1Generation of B Cell-Deficient Pigs by Highly Efficient CRISPR/Cas9-Mediated Gene Targeting显示文摘Generating B cell-deficient mutant is the first step to produce human antibody repertoires in large animal models. In this study, we applied the clustered regularly interspaced short palindromic repeat(CRISPR)/CRISPR-associated(Cas) system to target the JHregion of the pig Ig M heavy chain gene which is crucial for B cell development and differentiation. Transfection of Ig M-targeting Cas9 plasmid in primary porcine fetal fibroblasts(PFFs) enabled inducing gene knock out(KO) in up to 53.3% of colonies analyzed, a quarter of which harbored biallelic modification, which was much higher than that of the traditional homologous recombination(HR). With the aid of somatic cell nuclear transfer(SCNT) technology, three piglets with the biallelic Ig M heavy chain gene mutation were produced. The piglets showed no antibody-producing B cells which indicated that the biallelic mutation of the Ig M heavy chain gene effectively knocked out the function of the Ig M and resulted in a B cell-deficient phenotype. 2A unifying approach in simulating the shot peening process using a 3D random representative volume finite element model显示文摘Using a modified 3D random representative volume(RV)finite element model,the effects of model dimensions(impact region and interval between impact and representative regions),model shapes(rectangular,square,and circular),and peening-induced thermal softening on resultant critical quantities(residual stress,Almen intensity,coverage,and arc height)after shot peening are systematically examined.A new quantity,i.e.,the interval between impact and representative regions,is introduced and its optimal value is first determined to eliminate any boundary effect on shot peening results.Then,model dimensions are respectively assessed for all model shapes to reflect the actual shot peening process,based on which shape-independent critical shot peening quantities are obtained.Further,it is found that thermal softening of the target material due to shot peening leads to variances of the surface residual stress and arc height,demonstrating the necessity of considering the thermal effect in a constitutive material model of shot peeing.Our study clarifies some of the finite element modeling aspects and lays the ground for accurate modeling of the SP process.Dianyin HU Ye GAO Fanchao MENG Jun SONG Yanfei WANG Mengxi REN Rongqiao WANG 2017Chinese Journal of Aeronautics2017,30,4:13
3Transcriptional landscape of rice roots at the single-cell resolution显示文摘There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in monocots.Despite this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model Arabidopsis.However,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this clade.Here,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice cultivars.Using combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both cultivars.Importantly,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice cultivars.In addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and rice.These species-specific features emphasize the importance of analyzing tissues across diverse model species,including rice.Taken together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants.Qing Liu Zhe Liang Dan Feng Sanjie Jiang Yifan Wang Zhuoying Du Ruoxi Li Guihua Hu Pingxian Zhang Yanfei Ma Jan ULohmann Xiaofeng Gu 2021Molecular Plant2021,14,3:9

5An improved imaging algorithm for fixed-receiver bistatic SAR显示文摘Based on the signal model of fixed-receiver bistatic SAR, this paper analyzes the characteristics of azimuth-variant, which includes variation of range migration curve and variation of Doppler rate caused by different azimuth position of targets. Then, an extended nonlinear chirp scaling (NLCS) algorithm is proposed, with its flowchart and phase compensation functions being provided. 6Epigallocatechin-3-gallate inhibits angiotensin Il-induced cardiomyocyte hypertrophy via regulating Hippo signaling pathway in H9c2 rat cardiomyocytes显示文摘AngiotensinⅡ(All) has been well known to induce cardiomyocyte hypertrophy. Epigallocatechin-3-gallate (EGCG) is the main active comp on ent of green tea and it has bee n show n to exhibit strong cardioprotective potential, although the underlying molecular mechanisms remain unclear. In this study, we investigated the role and mechanism of EGCG in preventing All-induced cardiomyocyte hypertrophy using rat H9c2 cardiomyocytes cells. Reactive oxygen species assay, cell size, and mRNA expression of cardiac hypertrophy markers ANP and BNP were assessed in response to All treatment. In addition, expression of proteins involved in Hippo signaling pathway were determined by western blot analysis. We found that All treatment resulted in significant upregulation of ANP and BNP expression levels and increase in H9c2 cell size, which were markedly attenuated by EGCG treatment. Furthermore, our results suggested that EGCG inhibited Allinduced cardiac hypertrophy via regulating the Hippo signaling pathway. 7Efficacy of Carbamazepine Combined with Botulinum Toxin A in the Treatment of Blepharospasm and Hemifacial Spasm显示文摘Purpose:To observe the efficacy of the combined treatment of carbamazepine and botulinum toxin A for blepharospasm and hemifacial spasm. Methods:Fifty-eight patients with either blepharospasm or hemifacial spasm were randomly divided into treatment and control groups. In the treatment group, 30 patients were administered with local intramuscular injections of botulinum toxin A and oral carbamazepine 100 mg/time,3 times/day for 60 days. Twenty-eight subjects in the control group under-went local intramuscular injections of botulinum toxin A only. Results:After combined treatment, the complete remission rate was 90%, which was significantly higher than that of the of the control group (67.9% , P<0.05,χ2 =4.733). However, no statistical significance was noted regarding the duration of therapeutic effects between the treatment group (range 14~40 weeks; 19.2 weeks on average) and control group (range 12~36 weeks; 18 weeks on average). 8Identification of a potential tumor suppressor gene, UBL3, in non-small cell lung cancer显示文摘Objective: Oncogenes have been shown to be drivers of non-small cell lung cancer(NSCLC), yet the tumor suppressing genes involved in lung carcinogenesis remain to be systematically investigated. This study aimed to identify tumor suppressing ubiquitin pathway genes(UPGs) that were critical to lung tumorigenesis.Methods: The 696 UPGs were silenced by an siRNA screening in NSCLC cells;the potential tumor suppressing UPGs were analyzed, and their clinical significance was investigated.Results: We reported that silencing of 11 UPGs resulted in enhanced proliferation of NSCLC cells, and four UPGs(UBL3, TRIM22, UBE2 G2, and MARCH1) were significantly downregulated in tumor samples compared to that in normal lung tissues and their expression levels were positively associated with overall survival(OS) of NSCLC patients. Among these genes, UBL3 was the most significant one. UBL3 expression was decreased in tumor samples compared to that in paired normal lung tissues in 59/86(68.6%) NSCLCs, was correlated with TNM stage and sex of NSCLC patients, and was significantly higher in non-smoking patients than in smoking patients. 9Minimizing Energy Cost for Green Data Center by Exploring Heterogeneous Energy Resource显示文摘With the deteriorating effects resulting from global warming in many areas, geographically distributed data centers contribute greatly to carbon emissions, because the major energy supply is fossil fuels. Considering this issue, many geographically distributed data centers are attempting to use clean energy as their energy supply, such as fuel cells and renewable energy sources. However, not all workloads can be powered by a single power sources, since different workloads exhibit different characteristics. In this paper, we propose a fine-grained heterogeneous power distribution model with an objective of minimizing the total energy costs and the sum of the energy gap generated by the geographically distributed data centers powered by multiple types of energy resources. In order to achieve these two goals, we design a two-stage online algorithm to leverage the power supply of each energy source. In each time slot, we also consider a chance-constraint problem and use the Bernstein approximation to solve the problem. 10EMC10 governs male fertility via maintaining sperm ion balance显示文摘Infertility is a severe public health problem worldwide that prevails up to 15% in reproductive-age couples,and male infertility accounts for half of total infertility.Studies on genetically modified animal models have identifed lots of genes involved in the pathogenesis of male infertility.The underlying causes,however,remain largely unclear.In this study,we provide evidence that EMCIO,one subunit of endoplasmic reticulum (ER)membrane protein complex (EMC),is required for male fertility.EMC10 is significantly decreased in spermatozoa from patients with asthenozoospermia and positively associated with human sperm motility. Male mice lacking Emc10 gene are completely sterile.Emc10-null spermatozoa exhibit multiple defects including abnormal morphology,decreased motility,impaired capacitation,and impotency of acrosome reaction,thereby which are incapable of fertilizing intact or ZP-free oocytes.However,intracytoplasmic sperm injection could rescue this defect caused by EMC10 deletion. 11DIW1 encoding a cladeⅠPP2C phosphatase negatively regulates drought tolerance by de-phosphorylating TaSnRK1.1 in wheat显示文摘Drought seriously impacts wheat production(Triticum aestivum L.),while the exploitation and utilization of genes for drought tolerance are insufficient.Leaf wilting is a direct reflection of drought tolerance in plants.Clade A PP2Cs are abscisic acid(ABA)co-receptors playing vital roles in the ABA signaling pathway,regulating drought response.However,the roles of other clade PP2Cs in drought tolerance,especially in wheat,remain largely unknown.Here,we identified a gain-of-function drought-induced wilting 1(DIW1)gene from the wheat Aikang 58 mutant library by map-based cloning,which encodes a cladeⅠprotein phosphatase 2C(TaPP2C158)with enhanced protein phosphatase activity.Phenotypic analysis of overexpression and CRISPR/Cas9 mutant lines demonstrated that DIW1/TaPP2C158 is a negative regulator responsible for drought resistance.We found that TaPP2C158 directly interacts with TaSnRK1.1 and de-phosphorylates it,thus inactivating the TaSnRK1.1–Ta AREB3 pathway.TaPP2C158 protein phosphatase activity is negatively correlated with ABA signaling.Association analysis suggested that C-terminal variation of TaPP2C158 changing protein phosphatase activity is highly correlated with the canopy temperature,and seedling survival rate under drought stress.Our data suggest that the favorable allele with lower phosphatase activity of TaPP2C158 has been positively selected in Chinese breeding history.This work benefits us in understanding the molecular mechanism of wheat drought tolerance,and provides elite genetic resources and molecular markers for improving wheat drought tolerance.Jingyi Wang Chaonan Li Long Li Lifeng Gao Ge Hu Yanfei Zhang Matthew P.Reynolds Xueyong Zhang Jizeng Jia Xinguo Mao Ruilian Jing 2023Journal of Integrative Plant Biology2023,65,8:1
12Remote plasma-enhanced atomic layer deposition of gallium oxide thin films with NH_3 plasma pretreatment显示文摘High quality gallium oxide(Ga_2O_3) thin films are deposited by remote plasma-enhanced atomic layer deposition(RPEALD) with trimethylgallium(TMG) and oxygen plasma as precursors. By introducing in-situ NH3 plasma pretreatment on the substrates, the deposition rate of Ga_2O_3 films on Si and GaN are remarkably enhanced, reached to 0.53 and 0.46 ?/cycle at 250 °C,respectively. The increasing of deposition rate is attributed to more hydroxyls(–OH) generated on the substrate surfaces after NH3 pretreatment, which has no effect on the stoichiometry and surface morphology of the oxide films, but only modifies the surface states of substrates by enhancing reactive site density. Ga_2O_3 film deposited on GaN wafer is crystallized at 250 °C, with an epitaxial interface between Ga_2O_3 and GaN clearly observed. 15Novel discovery of schisandrin A regulating the interplay of autophagy and apoptosis in oligoasthenospermia by targeting SCF/c-kit and TRPV1 via biosensors显示文摘Oligoasthenospermia is the primary cause of infertility.However,there are still enormous challenges in the screening of critical candidates and targets of oligoasthenospermia owing to its complex mechanism.In this study,stem cell factor(SCF),c-kit,and transient receptor potential vanilloid 1(TRPV1)biosensors were successfully established and applied to studying apoptosis and autophagy mechanisms.Interestingly,the detection limit reached 2.787×10^(-15)g/L,and the quantitative limit reached 1.0×10^(-13)g/L.Furthermore,biosensors were used to investigate the interplay between autophagy and apoptosis.Schisandrin A is an excellent candidate to form a system with c-kit similar to SCF/c-kit with a detection constant(K_(D))of 5.701×10^(-11)mol/L,whereas it had no affinity for SCF.In addition,it also inhibited autophagy in oligoasthenospermia through antagonizing TRPV1 with a K_(D) of up to 4.181×10^(-10)mol/L.In addition,in vivo and in vitro experiments were highly consistent with the biosensor.In summary,high-potency schisandrin A and two potential targets were identified,through which schisandrin A could reverse the apoptosis caused by excessive autophagy during oligoasthenospermia.Our study provides promising insights into the discovery of effective compounds and potential targets via a well-established in vitro-in vivo strategy.Lijuan Ma Boyi Li Jinchen Ma Chunyuan Wu Nan Li Kailin Zhou Yun Yan Mingshuang Li Xiaoyan Hu Hao Yan Qi Wang Yanfei Zheng Zhisheng Wu 2023Acta Pharmaceutica Sinica B2023,13,6:1
16Study on cyclic characteristics of the solar-powered adsorption cooling system显示文摘According to the typical variable heat source of solar energy and the unsteady adsorption process of adsorption chiller,the research of cyclic transient characteristics of the solar-powered adsorption cooling system is presented in this work.A mathematic model of the whole system including the model of adsorption chiller,which reveals the transient operation process of the solar-powered adsorption cooling system,is developed and verified by experimental data.On the basis of the simulated results,the transient characteristics and the overall performance of the system,not only in the traditional open cycle mode but also in closed cycle mode,are both analyzed theoretically.Furthermore,the influence of parameters matching of components configuration and operation process on the cyclic characteristics of the system,such as the solar collector area,the water tank capacity and the chiller startup temperature,are discussed.The research in this work may play a very important role in optimizing the system cyclic process and improving the system adaptability especially under the condition of variable heat source.JingYi Wu Sheng Li YanFei Hu 2009Science China(Technological Sciences)2009,52,6:1

18Hierarchical strain band formation and mechanical behavior of a heterostructured dual-phase material显示文摘Dispersive strain bands have been reported as a characteristic deformation feature of heterostructured materials,which helps to improve ductility.However,their formation mechanism is still not well understood.Here we report the formation of dispersed strain bands through dual-level hierarchical strain banding and its effect on the mechanical behavior of a heterostructured Fe-40Cu model material.Specifically,deformation started by the formation and propagation of dispersed microscale strain bands in the heterostructured Fe-40Cu material.High strain gradient was generated within the microscale strain bands during their propagation and was accommodated by the accumulation of geometrically necessary disloca-tions(GNDs).The dispersed microscale strain bands were not uniformly distributed,but instead grouped together to form macroscale strain bands that were uniformly distributed over the entire gage section to accommodate the majority of the applied strain.The formation of this dual-level hierarchical strain bands prevented the formation of large strain localization to fail the sample prematurely.It was also found that increasing the strain hardening capacity of soft copper zones provides more room for the accumulation of GNDs,resulting in higher constraint to microscale strain band propagation and consequently higher ductility.These observations suggest the possibility of tailoring microscale strain bands to optimize tensile performance of heterostructured materials.Zhongkai Li Yujie Liu Yanfei Wang Jun Hu Lei Xu Jianjun Wang Chunming Liu Yuntian Zhu 2023Journal of Materials Science & Technology2023,,31:0
19Strong phonon-magnon coupling of an O/Fe(001) surface显示文摘Using density functional calculations,we elucidate the interaction between magnons and phonons on Fe(001)and O/Fe(001)surfaces.The effective Heisenberg exchange parameters between neighbor sites(Ji)were derived by fitting the ab initio spin spiral dispersion relation to a classical Heisenberg model.In bulk Fe,J5bhas a larger amplitude than J2b-J4b.Surface magnons were softer on the Fe(001)surface than in bulk,but were strengthened on the O/Fe(001)surface through strong interactions between the O and Fe atoms.The four calculated high-energy phonon modes of O/Fe(001)excellently agreed with the experimental measurements.The J1in O/Fe(001)decreased significantly with increasing phonon amplitude.Phonon-magnon coupling occurred more easily on O/Fe(001)than on Fe(001).The softening of the magnon on the O/Fe(001)with phonon amplitude was attributed to the reduced amplitude of longitudinal excitations.Magnetic moment of the Fe atom of the second layer(Fe2)was especially sensitive to phonon amplitude,regardless of the motion direction of the Fe2 atoms.Yan Zhu YanFei Pan JiYu Fan ChunLan Ma Jun Hu XinYuan Wei KaiCheng Zhang HongBin Zhang 2020Science China(Physics,Mechanics & Astronomy)2020,63,11:0
20Brittle culm 25, which encodes an UDP-xylose synthase, affects cell wall properties in rice显示文摘Because plant mechanical strength influences plant growth and development,the regulatory mechanisms underlying cell-wall synthesis deserve investigation.Rice mutants are useful for such research.We have identified a novel brittle culm 25(bc25)mutant with reduced growth and partial sterility.BC25 encodes an UDP-glucuronic acid decarboxylase involved in cellulose synthesis and belongs to the UXS family.A single-nucleotide mutation in BC25 accounts for its altered cell morphology and cellwall composition.Transmission electron microscopy analysis showed that the thickness of the secondary cell wall was reduced in bc25.Monosaccharide analysis revealed significant increases in content of rhamnose and arabinose but not of other monosaccharides,indicating that BC25 was involved in xylose synthesis with some level of functional redundancy.Enzymatic assays suggested that BC25 functions with high activity to interconvert UDP-glucuronic acid(UDP-Glc A)and UDP-xylose.GUS staining showed that BC25 was ubiquitously expressed with higher expression in culm,root and sheath,in agreement with that shown by quantitative real-time(q RT)-PCR.RNA-seq further suggested that BC25 is involved in sugar metabolism.We conclude that BC25 strongly influences rice cell wall formation.Siliang Xu Mengchen Zhang Junhua Ye Dongxiu Hu Yuanyuan Zhang Zhen Li Junrong Liu Yanfei Sun Shan Wang Xiaoping Yuan Yue Feng Qun Xu Xinghua Wei Dali Zeng Yaolong Yang 2023The Crop Journal2023,11,3:0
