1Akt Inhibitor Perifosine Prevents Epileptogenesis in a Rat Model of Temporal Lobe Epilepsy显示文摘Accumulating data have revealed that abnormal activity of the mTOR(mammalian target of rapamycin)pathway plays an important role in epileptogenesis triggered by various factors. We previously reported that pretreatment with perifosine, an inhibitor of Akt(also called protein kinase B), abolishes the rapamycin-induced paradoxical increase of S6 phosphorylation in a rat model induced by kainic acid(KA). Since Akt is an upstream target in the mTOR signaling pathway, we set out to determine whether perifosine has a preventive effect on epileptogenesis. Here, we explored the effect of perifosine on the model of temporal epilepsy induced by KA in rats and found that pretreatment with perifosine had no effect on the severity or duration of the KA-induced status epilepticus. However, perifosine almost completely inhibited the activation of p-Akt and p-S6 both acutely and chronically following the KA-induced status epilepticus.Perifosine pretreatment suppressed the KA-induced neuronal death and mossy fiber sprouting. The frequency of spontaneous seizures was markedly decreased in rats pretreated with perifosine. Accordingly, rats pretreated with perifosine showed mild impairment in cognitive functions. 2Effect of emodin on Aquaporin 5 expression in rats with sepsis-induced acute lung injury显示文摘OBJECTIVE: To investigate the effects of emodin on aquaporin 5(AQP5) expression in rats with sepsis-induced acute lung injury.METHODS: We divided 60 adult male Sprague-Dawley rats, weighing 200-230 g, into four groups: control, sham surgery, model and emodin groups(n = 15 for each). We created a sepsis model with cecal ligation and puncture; the sham surgery group had their cecums replaced after exposure outside the abdominal cavity. Each group was further divided into three subgroups(n = 5 for each)and expressions of AQP5 m RNA and proteins in lung tissue were measured by real-time fluorescence polymerase chain reaction and western blot at 6, 12 and 24 h after surgery.RESULTS: AQP5 expression did not change over time in the control group and sham surgery group,but decreased over time in the model group. The lowest expression was found in 12-h subgroup,which significantly differed from the 6-h subgroup(P < 0.01). Compared with the model group, AQP5 expression in the emodin group was significantly higher in all the subgroups(all P <0.01). Expressions in the 12-h subgroup were the highest, and significantly differed from the other subgroups. We found that lung tissue damage,such as pulmonary edema, alveolar damage and the exudation of red blood cells in pulmonary interstitium and alveolar, was significantly milder in the emodin group under light microscope than the model group.CONCLUSION: AQP5 expression was significantly down-regulated in rats with sepsis-induced acute lung injury induced by cecal ligation and puncture.Early prophylactic use of emodin can significantly enhance the AQP5 expression, thus effectively reducing the degree of pulmonary edema in septic rats.Sun Yanni Sun Lijing Liu Shuqing Song Juan Cheng Jiejing Liu Jun 2015Journal of Traditional Chinese Medicine2015,35,6:5
3Somatic Mutations Confer Severe Peripheral Neuropathy in POEMS Syndrome-Associated Multicentric Castleman Disease显示文摘Dear Editor,POEMS-associated multicentric Castleman disease(POEMS-MCD)is a subtype of MCD that presents POEMS symptoms(polyneuropathy,organomegaly,endocrinopathy,M protein,and skin changes)as well as the manifestations of MCD.Among these symptoms,polyneuropathy is a critical feature.However,its mechanism remains poorly understood.The present study presents a case of severe peripheral neuropathy in a POEMSMCD patient.Whole-exome sequencing(WES),threedimensional modeling,and protein stability analysis demonstrated that the mutation V49G in PDLIM5 resulted in instability of the protein,which may play a key role in the pathogenesis of peripheral neuropathy in POEMSMCD.Qingqing Lin Juying Wei Jiejing Qian Liangshun You Wenbin Qian 2020Neuroscience Bulletin2020,36,6:3
4The oral microbiome of pregnant women facilitates gestational diabetes discrimination显示文摘The oral microbiota plays an important role in the development of various diseases,whereas its association with gestational diabetes mellitus (GDM) remains largely unclear.The aim of this study is to identify biomarkers from the oral microbiota of GDM patients by analyzing the microbiome of the saliva and dental plaque samples of 111 pregnant women.We find that the microbiota of both types of oral samples in GDM patients exhibits differences and significantly varies from that of patients with periodontitis or dental caries.Using bacterial biomarkers from the oral microbiota,GDM classification models based on support vector machine and random forest algorithms are constructed.The area under curve (AUC) value of the classification model constructed by combination of Lautropia and Neisseria in dental plaque and Streptococcus in saliva reaches 0.83,and the value achieves a maximum value of 0.89 by adding clinical features.These findings suggest that certain bacteria in either saliva or dental plaque can effectively distinguish women with GDM from healthy pregnant women,which provides evidence of oral microbiome as an informative source for developing noninvasive biomarkers of GDM.Xiaoqing Li Jiayong Zheng Xiuling Ma Bing Zhang Jinyang Zhang Wenhuan Wang Congcong Sun Yeping Wang Jianqiong Zheng Haiying Chen Jiejing Tao Hai Wang Fengyi Zhang Jinfeng Wang Hongping Zhang 2021Journal of Genetics and Genomics2021,48,1:2
5Pseudomonas mendocina LYX:A novel aerobic bacterium with advantage of removing nitrate high effectively by assimilation and dissimilation simultaneously显示文摘The problem of nitrate accumulation in aerobic tank and total nitrogen excessive discharge in effluent was very common in traditional livestock and poultry farming wastewater treatment systems owing to the lengthy process flow and low process control level.A strain LYX of aerobic bacterium was isolated from the activated sludge of a wastewater treatment system in a pig farm,which could remove nitrate effectively in aerobic tank and was identified Pseudomonas mendocina by 16S rRNA sequencing.Under the condition of nitrate as the sole nitrogen source,this strain removed over 90%of NO_(3)^(−)-N with an initial concentration of 110 mg/L under aerobic conditions within 48 hours.Among them,37.9%of NO_(3)^(−)-N was assimilated into Bio-N,about 51.9%was reduced to gaseous nitrogen and less than 0.5%of nitrogen was replaced by NO_(3)^(−)-N and NH_(4)^(+)-N,9.7%NO_(3)^(−)-N remained in the effluent at the end.At the same time,four key genes(napA,nirK,norB and nosZ)related to nitrate nitrogen removal were expressed during the denitrification process of P.mendocina LYX,in which the transcription level of the indicator genes of this aerobic denitrifying bacterium(napA)was the highest.In addition,it was found with the 15N tracer technique that inoculation of this strain on sludge increased the amount of nitrogen loss from 9.26 nmol N/(g·h)to 23.835 nmol N/(g·h).Therefore,P.medocina LYX is a potential bioagent for advanced nitrogen removal by assimilating and reducing nitrate simultaneously in aerobic tanks.Yuxin Li Jiayin Ling Pengcheng Chen Jinliang Chen Ruizhi Dai Jinsong Liao Jiejing Yu Yanbin Xu 2021Frontiers of Environmental Science & Engineering2021,15,4:2
6A New κ-Carrageenase Cgk S from Marine Bacterium Shewanella sp. Kz7显示文摘A new κ-carrageenase gene cgk S was cloned from marine bacterium Shewanella sp. Kz7 by using degenerate and site-finding PCR. The gene was comprised of an open reading frame of 1224 bp, encoding 407 amino acid residues, with a signal peptide of 24 residues. Based on the deduced amino acid sequence, the κ-carrageenase Cgk S was classified into the Glycoside Hydrolase family 16. The cgk S gene was expressed in Escherichia coli, and the recombinant enzyme was purified to homogeneity with a specific activity of 716.8 U mg-1 and a yield of 69%. Recombinant Cgk S was most active at 45℃ and p H 8.0. It was stable at p H 6.0–9.0 and below 30℃. The enzyme did not require Na Cl for activity, although its activity was enhanced by Na Cl. 13New key pre-distribution scheme using symplectic geometry over finite fields for wireless sensor networks显示文摘To achieve secure communication in wireless sensor networks(WSNs), where sensor nodes with limited computation capability are randomly scattered over a hostile territory, various key pre-distribution schemes(KPSs) have been proposed. In this paper, a new KPS is proposed based on symplectic geometry over finite fields. A fixed dimensional subspace in a symplectic space represents a node, all 1-dimensional subspaces represent keys and every pair of nodes has shared keys. But this naive mapping does not guarantee a good network resiliency. Therefore, it is proposed an enhanced KPS where two nodes have to compute a pairwise key, only if they share at least q common keys. This approach enhances the resilience against nodes capture attacks. 20Artificial intelligence-based medical image segmentation for 3D printing and naked eye 3D visualization显示文摘Image segmentation for 3D printing and 3D visualization has become an essential component in many fields of medical research,teaching,and clinical practice.Medical image segmentation requires sophisticated computerized quantifications and visualization tools.Recently,with the development of artificial intelligence(AI)technology,tumors or organs can be quickly and accurately detected and automatically contoured from medical images.This paper introduces a platform-independent,multi-modality image registration,segmentation,and 3D visualization program,named artificial intelligence-based medical image segmentation for 3D printing and naked eye 3D visualization(AIMIS3D).YOLOV3 algorithm was used to recognize prostate organ from T2-weighted MRI images with proper training.Prostate cancer and bladder cancer were segmented based on U-net from MRI images.CT images of osteosarcoma were loaded into the platform for the segmentation of lumbar spine,osteosarcoma,vessels,and local nerves for 3D printing.Breast displacement during each radiation therapy was quantitatively evaluated by automatically identifying the position of the 3D printed plastic breast bra.Brain vessel from multimodality MRI images was segmented by using model-based transfer learning for 3D printing and naked eye 3D visualization in AIMIS3D platform.Guang Jia Xunan Huang Sen Tao Xianghuai Zhang Yue Zhao Hongcai Wang Jie He Jiaxue Hao Bo Liu Jiejing Zhou Tanping Li Xiaoling Zhang Jinglong Gao 2022Intelligent Medicine2022,2,1:0
