| 1 | Antidiabetic Effects of Gegen Qinlian Decoction via the Gut Microbiota Are Attributable to Its Key Ingredient Berberine显示文摘Gegen Qinlian Decoction(GQD),a traditional Chinese medicine(TCM)formula,has long been used for the treatment of common metabolic diseases,including type 2 diabetes mellitus.However,the main limitation of its wider application is ingredient complexity of this formula.Thus,it is critically important to identify the major active ingredients of GQD and to illustrate mechanisms underlying its action.Here,we compared the effects of GQD and berberine,a hypothetical key active pharmaceutical ingredient of GQD,on a diabetic rat model by comprehensive analyses of gut microbiota,short-chain fatty acids,proinflammatory cytokines,and ileum transcriptomics.Our results show that berberine and GQD had similar effects on lowering blood glucose levels,modulating gut microbiota,inducing ileal gene expression,as well as relieving systemic and local inflammation.As expected,both berberine and GQD treatment significantly altered the overall gut microbiota structure and enriched many butyrate-producing bacteria,including Faecalibacterium and Roseburia,thereby attenuating intestinal inflammation and lowering glucose.Levels of short-chain fatty acids in rat feces were also significantly elevated after treatment with berberine or GQD.Moreover,concentration of serum proinflammatory cytokines and expression of immune-related genes,including Nfkb1,Stat1,and Ifnrg1,in pancreatic islets were significantly reduced after treatment.Our study demonstrates that the main effects of GQD can be attributed to berberine via modulating gut microbiota.The strategy employed would facilitate further standardization and widespread application of TCM in many diseases. | Xizhan Xu Zezheng Gao Fuquan Yang Yingying Yang Liang Chen Lin Han Na Zhao Jiayue Xu Xinmiao Wang Yue Ma Lian Shu Xiaoxi Hu Na Lyu Yuanlong Pan Baoli Zhu Linhua Zhao Xiaolin Tong Jun Wang | 2020 | Genomics, Proteomics & Bioinformatics2020,18,6: | 29 |
| 2 | The prognostic and therapeutic implications of circulating tumor cell phenotype detection based on epithelial-mesenchymal transition markers in the first-line chemotherapy of HER2-negative metastatic breast cancer显示文摘Background:Epithelial-mesenchymal transition(EMT)is implicated in the metastatic process and presents a chal-lenge to epithelial cell adhesion molecule-based detection of circulating tumor cells(CTCs),which have been demon-strated to be a prognostic indicator in metastatic breast cancer.Although evidence has indicated that heterogeneity of CTCs based on EMT markers is associated with disease progression,no standard recommendations have been established for clinical practice.This study aimed to evaluate the prognostic significance of dynamic CTC detection based on EMT for metastatic breast cancer patients.Methods:We enrolled 108 human epidermal growth factor receptor 2-negative metastatic breast cancer patients from the prospective phase III CAMELLIA study and applied the CanPatrol CTC enrichment technique to identify CTC phenotypes(including epithelial CTCs,biphenotypic epithelial/mesenchymal CTCs,and mesenchymal CTCs)in peripheral blood samples.Receiver operating characteristic curve analyses of total CTC count and the proportion of mesenchymal CTCs for predicting the 1-year progression-free survival(PFS)rate were conducted to determine the optimal cut-off values,and Kaplan-Meier analysis and Cox proportional hazards regression analysis were performed to investigate the prognostic value of the cut-off values of both total CTC count and the proportion of mesenchymal CTCs in combination.Results:For predicting the 1-year PFS rate,the optimal cut-off value of total CTC count was 9.5(Area under the curve[AUC]=0.538,95%confidence interval[CI]=0.418-0.657),and that of the proportion of mesenchymal CTCs was 10.7%(AUC=0.581,95%CI=0.463-0.699).We used the two cut-off values in combination to forecast PFS in which the total CTC count was equaled to or exceeded 10/5 mL with the proportion of mesenchymal CTCs surpassed 10.7%.Patients who met the combined criteria had significantly shorter median PFS than did those who did not meet the criteria(6.2 vs.9.9 months,P=0.010).A nomogram was constructed based on the criteria and significant clinicopatho-logical characteristics with a C-index of 0.613(P=0.010).Conclusions: The criteria, which combine the total CTC count and the proportion of mesenchymal CTCs, may be used to monitor therapeutic resistance and predict prognosis in patients with metastatic breast cancer. | Xiuwen Guan Fei Ma Chunxiao Li Shiyang Wu Shangying Hu Jiefen Huang Xiaoying Sun Jiayu Wang Yang Luo Ruigang Cai Ying Fan Qiao Li Shanshan Chen Pin Zhang Qing Li Binghe Xu | 2019 | Cancer Communications2019,39,1: | 15 |

| 4 | Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response. | Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan | 2021 | Cancer Biology & Medicine2021,18,3: | 11 |
| 5 | CMHX008,a PPAR γ partial agonist,enhances insulin sensitivity with minor influences on bone loss显示文摘Traditional thiazolidinediones(TZDs),such as rosiglitazone,are peroxisome proliferator-activated receptor g(PPARg)potent agonists that can be used to treat type 2 diabetes but carry unwanted effects,including increased risk for fracture.The present work aimed to compare the insulin-sensitizing efficacies and bone-loss side effects of CMHX008,a novel TZDs-like PPARg partial agonist,with those of rosiglitazone.A TR-FRET PPARg competitive binding assay was used to compare the binding affinity between CMHX008 and rosiglitazone.Mice were administered vehicle,CMHX008 or rosiglitazone for 16 weeks.Mesenchymal stem cells(MSCs)were used to examine differences in differentiation into osteoblasts after compounds treatment.TR-FRET showed lower affinity to PPARg by CMHX008 compared with rosiglitazone.Mice treated with CMHX008 showed insulin sensitization similar to that of mice treated with rosiglitazone,which was related to the significant inhibition of PPARg Ser273 phosphorylation and improved insulin sensitivity by facilitating the phosphorylation of insulin receptor and Akt in adipose tissues.Micro-CT and histomorphometric analyses demonstrated that the degree of trabecular bone loss after treatment with CMHX008 was weaker than that observed with rosiglitazone,as evidenced by consistent changes in BV/TV,Tb.N,Tb.Th,Tb.Sp,and the mineral apposition rate.MSCs treated with CMHX008 showed higher ALP activity and mRNA levels of bone formation markers than did cells treated with rosiglitazone in the osteoblast differentiation test.Thus,CMHX008 showed insulin-sensitizing effects similar to those of rosiglitazone with a lower risk of bone loss,suggesting that PPARg sparing eliminates the skeletal side effects of TZDs while maintaining their insulin-sensitizing properties. | Yi Hou Xuemei Cao Xiangnan Hu Xinyu Li Xiaoqin Shi Hongying Wang Chuan Peng Jiayu Li Jibin Li Qifu Li Chaodong Wu Xiaoqiu Xiao | 2018 | Genes & Diseases2018,5,3: | 6 |

Two different pre-coating structures, including porous B_4C–C and dense C/B, are designed by slurry dip and chemical vapor deposition(CVD) process respectively. The coating prepared by reactive melt infiltration(RMI) into B_4C–C presents a flat and smooth surface with a three-layer cross-sectional structure, namely interior SiC transition layer, gradient ZrB_2–ZrC–SiC layer, and ZrB_2–ZrC exterior layer. In comparison, the coating prepared by RMI into C/B shows a more granular surface with a different three-layer cross-sectional structure, interior unreacted B–C pre-coating layer, middle SiC layer, and exterior ZrB_2–ZrC–ZrSi_2 layer. | 7 | Effectiveness of vegetation on phosphorus removal from reclaimed water by a subsurface flow wetland in a coastal area显示文摘This work was conducted to evaluate the effectiveness and influence factors of vegetation on phosphorus (P) removal from reclaimed water in constructed wetlands.Comparisons were conducted between one pilot scale subsurface flow wetland (P-SSFW) and two demonstration subsurface flow wetlands,which were series-wound and named as first subsurface flow wetland (F-SSFW),and second subsurface flow wetland (S-SSFW),respectively.The three wetlands had the same vegetation and substrate,but different pH values,total dissolved solids (TDS) and P loads.Results showed that the P content in the vegetation shoots of the F-SSFW was 2.16 mg/g,while 2.31 mg/g in the S-SSFW and 2.69 mg/g in the P-SSFW.These differences were likely caused by the higher pH and TDS in the reclaimed water.The P content also differed among the tissues of the plant,which were 5.94-6.44 mg/g,2.20-2.77 mg/g,1.31-1.46 mg/g and 1.53-1.88 mg/g in the flowers,leaves,stems,and roots,respectively.The greatest discrepancy was observed in the leaves,indicating that the environment of the wetlands had the greatest influence on the leaves.When the total phosphorus (TP) load was lower,the proportion of P removed by vegetation assimilation was 16.17% in the P-SSFW,12.90% in the F-SSFW and 13.29% in the S-SSFW.However,the relative removal efficiency by vegetation among the three wetlands did not vary greatly from that observed in other studies.Moreover,the influence of pH,TDS and TP load was not as great as the influence of the vegetation species,type of substrate,influent style or climate. | Baoqing Shan Liang Ao Chunming Hu Jiayu Song | 2011 | Journal of Environmental Sciences2011,23,10: | 6 |
| 8 | High performance integrated photonic circuit based on inverse design method显示文摘The basic indexes of all-optical integrated photonic circuits include high-density integration,ultrafast response and ultralow energy consumption.Traditional methods mainly adopt conventional micro/nano-structures.The overall size of the circuit is large,usually reaches hundreds of microns.Besides,it is difficult to balance the ultrafast response and ultra-low energy consumption problem,and the crosstalk between two traditional devices is difficult to overcome.Here,we propose and experimentally demonstrate an approach based on inverse design method to realize a high-density,ultrafast and ultra-low energy consumption integrated photonic circuit with two all-optical switches controlling the input states of an all-optical XOR logic gate.The feature size of the whole circuit is only 2.5μm×7μm,and that of a single device is 2μm×2μm.The distance between two adjacent devices is as small as 1.5μm,within wavelength magnitude scale.Theoretical response time of the circuit is 150 fs,and the threshold energy is within 10 fJ/bit.We have also considered the crosstalk problem.The circuit also realizes a function of identifying two-digit logic signal results.Our work provides a new idea for the design of ultrafast,ultra-low energy consumption all-optical devices and the implementation of high-density photonic integrated circuits. | Huixin Qi Zhuochen Du Xiaoyong Hu Jiayu Yang Saisai Chu Qihuang Gong | 2022 | Opto-Electronic Advances2022,5,10: | 5 |

| 10 | Machine-learning-based high-resolution DOA measurement and robust directional modulation for hybrid analog-digital massive MIMO transceiver显示文摘At hybrid analog-digital(HAD)transceiver,an improved HAD estimation of signal parameters via rotational invariance techniques(ESPRIT),called I-HAD-ESPRIT,is proposed to measure the direction of arrival(DOA)of a desired user,where the phase ambiguity due to HAD structure is dealt with successfully.Subsequently,a machine-learning(ML)framework is proposed to improve the precision of measuring DOA.Meanwhile,we find that the probability density function(PDF)of DOA measurement error(DOAME)can be approximated as a Gaussian distribution by the histogram method in ML.Then,a slightly large training data set(TDS)and a relatively small real-time set(RTS)of DOA are formed to predict the mean and variance of DOA/DOAME in the training stage and real-time stage,respectively.To improve the precisions of DOA/DOAME,three weight combiners are proposed to combine the-maximum-likelihood-learning outputs of TDS and RTS.Using the mean and variance of DOA/DOAME,their PDFs can be given directly,and we propose a robust beamformer for directional modulation(DM)transmitter with HAD by fully exploiting the PDF of DOA/DOAME,especially a robust analog beamformer on RF chain.Simulation results show that:(1)the proposed I-HAD-ESPRIT can achieve the HAD Cramer-Rao lower bound(CRLB);(2)the proposed ML framework performs much better than the corresponding real-time one without training stage;(3)the proposed robust DM transmitter can perform better than the corresponding non-robust ones in terms of secrecy rate. | Zhihong ZHUANG Ling XU Jiayu LI Jinsong HU Linlin SUN Feng SHU Jiangzhou WANG | 2020 | Science China(Information Sciences)2020,63,8: | 5 |

| 12 | Development of high-resolution chloroplast markers for intraspecific phylogeographic studies of Phaeocystis globosa显示文摘Phaeocystis globosa is an important harmful algal bloom causative species distributing widely in temperate and tropical coastal waters in the world.The morphological,physiological,and biochemical characteristics are different among geographic strains,which can not be distinguished with nuclear ribosomal DNA markers at present.Therefore,the genetic distance and phylogeographic relationships of nuclear 28S rDNA D1–D2 and ITS regions,and three chloroplast intergenic spacers(petN-trnS1,trnM1-psbA,and rbcS-rpl27)were analyzed and compared among 13 strains of P.globosa isolated from the Pacific Ocean and Atlantic Ocean in this study.In addition,the nucleotide polymorphisms of 28S rDNA D1–D2,ITS,and rbcS-rpl27 regions were evaluated in two P.globosa strains.The various levels of nucleotide polymorphism were in the nuclear 28S rDNA D1–D2 region and ITS region,but no polymorphism was in the chloroplast rbcS-rpl27 intergenic spacer.A reasonable intraspecific phylogeographic relationship was presented by rbcS-rpl27 intergenic spacer,which had the strongest distinction to geographic strains compared to those of 28S rDNA D1–D2 and ITS regions.In the phylogenetic tree of rbcS-rpl27 intergenic spacer,the two strains from the North Sea of the Atlantic Ocean were divided firstly from the species of P.globosa,and then formed an independent clade,while the other Atlantic strains and all of Pacific strains joined up to build the other clade.It was implied that at least two genetically distant populations of P.globosa existed in the Atlantic coastal regions.This study provided a high-resolution chloroplast marker to analyze intraspecific phylogeographic populations of P.globosa,and preliminarily clarified the genetic relationships of the Pacific and Atlantic strains of P.globosa. | Qingchun ZHANG Zhuang NIU Jinxiu WANG Chao LIU Fanzhou KONG Xiaokun HU Jiayu ZHAO Rencheng YU | 2021 | Journal of Oceanology and Limnology2021,39,2: | 3 |

| 14 | Organosulfur compounds induce cytoprotective autophagy against apoptosis by inhibiting mTOR phosphorylation activity in macrophages显示文摘Organosulfur compounds (OSCs)are the bioactive components of garlic.Some OSCs have apoptotic or autophagy-inducing effects.Autophagy plays roles in both cytoprotection and apoptosis-related cell death,and the interaction between autophagy and apoptosis is important in the modulation of immune responses.The mechanism of an OSC-mediated effect via the interaction of autophagy and apoptosis is unknown.In this study,the effects of five OSC compounds on autophagy in the macrophage cell line RAW264.7 and primary macrophages were investigated.We found that S-allylcysteine (SAC),diallyl disulde (DADS)and diallyl tetrasulfide (DTS)treatment increased the number of autophagosomes of RAW264.7 cells,inhibited the phosphorylation of ribosomal protein S6 kinase beta-1 (p70S6K/S6K1)which is a substrate of mammalian target of rapamycin (roTOR),and significantly enhanced autophagy flux.The induction of autophagy by SAC,DADS and DTS was inhibited by stably knocking down the expression of autophagy-related gene 5 (ATG5)with short hairpin RNA (shRNA).Further experiments confirmed that SAC,DADS and DTS also induced apoptosis in RAW264.7 cells.The induction of apoptosis and Caspase 3 activity by SAC,DADS and DTS were increased by stably knocking down of ATG5 expression with shRNA in RAW264.7 cells or treating with 5 mM 3-MA in primary macrophages.Our results suggest that SAC,DADS and DTS induce both autophagy and apoptosis.The autophagy induction protects macrophages from apoptosis by inhibiting mTOR phosphorylation activity to maintain the mass of immune cells. | Yanyang Wu Yongquan Hu Haiyan Zhou Jiayu Zhu Zhongyi Tong Si Qin Dongbo Liu | 2018 | Acta Biochimica et Biophysica Sinica2018,50,11: | 2 |

| 19 | Active oxygen species and oxidation mechanism over Ce-doped LaMn_(0.8)Ni_(0.2)O_(3)/hierarchical ZSM-5 in pentanal oxidation显示文摘Hierarchical ZSM-5(HZ)molecular sieves based on fly ash were synthesized using a method combining water heat treatment with step-by-step calcination.The coupling catalysts between La_(1-x)Ce_(x)Mn_(0.8)-Ni_(0.2)O_(3)(x≤0.5)perovskites and HZ were prepared through the impregnation method,which were characterized by X-ray diffraction(XRD),scanning electron microscopy(SEM),high-resolution transmission electron microscopy(HRTEM),N_(2)adsorption,X-ray photoelectron spectroscopy(XPS),NH3-temperature programmed desoprtion(NH3-TPD),H_(2)-temperature programmed reduction(H_(2)-TPR)and O_(2)-TPD techniques and investigated regarding pentanal oxidation at 120-390℃to explore the effects of Ce doping on the catalytic activity and the active oxygen species of the coupling catalysts,meanwhile,the reaction mechanism and pathway of pentanal oxidation were also studied.The results reveal that Ce substitution at La sites can change the electronic interactions between all the elements and promote the electronic transfer among La,Ce,Ni,Mn and HZ,influencing directly the physicochemical characteristics of the catalysts.Moreover,the amount and transfer ability of surface adsorbed oxygen(O_(2)-and O-)regarded as the reactive oxygen species and the low temperature reducibility are the main influence factors in pentanal oxidation.Additionally,La_(0.8)Ce_(0.2)Mn_(0.8)Ni_(0.2)O_(3)/HZ exhibits the best catalytic activity and deep oxidation capacity as well as a better water resistance due to its larger amount of surface adsorbed oxygen species and higher low temperature reducibility.What’s more,appropriate Ce substitution can significantly enhance the amount of O_(2)-ions,which can distinctly enhance the catalytic activity of the catalyst,and moderate acid strength and appropriate acid amount can also facilitate the improvement of the pentanal oxidation activity.It is found that there is a synergic catalytic effect between surface acidity and redox ability of the catalyst.According to the in situ DRIFTS and GC/MS analyses,pentanal can be oxidized gradually to CO_(2)and H_(2)O by the surface oxygen species with the form of adsorption in air following the Langmuir-Hinshelwood(L-H)reaction mechanism.Two reaction pathways for the pentanal oxidation process are proposed,and the conversion of the formates to carbonates may be one of the main rate-determining steps. | Jian Li Yingjie Shi Xiaoheng Fu Yun Shu Jiayu Huang Jinwei Zhu Gang Tian Jingnan Hu | 2021 | Journal of Rare Earths2021,39,9: | 2 |
