1Construction of targeted plasmid vector pcDNA3.1-Egr.1p-p16 and its expression in pancreatic cancer JF305 cells induced by radiation in vitro显示文摘AIM: To construct pcDNA3.1-Egr.1p-p16 recombinant plasmid and investigate the expression of p16 in pancreatic cancer JF305 cells induced by radiation and the feasibility of gene radiotherapy for pancreatic carcinoma. METHODS: Human p16 cDNA was ligated to the downstream of Egr-1 promotor to construct pcDNA3.1-Egr.1p-p16 plasmid by restriction enzyme digested. The recombined plasmids were transfected into pancreatic cancer JF305 cells with lipofectamine. p16 mRNA level was detected by RT-PCR. The expression of p16 after different doses of X-ray radiation was detected by Western blot technique. Cell survival was assessed by clonogenic assays and cell viability was analysed by trypen blue exclusion. Flow cytometry was performed to study the apoptosis of JF305 cells. RESULTS: Restriction enzyme digestion showed the correctly constructed pcDNA3.1-Egr.1p-p16. The p16 expression in cells transfected with pcDNA3.1-Egr.1p-p16 induced by different doses of radiation was higher than that in the control group (P < 0.05). Eight hours after 2 Gy X-ray radiation, the expression reached its peak(87.00 ng/L), and was signifi cantly higher than that in the control group (P < 0.0.5). Clonogenic analysis and trypan blue extraction test showed that the pcDNA3.1-Egr.1p-p16 transfer enhanced radiation-induced cell killing in p16-null JF305 cell lines. The induction of apoptosis was lower in combined transfection and irradiation group than that in irradiation alone. CONCLUSION: X-ray can induce the recombinant plasmid pcDNA3.1-Egr.1p-p16 expression in JF305 cells. 2Cadmium pollution from phosphate fertilizers in arable soils and crops:an overview显示文摘The application of mineral and organic phosphorus fertilizers to arable land has greatly increased crop yield to meet the world food demand.On the other hand,impurities in these fertilizers,such as heavy metals,are being added to agricultural soils,resulting both from the raw materials themselves and the processes used to obtain the final product.Cadmium,a non-essential and toxic heavy metal,has been found in relatively high amounts in common P fertilizers obtained from sediments.This metal poses a high risk for soil fertility,crop cultivation,and plants in general.Furthermore,human health might be compromised by the cadmium concentrations in agricultural and livestock products,due to the bioaccumulation effect in the food web.The accumulation in the different matrixes is the result of the high mobility and flexible availability of this harmful metal.This review summarizes risks to human health,the factors influencing cadmium movement in soils and crop uptake,as well as common plant responses to its toxicity.In addition,it summarizes cadmium balances in soils,trends,long-term experiments,and further studies.Cadmium inputs and outputs in arable soil,together with their calculated concentrations,are compared between two different regions:the European countries(in particular Germany)and China.The comparison appears useful because of the different proportions in the inputs and outputs of cadmium,and the diverse geographical,environmental and social factors.Moreover,these variables and their influences on cadmium contamination improve the understanding of the pollution from phosphate fertilizers and will help to establish future mitigation policies.Andrea Giovanna NINO-SAVALA Zhong ZHUANG Xin MA Andreas FANGMEIER Huafen LI Aohan TANG Xuejun LIU 2019Frontiers of Agricultural Science and Engineering2019,6,4:2

5Emergence of a novel reassortant avian influenza virus(H10N3) in Eastern China with high pathogenicity and respiratory droplet transmissibility to mammals显示文摘Decades have passed since the first discovery of H10-subtype avian influenza virus(AIV) in chickens in 1949,and it has been detected in many species including mammals such as minks,pigs,seals and humans.Cases of human infections with H10N8viruses identified in China in 2013 have raised widespread attention.Two novel reassortant H10N3 viruses were isolated from chickens in December 2019 in eastern China during routine surveillance for AIVs.The internal genes of these viruses were derived from genotype S(G57) H9N2 and were consistent with H5N6,H7N9 and H10N8,which cause fatal infections in humans.Their viral pathogenicity and transmissibility were further studied in different animal models.The two H10N3 isolates had low pathogenicity in chickens and were transmitted between chickens via direct contact.These viruses were highly pathogenic in mice and could be transmitted between guinea pigs via direct contact and respiratory droplets.More importantly,these viruses can bind to both human-type SAα-2,6-Gal receptors and avian-type SAα-2,3-Gal receptors.Asymptomatic shedding in chickens and good adaptability to mammals of these H10N3 isolates would make it easier to transmit to humans and pose a threat to public health.Kaituo Liu Pingyun Ding Yuru Pei Ruyi Gao Wenwen Han Huafen Zheng Zhuxing Ji Miao Cai Jinyuan Gu Xiuli Li Min Gu Jiao Hu Xiaowen Liu Shunlin Hu Pinghu Zhang Xiaobo Wang Xiaoquan Wang Xiufan Liu 2022Science China(Life Sciences)2022,65,5:1
6Source-specific risk assessment for cadmium in wheat and maize:Towards an enrichment model for China显示文摘Cadmium (Cd) pollution of agricultural soil is of public concern due to its high potential toxicity and mobility.This study aimed to reveal the risk of Cd accumulation in soil and wheat/maize systems,with a specific focus on the source-specific ecological risk,human health risk and Cd enrichment model.For this we investigated more than 6100 paired soil and grain samples with 216 datasets including soil Cd contents,soil pH and grain Cd contents of 85 sites from China.The results showed that mining activities,sewage irrigation,industrial activities and agricultural practices were the critical factors causing Cd accumulation in wheat and maize cultivated sites.Thereinto,mining activities contributed to a higher Cd accumulation risk in the southwest China and Middle Yellow River regions;sewage irrigation influenced the Cd accumulation in the North China Plain.In addition,the investigated sites were classified into different categories by comparing their soil and grain Cd contents with the Chinese soil screening values and food safety values,respectively.Cd enrichment models were developed to predict the Cd levels in wheat and maize grains.The results showed that the models exhibited a good performance for predicting the grain Cd contents among safe and warning sites of wheat (R2=0.61 and 0.72,respectively);while the well-fitted model for maize was prone to the overestimated sites (R2=0.77).This study will provide national viewpoints for the risk assessments and prediction of Cd accumulation in soil and wheat/maize systems.Zhong Zhuang Qiqi Wang Siyu Huang Andrea Giovanna NinoSavala Yanan Wan Huafen Li Andreas H.Schweiger Andreas Fangmeier Jürgen Franzaring 2023Journal of Environmental Sciences2023,,3:1

13Enhanced pathogenicity and transmissibility of H9N2 avian influenza virus in mammals by hemagglutinin mutations combined with PB2-627K显示文摘H9N2 avian influenza viruses(AIVs)circulate globally in poultry and have become the dominant AIV subtype in China in recent years.Previously,we demonstrated that the H9N2 virus(A/chicken/Eastern China/SDKD1/2015)naturally harbors a mammalian-adaptive molecular factor(627K)in the PB2 protein and is weakly pathogenic in mice.Here,we focused on new markers for virulence in mammals.A mouse-adapted H9N2 virus was serially passaged in mice by infecting their lungs.As expected,infected mice showed clinical symptoms and died at passage six.A comparison between the wild-type and mouse-adapted virus sequences identified amino acid substitutions in the hemagglutinin(HA)protein.H9N2 viruses with the T187P t M227L double mutation exhibited an increased affinity to human-type(SAα2,6Gal)receptors and significantly enhanced viral attachment to mouse lung tissues,which contributed to enhancing viral replication and virulence in mice.Additionally,HA with the T187P t M227L mutation enabled H9N2 viral transmission in guinea pigs via direct contact.AIV pathogenicity in mice is a polygenic trait.Our results demonstrated that these HA mutations might be combined with PB2-627K to significantly increase H9N2 virulence in mice,and this enhanced virulence was achieved in other H9N2 AIVs by generating the same combination of mutations.In summary,our study identified novel key elements in the HA protein that are required for H9N2 pathogenicity in mice and provided valuable insights into pandemic preparedness against emerging H_(9)N_(2)strains.Kaituo Liu Yaqian Guo Huafen Zheng Zhuxing Ji Miao Cai Ruyi Gao Pinghu Zhang Xiaowen Liu Xiulong Xu Xiaoquan Wang Xiufan Liu 2023Virologica Sinica2023,38,1:0

15Optimization on Extraction Conditions of Flavonoids from Edgeworthia chrysantha Lind1.显示文摘[Objectives]The extraction conditions of flavonoids from flowers of Edgeworthia chrysantha Lind1.were optimized.[Methods]With ethanol as an extraction agent,firstly by single-factor tests,the best levels of ethanol concentration,material-to-liquid ratio,extraction temperature and extraction time were determined.Then the optimal extraction conditions were determined through the quadratic orthogonal regression rotatable design.[Results]The optimal extraction conditions for flavonoids from E.chrysantha were∶material-to-liquid ratio 1∶15.4,extraction temperature 75℃,extraction time 120 min and ethanol concentration 75%,with which the highest yield of flavonoids was 6.02 mg/g.[Conclusions]This study provides a theoretical basis for the further development and utilization of E.chrysantha flowers.Pengfei CAO Huafen WU Yinhua CHEN 2020Agricultural Biotechnology2020,9,4:0
16Translation:Management of Coronavirus Disease 2019(COVID-19):Experience in Zhejiang Province,China显示文摘The current epidemic situation of coronavirus disease 2019(COVID-19)still remains severe.As the National Clinical Research Center for Infectious Diseases,The First Affiliated Hospital of the Zhejiang University School of Medicine is the primary medical care center for COVID-19 in Zhejiang Province.Based on the present expert consensus carried out by the National Health Commission and National Administration of Traditional Chinese Medicine,our team summarized and established an effective treatment strategy centered on“Four-Anti and Two-Balance”for clinical practice.The“Four-Anti and Two-Balance”strategy includes antivirus,anti-shock,antihypoxemia,and anti-secondary infection,and maintaining of water,electrolyte and acid/base balance and microecological balance.Simultaneously,an integrated multidisciplinary personalized treatment is recommended to improve therapeutic effects.The importance of early viral detection,dynamic monitoring of inflammatory indexes,and chest radiographs has been emphasized in clinical decision-making.Sputum was observed with the highest positive rate by reverse transcription-polymerase chain reaction(RTPRC).Viral nucleic acids could be detected in 10%of the patients’blood samples at the acute phase and 50%of patients had positive RT-PCR results in their feces.We also isolated live viral strains from feces,indicating potential infectiousness of feces.Dynamic cytokine detection was necessary to timely identify cytokine storms and for the application of the artificial liver blood purification system.The“Four-Anti and Two-Balance”strategy effectively increased cure rates and reduced mortality.Early antiviral treatment alleviated disease severity and prevented illness progression.We found that lopinavir/ritonavir combined with abidol showed antiviral effects against COVID-19.Shock and hypoxemia were usually caused by cytokine storms.The artificial liver blood purification system was able to rapidly remove inflammatory mediators and block the cytokine storm.Moreover,it also contributed to the balance of fluids,electrolytes,and acids/bases and thus improved treatment efficacy during critical illness.For cases of severe illness,early and also short periods of moderate glucocorticoid administration was supported.Patients with an oxygenation index below 200mmHg were transferred to the intensive care unit.Conservative oxygen therapy was preferred and noninvasive ventilation(NIV)was not recommended.Patients with mechanical ventilation were strictly supervised with cluster ventilator-associated pneumonia prevention strategies.Antimicrobial prophylaxis was prescribed rationally and was not recommended,except for patients with a long course of disease,repeated fever,and elevated procalcitonin,similarly secondary fungal infections were of concern.Some patients with COVID-19 showed intestinal microbial dysbiosis with decreased genus such as Lactobacillus and Bifidobacterium.Nutritional and gastrointestinal function should;therefore,be assessed for all patients.Nutritional support and application of prebiotics or probiotics were suggested to regulate the balance of intestinal microbiota and reduce the risk of secondary infections due to bacterial translocation.Anxiety and fear were common in patients with COVID-19.Therefore,we established a dynamic assessment and warning for psychological crises.We also integrated Chinese medicine in the treatment to promote rehabilitation.We optimized nursing processes for severe patients to promote their rehabilitation.Since viral clearance patterns after severe acute respiratory syndrome coronavirus 2 infections remained unclear,2 weeks quarantine for discharged patients was required,and a regular following-up was also needed.These Zhejiang experiences and suggestions have been implemented in our center and achieved good results.However,since COVID-19 was a newly emerging disease,more work is warranted to further improve strategies of prevention,diagnosis,and treatment for COVID-19.Kaijin Xu Hongliu Cai Yihong Shen Qin Ni Yu Chen Shaohua Hu Jianping Li Huafen Wang Liang Yu He Huang Yunqing Qiu Guoqing Wei Qiang Fang Jianying Zhou Jifang Sheng Tingbo Liang Lanjuan Li 2020Infectious Microbes & Diseases2020,2,2:0
17Influence of metal-contamination on distribution in subcellular fractions of the earthworm (Metaphire californica) from Hunan Province, China显示文摘Earthworms have the ability to accumulate of heavy metals,however,there was few studies that addressed the metals in earthworm at subcellular levels in fields.The distributions of metals(Cd,Cu,Zn,and Pb) in subcellular fractions(cytosol,debris,and granules) of earthworm Metaphire californica were investigated.The relationship between soil metals and earthworms were analyzed to explain its high plasticity to inhabit in situ contaminated soil of Hunan Province,south China.The concentration of Cd in subcellular compartments showed the same pattern as Cu in the order of cytosol > debris > granules.The distribution of Zn and Pb in earthworms indicated a similar propensity for different subcellular fractions that ranked as granules > debris > cytosol for Zn,and granules > cytosol > debris for Pb.The internal metal concentrations in earthworms increased with the soil metals(p < 0.05).Significant positive correlations were found between soil Cd and Cd concentrations in cytosol and debris(p < 0.01).Moreover,the soil Pb concentration significantly influenced the Pb concentrations in cytosol and debris(p < 0.01),similar to that of Cd.The soil Cu concentrations was only associated with the Cu in granules(p < 0.05).Soil Zn concentrations correlated with the Zn concentrations in each subcellular fraction(p < 0.05).Our results provide insights into the variations of metals partitioning in earthworms at subcellular levels and the relationships of soil metals,which could be one of the detoxification strategies to adapt the long-term contaminated environment.Kun Wang Yuhui Qiao Huiqi Zhang Shizhong Yue Huafen Li Xionghui Ji Longsheng Liu 2018Journal of Environmental Sciences2018,30,11:0
