1The m^(6)A methylome of SARS-CoV-2 in host cells显示文摘The newly identified Severe Acute Respiratory Syndrome Coronavirus 2(SARS-CoV-2)has resulted in a global health emergency because of its rapid spread and high mortality.The molecular mechanism of interaction between host and viral genomic RNA is yet unclear.We demonstrate herein that SARS-CoV-2 genomic RNA,as well as the negative-sense RNA,is dynamically N6-methyladenosine(m^(6)A)-modified in human and monkey cells.Combined RIP-seq and miCLIP analyses identified a total of 8 m^(6)A sites at single-base resolution in the genome.Especially,epidemic strains with mutations at these identified m^(6)A sites have emerged worldwide,and formed a unique cluster in the US as indicated by phylogenetic analysis.Further functional experiments showed that m^(6)A methylation negatively regulates SARS-CoV-2 infection.SARS-CoV-2 infection also triggered a global increase in host m^(6)A methylome,exhibiting altered localization and motifs of m^(6)A methylation in mRNAs.Altogether,our results identify m^(6)A as a dynamic epitranscriptomic mark mediating the virus–host interaction.Jun’e Liu Yan-Peng Xu Kai Li Qing Ye Hang-Yu Zhou Hanxiao Sun Xiaoyu Li Liu Yu Yong-Qiang Deng Rui-Ting Li Meng-Li Cheng Bo He Jia Zhou Xiao-Feng Li Aiping Wu Chengqi Yi Cheng-Feng Qin 2021Cell Research2021,31,4:5
2AI-Enhanced Constellation Design for NOMA System: A Model Driven Method显示文摘Grant-free Non-orthogonal Multiple Access(GF-NOMA)is a promising technology for massive access users and sporadic small-packet transmission for Beyond the 5th Generation mobile communication system(B5G)/the 6th Generation mobile communication system(6G).One of the key aspects in GF-NOMA system is the signature/constellation design.However,due to the channel variation and random activation of users,conventional optimization approaches seem unsuitable for such complex models.In this paper,as an initial attempt,we propose a human intelligence(HI)-guided artificial intelligence(AI)-enhanced signature/constellation design method.By separate design of modulation and power allocation inspired by prior knowledge,the proposed deep neuron network(DNN)for NOMA signature/constellation design not only has smaller size of DNN and less training data,but also has stronger interpretability.In the last section,via simulations we demonstrate that in terms of bit error rate,the proposed scheme can achieve significant performance gain over the conventional NOMA schemes.Sen Wang Hanxiao Yu Yifei Yuan Guangyi Liu Zesong Fei 2020China Communications2020,17,11:3
3Mixture Toxicities of Three Pesticides Having Different Time-Toxicity Profiles显示文摘In the environment,chemical pollutants always exist as mixtures.To evaluate mixture toxicity,we should ex-amine the concentration ratios and the concentration levels of mixture components as well as the interactive time between the components and testing organism.Selecting three pesticides,metribuzin(MET),dodine(DOD)and ri-domil(RID),as mixture components,Vibrio qinghaiensis sp.-Q67 as testing organism,and 96-well microplate as exposure experiment carrier,the luminescence inhibition toxicities of the pesticides and their binary mixtures at six exposure time points(0.25,2,4,8,12,and 16 h)were determined.The results showed that the three pesticides have different time-toxicity profiles.The toxicity of MET increases over time,and those of DOD and RID show a slight decrease or increase in the first 4 h,respectively,and then remain unchanged to 16 h.The toxicities of the binary mixtures consisting of the three pesticides depend on not only the mixture composition but also the exposure time.The time-toxicity profiles of all rays in MET-DOD mixture system are U-shaped curves,those in MET-RID system are basically monotonically increased and those in DOD-RID system are basically monotonically decreased.How-ever,our findings indicate that for the test substances,mixture toxicity can be predicted by the extended concentra-tion addition model.Mo Yu Shushen Liu Mengchao Wang Fu Chen Hanxiao Tang 2014Chinese Journal of Chemistry2014,32,6:2
4Comparative population genomics dissects the genetic basis of seven domestication traits in jujube显示文摘Jujube(Ziziphus jujuba Mill.)is an important perennial fruit tree with a range of interesting horticultural traits.It was domesticated from wild jujube(Ziziphus acidojujuba),but the genomic variation dynamics and genetic changes underlying its horticultural traits during domestication are poorly understood.Here,we report a comprehensive genome variation map based on the resequencing of 350 accessions,including wild,semi-wild and cultivated jujube plants,at a>15×depth.Using the combination of a genome-wide association study(GWAS)and selective sweep analysis,we identified several candidate genes potentially involved in regulating seven domestication traits in jujube.For fruit shape and kernel shape,we integrated the GWAS approach with transcriptome profiling data,expression analysis and the transgenic validation of a candidate gene to identify a causal gene,ZjFS3,which encodes an ethyleneresponsive transcription factor.Similarly,we identified a candidate gene for bearing-shoot length and the number of leaves per bearing shoot and two candidate genes for the seed-setting rate using GWAS.In the selective sweep analysis,we also discovered several putative genes for the presence of prickles on bearing shoots and the postharvest shelf life of fleshy fruits.This study outlines the genetic basis of jujube domestication and evolution and provides a rich genomic resource for mining other horticulturally important genes in jujube.Mingxin Guo Zhongren Zhang Yanwei Cheng Sunan Li Peiyin Shao Qiang Yu Junjie Wang Gan Xu Xiaotian Zhang Jiajia Liu Linlin Hou Hanxiao Liu Xusheng Zhao 2020Horticulture Research2020,7,1:2
5MAT1 correlates with molecular subtypes and predicts poor survival in breast cancer显示文摘Objective:Menage a trois 1(MAT1)is a targeting subunit of cyclin-dependent kinase-activating kinase and general transcription factor IIH kinase,which modulates cell cycle,transcription and DNA repair.Its dysregulation is responsible for diseases including cancers.To further explore the role of MAT1 in breast cancer,we investigated the pathways in which MAT1 might be involved,the association between MAT1 and molecular subtypes,and the role of MAT1 in clinical outcomes of breast cancer patients.Methods:We conducted immunohistochemistry staining on tissue microarray and immunofluorescence staining on sections of MAT1 stable breast cancer cells.Also,we performed Kyoto Encyclopedia of Genes and Genomes pathway analysis,correlation analysis and prognosis analysis on public databases.Results:MAT1 was involved in multiple pathways including normal physiology signaling and disease-related signaling.Furthermore,MAT1 positively correlated with the protein status of estrogen receptor and progesterone receptor,and was enriched in luminal-type and human epidermal growth factor receptor 2-enriched breast cancer in comparison with basal-like subtype at both m RNA and protein levels.Correlation analysis revealed significant association between MAT1 m RNA amount and epithelial markers,mesenchymal markers,cancer stem cell markers,apoptosis markers,transcription markers and oncogenes.Consistently,the results of immunofluorescence stain indicated that MAT1 overexpression enhanced the protein abundance of epidermal growth factor receptor,vimentin,sex determining region Y-box 2 and sine oculis homeobox homolog 1.Importantly,Kaplan-Meier Plotter analysis reflected that MAT1 could serve as a prognostic biomarker predicting worse relapse-free survival and metastasis-free survival.Conclusions:MAT1 is correlated with molecular subtypes and is associated with unfavorable prognosis for breast cancer patients.Hanxiao Xu Xianguang Bai Shengnan Yu Qian Liu Richard G Pestell Kongming Wu 2018Chinese Journal of Cancer Research2018,30,3:2
6Clinical outcomes of atezolizumab in combination with etoposide/platinum for treatment of extensive-stage small-cell lung cancer:A real-world,multicenter,retrospective,controlled study in China显示文摘Objective:Atezolizumab along with chemotherapy has prolonged the survival of patients with extensive-stage small-cell lung cancer(ES-SCLC)worldwide,although real-world(RW)data are lacking in China.This study was designed to evaluate the efficacy and clinical outcomes of atezolizumab plus etoposide/platinum(EP).Methods:Data obtained in this retrospective study were captured from six oncology units of five medical facilities from January 2019 to April 2022.For first-line treatments,atezolizumab combined with EP vs.EP alone,we primarily evaluated progression-free survival(PFS);other efficacy indicators,including overall survival(OS),objective response rate(ORR),and patterns of SCLC progression and adverse events(AEs)were assessed.Results:The primary analysis included data from 225 patients,of whom 133 received EP along with atezolizumab(atezolizumab group)and 92 received EP alone(EP group).The PFS duration of the atezolizumab group[7.10 months;95%confidence interval(95%CI),6.53-9.00]exceeded that of the EP group(6.50 months;95%CI,4.83-7.53).Overall,the hazard ratio(HR)was 0.69(95%CI,0.49-0.97)(P=0.029);particularly,the HR was 0.54(95%CI,0.36-0.80)among patients undergoing≥4 chemotherapy cycles and 0.33(95%CI,0.20-0.56)among individuals with atezolizumab maintenance.The ORR and disease-control rate(DCR)were similar between the two groups.Because of incomplete OS data,the median OS was not determined for either group.Bone marrow suppression was the most common AE detected(58.6%)in the atezolizumab group.Immune-related AEs occurred in 19 patients in the atezolizumab group(14.3%),with only one case of grade 3 encephalitis.Conclusions:This RW study in China demonstrated improved clinical outcomes of atezolizumab along with EP for ES-SCLC,particularly in the chemosensitive population.These results align with the results of the IMpower133 study,although the impact of this treatment modality on OS warrants additional follow-up studies.Hanxiao Chen Xiangjuan Ma Jie Liu Yu Yang Yong Fang Liping Wang Jian Fang Jun Zhao Minglei Zhuo 2022Chinese Journal of Cancer Research2022,34,4:1
7Analysis of irregular repetition spatially-coupled slotted ALOHA显示文摘Contention-based access is a promising technology for massive and sporadic transmissions. In this paper, we propose a novel contention-based multiple access scheme, named irregular repetition spatiallycoupled slotted ALOHA(IRSC-SA), motivated by the spatial coupling and irregular repetition techniques.There are different classes of users and slots in IRSC-SA, which result in unequal protection for different users. 8Magnetic Fields Affect Alcoholic Liver Disease by Liver Cell Oxidative Stress and Proliferation Regulation显示文摘It is well known that alcohol consumption leads to substantially increased free radical levels and health risks,which lacks effective treatment besides alcohol abstinence.Here,we compared different static magnetic field(SMF)settings and found that a downward quasi-uniform SMF of~0.1 to 0.2 T could effectively alleviate alcohol-induced liver damage and lipid accumulation and improve hepatic function.SMFs of two different directions can reduce the inflammation,reactive oxygen species levels,and oxidative stress in the liver,while the downward SMF had more obvious effects.Moreover,we found that the upward direction SMF of~0.1 to 0.2 T could inhibit DNA synthesis and regeneration in hepatocytes,which caused detrimental effects on the lifespan of'heavy drinking'mice.In contrast,the downward SMF prolongs survival of'heavy drinking'mice.On one hand,our study shows that~0.1 to 0.2 T moderate quasi-uniform SMFs with a downward direction have great promises to be developed into a physical method to reduce alcohol-induced liver damage;on the other hand,although the internationally recognized upper limit for SMF public exposure is 0.4 T,people should also pay extra attention to SMF strength,direction,and inhomogeneity that could generate harmful effects on specific severe pathological conditions.Chao Song Hanxiao Chen Biao Yu Lei Zhang Junjun Wang Chuanlin Feng Xingxing Yang Xiaofei Tian Yixiang Fan Xinmiao Ji Hua Wang Can Xie Xin Zhang 2023Research2023,,4:0
9Evaluating the quality consistency of compound Liangmianzhen buccal tablets during accessories replacement by HPLC/UV related with its active profiling and UV dissolution profiles显示文摘Compound Liangmianzhen buccal tablet is a drug that exhibits notable effect on cough and diphtheria.Since the accessory has been changed,along with the difficulties in traditional Chinese drug-evaluation,it is imperative to set up a reliable method to evaluate the drug and ensure the quality consistency.First,the content of the Compound Liangmianzhen buccal tablets reflected by fingerprints were evaluated by the comprehensive linearly quantified fingerprint method and systematic quantified fingerprint method,in which thirteen fingerprint peaks was selected and gallic acid was chosen as the reference peak.The results of the two methods showed all batches in grades 1–3 of above good level with the qualitative similarities of Sl and Sm above 0.90,and the quantitative ones of Pl%and Pm%from 80%–120%,indicating all samples far qualified.Second,the measurement of antioxidant effect based on DPPH free radical clearance test has demonstrated the consistency between the old prescription group and the new ones and further dug up the major antioxidant compositions.Third,dissolution profiles at 190 to 400 nm that evaluate the curative effect consistency were evaluated by f2 and the correlation of the dissolution curve.In conclusion,the combination of 3 kinds of analysis methods can comprehensively assess the quality of the traditional Chinese drug and be served as a routine procedure to evaluate the herbal medicine consistency.Yue Yu Hanxiao Chi Guoxiang Sun Fangliang Yang Wanyang Sun Jiayue Hu Yanhua Zhang 2020Asian Journal of Traditional Medicines2020,15,1:0
10A columnar multi-layer sliding triboelectric nanogenerator for water wave energy harvesting independent of wave height and direction显示文摘The ocean,with its highly variable and complex meteorological conditions,harbors enormous renewable resources.Triboelectric nanogenerators(TENGs),which possess unique advantages,show exciting prospects in water wave energy collection.How to design and optimize TENGs to cover all characteristic water wave energies and achieve efficient energy utilization is emergent.In this paper,we carefully designed and fabricated a columnar multi-layer sliding TENG(CMLS-TENG)that can harvest water wave energy independent of wave height and direction.Drive rods with a hollow acrylic spherical shell were introduced to deliver wave energy,ensuring that the CMLS-TENG can work in all directions from 0°to 360°.Based on the sliding structure,switching the optimized CMLS-TENG is independent of wave heights.The optimized CMLS-TENG can achieve a total power density of 730 mW/m^(3) at a wave height of only 4.8 cm regardless of wave direction,which can illuminate multiple light-emitting diodes(LEDs)to provide lighting and provide power to a watch and a hygrometer for temperature and humidity monitoring.This work provides new choices and hopes for the effective collection of full-range water wave energy.Xue Miao Hanxiao Yang Zekun Li Meifei Cheng Yilin Zhao Lingyu Wan Aifang Yu Junyi Zhai 2024Nano Research2024,17,4:0
