in controls but not in knockout mice.Furthermore,these findings were confirmed in a human sample in which we found significantly decreased gray and white matter in sensorimotor cortico-striatal networks in schizophrenic patients.The psychiatric risk alleles of the GABA pathway also displayed a significant negative correlation with the GMVs of the somatosensory cortex in patients.Our study identified that ErbB4 ablation in parvalbumin interneurons induced GABAergic dysregulation,providing valuable mechanistic insights into the sensorimotor cortico-striatal community structure deficits associated with schizophrenia.Chengcheng Zhang Peiyan Ni Yikang Liu Yang Tian Jinxue Wei Bo Xiang Liansheng Zhao Xiaojing Li Xiaohong Ma Wei Deng Wanjun Guo Rongjun Ni Yamin Zhang Qiang Wang Hailiang Huang Nanyin Zhang Tao Li 2020Neuroscience Bulletin2020,36,2:9
3Effects of Ti additive on HPHT diamond synthesis in Fe-Ni-C system显示文摘Diamond crystals with low nitrogen concentration were synthesized from the Fe-Ni-C system with Ti additive at high pressure and high temperature (HPHT) in a china-type cubic high pressure apparatus (CHPA). The synthesis pressure range was 4.8-5.2 GPa, and the temperature range was 1420-1600 K. The lowest synthesis pressure for diamond fell first and then rose with the increase of Ti additive. The color, shape, surface morphology and nitrogen impurity concentration of the synthesized diamond crystals were characterized using optical microscopy (OM), scanning electron microscopy (SEM) and micro Fourier transform infrared (FTIR) spectrometry. The results show that the Ti additive has significant effects on color, growth rate, crystal shape, surface morphology and nitrogen impurity con- centration of the synthesized diamond crystals. The color of diamond crystals synthesized without Ti additive is yellow, while that with Ti additive becomes light and nearly colorless. The growth rate without Ti additive is higher than that with Ti additive. The crystal shapes of as-grown diamond crystals vary with the increase of Ti additive. The {111} crystal faces become dominant and some {311} crystal faces appear with the increase of Ti additive. 4Genomic and transcriptomic analyses enable the identification of important genes associated with subcutaneous fat deposition in Holstein cows显示文摘Subcutaneous fat deposition has many important roles in dairy cattle,including immunological defense and mechanical protection.The main objectives of this study are to identify key candidate genes regulating subcutaneous fat deposition in high-producing dairy cows by integrating genomic and transcriptomic datasets.A total of 1654 genotyped Holstein cows are used to perform a genome-wide association study(GWAS)aiming to identify genes associated with subcutaneous fat deposition.Subsequently,weighted gene co-expression network analyses(WGCNA)are conducted based on RNA-sequencing data of 34 cows and cow yield deviations of subcutaneous fat deposition.Lastly,differentially expressed(DE)m RNA,lnc RNA,and differentially alternative splicing genes are obtained for 12 Holstein cows with extreme and divergent phenotypes for subcutaneous fat deposition.Forty-six protein-coding genes are identified as candidate genes regulating subcutaneous fat deposition in Holstein cattle based on GWAS.Eleven overlapping genes are identified based on the analyses of DE genes and WGCNA.Furthermore,the candidate genes identified based on GWAS,WGCNA,and analyses of DE genes are significantly enriched for pathways involved in metabolism,oxidative phosphorylation,thermogenesis,fatty acid degradation,and glycolysis/gluconeogenesis pathways.Integrating all findings,the NID2,STARD3,UFC1,DEDD,PPP1R1B,and USP21 genes are considered to be the most important candidate genes influencing subcutaneous fat deposition traits in Holstein cows.This study provides novel insights into the regulation mechanism underlying fat deposition in high-producing dairy cows,which will be useful when designing management and breeding strategies.Hailiang Zhang Siyuan Mia Luiz F.Brito Lirong Hu Lei Wang Longgang Ma Qing Xu Gang Guo Ying Yu Yachun Wang 2023Journal of Genetics and Genomics2023,50,6:1
5Multi-Scale Design and Optimization of Composite Material Structure for Heavy-Duty Truck Protection Device显示文摘In this paper,to present a lightweight-developed front underrun protection device(FUPD)for heavy-duty trucks,plain weave carbon fiber reinforced plastic(CFRP)is used instead of the original high-strength steel.First,the mechanical and structural properties of plain carbon fiber composite anti-collision beams are comparatively analyzed from a multi-scale perspective.For studying the design capability of carbon fiber composite materials,we investigate the effects of TC-33 carbon fiber diameter(D),fiber yarn width(W)and height(H),and fiber yarn density(N)on the front underrun protective beam of carbon fiber compositematerials.Based on the investigation,a material-structure matching strategy suitable for the front underrun protective beam of heavy-duty trucks is proposed.Next,the composite material structure is optimized by applying size optimization and stack sequence optimization methods to obtain the higher performance carbon fiber composite front underrun protection beam of commercial vehicles.The results show that the fiber yarn height(H)has the greatest influence on the protective beam,and theH1matching scheme for the front underrun protective beamwith a carbon fiber composite structure exhibits superior performance.The proposed method achieves a weight reduction of 55.21% while still meeting regulatory requirements,which demonstrates its remarkable weight reduction effect.Yanhui Zhang Lianhua Ma Hailiang Su Jirong Qin Zhining Chen Kaibiao Deng 2024Computer Modeling in Engineering & Sciences2024,139,5:0
6Misalignment Tolerance in One-side and Symmetric Loading Hopkinson Pressure Bar Experiments显示文摘The split-Hopkinson pressure bar(SHPB)is a widely used experimental technique for studying the mechanical properties of materials at high strain rates.There are two kinds of loading methods applied in the SHPB technique,namely one-side loading and symmetric loading.However,the experimental accuracy of the two loading methods is affected by the interface contact.The present study focused on the inadequate contact caused by the misalignment of the pressure bars.The commercial software ABAQUS was used for simulations.The result shows that the inadequate contact caused by the alignment of the bars has a non-negligible effect on the calculated results.Compared with the one-side loading Hopkinson pressure bar,the symmetric loading Hopkinson pressure bar has a more relaxed requirement for the alignment of the bars.The conclusion arrived at in this paper can help researchers to make a reasonable choice between one-side and symmetric loading Hopkinson pressure bars according to actual requirements.Hailiang Nie Weifeng Ma Xueliang He Ke Wang Junjie Ren Jun Cao Wei Dang 2022Acta Mechanica Solida Sinica2022,35,2:0

9A lncRNA from an inflammatory bowel disease risk locus maintains intestinal host-commensal homeostasis显示文摘Inflammatory bowel diseases(IBD)are known to have complex,genetically influenced etiologies,involving dysfunctional interactions between the intestinal immune system and the microbiome.Here,we characterized how the RNA transcript from an IBD-associated long non-coding RNA locus(“CARINH-Colitis Associated IRF1 antisense Regulator of Intestinal Homeostasis”)protects against IBD.We show that CARINH and its neighboring gene coding for the transcription factor IRF1 together form a feedforward loop in host myeloid cells.The loop activation is sustained by microbial factors,and functions to maintain the intestinal host-commensal homeostasis via the induction of the anti-inflammatory factor IL-18BP and anti-microbial factors called guanylate-binding proteins(GBPs).Extending these mechanistic insights back to humans,we demonstrate that the function of the CARINH/IRF1 loop is conserved between mice and humans.Genetically,the T allele of rs2188962,the most probable causal variant of IBD within the CARINH locus from the human genetics study,impairs the inducible expression of the CARINH/IRF1 loop and thus increases genetic predisposition to IBD.Our study thus illustrates how an IBD-associated lncRNA maintains intestinal homeostasis and protects the host against colitis.Hongdi Ma Taidou Hu Wanyin Tao Jiyu Tong Zili Han Dietmar Herndler-Brandstetter Zheng Wei Ruize Liu Tingyue Zhou Qiuyuan Liu Xuemei Xu Kaiguang Zhang Rongbin Zhou Judy H.Cho Hua-Bing Li Hailiang Huang Richard A.Flavell Shu Zhu 2023Cell Research2023,33,5:0
10Single-atomic Mn sites coupled with Fe_(3)C nanoparticles encapsulated in carbon matrixes derived from bimetallic Mn/Fe polyphthalocyanine conjugated polymer networks for accelerating electrocatalytic oxygen reduction显示文摘The construction of robust coupling catalysts for accelerating electrocatalytic oxygen reduction reaction(ORR)through the modulation of the electronic structure and local atomic configuration is critical but remains challenging.Herein,we report a facile and effective isolation-polymerization-pyrolysis(IPP)strategy for high-precision synthesis of single-atomic Mn sites coupled with Fe_(3)C nanoparticles encapsulated in N-doped porous carbon matrixes(Mn SAs/Fe_(3)C NPs@NPC)catalyst derived from predesigned bimetallic Fe/Mn polyphthalocyanine(FeMn-BPPc)conjugated polymer networks by solid-phase reaction approach.Benefiting from the synergistic effects between the single-atomic Mn-N_(4)sites and Fe_(3)C NPs as well as the confinement effect of NPC,the Mn SAs/Fe_(3)C NPs@NPC catalyst exhibited excellent electrocatalytic activity and stability for ORR.The assembled Znair battery displayed larger power density of 186 mW·cm^(−2)than that of Pt/C+Ir/C-based battery.It also exhibits excellent stability without obvious voltage change after 106 cycles with 36 h.Combing in-situ Raman spectra with in-situ attenuated total reflectance surface-enhanced infrared absorption spectroscopy(ATR-SEIRAS)characterization results indicated that the Mn-N_(4)site as an active site for the O_(2)adsorption-activation process,which effectively facilitates the generation of key*OOH intermediates and*OH desorption to promote the multielectron reaction kinetics.Theoretical calculation reveals that the excellent electrocatalytic performance originates from the charge redistribution and the d orbital shift resulting from Mn-Fe bond,which buffers the activity of ORR through the electron reservoir capable of electron donation or releasing.This work paves a novel IPP strategy for constructing high-performance coupling electrocatalyst towards the ORR for energy conversion devices.Yuan Pan Min Li Wanliang Mi Minmin Wang Junxi Li Yilin Zhao Xuelu Ma Bin Wang Wei Zhu Zhiming Cui Hailiang Yin Yunqi Liu 2022Nano Research2022,15,9:0
