2Effects of Pinus massoniana bark extract on cell proliferation and apoptosis of human hepatoma BEL-7402 cells显示文摘AIM: To study the effects of Pinus massoniana bark extract (PMBE) on cell proliferation and apoptosis of human hepatoma BEL-7402 cells and to elucidate its molecular mechanism.METHODS: BEL-7402 cells were incubated with various concentrations (20-200 μg/mL) of PMBE for different periods of time. After 48 h, cell proliferation was determined by 3-(4,5-dimethyl-thiazolyl-2)-2,5-diphenyl tetrazolium bromide (MTT) assay. Apoptosis was evaluated by morphological observation, agarose gel electrophoresis,and flow cytometry analysis. Possible molecular mechanisms were primarily explored through immunohistochemical staining.RESULTS: PMBE (20-200 μg/mL) significantly suppressed BEL-7402 cell proliferation in a time- and dose-dependent manner. After treatment of BEL-7402 cells with 160 μg/mL PMBE for 24, 48, or 72 h, a typical apoptotic 'DNA ladder'was observed using agarose gel electrophoresis. Nuclear condensation and boundary aggregation or split, apoptotic bodies were seen by fluorescence and electron microscopy.Sub-G1 curves were displayed by flow cytometry analysis.PMBE decreased the expression levels of Bcl-2 protein in a time-dependent manner after treatment of cells with 160 μg/mL PMBE.CONCLUSION: PMBE suppresses proliferation of BEL-7402 cells in a time- and dose-dependent manner and induces cell apoptosis by possibly downregulating the expression of the bcl-2 gene.Ying-Yu Cui Heng Xie Kang-Biao Qi Yan-Ming He Jin-Fa Wang 2005World Journal of Gastroenterology2005,11,34:10
3EFFECTS OF VITAMIN D RECEPTOR GENE POLYMORPHISMS ON SUSCEPTIBILITY TO TYPE 1 DIABETES MELLITUS显示文摘Objective To investigate the influence of vitamin D receptor (VDR) gene polymorphisms on susceptibility to type 1 diabetes mellitus (T1DM) in the Chinese Han population. Method One hundred and thirty-six Chinese Han people, including 54 T1DM patients and 82 unrelated healthy subjects as control were genotyped by polymerase chain reaction-restriction fragment length polymorphism for three restriction sites in the VDR gene, which were ApaI, TaqI, and BamI. Results The frequency of B allele of BsmI site in VDR gene was significantly higher in T1DM patients than in healthy subjects (P = 0.033) while no difference was found between the two groups in the distribution of ApaI and TaqI polymorphisms. 4In Situ Coupling Strategy for Anchoring Monodisperse Co_9S_8 Nanoparticles on S and N Dual?Doped Graphene as a Bifunctional Electrocatalyst for Rechargeable Zn–Air Battery显示文摘An in situ coupling strategy to prepare Co_9S_8/S and N dual?doped graphene composite(Co_9S_8/NSG) has been proposed. The key point of this strategy is the function?oriented design of organic compounds. Herein, cobalt porphyrin derivatives with sulfo groups are employed as not only the coupling agents to form and anchor Co_9S_8 on the graphene in situ, but also the heteroatom?doped agent to generate S and N dual?doped graphene. The tight coupling of multiple active sites endows the composite materials with fast electrochemical kinetics and excellent stability for both oxygen reduction reaction(ORR) and oxygen evolution reaction(OER). The obtained electrocatalyst exhibits better activity parameter(ΔE = 0.82 V) and smaller Tafel slope(47.7 mV dec^(-1) for ORR and 69.2 mV dec^(-1) for OER) than commercially available Pt/C and RuO_2. Most importantly, as electrocatalyst for rechargeable Zn–air battery, Co_9S_8/NSG displays low charge–discharge voltage gap and outstanding long?term cycle stability over 138 h compared to Pt/C–RuO_2. To further broaden its application scope, a homemade all?solid?state Zn–air battery is also prepared, which displays good charge–discharge performance and cycle performance. 7The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. 8RELATIONSHIP BETWEEN HEMOGLOBIN A1C AND BLOOD GLUCOSE THROUGHOUT THE DAY IN WELL-GLYCEMIC-CONTROLLED MEDICAL NUTRITION THERAPY ALONE TYPE 2 DIABETIC PATIENTS显示文摘Objective To investigate the relationship between glycosylated hemoglobin A1c (HbA1c) and blood glucose levels of eight different points throughout the day in well-glycemic-controlled medical nutrition therapy (MNT) alone type 2 diabetic patients. Methods Data were collected as capillary blood glucose value of eight different sample points among sixteen observing days in thirty MNT alone type 2 diabetic patients. The correlation between HbA1c and capillary blood glucose value was evaluated by Pearson’s correlation method. Results The r-values between HbA1c and capillary blood glucose of 3:00, 6:00, and bedtime (22:00-23:00) were 0.81, 0.79, and 0.78, respectively(P<0.001). The best correlation was found between the mean value of 8-point blood glucose value throughout the day and HbA1c (r=0.84, P<0.001). 9Atomic Fe-Zn dual-metal sites for high-efficiency pH-universal oxygen reduction catalysis显示文摘An effective electrocatalyst being highly active in all pH range for oxygen reduction reaction(ORR)is crucial for energy conversion and storage devices.However,most of the high-efficiency ORR catalysis was reported in alkaline conditions.Herein,we demonstrated the preparation of atomically dispersed Fe-Zn pairs anchored on porous N-doped carbon frameworks(Fe-Zn-SA/NC),which works efficiently as ORR catalyst in the whole pH range.It achieves high half-wave potentials of 0.78,0.85 and 0.72 V in 0.1 M HClO4,0.1 M KOH and 0.1 M phosphate buffer saline(PBS)solutions,respectively,as well as respectable stability.The performances are even comparable to Pt/C.Furthermore,when assembled into a Zn-air battery,the high power density of 167.2 mWcm−2 and 120 h durability reveal the feasibility of Fe-Zn-SA/NC in real energy-related devices.Theoretical calculations demonstrate that the superior catalytic activity of Fe-Zn-SA/NC can be contributed to the lower energy barriers of ORR at the Fe-Zn-N6 centers.This work demonstrates the potential of Fe-Zn pairs as alternatives to the Pt catalysts for efficient catalytic ORR and provides new insights of dual-atom catalysts for other energy conversion related catalytic reactions.Jie Xu Shuhua Lai Defeng Qi Min Hu Xianyun Peng Yifan Liu Wei Liu Guangzhi Hu Heng Xu Fan Li Chao Li Jia He Longchao Zhuo Jiaqiang Sun Yuan Qiu Shusheng Zhang Jun Luo Xijun Liu 2021Nano Research2021,14,5:6
10EFFECTS OF GLUCAGON ON ISLET β CELL FUNCTION IN PATIENTS WITH DIABETES MELLITUS显示文摘Objective To evaluate islet β cell response to intravenous glucagon(a non-glucose secretagogue)stimulation in diabetes mellitus.Methods Nineteen patients with type 1 diabetes(T1D)and 131 patients with type 2 diabetes(T2D)were recruited in this study.T2D patients were divided into two groups according to therapy:36 cases treated with insulin and 95 cases treated with diet or oral therapy.The serum C-peptide levels were determined at fasting and six minutes after intravenous injection of 1 mg of glucagon.Results Both fasting and 6-minute post-glucagon-stimulated C-peptide levels in T1D patients were significantly lower than those of T2D patients(0.76±0.36 ng/mL vs.1.81±0.78 ng/mL,P<0.05;0.88±0.42 ng/mL vs.3.68±0.98 ng/mL,P<0.05).In T1D patients,the C-peptide level after injection of glucagon was similar to the fasting level.In T2D,patients treated with diet or oral drug had a significantly greater fasting and stimulated C-peptide level than those patients received insulin therapy(2.45±0.93 ng/mL vs.1.61±0.68 ng/mL,P<0.05;5.26±1.24 ng/mL vs.2.15±0.76 ng/mL,P<0.05).The serum C-peptide level after glucagon stimulation was positively correlated with C-peptide levels at fasting in all three groups(r=0.76,P<0.05).Conclusions The 6-minute glucagon test is valuable in assessing the function of islet β cell in patients with diabetes mellitus.It is helpful for diagnosis and treatment of diabetes mellitus.Tong Wang Xin-hua Xiao Wen-hui Li Heng Wang Qi Sun Tao Yuan Guo-hua Yang 2008Chinese Medical Sciences Journal2008,23,2:5
11Ozone vertical variations during a typhoon derived from the OMI observations and reanalysis data显示文摘Typhoon is considered to play a key role in the dynamical exchange processes taking place between the troposphere and stratosphere.In this paper,the impact of typhoon on the ozone distribution in the upper troposphere and middle stratosphere is investigated using ozone profiles measured by Aura’s Ozone Monitoring Instrument and meteorological fields from reanalysis data.During the passage of Typhoon Hai-Tang in 2005 over the western North Pacific,low values of ozone column above 200 hPa and ozone mixing ratio between the upper troposphere and the middle stratosphere(from 200 to 50 hPa)are observed right above the typhoon’s track,a result due to the strong upward propagation of air associated with overshooting convection in typhoon.A comparative analysis of different stages of Hai-Tang suggests that in the region where typhoon has higher intensity the troposphere-to-stratosphere transport is enhanced.These results indicated that the typhoon has a significant impact on the ozone variation in the upper troposphere and the middle stratosphere.FU YunFei XIAN Tao Lü DaRen LIU GuoSheng HENG ZhiWei SUN Liang LIU Qi WANG Yu YANG YuanJian 2013Chinese Science Bulletin2013,58,32:5

13The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. 14Chip formation dependence of machining velocities in nano-scale by molecular dynamics simulations显示文摘In this study, molecular dynamics simulations were carried out to study the effect of machining velocities on the mechanism of chip formation in nano-metric copper. A wide range of cutting velocities was performed from 10 to 2000 m/s, and the microstructure's evolution from a crystalline state to an amorphous state was studied. At the low machining velocity, dislocations were generated from the surface in front of the tool, and the immobile dislocation deduced by the cross slip of dislocation was observed. At the high machining velocity, no crystal dislocation nucleated, but instead disorder atoms were found near the tool. 17Greenhouse gas emissions from a constructed wetland for municipal sewage treatment显示文摘The fluxes of greenhouse gases (methane and nitrous oxide) emission from a constructed wetland in the Eastern China as municipal sewage treatment were measured from June 1999 to August 2000 by the closed chamber method. The constructed wetland for municipal sewage treatment is a significant source of methane, up to 976 6×10 6 g CH 4/a, which was emitted from the constructed wetland with the area of 495000 m 2 and wastewater loading rate of 12000 m 3/d. Its daily mean methane flux reached 5 22 g CH 4/(m 2·d), 250 times as much as that in natural wetland in the same latitude region. 227 8 mg CH 4 was produced from the treatment of 1 liter wastewater, up to 700—1000 times as much as that in the secondary treatment. 20A collaborative strategy for enhanced reduction of Cr(Ⅵ) by Fe(0) in the presence of oxalate under sunlight:Performance and mechanism显示文摘Nanometer-size zero-valent iron(NZVI)is an efficient reducing agent,but its surface is easily passivated with an oxide layer,leading to reaction inefficiency.In our study,oxalate(OA)was introduced into this heterogeneous system of NZVI,which could form ferrioxalate complexes with the NZVI surface-bound Fe3+and dissolved Fe3+in the solution.Photolysis of ferrioxalate complexes can facilitate the generation of Fe2+from Fe3+and CO2·-radical,both species have strong reduction capacity.Hence,a'photo-oxalate-Fe(0)'system through sunlight induction was established,which not only prohibited the formation of a surface passivation layer,but also displayed a synergetic mechanism of ferrioxalate photolysis to enhance reduction,exhibiting remarkably higher degradation activity(several times faster)toward the model pollutant Cr(Ⅵ)than the mechanism with NZVI alone.Factor tests suggested that both NZVI dosage and OA content markedly affected the reduction rate.Low pH was beneficial to the reduction efficiency.Moreover,recyclability experiment showed that the reduction rate decreased from 0.21706 to 0.03977 min-1 after three cycles of reuse due to the NZVI losing reaction activity generally,but the system still maintained considerable reduction capacity.Finally,a mechanism was revealed whereby NZVI would transform to Fe oxides after the exhaustion of its reductive power,and the photolysis of ferrioxalate to promote the cycling of iron species played the predominant role in providing extra reduction ability.These features confirm that introduction of OA into Cr(Ⅵ)reduction by NZVI through sunlight induction is advantageous and promising.Qi Zhou Yang Li Heng Yang Yun Gao 2020Journal of Environmental Sciences2020,32,4:3
