1Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. 4Effect of high glucose levels on the calcification of vascular smooth muscle cells by inducing osteoblastic differentiation and intracellular calcium deposition via BMP-2/Cbfα-1 pathway显示文摘In this paper,we investigate the effect and the possible mechanism of high glucose levels on the calcification of human aortic smooth muscle cells (HASMCs).HASMCs were divided into four groups: normal glucose group (NG),osmolality control group (OC),high glucose group (HG,HASMCs culture medium containing 30 mmol/L glucose),and high glucose plus recombinant human Noggin protein (bone morphogenetic protein-2 (BMP-2) antagonist) group (HN).The mRNA levels and the protein expressions of BMP-2 and core binding factor alpha-1 (Cbfα-1) were measured by real-time quantitative polymerase chain reaction (PCR) and Western blot.After induced by 10 mmol/L β-glycerol phosphoric acid,cells were harvested for assessments of alkaline phosphatase (ALP) activities at Days 1,2,and 3,and intracellular calcium contents at Days 7 and 14,respectively.High glucose levels increased the mRNA levels and the protein expressions of BMP-2 and Cbfα-1 (P<0.05).The expression of Cbfα-1 was partially blocked by Noggin protein (P<0.05),while BMP-2 was not (P>0.05).After being induced by β-glycerol phosphoric acid,high glucose levels increased the ALP activity [(48.63±1.03) vs.(41.42±2.28) U/mg protein,Day 3;P<0.05] and the intracellular calcium content [(2.76±0.09) vs.(1.75±0.07) μmol/mg protein,Day 14;P<0.05] in a time-dependent manner when compared with the NG group,while the ALP activity could not be blocked by Noggin protein [(48.63±1.03) vs.(47.37±0.97) U/mg protein,Day 3;P>0.05].These results show that high glucose levels can evoke the calcification of HASMCs by inducing osteoblastic trans-differentiation and intracellular calcium deposition via the BMP-2/Cbfα-1 pathway,which can be partially blocked by Noggin protein.Fang LIU Hui ZHONG Jing-yuan LIANG Ping FU Zhi-juan LUO Li ZHOU Rong GOU Jun HUANG 2010Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2010,11,12:12

6LORELEI-LIKE GPI-ANCHORED PROTEINS 2/3 Regulate Pollen Tube Growth as Chaperones and Coreceptors for ANXUR/BUPS Receptor Kinases in Arabidopsis显示文摘Pollen tube growth is crucial for successful fertilization.In Arabidopsis thaliana,the ANXUR(ANX)/BUPS receptor kinase complex controls and maintains pollen tube growth in response to autocrine rapid alkalin-ization factor 4/19(RALF4/19)signaling;however,the molecular and cellular mechanisms underlying the ANX/BUPS-mediated regulation of pollen tube growth remain unclear.In this study,we found that pollen-specific LORELEI-like GPI-anchored proteins 2 and 3(LLG2/3)promote pollen tube growth in vitro and in vivo.LLG2/3 interacte with ANX/BUPS in a RALF4-concentration-dependent manner,suggesting that ANX/BUPS and LLG2/3 might form a receptor-coreceptor complex for perceiving RALF peptide signals.Disruption of the ANX/BUPS-LLG2/3 interaction led to the cytoplasmic retention of ANX1/2,in either llg2/3 knockdown mutants or in anx1/2 mutants lacking the J region,which mediates the ANX/BUPS-LLG2/3 interaction.Moreover,we found that RALF4 induced the production of reactive oxygen species(ROS),which stimulate pollen tube growth and reduce pollen burst rate.ROS levels are reduced in the pollen tubes of LLG2/3 RNAi lines,and application of exogenous H2O2 could partially rescue the defective pollen tube growth of LLG2/3 RNAi lines.Taken together,our study identifies LLG2/3 as novel regulatory components of pollen tube growth,and shows that they chaperone ANX/BUPS for secretion to the apical plasma membrane of pollen tube and act as coreceptors of ANX/BUPS in the acti-vation of ROS production for promoting pollen tube growth.Hanqian Feng Chen Liu Rong Fu Minmin Zhang Hui Li Lianping Shen Qiqi Wei Xiang Sun Lin Xu Bin Ni Chao Li 2019Molecular Plant2019,12,12:10
7Evaluation of surgical procedure selection based on intraoperative free portal pressure measurement in patients with portal hypertension显示文摘BACKGROUND:Various surgical procedures can be used to treat liver cirrhosis and portal hypertension.How to select the most appropriate procedure for patients with portal hypertension has become a difficult problem.This study aimed to analyze the relationship between the value of intraoperative free portal pressure(FPP)and postoperative complications,and to explore the significance of intraoperative FPP measurement with respect to surgical procedure selection.METHODS:The clinical data of 187 patients with portal hypertension who received pericardial devascularization and proximal splenorenal shunt combined with devascularization(combined operation)at the Department of General Surgery in our hospital from January 2001 to September 2008 were retrospectively analyzed.Among the patients who received pericardial devascularization,those with a postoperative FPP ≥22 mmHg were included in a high-pressure group(n=68), and those with FPP<22 mmHg were in a low-pressure group(n=49).Seventy patients who received the combined operation comprised a combined group.The intraoperative FPP measurement changes at different times,and the incidence of postoperative complications in the three groups of patients were compared.RESULTS:The postoperative FPP value in the high-pressure group was 27.5±2.3 mmHg,which was significantly higher than that of the low-pressure(20.9±1.8 mmHg)or combined groups(21.7±2.5 mmHg).The rebleeding rate in the high-pressure group was significantly higher than that in the low-pressure and combined groups.The incidence rates of postoperative hepatic encephalopathy and liver failure were not statistically different among the three groups.The mortality due to rebleeding in the low-pressure and combined groups(0.84%) was significantly lower than that of the high-pressure group.CONCLUSIONS:The study demonstrates that FPP is a critical measurement for surgical procedure selection in patients with portal hypertension.A FPP value≥22 mmHg after splenectomy and devascularization alone is an important indicator that an additional proximal splenorenal shunt needs to be performed.Sun, Yong-Wei Chen, Wei Luo, Meng Hua, Rong Liu, Wei Huo, Yan-Miao Wu, Zhi-Yong Cao, Hui 2010Hepatobiliary & Pancreatic Diseases International2010,9,3:9
8Correlation of Seven Biological Factors(Hsp90α,p53,MDM2,Bcl-2,Bax,Cytochrome C,and Cleaved caspase3)with Clinical Outcomes of ALK+ Anaplastic Large-cell Lymphoma显示文摘Objective To explore correlation of seven apoptosis‐related proteins (Hsp90α,p53,MDM2,Bcl‐2,Bax,Cytochrome C,and Cleaved caspase3) with clinical outcomes of ALK+ anaplastic large‐cell lymphoma (ALCL).Methods Using immunohistochemistry and immunofluorescence double staining methods,the expressions of these seven apoptosis‐associated proteins were studied to clarify their relationship with clinical outcomes of 36 ALK+ and 25 ALK‐ systemic ALCL patients enrolled between 1996 and 2006.The relationship of these apoptosis‐regulating proteins with NPM‐ALK status was also evaluated with the tyrosine inhibitor herbimycin A (HA) in vitro by immunocytochemistry,Western blotting and flow cytometric assays.Results The presence of Hsp90α‐,MDM2‐,Bax‐,Cytochrome C,and Cleaved caspase3‐positive tumor cells was found significantly different in ALK+ and ALK‐ ALCLs ,which was correlated with highly favorable clinical outcome.The Bcl‐2‐ and p53‐positive tumor cells were found in groups of patients with unfavorable prognosis.Inhibition of NPM‐ALK by HA could reactivate the p53 protein and subsequent apoptosis‐related proteins and therefore induced apoptosis in ALK+ ALCL cells.Conclusion Our results suggest that these seven proteins might be involved in apoptosis regulation and associated with clinical outcome of ALK+ systemic ALCLs.We also reveal a dynamic chain relation that NPM‐ALK regulates p53 expression and subsequent apoptosis cascade in ALK+ ALCLs.LI Hui Ling HUANG Xue Ping ZHOU Xin Hua JI Tian Hai WU Zi Qing WANG Zhi Qiang JIANG Hui Yong LIU Fan Rong ZHAO Tong 2011Biomedical and Environmental Sciences2011,24,6:9
9Nasopharyngeal adenoid cystic carcinoma: magnetic resonance imaging features in ten cases显示文摘Nasopharyngeal adenoid cystic carcinoma (NACC) is a rare malignancy with high local invasiveness. To date, there is no consensus on the imaging characteristics of NACC. To address this, we retrospectively reviewed 10 cases of NACC and summarized the magnetic resonance imaging (MRI) features. MR images of 10 patients with histologically validated NACC were reviewed by two experienced radiologists. The location, shape, margin, signal intensity, lesion texture, contrast enhancement patterns, local invasion, and cervical lymphadenopathy of all tumors were evaluated. Clinical and pathologic records were also reviewed. No patients were positive for antibodies against Epstein-Barr virus (EBV). The imaging patterns of primary tumors were classified into two types as determined by location, shape, and margin. Of all patients, 7 had tumors with a type 1 imaging pattern and 3 had tumors with a type 2 imaging pattern. The 4 tubular NACCs were all homogeneous tumors, whereas 3 (60%) of 5 cribriform NACCs and the sole solid NACC were heterogeneous tumors with separations or central necrosis on MR images. Five patients had perineural infiltration and intracranial involvement, and only 2 had cervical lymphadenopathy. Based on these results, we conclude that NACC is a local, aggressive neoplasm that is often negative for EBV infection and associated with a low incidence of cervical lymphadenopathy. 11Mechanism of acupuncture regulating visceral sensation and mobility显示文摘Chinese ancient medical scientists have long focused on the internal and external contacts between acupoints on the surface of the body and the viscera.The Miraculous Pivot(it is one of the earliest medical classics in China)stated,“Twelve regular channels belong to the zang-fu organs internally,and connect to the extremities and joints externally.”Traditional Chinese medicine considers acupoints as defined areas where the Qi of viscera and meridians are transfused.These include the reaction points of visceral diseases on the body surface as well as the acupuncture trigger points that promote the flow of Qi and blood,and regulate visceral function.Chinese ancient medical scientists classified the specificity of the main acupoints in the body based on the meridian doctrine,which has been instructing clinical application for about 2000 years.Laws on the domino effect of acupoints have mainly focused on conclusions to clinical experiences.Indications of some acupoints exceed the practical paradigm since the excessive extension occurred during theory derivation.The current research direction on acupuncture focuses on three aspects:the effectiveness of acupuncture and moxibustion;the relevances and associations between meridians and viscera;and the physical and chemical properties and relevant physical basis of acupoints.The relevance between meridians and viscera is the central theory in the meridian doctrine,and acupoints are regarded as an important link in the relationship between meridians and viscera.Specific relationships between acupoints and target organs exist.Stimulating different acupoints on the body surface can help deal with different diseases,especially visceral diseases.In addition,acupoints have a dual function of reflecting and treating visceral diseases.There is no systemic research available on acupoint specificity,despite current knowledge and clinical experiences,which results in a weak foundation for acupuncture theory.This study focuses on the relevance and associations between meridians and viscera.A summary of the mechanisms of acupuncture regulating visceral sensation and mobility and the specific relationships between acupoints and their target organs are presented in this review.Peijing Rong Bing Zhu Yuqing Li Xinyan Gao Hui Ben Yanhua Li Liang Li Wei He Rupeng Liu Lingling Yu 2011Frontiers of Medicine2011,5,2:8

13Exosomal piRNA profiling revealed unique circulating piRNA signatures of cholangiocarcinoma and gallbladder carcinoma显示文摘Cholangiocarcinoma(CCA)and gallbladder carcinoma(GBC)are biliary tract cancers with poor five-year survival and high recurrence rates.Both CCA and GBC patients suffer from lack of circulating diagnostic biomarkers at the early stage.Extracellular vesicles,especially exosomes,have been emerged as promising diagnostic sources for cancers due to easy and quick accessibility.Hence,identification of exosomal biomarkers provides a novel strategy for CCA and GBC diagnosis.Here,five CCA patients and four GBC patients were enrolled for exosomal small RNA sequencing.Our data showed that exosomal piwi-interacting RNA(piRNA)populations were altered in the plasma of CCA and GBC patients.In comparison to healthy individuals,694 and 323 piRNAs were upregulated in CCA and GBC,respectively,while 36 and 191 piRNAs were down regulated.Interestingly,sequencing results predicted that piR-2660989,piR-10506469,piR-20548188,piR-10822895,piR-hsa-23209,and piR-18044111 were upregulated in both CCA and GBC plasma.Importantly,we further included blood samples from 50 health individuals,40 CCA patients,and 25 GBC patients and found that piR-10506469 were significantly increased in the exosomes of plasma from both CCA and GBC patients.Moreover,we analyzed the expression levels of differentially expressed exosomal piRNAs in the plasma of CCA and GBC patient before and after surgeries and found that piR-10506469 and piR-20548188 were significantly decreased in patients underwent surgeries.Taken together,our data revealed that exosomal piRNAs those are differentially expressed in CCA and GBC plasma may serve as potential biomarkers for the diagnosis of CCA and GBC.Xinjin Gu Chen Wang Hui Deng Chong Qing Rong Liu Sanhong Liu Xinying Xue 2020Acta Biochimica et Biophysica Sinica2020,52,5:7
14A modified method for locating parapharyngeal space neoplasms on magnetic resonance images: implications for differential diagnosis显示文摘The parapharyngeal space(PPS) is an inverted pyramid-shaped deep space in the head and neck region, and a variety of tumors, such as salivary gland tumors, neurogenic tumors, nasopharyngeal carcinomas with parapharyngeal invasion, and lymphomas, can be found in this space. The differential diagnosis of PPS tumors remains challenging for radiologists. This study aimed to develop and test a modified method for locating PPS tumors on magnetic resonance(MR) images to improve preoperative differential diagnosis. The new protocol divided the PPS into three compartments: a prestyloid compartment, the carotid sheath, and the areas outside the carotid sheath. PPS tumors were located in these compartments according to the displacements of the tensor veli palatini muscle and the styloid process, with or without blood vessel separations and medial pterygoid invasion. This protocol, as well as a more conventional protocol that is based on displacements of the internal carotid artery(ICA), was used to assess MR images captured from a series of 58 PPS tumors. The consequent distributions of PPS tumor locations determined by both methods were compared. Of all 58 tumors, our new method determined that 57 could be assigned to precise PPS compartments. Nearly all(13/14; 93%) tumors that were located in the pre-styloid compartment were salivary gland tumors. All 15 tumors within the carotid sheath were neurogenic tumors. The vast majority(18/20; 90%) of trans-spatial lesions were malignancies. However, according to the ICA-based method, 28 tumors were located in the pre-styloid compartment, and 24 were located in the post-styloid compartment, leaving 6 tumors that were difficult to locate. Lesions located in both the pre-styloid and the post-styloid compartments comprised various types of tumors. 17New evidence for the links between the local water cycle and the underground wet sand layer of a mega-dune in the Badain Jaran Desert, China显示文摘Scientists and the local government have great concerns about the climate change and water resources in the Badain Jaran Desert of western China. A field study for the local water cycle of a lake-desert system was conducted near the Noertu Lake in the Badain Jaran Desert from 21 June to 26 August 2008. An underground wet sand layer was observed at a depth of 20–50 cm through analysis of datasets collected during the field experiment. Measurements unveiled that the near surface air humidity increased in the nighttime. The sensible and latent heat fluxes were equivalent at a site about 50 m away from the Noertu Lake during the daytime, with mean values of 134.4 and 105.9 W/m2 respectively. The sensible heat flux was dominant at a site about 500 m away from the Noertu Lake, with a mean of 187.7 W/m2, and a mean latent heat flux of only 26.7 W/m2. There were no apparent differences for the land surface energy budget at the two sites during the night time. The latent heat flux was always negative with a mean value of –12.7 W/m2, and the sensible heat flux was either positive or negative with a mean value of 5.10 W/m2. A portion of the local precipitation was evaporated into the air and the top-layer of sand dried quickly after every rainfall event, while another portion seeped deep and was trapped by the underground wet sand layer, and supplied water for surface psammophyte growth. With an increase of air humidity and the occurrence of negative latent heat flux or water vapor condensation around the Noertu Lake during the nighttime, we postulated that the vapor was transported and condensed at the lakeward sand surface, and provided supplemental underground sand pore water. 18The Association of Socioeconomic Status with the Burden of Cataract-related Blindness and the Effect of Ultraviolet Radiation Exposure: An Ecological Study显示文摘Objective To assess the association of socioeconomic status with the burden of cataract blindness in terms of year lived with disability(YLD) rates and to determine whether ultraviolet radiation(UVR) levels modify the effect of socioeconomic status on this health burden.Methods National and subnational age-standardized YLD rates associated with cataract-related blindness were derived from the Global Burden of Disease(GBD) study 2017. The human development index(HDI) from the Human Development Report was used as a measure of socioeconomic status.Estimated ground-level UVR exposure was obtained from the Ozone Monitoring Instrument(OMI)dataset of the National Aeronautics and Space Administration(NASA).Results Across 185 countries, socioeconomic status was inversely associated with the burden of cataract blindness. Countries with a very high HDI had an 84% lower age-standardized YLD rate [95%confidence interval(CI): 60%–93%, P < 0.001] than countries with a low HDI;for high-HDI countries, the proportion was 76%(95% CI: 53%–88%, P < 0.001), and for medium-HDI countries, the proportion was48%(95% CI: 15%–68%, P = 0.010;P for trend < 0.001). The interaction analysis showed that UVR exposure played an interactive role in the association between socioeconomic status and cataract blindness burden(P value for interaction = 0.047).Conclusion Long-term high-UVR exposure amplifies the association of poor socioeconomic status with the burden of cataract-related blindness.
