| 2 | Efficiency and Inheritance of Targeted Mutagenesis in Maize Using CRISPR-Cas9显示文摘CRISPR/Cas(clustered regularly interspaced short palindromic repeats/CRISPR-associated proteins) is an adaptive immune system in bacteria and archaea to defend against invasion from foreign DNA fragments. Recently, it has been developed as a powerful targeted genome editing tool for a wide variety of species. However, its application in maize has only been tested with transiently expressed somatic cells or with a limited number of stable transgenic T0 plants. The exact efficiency and specificity of the CRISPR/Cas system in the highly complex maize genome has not been documented yet. Here we report an extensive study of the well-studied type II CRISPR-Cas9 system for targeted genome editing in maize, with the codon-optimized Cas9 protein and the short non-coding guide RNA generated through a functional maize U6 sn RNA promoter. Targeted gene mutagenesis was detected for 90 loci by maize protoplast assay, with an average cleavage efficiency of 10.67%. Stable knockout transformants for maize phytoene synthase gene(PSY1) were obtained. Mutations occurred in germ cells can be stably inherited to the next generation. Moreover, no off-target effect was detected at the computationally predicted putative off-target loci. No significant difference between the transcriptomes of the Cas9 expressed and non-expressed lines was detected. | 8 | Defective Kernel 39 encodes a PPR protein required for seed development in maize显示文摘RNA editing is a posttranscriptional process that is important in mitochondria and plastids of higher plants. All RNA editing-specific trans-factors reported so far belong to PLS-class of pentatricopeptide repeat(PPR)proteins. Here, we report the map-based cloning and molecular characterization of a defective kernel mutant dek39 in maize. Loss of Dek39 function leads to delayed embryogenesis and endosperm development, reduced kernel size, and seedling lethality. Dek39 encodes an E subclass PPR protein that targets to both mitochondria and chloroplasts, and is involved in RNA editing in mitochondrial NADH dehydrogenase3(nad3) at nad3-247 and nad3-275. C-to-U editing of nad3-275 is not conserved and even lost in Arabidopsis, consistent with the idea that no close DEK39 homologs are present in Arabidopsis. However, the amino acids generated by editing nad3-247 and nad3-275 are highly conserved in many other plant species, and the reductions of editing at these two sites decrease the activity of mitochondria NADH dehydrogenase complex I,indicating that the alteration of amino acid sequence is necessary for Nad3 function. | 9 | Effects of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze-Yellow rivers, Tibetan Plateau of China显示文摘Improving our knowledge of the effects of environmental factors(e.g.soil conditions,precipitation and temperature) on belowground biomass in an alpine grassland is essential for understanding the consequences of carbon storage in this biome.The object of this study is to investigate the relative importance of soil nutrients and climate factors on belowground biomass in an alpine meadow in the source region of the Yangtze and Yellow rivers,Tibetan Plateau.Soil organic carbon(SOC),total nitrogen(TN) and total phosphorous(TP) contents and belowground biomass were measured at 22 sampling sites across an alpine meadow on the Tibetan Plateau.We analyzed the data by using the redundancy analysis to determine the main environmental factors affecting the belowground biomass and the contribution of each factor.The results showed that SOC,TN and TP were the main factors that influenced belowground biomass,and the contribution of SOC,TN and TP on biomass was in the range of 47.87%–72.06% at soil depths of 0–30 cm.Moreover,the combined contribution of annual mean temperature(AMT) and mean annual precipitation(MAP) on belowground biomass ranged from 0.92% to 4.10%.A potential mechanism for the differences in belowground biomass was caused by the variations in soil nitrogen and phosphorous,which were coupled with SOC.A significant correlation was observed between MAP and soil nutrients(SOC,TN and TP) at the soil depth of 0–10 cm(P<0.05).We concluded that precipitation is an important driving force in regulating ecosystem functioning as reflected in variations of soil nutrients(SOC,TN and TP) and dynamics of belowground biomass in alpine grassland ecosystems. | WANG Haiming SUN Jian LI Weipeng WU Jianbo CHEN Youjun LIU Wenhui | 2016 | Journal of Arid Land2016,8,6: | 5 |

| 11 | Enhanced ozonation degradation of atrazine in the presence of nano-ZnO: Performance, kinetics and effects显示文摘Enhanced ozonation degradation of atrazine(ATZ) with nano-ZnO(nZnO) as catalyst and the influences of the operational parameters have been investigated through semi-continuous experiments in this study. The results demonstrated that the combination of ozone(O_3) and nZnO showed an obvious synergetic effect and the ATZ degradation conformed to pseudo-first-order kinetics. An improvement of ATZ degradation efficiency by 41.8% and pseudo-first-order rate constant by more than a factor of four was obtained in the O_3/nZnO process after 5 min of reaction compared to O_3 alone. Meanwhile, the degradation efficiency of ATZ was gradually enhanced with increasing nZnO dosage and initial pH in the range from 3.0 to 8.0, and a higher amount of ATZ was degraded when the initial concentration of ATZ rose from 0.5 to 5 mg/L. Additionally, sulfate ion, chloride ion, nitrate ion and low concentrations of humic acid substances led to enhancement of the ATZ degradation. The notable decrease of ATZ removal efficiency observed in the presence of radical scavengers and the results of free radical tests indicated thatUOH is the dominant active radical species. | 19 | CD49a^(+)CD49b^(+) NK cells induced by viral infection reflect an activated state of conventional NK cells显示文摘Natural killer(NK) cells are important innate effectors that play a pivotal role in the defense against tumors and infections and participate in regulating adaptive immunity. Recent studies have revealed phenotypic and functional heterogeneity of NK cells.Here, using murine models of acute and chronic lymphocytic choriomeningitis virus infection, we observed that a CD49a^(+)CD49b^(+) NK cell subset emerged in the liver and other tissues, and underwent vigorous expansion following viral infection,before progressively decreasing in cell number. These viral infection-induced CD49a^(+)CD49b^(+) NK cells displayed an activated and mature phenotype. Moreover, compared with liver-resident NK cells and conventional NK(cNK) cells, CD49a^(+)CD49b^(+) NK cells showed increased functional competence, as evidenced by higher amounts of IFN-γ production and stronger cytotoxic capabilities during viral infection. Generation of these CD49a^(+)CD49b^(+) NK cells was shown to be independent of the T-bet transcription factor. Adoptive transfer experiments revealed that c NK cells could convert into CD49a^(+)CD49b^(+) NK cells following viral infection. | 20 | Synthesis of an ordered porous carbon with the dual nitrogen-doped interfaces and its ORR catalysis performance显示文摘Both nitrogen-doping feature and pore structure are critical factors for developing nitrogen-doped carbons based catalysts with a high performance toward oxygen reduction reaction(ORR).Herein,a simple one-step CVD of acetylene and acetonitrile vapor method using silanized SBA-15 as a template has been developed to synthesize an ordered porous carbon(OPC) with dual nitrogen-doped interfaces.The optimized sample as prepared with the CVD of 4 h at 750℃ contains two types of ordered mesopores that one type is the ordered cylindrical pores inheriting from the pores of SBA-15 and has a pore width of4.0~5.0 nm,the other type is the ordered quasi-hexagonal pores with a width of 3.0~4.0 nm produced by etching the pore walls of SBA-15.These two types of pores whose pore walls are built by the nitrogen doped carbon layers resulted by the CVD and thus it actually makes the dual nitrogen-doped interfaced OPC(DN-OPC).Meanwhile,DN-OPC contains a few of micropores and a large SSA of 1430 m~2/g.This dualordered pores and dual nitrogen-doped interfaces cannot only facilitate mass transport but also utilize the active sites of DN-OPC for ORR.Therefore,as metal-free ORR catalyst,DN-OPC exhibits a good activity close to commercial Pt/C catalyst,and an excellent durability and methanol tolerance. | Hongwei Zhao Yanqiu Zhang Li Lixiang Xin Geng Haiming Yang Weimin Zhou Chengguo Sun Baigang An | 2021 | Chinese Chemical Letters2021,32,1: | 2 |
