1The Distribution of Different Clades of Seneca Valley Viruses in Guangdong Province,China显示文摘Seneca Valley virus (SVV), a newly determined etiological agent of vesicular disease in swine, causes porcine idiopathic disease and occasional acute death in piglets. Recently, an increased number of SVV infection cases have been reported in the United States (US) and China, resulting in significant economic losses to the swine industry. The first identification of SVV in China was reported in Guangdong Province, a major swine producing province. The cases of SVV were continuously reported in Guangdong in 2015 and 2016. However, the spread of SVV in Guangdong in 2017 remains unknown.In this study, we determined two new SVV strains, CH-GD-2017-1 and CH-GD-2017-2, from Guangdong. The genetic analysis suggested that the two Guangdong strains showed different characteristics to previous Guangdong strains. They showed lower nucleotide similarity with strains isolated in 2015 and 2016, and were more similar to the US strains.Phylogenetic analyses indicated that the new strains were clustered in a different clade with previous Guangdong strains.We found 28 mutated amino acids in the new strains, compared with the first Guangdong strain, SVV CH-01-2015. In the geographic analysis, we found that the US and China reported more SVV cases than other countries, and most of the SVV cases were reported in east and central China—of which, Guangdong Province is one of the major epidemic regions. 2Increasing prevalence of antibiotic resistance genes in manured agricultural soils in northern China显示文摘Land application of manure tends to result in the dissemination of antibiotic resistance in the environment.In this study,the influence of long-term manure application on the enrichment of antibiotic resistance genes(ARGs)and mobile genetic elements(MGEs)in agricultural soils was investigated.All the analyzed eight ARGs(tetA,tetW,tetX,sull,sulll,ermF,aac(6')-Ib-cr and blaTEM)and two MGEs(intJJ and Tn916/1545)were detected in both the manured and control soils,with relative abundances ranging from 10^-6 to 10^-2 Compared with the control soil,the relative abundances of ARGs and MGEs in manured soils were enriched 1.0-18.1 fold and 0.6-69.1 fold,respectively.High-throughput sequencing analysis suggested that at the phylum level,the bacteria carrying intll and ermF might be mainly affiliated with Proteobacteria and Bacteroides,respectively.The dominant genera carrying intll and ermF cow\d be Pseudomonas and Bacteroides,independent of manure application.Correlation analysis revealed that ARGs had strong links with soil physicochemical properties(TC,TN,and OM),heavy metals(Cu,Zn and Pb)and MGEs,indicating that the profile and spread of ARGs might be driven by the combined impacts of multiple factors.In contrast,soil pH and C/N exhibited no significant relationships with ARGs.Our findings provide evidence that long-term manure application could enhance the prevalence and stimulate the propagation of antibiotic resistance in agricultural soils.Nan Wu Weiyu Zhang Shiyu Xie Ming Zeng Haixue Liu Jinghui Yang Xinyuan Liu Fan Yang 2020Frontiers of Environmental Science & Engineering2020,14,1:3
3Curcumin’s prevention of inflammation-driven early gastric cancer and its molecular mechanism显示文摘Worldwide,gastric cancer is the second leading cause of cancer deaths and the fifth most common malignant tumor.Gastric cancer is believed to be caused by a variety of factors,such as genetics,epigenetics,and environmental influences.Among the pathogenic factors,inflammation has been considered as one of the main risk factors for gastric cancer.There are currently limited ways to prevent gastric cancer.Although the combined application of aspirin and non-steroidal anti-inflammatory drugs can reduce the risk,it has great side effects and can easily cause gastric perforation or gastric bleeding.Therefore,an alternative plan is urgently needed.Curcumin is the yellow pigment in the rhizome of the plant turmeric.Current studies have found that curcumin has a protective effect on gastric mucosal damage caused by non-steroidal anti-inflammatory drugs,gastric mucosal damage in rats,and gastric mucosal damage caused by stress bleeding and Helicobacter pylori infection.Curcumin shows significant antiinflammatory and anti-cancer activities by regulating DNA methylation,histone modification,nuclear factor erythrocyte 2 related factor 2 and other related signal pathways.In this article,the latest evidence of curcumin for epigenetic changes in gastric cancer and its potential contribution to gastric cancer were discussed.Wensen Zhang Na Cui Jin Ye Bingyou Yang Yanping Sun Haixue Kuang 2022Chinese Herbal Medicines2022,14,2:3

6Adjunctive sepsis therapy with aminophylline(STAP):a randomized controlled trial显示文摘Background:Sepsis is a serious disease caused by infection.Aminophylline has anti-asthma and anti-inflammatory effects.We aimed to explore the safety and effect of aminophylline in sepsis.Methods:We conducted a clinical randomized controlled trial involving 100 patients diagnosed with sepsis within 48 h after intensive care unit(ICU)admission in two sites.All patients were randomized in a 1:1 ratio to receive standard therapy with or without aminophylline.The primary clinical outcome was all-cause mortality at 28 days.Results:From September 27,2018 to February 12,2020,we screened 277 septic patients and eventually enrolled 100 patients,with 50 assigned to the aminophylline group and 50 to the usual-care group.At 28 days,7 of 50 patients(14.0%)in the aminophylline group had died,compared with 16 of 50(32.0%)in the usual-care group(P=0.032).Cox regression showed that the aminophylline group had a lower hazard of death(hazard ratio=0.312,95%confidence interval:0.129–0.753).Compared with the usual-care group,patients in the aminophylline group had a longer survival time(P=0.039 by the log-rank test).The effects of aminophylline on vasopressor dose,oxygenation index,and sequential organ failure assessment score were timedependent with treatment.There were no significant differences in total hospitalization days,ICU hospitalization days,and rates of serious adverse events(all P>0.05).No adverse events were observed in the trial.Conclusions:Aminophylline as an adjunct therapy could significantly reduce the risk of death and prolong the survival time of patients with sepsis.Trial registration:ChiCTR.org.cn,ChiCTR1800019173.Ruifang Zhang Huan Liu Dongmei Dai Xianfei Ding Dong Wang Yan Wang Xuexiu Shi Shuguang Zhang Xiaoguang Duan Haixu Wang Yonggang Luo Shaohua Liu Bing Han Xiaojuan Zhang Yu Fang Jing Yang Wangbin Xu Tongwen Sun 2022Chinese Medical Journal2022,135,23:2

8Foot-and-Mouth Disease Virus Inhibits RIP2 Protein Expression to Promote Viral Replication显示文摘Receptors interaction protein 2(RIP2)is a specific adaptor molecule in the downstream of NOD2.The role of RIP2 during foot-and-mouth disease virus(FMDV)infection remains unknown.Here,our results showed that RIP2 inhibited FMDV replication and played an important role in the activation of IFN-βand NF-κB signal pathways during FMDV infection.FMDV infection triggered RIP2 transcription,while it reduced the expression of RIP2 protein.Detailed analysis showed that FMDV 2B,2C,3C^(pro),and L^(pro) proteins were responsible for inducing the reduction of RIP2 protein.3C^(pro) and L^(pro) are viral proteinases that can induce the cleavage or reduction of many host proteins and block host protein synthesis.The carboxyl terminal 105-C114 and 135-C144 regions of 2B were essential for reduction of RIP2.Our results also showed that the N terminal 1-61 region of 2C were essential for the reduction of RIP2.The 2C-induced reduction of RIP2 was dependent on inducing the reduction of poly(A)-binding protein 1(PABPC1).The interaction between RIP2 and 2C was observed in the context of viral infection,and the residues 1-61 were required for the interaction.These data clarify novel mechanisms of reduction of RIP2 mediated by FMDV.Huisheng Liu Qiao Xue Zixiang Zhu Fan Yang Weijun Cao Xiangtao Liu Haixue Zheng 2021Virologica Sinica2021,36,4:1
9Differential gene expression in porcine SK6 cells infected with wild-type and SAP domain-mutant foot-and-mouth disease virus显示文摘Foot-and-mouth disease virus(FMDV) is the causative agent of a highly contagious disease in livestock. The viral proteinaseL^(rop) of FMDV is involved in pathogenicity, and mutation of theL^(rop) SAP domain reduces FMDV pathogenicity in pigs. To determine the gene expression profiles associated with decreased pathogenicity in porcine cells, we performed transcriptome analysis using next-generation sequencing technology and compared differentially expressed genes in SK6 cells infected with FMDV containingL^(rop) with either a wild-type or mutated version of the SAP domain. This analysis yielded 1,853 genes that exhibited a ≥ 2-fold change in expression and was validated by real-time quantitative PCR detection of several differentially expressed genes. Many of the differentially expressed genes correlated with antiviral responses corresponded to genes associated with transcription factors, immune regulation, cytokine production, inflammatory response, and apoptosis. Alterations in gene expression profiles may be responsible for the variations in pathogenicity observed between the two FMDV variants. 12Nortriterpenoids from the fruit stalk of Schisandra chinensis (Turcz.) Baill.显示文摘Objective:The fruit stalk of Schisandra chinensis(Turcz.)Baill.(S.chinensis)has been found to contain bioactive components similar to the fruit of S.chinensis.Here,we report a recent discovery about new nortriterpenoids with a novel skeleton and anti-gastric cancer activity,which were isolated from the fruit stalk of S.chinensis.Methods:The chemical components of ethyl acetate extract from 70%ethanol extract from S.chinensis fruit stalk were separated,purified,and identified by liquid chromatography methods(silica gel,ODS,HPLC)and extensive spectroscopic analyses(NMR,IR,UV,MS,CD).Results:Two new nortriterpenoids,schilancitrilactone M and 25-hydroxyl schindilactone D(1 and 2),along with ten known nortriterpenoids(3-12)were isolated from the fruit stalk of S.chinensis.The isolated compounds were tested for their cytotoxic activities against MGC-803 cells,and the results showed that compounds 6-8 possessed significant activities with IC50 of 9.01,11.77,and 2.74μmol/L,respectively.Conclusion:Twelve nortriterpenoids including two new compounds were isolated from the fruit stalk of S.chinensis for the first time.Among them,compounds 6-8 showed significant anti-gastric cancer activities.We postulated that the fruit stalk of S.chinensis could be used as an anti-gastric cancer drug.Yan Liu Xiaohui Rong Yuanhang Chang Juan Pan Wei Guan Haixue Kuang Bingyou Yang 2022Frigid Zone Medicine2022,2,2:1
13Porcine Picornavirus 3C Protease Degrades PRDX6 to Impair PRDX6-mediated Antiviral Function显示文摘Peroxiredoxin-6(PRDX6)is an antioxidant enzyme with both the activities of peroxidase and phospholipase A2(PLA2),which is involved in regulation of many cellular reactions.However,the function of PRDX6 during virus infection remains unknown.In this study,we found that the abundance of PRDX6 protein was dramatically decreased in foot-and-mouth disease virus(FMDV)infected cells.Overexpression of PRDX6 inhibited FMDV replication.In contrast,knockdown of PRDX6 expression promoted FMDV replication,suggesting an antiviral role of PRDX6.To explore whether the activity of peroxidase and PLA2 was associated with PRDX6-mediated antiviral function,a specific inhibitor of PLA2(MJ33)and a specific inhibitor of peroxidase activity(mercaptosuccinate)were used to treat the cells before FMDV infection.The results showed that incubation of MJ33 but not mercaptosuccinate promoted FMDV replication.Meanwhile,overexpression of PRDX6 slightly enhanced type I interferon signaling.We further determined that the viral 3Cprowas responsible for degradation of PRDX6,and 3Cpro-induced reduction of PRDX6 was independent of the proteasome,lysosome,and caspase pathways.The protease activity of 3Cprowas required for induction of PRDX6 reduction.Besides,PRDX6 suppressed the replication of another porcine picornavirus Senecavirus A(SVA),and the 3Cproof SVA induced the reduction of PRDX6 through its proteolytic activity as well.Together,our results suggested that PRDX6 plays an important antiviral role during porcine picornavirus infection,and the viral 3Cproinduces the degradation of PRDX6 to overcome PRDX6-mediated antiviral function.Congcong Wang Huanhuan Feng Xiangle Zhang Kangli Li Fan Yang Weijun Cao Huisheng Liu Lili Gao Zhaoning Xue Xiangtao Liu Zixiang Zhu Haixue Zheng 2021Virologica Sinica2021,36,5:1
14A bibliometrics visualization analysis and hotspots prediction for natural product on osteoporosis research from 2000 to 2021显示文摘Aim The purpose of this study was to explore the application status of natural products in osteoporosis research and explore its research hotspots and future research directions by analyzing the literature in the field of natural products therapy for osteoporosis from 2000 to 2021.Method Publications on natural products in osteoporosis research were extracted from the Web of Science core collection database.Bibliometrics package in R language,VOSviewer and CiteSpace software were used for visualization analysis.Result From 2000 to 2021,the number of articles on natural products in osteoporosis research was 1596,and the total number of articles increased rapidly.China was the driving force in this field,and the top-contributing institution was Guangzhou University of Chinese Medicine.Most publications were published in the Journal of Ethnopharmacology.While J.K.Xu and Y.Zhang were the most cited and co-cited author.Countries,research institutions and authors in different regions cooperate closely.The hot topics included osteoporosis,differentiation,expression,in vitro,cell,osteogenesis and osteogenesis were the research frontiers in recent years.Conclusion This study conducted a bibliometric visualization analysis on the research status,research hotspots and development trend of natural products in osteoporosis for the first time.In addition,this paper also puts forward suggestions and prospects for the future development of this field,which will help researchers to open up new directions for future research in this field.Ajiao Hou Lin Yang Jiahao Lü Liu Yang Haixue Kuang Hai Jiang 2022Journal of Future Foods2022,2,4:1

17FoxJ1 inhibits African swine fever virus replication and viral S273R protein decreases the expression of FoxJ1 to impair its antiviral effect显示文摘African swine fever(ASF)is a highly pathogenic swine infectious disease that affects domestic pigs and wild boar,which is caused by the African swine fever virus(ASFV).ASF has caused huge economic losses to the pig industry and seriously threatens global food security and livestock health.To date,there is no safe and effective commercial vaccine against ASF.Unveiling the underlying mechanisms of ASFV-host interplay is critical for developing effective vaccines and drugs against ASFV.In the present study,RNA-sequencing,RT-qPCR and Western blotting analysis revealed that the transcriptional and protein levels of the host factor FoxJ1 were significantly down-regulated in primary porcine alveolar macrophages(PAMs)infected by ASFV.RT-qPCR analysis showed that overexpression of FoxJ1 upregulated the transcription of type I interferon and interferon stimulating genes(ISGs)induced by poly(dA:dT).FoxJ1 revealed a function to positively regulate innate immune response,therefore,suppressing the replication of ASFV.In addition,Western blotting analysis indicated that FoxJ1 degraded ASFV MGF505-2R and E165R proteins through autophagy pathway.Meanwhile,RT-qPCR and Western blotting analysis showed that ASFV S273R inhibited the expression of FoxJ1.Altogether,we determined that FoxJ1 plays an antiviral role against ASFV replication,and ASFV protein impairs FoxJ1-mediated antiviral effect by degradation of FoxJ1.Our findings provide new insights into the antiviral function of FoxJ1,which might help design antiviral drugs or vaccines against ASFV infection.Caina Ma Shasha Li Fan Yang Weijun Cao Huisheng Liu Tao Feng Keshan Zhang Zixiang Zhu Xiangtao Liu Yonghao Hu Haixue Zheng 2022Virologica Sinica2022,37,3:1

20Petroleum exploration breakthrough and geological significance in Cretaceous Yingcheng and Denglouku formations of Shuangcheng area, northern Songliao Basin, NE China显示文摘Based on drilling geological,geochemical,geophysical and production test data,the characteristics of source rocks,reservoir rocks and caprocks,as well as the process of hydrocarbon generation,trap evolution and oil accumulation of the oil-bearing assemblage composed of the Cretaceous Yingcheng Formation(K1yc)and Denglouku Formation(K1d)in the Shuangcheng area,northern Songliao Basin,NE China,were analyzed by using the research methods for petroleum systems.The source rocks mainly exist in K1yc,with the organic matters mainly originated from lower aquatic organisms and algae,and reaching the grade of high-quality source rock in terms of organic abundance.The crude oil,which is characterized by low density,high freezing point and high wax content,is believed to have generated by the K1yc source rocks.The reservoir rocks include K1d sandstones and K1yc glutenites.The mudstone in the fourth member of K1d serves as the direct caprock of the oil reservoir.The oil was generated during the period between Yaojia Formation and Nenjiang Formation,and then accumulated during the periods of Nenjiang Formation and Paleogene–Neogene.The traps evolved in three stages:the late Yingcheng Formation,the late Quantou Formation and the late Nenjiang Formation,forming structural and structural-lithologic reservoirs.It is concluded that good source-reservoir-caprock assemblage,late hydrocarbon charging,short migration distance and stable tectonic setting are favorable factors for the formation of oil reservoirs.LIU Chao FU Xiaofei LI Yangcheng WANG Haixue SUN Lidong LU Jiamin LI Junhui SUN Youhai SHI Lidong HU Huiting YUAN Hongqi YANG Zicheng 2023Petroleum Exploration and Development2023,50,1:0
