| 1 | Sodium oligomannate therapeutically remodels gut microbiota and suppresses gut bacterial amino acids-shaped neuroinflammation to inhibit Alzheimer's disease progression显示文摘Recently,increasing evidence has suggested the association between gut dysbiosis and Alzheimer's disease(AD)progression,yet the role of gut microbiota in AD pathogenesis remains obscure.Herein,we provide a potential mechanistic link between gut microbiota dysbiosis and neuroinflammation in AD progression.Using AD mouse models,we discovered that,during AD progression,the alteration of gut microbiota composition leads to the peripheral accumulation of phenylalanine and isoleucine,which stimulates the differentiation and proliferation of pro-inflammatory T helper 1(Thl)cells.The brain-infiltrated peripheral Th1 immune cells are associated with the Ml microglia aaivation,contributing to AD-associated neuroinflammation.Importantly,the elevation of phenylalanine and isoleucine concentrations and the increase of Th1 cell frequency in the blood were also observed in two small independent cohorts of patients with mild cognitive impairment(MCI)due to AD.Furthermore,GV-971,a sodium oligomannate that has demonstrated solid and consistent cognition improvement in a phase 3 clinical trial in China,suppresses gut dysbiosis and the associated phenylalanine/isoleucine accumulation,harnesses neuroinflammation and reverses the cognition impairment.Together,our findings highlight the role of gut dysbiosis-promoted neuroinflammation in AD progression and suggest a novel strategy for AD therapy by remodelling the gut microbiota. | Xinyi Wang Guangqiang Sun Teng Feng Jing Zhang Xun Huang Tao Wang Zuoquan Xie Xingkun Chu Jun Yang Huan Wang Shuaishuai Chang Yanxue Gong Lingfei Ruan Guanqun Zhang Siyuan Yan Wen Lian Chen Du Dabing Yang Qingli Zhang Feifei Lin Jia Liu Haiyan Zhang Changrong Ge Shifu Xiao Jian Ding Meiyu Geng | 2019 | Cell Research2019,29,10: | 192 |
| 2 | Early kidney injury during long-term adefovir dipivoxil therapy for chronic hepatitis B显示文摘AIM: To evaluate urine β2-microglobulin(β2-M), retinol-binding protein(RBP) excretion, and renal impairment with adefovir dipivoxil(ADV) for chronic hepatitis B. METHODS: We enrolled 165 patients with chronic hepatitis B infection who were treated with ADV monotherapy(n = 90) or ADV plus lamivudine combination therapy(n = 75). An additional 165 chronic hepatitis B patients treated with entecavir were recruited as controls. We detected serum creatinine, urine β2-M, and RBP levels, and estimated the glomerular filtration rate(e GFR) at the initiation of antiviral therapy and every 6 mo for a period of five years. RESULTS: Urine β2-M abnormalities were observed in patients during the first(n = 3), second(n = 7), third(n = 11), fourth(n = 16), and fifth(n = 21) year of ADV treatment. Urinary RBP abnormalities were observed in patients during the first(n = 2), second(n = 8), third(n = 12), fourth(n = 15), and fifth(n = 22) year of ADV treatment. e GFR decreased 20%-30% from baseline in 20 patients, 30%-50% in 12 patients, and > 50% in 3 patients during the five years of treatment. Further analysis indicated that decreases in e GFR of ≥ 30% relative to the baseline level correlated significantly with urine RBP and β2-M abnormalities. In contrast, both serum creatinine and e GFR remained stable in patients treated with entecavir, and only one of these patients developed a urine β2-M abnormality, and two developed urine RBP abnormalities during the five years of treatment. | 3 | Soil pH impact on microbial tetraether lipids and terrestrial input index (BIT) in China显示文摘The distributions of microbial tetraether lipids i.e. glycerol dialkyl glycerol tetraethers (GDGTs) were investigated in a variety of Chinese soils featured by a wide range of pH values in different climate zones. The bacterial branched GDGTs and archaeal isoprenoid GDGTs were found to be widely distributed in Chinese soils and their relative abundance was controlled primarily by soil pH. The bacterial bGDGTs usually dominate over archaeal iGDGTs in abundance when the soil pH is below 8.0. However archaeal iGDGTs become dominant and the abundance of halophilic archaea characterized by GDGT V without cyclopentyl ring increases dramatically in the alkaline soil with pH greater than 8.0. The relative abundance of archaeal iGDGTs to bacterial bGDGTs was defined here as RAI index. The RAI index decreases with elevated soil pH and a linear correlation is found between them indicating that the relative abundance of archaeal iGDGTs to bacterial bGDGTs increases with increased pH. The RAI index thus could be a novel proxy of the pH values of paleosol. The terrestrial input index BIT (Branched and Isoprenoid Tetraethers Index) decreases with increased soil pH suggesting that soil pH should be taken into consideration when the terrestrial contribution to marine sediments is estimated by BIT index. | 6 | A general end-to-end diagnosis framework for manufacturing systems显示文摘The manufacturing sector is envisioned to be heavily influenced by artificial-intelligence-based technologies with the extraordinary increases in computational power and data volumes. A central challenge in the manufacturing sector lies in the requirement of a general framework to ensure satisfied diagnosis and monitoring performances in different manufacturing applications. Here, we propose a general data-driven,end-to-end framework for the monitoring of manufacturing systems. This framework, derived from deep-learning techniques, evaluates fused sensory measurements to detect and even predict faults and wearing conditions. This work exploits the predictive power of deep learning to automatically extract hidden degradation features from noisy, time-course data. We have experimented the proposed framework on 10 representative data sets drawn from a wide variety of manufacturing applications. | 7 | Masked Sentence Model Based on BERT for Move Recognition in Medical Scientific Abstracts显示文摘Purpose:Mo ve recognition in scientific abstracts is an NLP task of classifying sentences of the abstracts into different types of language units.To improve the performance of move recognition in scientific abstracts,a novel model of move recognition is proposed that outperforms the BERT-based method.Design/methodology/approach:Prevalent models based on BERT for sentence classification often classify sentences without considering the context of the sentences.In this paper,inspired by the BERT masked language model(MLM),we propose a novel model called the masked sentence model that integrates the content and contextual information of the sentences in move recognition.Experiments are conducted on the benchmark dataset PubMed 20K RCT in three steps.Then,we compare our model with HSLN-RNN,BERT-based and SciBERT using the same dataset.Findings:Compared with the BERT-based and SciBERT models,the F1 score of our model outperforms them by 4.96%and 4.34%,respectively,which shows the feasibility and effectiveness of the novel model and the result of our model comes closest to the state-of-theart results of HSLN-RNN at present.Research limitations:The sequential features of move labels are not considered,which might be one of the reasons why HSLN-RNN has better performance.Our model is restricted to dealing with biomedical English literature because we use a dataset from PubMed,which is a typical biomedical database,to fine-tune our model.Practical implications:The proposed model is better and simpler in identifying move structures in scientific abstracts and is worthy of text classification experiments for capturing contextual features of sentences.Originality/value:T he study proposes a masked sentence model based on BERT that considers the contextual features of the sentences in abstracts in a new way.The performance of this classification model is significantly improved by rebuilding the input layer without changing the structure of neural networks. | Gaihong Yu Zhixiong Zhang Huan Liu Liangping Ding | 2019 | Journal of Data and Information Science2019,4,4: | 14 |
| 8 | Origin of Tafoni in the Late Cretaceous Aeolian Sandstones,Danxiashan UNESCO Global Geopark,South China显示文摘Tafoni occur generally in granular rocks around the world, but their origin remains controversial. In this study,the roles of lithology, micro–climate, and organism in tafoni development are investigated in the Danxiashan UNESCO Global Geopark of South China. The Jinshiyan Temple and Luyitang sites along the Jinjiang River are chosen to conduct tafoni morphometry, bedrock petrography, and micro–climate analyses. The research methods used in this study include field observations and measurements, Voronoi diagrams, polarizing microscopic and scanning electron microscopic(SEM)image analysis, and meteorological detection. The tafoni are mostly 2–10 cm in length and elliptical in shape. The Late Cretaceous Jinshiyan sandstones are characterized by a grain–to–grain contact fabric and moderate sorting with a high proportion of soluble grains and cements. The Voronoi diagrams developed through the Arc GIS software are largely consistent with the tafoni openings. Moreover, owing to exposure to solar insolation, the external temperature values are higher than the internal ones, while the external relative humidity values are lower than the internal ones. Therefore, the permeable Jinshiyan sandstones are fundamental for tafoni development, while the abundant moisture from the Jinjiang River and frequent rains is favorable for salt weathering in tandem with biological activities. | 9 | Distribution of microbial fatty acids and fatty alcohols in soils from an altitude transect of Mt. Jianfengling in Hainan, China: Implication for paleoaltimetry and paleotemperature reconstruction显示文摘The temperature gradient along the altitude transect of Mt.Jianfengling provides a good opportunity to establish and evaluate the microbial lipid-based environmental proxies.The soils collected from 14 different altitudes of Mt.Jianfengling contain abundant microbial fatty acids and fatty alcohols,including iso/anteiso fatty acids(i/aC12:0–i/aC19:0),10-Me-C16:0 fatty acid,iso/anteiso fatty alcohols(i/aC13–iC26),10-Me-C16:0 fatty alcohol and unsaturated fatty alcohols,which can indicate a strong microbial activity in the Jianfengling soils.The branched and unsaturated fatty alcohols can be only detected when saponification is performed,implying that these lipids are present as the constituents of bacterial wax esters in the soils.The ratio of aC15/iC15 fatty acids is positively correlated with altitude,suggesting that the decrease in temperature can induce the increase in the relative abundance of anteiso C15 fatty acid.In contrast,the ratio of aC15/iC15 fatty alcohols and of aC15/nC15 fatty alcohols both decrease with increased altitude or decreased temperature.Similarly,the ratio of nC18:1/nC18:0 fatty alcohols also decreases with decreased temperature,which is opposite to the previous observation that unsaturation of fatty acids in microorganism increases in response to decreased temperature.Besides,the average chain length(ACL)of long chain fatty alcohols(C22–C30)from leaf waxes and carbon preference index(CPI)of all n-fatty alcohols are also significantly correlated with altitude or mean annual temperature,demonstrating their potential for paleoclimate reconstruction.The correlation of microbial fatty acids and alcohols as well as ACL and CPI of plant wax-derived fatty alcohols with altitude may provide novel ways to reconstruct paleotemperature and paleoaltimetry. | YANG Huan DING WeiHua XIE ShuCheng | 2014 | Science China Earth Sciences2014,57,5: | 8 |
| 10 | Efficacy and safety of vitrectomy with internal limiting membrane peeling for diabetic macular edema: a Meta-analysis显示文摘AIM: To evaluate the efficacy and safety of vitrectomy with internal limiting membrane(ILM) peeling for diabetic macular edema(DME). METHODS: The PubMed, Embase, Web of Science, Cochrane, SionMed, ClinicalTrials.gov, CNKI databases and Wanfang databases, published until Oct. 2017, were searched to identify studies comparing the clinical outcomes following vitrectomy with and without ILM peeling, for treating DME. Pooled results were expressed as odds ratios(ORs) with corresponding 95% confidence intervals(CI) for vitrectomy with and without ILM peeling with regard to best corrected visual acuity(BCVA), central macular thickness(CMT), and complication incidents. RESULTS: A total of 14 studies involving 857 eyes were included of which three studies were Chinese and the rests were English literatures. Meta-analysis indicated that compared with vitrectomy alone, vitrectomy with ILM peeling could improve BCVA more obviously(OR=1.66, 95%CI: 1.12-2.46, P=0.01) and had higher rate of CMT reduction(OR=3.89, 95%CI: 1.37-11.11, P=0.01). There were significant statistical differences between the two surgical methods for both BCVA and CMT(P<0.05). For the incidence of intraoperative and postoperative complications, the incidence of epiretinal membrane(ERM) was slightly lower in the ILM peeling group than the group without ILM peeling(OR=0.38, 95%CI: 0.07-2.00, P=0.25), although insignificant statistically. Other incidences of overall complications, iatrogenic peripheral retinal break and increased intraocular pressure indicated no significant difference between two groups(OR=1.19, 95%CI: 0.82-1.73, P=0.36; OR=1.21, 95%CI: 0.66-2.21, P=0.53; OR=1.34, 95%CI: 0.75-2.40, P=0.32). CONCLUSION: Vitrectomy is effective for DME and the effect can be improved by additional ILM peeling, especially for anatomical efficacy, without increasing the incidence of intraoperative and postoperative complications. | 11 | Potential predictors for mental stress-induced myocardial ischemia in patients with coronary artery disease显示文摘Background: Mental stress-induced myocardial ischemia (MSIMI) is closely associated with adverse cardiac events in patients with coronary artery disease (CAD) and we aimed to determine whether biomarkers and blood pressure could be potential predictors of MSIMI. Methods: This study enrolled 82 patients with documented CAD between June 1, 2017 and November 9, 2017. Patient blood samples were obtained at resting period and at the end of mental arithmetic. Then, patients were assigned to MSIMI positive group and MSIMI negative group. The main statistical methods included linear regression, receiver operating characteristic (ROC) curves, and logistic regression. Results: Patients with CAD with MSIMI had significantly greater median resting N-terminal pro-brain natriuretic peptide (NT-proBNP, 141.02 [45.85–202.76] pg/mL vs. 57.95 [27.06–117.64] pg/mL;Z =-2.23, P = 0.03) and mean systolic blood pressure (SBP)(145.56 ± 16.87 mmHg vs. 134.92 ± 18.16 mmHg, Z =-2.13, P = 0.04) when compared with those without MSIMI. After 5-min mental stress task, those who developed MSIMI presented higher elevation of median post-stressor high sensitivity cardiac troponin I (hs-cTnI, 0.020 [0.009–0.100] ng/mL vs. 0.009 [0.009–0.010] ng/mL;Z =-2.45, P = 0.01), post-stressor NT-proBNP (138.96 [39.93–201.56] pg/mL vs. 61.55 [25.66–86.50] pg/mL;Z =-2.15, P = 0.03) compared with those without MSIMI. Using the ROC curves, and after the adjustment for basic characteristics, the multiple logistic regression analysis showed that patients presenting a post-stressor hs-cTnI ≥ 0.015 ng/mL had seven-fold increase in the risk of developing MSIMI (odds ratio [OR]: 7.09;95% confidence interval [CI]: 1.65–30.48;P = 0.009), a rest NT-proBNP ≥ 80.51 pg/mL had nearly eight-fold increase (OR: 7.85;95% CI: 1.51–40.82;P = 0.014), a post-stressor NT-proBNP ≥ 98.80 pg/mL had 35-fold increase (OR: 34.96;95% CI: 3.72– 328.50;P = 0.002), a rest SBP ≥ 129.50 mmHg had 11-fold increase (OR: 11.42;95% CI: 1.21–108.17;P = 0.034). | 12 | Weakened carbon and nitrogen metabolisms under post-silking heat stress reduce the yield and dry matter accumulation in waxy maize显示文摘Post-silking high temperature is one of the abiotic factors that affects waxy maize(Zea mays L. sinensis Kulesh) growth in southern China. We conducted a pot trial in 2016–2017 to study the effects of post-silking daytime heat stress(35°C) on the activities of enzymes involved in leaf carbon and nitrogen metabolisms and leaf reactive oxygen species(ROS) and water contents. This study could improve our understanding on dry matter accumulation and translocation and grain yield production. Results indicated that decreased grain number and weight under heat stress led to yield loss, which decreased by 20.8 and 20.0% in 2016 and 2017, respectively. High temperature reduced post-silking dry matter accumulation(16.1 and 29.5% in 2016 and 2017, respectively) and promoted translocation of pre-silking photoassimilates stored in vegetative organs, especially in leaf. The lower leaf water content and chlorophyll SPAD value, and higher ROS(H2O2 and O2^-·) content under heat stress conditions indicated accelerated senescent rate. The weak activities of phosphoenolpyruvate carboxylase(PEPCase), Ribulose-1,5-bisphosphate carboxylase(Ru BPCase), nitrate reductase(NR), and glutamine synthase(GS) indicated that leaf carbon and nitrogen metabolisms were suppressed when the plants suffered from a high temperature during grain filling. Correlation analysis results indicated that the reduced grain yield was mainly caused by the decreased leaf water content, weakened NR activity, and increased H2O2 content. The increased accumulation of grain weight and post-silking dry matter and the reduced translocation amount in leaf was mainly due to the increased chlorophyll SPAD value and NR activity. Reduced PEPCase and Ru BPCase activities did not affect dry matter accumulation and translocation and grain yield. In conclusion, post-silking heat stress down-regulated the leaf NR and GS activities, increased the leafwater loss rate, increased ROS generation, and induced pre-silking carbohydrate translocation. | 17 | Evaluation of Chinese rice varieties resistant to the root-knot nematode Meloidogyne graminicola显示文摘The root-knot nematode Meloidogyne graminicola, which is distributed worldwide, is considered a major constraint on rice production in Asia. The present study used the root gall index and number of nematodes inside the roots to evaluate resistance/susceptibility to M. graminicola in different subpopulations of Oryza sativa(aus, hybrid aus, indica, hybrid indica, temperate japonica, tropical japonica). Nematode development in highly resistant varieties was also evaluated. Analyses of randomly selected 35 varieties showed the number of M. graminicola nematodes inside the roots correlated very strongly(r=0.87, P≤0.05) with the nematode gall index, and the results from pot and field experiments revealed similar rankings of the varieties for resistance/susceptibility. Among the 136 tested varieties, temperate japonica displayed the highest gall index, followed by tropical japonica, indica, hybrid indica, aus, and hybrid aus. Zhonghua 11(aus), Shenliangyou 1(hybrid aus) and Cliangyou 4418(hybrid indica) were highly resistant to M. graminicola under both pot and field conditions. Further examination of nematode development suggested that compared to susceptible rice, M. graminicola penetrated less often into highly resistant varieties and more frequently failed to develop into females. | 18 | Endothelial cell injury with inflammatory cytokine and coagulation in patients with sepsis显示文摘BACKGROUND:Current studies on CD62 P have focused mainly on cardiovascular diseases,while only few studies have evaluated the effects of CD62 P on the development of sepsis and the association between endothelial cell injury with inflammation and coagulation.This study attended to explore the association between endothelial cell injury with inflammation and coagulation by evaluating the expression of soluble CD62P(s-CD62P) in plasma and its mechanism in patients with sepsis,thus to provide the evidence of effective treatment of sepsis with anti-adhesion therapy targeted CD62 P.METHODS:A total of 70 critically ill patients with systemic inflammatory response syndrome(SIRS) admitted to intensive care unit(ICU) between September 2009 and February 2010 were enrolled for a prospective and control study.According to the diagnostic criteria of sepsis/SIRS,the patients were divided into two groups:a sepsis group(n=38) and a SIRS group(n=32).Another 20 healthy volunteers served as a control group.Patients in the sepsis group and SIRS group were matched by clinical signs of high blood pressure,diabetes and its complications.The demographics of the patients including age,sex,body mass index(BMI),smoking and alcohol addict were compared among the groups.Six mL peripheral blood samples were collected within 24-hour admission in ICU for enzymelinked immunosorbent assay(ELISA) to detect the plasma levels of S-CD62 P,TNF-α,and hs-CRP.And variables of coagulation function such as platelet(PLT),prothrombin(PT),activated partial thromboplastin time(APTT),D-dimer and antithrombin-Ⅲ(AT-Ⅲ) were analyzed during 24 hours after admission to ICU.Meanwhile sequential organ failure assessment(SOFA) score of critically ill patients was evaluated.Data were expressed as meanistandard deviation and were statistically analyzed by using SPSS 17.0statistical software.The differences in plasma levels of S-CD62 P of patients in each group were analyzed by ANOVA and the Kruskal-Wallis test.The relations between S-CD62 P and inflammatory cytokines as well as with coagulation were determined by Pearson's product moment correlation coefficient analysis.Changes were considered as statistically significant if P value was less than 0.05.RESULTS:Compared with the control group and SIRS group,the sepsis group demonstrated significantly higher levels of S-CD62 P,TNF-a and highly sensitive C-reactive protein(hs-CRP)(PO.05).The plasma levels of D-dimer,PT,and APTT in the sepsis and SIRS groups were significantly higher than those in the control group,while the platelet count and the activity of AT-Ⅲ were obviously lower(P<0.05).In the sepsis group,the plasma levels of hs-CRP and TNF-a were positively correlated with PT,APTT,and D-dimer,and negatively correlated with AT-Ⅲ and PLT(P<0.05).The plasma levels of S-CD62 P were significantly correlated with the plasma levels of TNF-a,hs-CRP,D-dimer,PT,and APTT,whereas they were correlated negatively well with PLT and AT-Ⅲ(P<0.05).CONCLUSIONS:The concentration of plasma S-CD62 P is elevated as a early biomarker in patients with sepsis,and it serves as one of the pathogenic factors responsible for endothelial cell damage.Coagulation and mediators of inflammation promote each other,aggravating the severity of sepsis.Plasma S-CD62 P may be an important factor for the development of coagulation and inflammatory reaction. | Huan Ding Xiang-yuan Cao Xi-gang Ma Wen-jie Zhou | 2013 | World Journal of Emergency Medicine2013,4,4: | 4 |
| 19 | Upregulation of UBAP2L in Bone Marrow Mesenchymal Stem Cells Promotes Functional Recovery in Rats with Spinal Cord Injury显示文摘Post-translational modifications of cellular proteins with ubiquitin or ubiquitin-like proteins regulate many cellular processes,such as cell proliferation,differentiation,apoptosis, signal transduction,intercellular immune recognition,inflammatory response,stress response,and DNA repair.Nice4/UBAP2L is an important member in the family of ubiquitin-like proteins,and its biological function remains unknown.This study aimed to investigate the effect of UBAP2L on spinal cord injury (SCI).At first,rat bone marrow mesenchymal stem cells (BMSCs)were infected with adeno-associated virus to induce over-expression of Nice4.Subsequently,the infected BMSCs were transplanted into rats suffering from semi-sectioned SCI.The results showed that the over-expression of Nice4 significantly promoted the proliferation and differentiation of BMSCs. In addition,the transplantation of infected BMSCs into the injured area of SCI rats improved the function repair of SCI.Importantly,the immunohistochemical and hematoxylin-eosin staining and RT-PCR results showed that the number of neuronal cells,oligodendrocytes,and astrocytes was significantly increased in the injured area,along with significantly upregulated expression ofcyclin D1 and p38 mitogen-activated protein kinase (MAPK).Meanwhile,the expression of caspase 3 protein was significantly down-regulated.In conclusion,the over-expression of Nice4 gene can promote the functional recovery in SCI rats by promoting cell proliferation and inhibiting apoptosis.
