| 2 | Effect of UTRs from TMV-RNA on the expression of foreign gene in transgenic plants显示文摘To investigate the effects of 5’and 3’ untranslated region (UTR) from tobacco mosaicvirus (TMV) on expression of foreign genes, four plant-expression vectors pBG437, pBG438, pBG440 and pBG440ANOS containing expression cassettes of GUS-NOS; Ω-GUS-NOS, Ω-GUS-3’ UTR-NOS and Ω-GUS-3’ UTR downstream of CaMV 35S promoter with double enhancer sequences respectively have been constructed. Results from a large number of transgenic tobacco plants show that the GUS activity of pBG440 transformed plants is the highest, being 5-fold that of pBG437 and 1.6-fold that of pBG438. Similar results have been obtained in transient expression by injection of Agrobacterium tumefaciens harbouring these constructs into N. benthamiana leaves. | 4 | Construction of a new plant expression vector containing two insect resistant genes and its expression in transgenic tobacco plants显示文摘A new plant expression vector (pBS29K-BA) containing two insect resistant genes, a synthetic chimeric gene BtS29K encoding the activated insecticidal protein Cry1Ac and a gene API-BA encoding the arrowhead (Sagittaria sagittifolia L.) proteinase inhibitor (API) A and B, is constructed. Transgenic tobacco plants expressing these two genes are obtained through Agrobacterium-mediated transformation of tobacco leaf discs. The average expression levels of Cry1Ac and API-BA proteins in transgenic plants are of 3.2 μg and 4.9 μg per gram fresh leaf respectively. | 5 | Transfer learning: A new aerodynamic force identification network based on adaptive EMD and soft thresholding in hypersonic wind tunnel显示文摘The aerodynamic test in the pulse combustion wind tunnel is very important for the design, evaluation and optimization of aerodynamic characteristics of the hypersonic aircraft.The test accuracy even affects the success or failure of hypersonic aircraft development. In the aerodynamic test of pulse combustion wind tunnel, the aerodynamic signal is disturbed by the inertial force signal, which seriously affects the test accuracy of aerodynamic force. Aiming at the above problems, this paper innovatively proposes an aerodynamic intelligent identification method, that is the transfer learning network based on adaptive Empirical Modal Decomposition(EMD) and Soft Thresholding(TLN-AE&ST). Compared with the existing aerodynamic intelligent identification model based on deep learning technology, this study introduces the transfer learning idea into the aerodynamic intelligent identification model for the first time. The TLN-AE&ST effectively alleviates the problem of scarcity of training samples for intelligent models due to the high cost of wind tunnel tests, and provides a new idea for further implementation of deep learning technology in the field of wind tunnel aerodynamic testing. And this study designed residual attention block with soft threshold and dense block with adaptive EMD in TLN-AE&ST model. Residual attention block with soft threshold module can more effectively suppress the influence of instrument noise signal on model training effect. Dense block with adaptive EMD makes the deep learning model no longer a black box to a certain extent, and has certain physical significance. | 8 | Single-cell analysis defines the lineage plasticity of stem cells in cervix epithelium显示文摘Information about the dynamic change and post-injury regeneration of cervical epithelium is relatively rare,even though it is tightly related to gynecologic malignancy.Here,using a feeder cell-based culturing system,we stably cloned mouse and human P63 and KRT5 expressing cells from the adult cervix as putative cervical stem/progenitor cells(CVSCs).When subjected to differentiation,the cultured cells gave rise to mature cervical epithelium by differentiating into squamous or glandular cells.The ability of endogenous mouse CVSCs to reconstitute cervical epithelium after injury was also evident from the genetic lineage tracing experiments.Single-cell transcriptomic analysis further classified the CVSCs into three subtypes and delineated their bi-lineage differentiation roadmap by pseudo-time analysis.We also tracked the real-time differentiation routes of two representing single CVSC lines in vitro and found that they recapitulated the predicted roadmap in pseudo-time analysis.Signaling pathways including Wnt,TGF-beta,Notch and EGFR were found to regulate the cervical epithelial hierarchy and implicated the different roles of distinct types of cells in tissue homeostasis and tumorigenesis.Collectively,the above data provide a cloning system to achieve stable in vitro culture of a bi-lineage stem/progenitor cell population in the cervix,which has profound implications for our understanding of the cervix stem/progenitor cell function in homeostasis,regeneration,and disease and could be helpful for developing stem cell-based therapies in future. | Zixian Zhao Yujia Wang Yingchuan Wu Dandan Li Ting Zhang Yu Ma Xiaoming Teng Wei Zuo | 2021 | Cell Regeneration2021,10,1: | 0 |
