1Antimony speciation in the environment:Recent advances in understanding the biogeochemical processes and ecological effects显示文摘Antimony(Sb) is a toxic metalloid, and its pollution has become a global environmental problem as a result of its extensive use and corresponding Sb-mining activities. The toxicity and mobility of Sb strongly depend on its chemical speciation. In this review, we summarize the current knowledge on the biogeochemical processes(including emission, distribution,speciation, redox, metabolism and toxicity) that trigger the mobilization and transformation of Sb from pollution sources to the surrounding environment. Natural phenomena such as weathering, biological activity and volcanic activity, together with anthropogenic inputs, are responsible for the emission of Sb into the environment. Sb emitted in the environment can adsorb and undergo redox reactions on organic or inorganic environmental media, thus changing its existing form and exerting toxic effects on the ecosystem. 3Combined obeticholic acid and apoptosis inhibitor treatment alleviates liver fibrosis显示文摘Obeticholic acid(OCA), the first FXR-targeting drug, has been claimed effective in the therapy of liver fibrosis. However, recent clinical trials indicated that OCA might not be effective against liver fibrosis, possibly due to the lower dosage to reduce the incidence of the side-effect of pruritus. Here we propose a combinatory therapeutic strategy of OCA and apoptosis inhibitor for combating against liver fibrosis. CCl4-injured mice, D-galactosamine/LPS(GalN/LPS)-treated mice and cycloheximide/TNFα(CHX/TNFα)-treated HepG2 cells were employed to assess the effects of OCA, or together with IDN-6556, an apoptosis inhibitor. OCA treatment significantly inhibited hepatic stellate cell(HSC)activation/proliferation and prevented fibrosis. Elevated bile acid(BA) levels and hepatocyte apoptosis triggered the activation and proliferation of HSCs. OCA treatment reduced BA levels but could not inhibit hepatocellular apoptosis. An enhanced anti-fibrotic effect was observed when OCA was co-administrated with IDN-6556. Our study demonstrated that OCA inhibits HSCs activation/proliferation partially by regulating BA homeostasis and thereby inhibiting activation of HSCs. 4Integration of A Deep Learning Classifier with A Random Forest Approach for Predicting Malonylation Sites显示文摘As a newly-identified protein post-translational modification, malonylation is involved in a variety of biological functions. Recognizing malonylation sites in substrates represents an initial but crucial step in elucidating the molecular mechanisms underlying protein malonylation. In this study, we constructed a deep learning(DL) network classifier based on long short-term memory(LSTM) with word embedding(LSTMWE) for the prediction of mammalian malonylation sites.LSTMWEperforms better than traditional classifiers developed with common pre-defined feature encodings or a DL classifier based on LSTM with a one-hot vector. The performance of LSTMWE is sensitive to the size of the training set, but this limitation can be overcome by integration with a traditional machine learning(ML) classifier. Accordingly, an integrated approach called LEMP was developed, which includes LSTMWEand the random forest classifier with a novel encoding of enhanced amino acid content. LEMP performs not only better than the individual classifiers but also superior to the currently-available malonylation predictors. Additionally, it demonstrates a promising performance with a low false positive rate, which is highly useful in the prediction application. 5Defect inspection of flip chip package using SAM technology and fuzzy C-means algorithm显示文摘Solder bumps are widely used in surface mount components, which provide electrical and mechanical connection between the chip/package and the substrate. As the solder bump getting smaller in dimension and pitch, it becomes more difficult to inspect the solder defects hidden in the IC package. In this paper, an intelligent inspection method using the scanning acoustic microscopy(SAM) and the fuzzy C-means(FCM) algorithm was investigated. A flip chip package of FA10 was chosen as the test sample. The SAM tests of FA10 were carried out in C-scan mode. The sub-image of every solder bump was segmented from the SAM image. The statistical features were then calculated and adopted for clustering of solder bumps using the FCM algorithm. The recognition results of FCM reached a high accuracy of 94.3%. 7Reconfigurable photovoltaic effect for optoelectronic artificial synapse based on ferroelectric p-n junction显示文摘Neuromorphic machine vision has attracted extensive attention on wide fields.However,both current and emerging strategies still suffer from power/time inefficiency,and/or low compatibility,complex device structure.Here we demonstrate a driving-voltage-free optoelectronic synaptic device using non-volatile reconfigurable photovoltaic effect based on MoTe_(2)/α-In_(2)Se_(3) ferroelectric p-n junctions.This function comes from the non-volatile reconfigurable built-in potential in the p-n junction that is related to the ferroelectric polarization inα-In_(2)Se_(3).Reconfigurable rectification behavior and photovoltaic effect are demonstrated firstly.Notably,the figure-of-merits for photovoltaic effect like photoelectrical conversion efficiency non-volatilely increases more than one order.Based on this,retina synapse-like vision functions are mimicked.Optoelectronic short-term and long-term plasticity,as well as basic neuromorphic learning and memory rule are achieved without applying driving voltage.Our work highlights the potential of ferroelectric p-n junctions for enhanced solar cell and low-power optoelectronic synaptic device for neuromorphic machine vision.Yanrong Wang Feng Wang Zhenxing Wang Junjun Wang Jia Yang Yuyu Yao Ningning Li Marshet Getaye Sendeku Xueying Zhan Congxin Shan Jun He 2021Nano Research2021,14,11:2
8Influence of Deposition Pressure on Properties of ZnO:Al Films Fabricated by RF Magnetron Sputtering显示文摘Transparent conductive aluminum doped zinc oxide(ZnO:Al,AZO) films were prepared on glass substrates by rf(radio frequency) magnetron sputtering from ZnO: 3wt% Al_2O_3 ceramic target. The effect of argon gas pressure(PAr) was investigated with small variations to understand the influence on the electrical, optical and structural properties of the films. Structural examinations using X-ray diffraction(XRD) and scanning electron microscopy(SEM) showed that the ZnO:Al thin films were(002) oriented. The resistivity values were measured by four-point probe with the lowest resistivity of 5.76×10^(-4) Ω?cm(sheet resistance=9.6 Ω/sq. for a thickness=600 nm) obtained at the PAr of 0.3 Pa. The transmittance was achieved from ultravioletvisible(UV-VIS) spectrophotometer, 84% higher than that in the visible region for all AZO thin films. 9The sensitivity of the array resistivity log to mud filtrate invasion and its primary five-parameter inversion for improved oil water recognition显示文摘In order to improve reservoir fluid recognition, the sensitivity of array resistivity response to the difference of the invasion properties in both oil-bearing layers and water layers is analyzed. Then the primary inversion is carried out based on the array resistivity log. The mud invasion process is numerically simulated based on the oil-water flow equation and water convection diffusion equation. The results show that the radial resistivity of a fresh mud-invaded oil-bearing layer presents complex distribution characteristics, such as nonlinear increase, increasing to decreasing and low resistivity annulus, and the resistive invasion profile of a water layer is monotonic. Under specific conditions, array resistivity log can reflect these changes and the array induction log is more sensitive. Nevertheless, due to the effect of factors like large invasion depth, reservoir physical and oil-bearing properties, the measured apparent resistivity may differ greatly from the actual mud filtrate invasion profile in an oil-bearing layer. We proposed a five-parameter formation model to simulate the complex resistivity distribution of fresh mud-invaded formation. Then, based on the principle of non-linear least squares, the measured array resistivity log is used for inversion with the Marquardt method. It is demonstrated that the inverted resistivity is typically non-monotonic in oil-bearing layers and is monotonic in water layers. 10Comparisons of stable isotopic fractionation in winter and summer at Baishui Glacier No.1,Mt.Yulong,China显示文摘Based on the data of δ18O in surface snow, snow pits, meltwater and the glacier-fed river water at Baishui Glacier No.1, Mt. Yulong, the isotopic fractionation behaviors in the typical monsoonal temperate glacier system in winter and summer were compared. The results indicate that the isotopic fractionation degree in summer is greater than that in winter, suggesting that the snow/ice melting is more intense in summer. Moreover, whenever it is in winter or summer, from surface snow to meltwater, and to glacier-fed river water, the gradient of δ18O with altitude gradually increases. 11Discovery of cutinitic liptobiolith in northwestern China and a comparative study with Luquan Devonian coal显示文摘Devonian cutinitic liptobiolith is a special cuticle-rich coal formed during the early stage of land plant evolution. In China, this special coal has previously only been reported in the Yangtze area, with the Luquan Devonian cutinitic liptobiolith of Yunnan Province being the most well-known. Recently, this type of coal was first discovered in the Bulonggoer Sag, to the northwest of the Junggar Basin, NW Xinjiang. Its geological age is regarded as the Givetian period of the late Middle Devonian,which is equivalent to the age of the Luquan Devonian coal. The cutinitic liptobiolith from NW Xinjiang has a distinct sheet-like texture resembling a stack of paper or leaf sheets in appearance, and so it is commonly called 'paper coal' or 'leaf coal'. Ribbonlike cutinite(thick-walled type) is the most abundant maceral in the coal(>60% and up to 80%, on a mineral-free basis), followed by ribbon-like collotelinite. The NW Xinjiang and Luquan cutinitic liptobioliths share similar maceral compositions;while their coal-forming plants and depositional conditions are distinct. The NW Xinjiang cutinitic liptobiolith was deposited in a regional fluvial or flood plain condition, with lycopsids as the dominant coal-forming plants. In contrast, the Luquan cutinitic liptobiolith formed in a coastal-lagoon environment, with primitive ferns as the predominant coal-forming plants, followed by lycopsids. The saturated fractions of the NW Xinjiang and Luquan liptobioliths are mainly composed of tetracyclic diterpanes. However, the tetracyclic diterpane compositions in the NW Xinjiang cutinitic liptobiolith are distinct from those in the Luquan Devonian coal:The former is dominated by atisanes and the latter is mainly composed of phyllocladanes. 18DNMT3A inhibits E2F1-induced arterial marker expression and impairs angiogenesis in human umbilical artery endothelial cells显示文摘Arterial marker genes EphrinB2 and HEY2 are essential for cardiovascular development and post-natal neovascularization.Our previous study confirmed.that E2F1 could activate the transcription of EphrinB2 and HEY2 in human mesenchymal stem cells;however,the detailed mechanism has not been resolved yet.In this study,we focused on the interaction between E2F1 and DNMT3A,a de novo DNA methyltransferase,on regulating the expression of EphrinB2 and HEY2,and explored the potential mechanisms.Gain-and loss-of-function experiments implicated the positive effect of E2F1 on the expression of EphrinB2 and HEY2 and tube formation in human umbilical artery endothelial cells.Accumulation of DNMT3A decreased the levels of EphrinB2 and HEY2,and impaired tube formation induced by E2F1,while inhibiting DNMT3A by RNA interference aug-mented their expression and angiogenesis in E2F 1-trasfected cells.We then asked whether the low expressions of EphrinB2 and HEY2 induced by DNMT3A are related to the methylation status of their promoters.Surprisingly,the methylation status of the CpG islands in the promoter region was not significantly affected by overexpression of exogenous DNMT3A.Furthermore,the inter-action between E2F1 and DNMT3A was confirmed by co-immunoprecipitation.DNMT3A could inhibit the transcription of EphrinB2 and HEY2 promoters by affecting the binding of E2F1 to its recognition sequences as revealed by luciferase reporter assay and chromatin immunoprecipita-tion.These results identified a novel mechanism underlying the cooperation of DNMT3A with E2F1 on regulating target gene expression,and revealed their roles in the angiogenic process.Kaiyue Su Ningning Lin Shouqiang Xie Yabo Han Zaiming Yang Hongmin Zhang Hongpeng He Sa Zhou Wenjian Ma Tongcun Zhang Nan Wang 2020Acta Biochimica et Biophysica Sinica2020,52,11:1

20BLM helicase inhibition synergizes with PARP inhibition to improve the radiosensitivity of olaparib resistant non-small cell lung cancer cells by inhibiting homologous recombination repair显示文摘Objective:We aimed to investigate the radiosensitizing efficacy of the poly-ADP-ribose polymerase(PARP)inhibitor,olaparib,and the Bloom syndrome protein(BLM)helicase inhibitor,ML216,in non-small cell lung cancer(NSCLC)cells.Methods:Radiosensitization of NSCLC cells was assessed by colony formation and tumor growth assays.Mechanistically,the effects of ML216,olaparib,and radiation on cell and tumor proliferation,DNA damage,cell cycle,apoptosis,homologous recombination(HR)repair,and non-homologous end joining(NHEJ)repair activity were determined.Results:Both olaparib and ML216 enhanced the radiosensitivities of olaparib-sensitive H460 and H1299 cells,which was seen as decreased surviving fractions and Rad51 foci,increased total DNA damage,andγH2AX and 53BP1 foci(P<0.05).The expressions of HR repair proteins were remarkably decreased in olaparib-treated H460 and H1299 cells after irradiation(P<0.05),while olaparib combined with ML216 exerted a synergistic radiosensitization effect on olaparib-resistant A549 cells.In addition to increases of double strand break(DSB)damage and decreases of Rad51 foci,olaparib combined with ML216 also increased pDNA-PKcs(S2056)foci,abrogated G2 cell cycle arrest,and induced apoptosis in A549 lung cancer after irradiation in vitro and in vivo(P<0.05).Moreover,Western blot showed that olaparib combined with ML216 and irradiation inhibited HR repair,promoted NHEJ repair,and inactivated cell cycle checkpoint signals both in vitro and in vivo(P<0.05).Conclusions:Taken together,these results showed the efficacy of PARP and BLM helicase inhibitors for radiosensitizing NSCLC cells,and supported the model that BLM inhibition sensitizes cells to PARP inhibitor-mediated radiosensitization,as well as providing the basis for the potential clinical development of this combination for tumors intrinsically resistant to PARP inhibitors and radiotherapy.Yangyang Kong Chang Xu Xiaohui Sun Hao Sun Xiaotong Zhao Ningning He Kaihua Ji Qin Wang Liqing Du Jinhan Wang Manman Zhang Yang Liu Yan Wang Qiang Liu 2022Cancer Biology & Medicine2022,19,8:1
