| 1 | Combined effects of p53 and MDM2 polymorphisms on susceptibility and surgical prognosis in hepatitis B virus-related hepatocellular carcinoma显示文摘The p53 signaling pathway works as a potent barrier to tumor progression.Two single nucleotide polymor-phisms(SNPs)in the gene loci of p53 pathway,p53 codon 72 Arg72Pro and MDM2 SNP309(T>G),have been shown to cause perturbation of p53 function,but the effect of the two SNPs on the risk of hepatocellular carcinoma(HCC)remains inconsistent.This study in-vestigated the influence of combined p53 Arg72Pro and MDM2 SNP309 on the risk of developing HCC in pa-tients with chronic hepatitis B virus infection,and evaluated the significance of the two combined SNPs on patient prognosis.In total,350 HCC patients,230 non-HCC patients,and 96 healthy controls were geno-typed for the p53 Arg72Pro and MDM2 SNP309.The combined p53 Pro/Pro and MDM2 G/G genotype was significantly associated with HCC risk(P=0.047).Mul-tivariate analysis indicated that combined p53 Pro/Pro and MDM2 G/G genotype was an independent factor affecting recurrence and survival(P<0.05).Patients with combined p53 Pro/Pro and MDM2 G/G genotypes had a poorer prognosis than other genotypes,P<0.01 for both disease-free survival(DFS)and overall survival(OS).DFS and OS rates also differed significantly be-tween Barcelona Clinic Liver Cancer(BCLC)stage A patients with combined p53 Pro/Pro and MDM2 G/G and other genotypes(P<0.05).Thus,the combined p53 Pro/Pro and MDM2 G/G genotype is associated with increased risk of developing HCC and is an independ-ent adverse prognostic indicator in early stage HCC. | Yun Yang Tian Xia Ning Li Jin Zhang Yuan Yang Wenming Cong Qiang Deng Ke Lan Weiping Zhou | 2013 | Protein & Cell2013,4,1: | 8 |
| 2 | Low-frequency sea level variation and its correlation with climate events in the Pacific显示文摘Global mean sea level(GMSL) has not only significant secular trend and seasonal variations,but also inter-annual and decadal variations.This paper reconstructs the time series of GMSL variations between 1948 and 2007 by combining satellite altimeter measurements and tide gauge observations.Based on the time series,the acceleration of GMSL rise in the second half of 20th century is estimated to be 0.010±0.009 mm/a 2,and multi-scale low frequency sea level oscillations including decadal variations are detected,and the high-rate of GMSL rise during 1993-2003 is locate in the ascending phase of low frequency oscillation.Then,using the reconstructed GMSL time series after removing the secular trend from satellite altimetry and tide gauges measurements,it shows that low frequency signal of sea level variation has strong correlations with the index of El Ni o/Southern Oscillation(ENSO) and Pacific Decadal Oscillation(PDO).But in several time periods,they have large difference due to effects of both high frequency occurrence of El Ni o and La Ni a phenomenon and short term switch of PDO events. | JIN TaoYong LI JianCheng JIANG WeiPing CHU YongHai | 2012 | Chinese Science Bulletin2012,57,27: | 6 |
| 3 | The heterogeneity of islet autoantibodies and the progression of islet failure in type 1 diabetic patients显示文摘Type 1 diabetes mellitus is heterogeneous in many facets. The patients suffered from type 1 diabetes present several levels of islet function as well as variable number and type of islet-specific autoantibodies. This study was to investigate prevalence and heterogeneity of the islet autoantibodies and clinical phenotypes of type 1 diabetes mellitus; and also discussed the process of islet failure and its risk factors in Chinese type 1 diabetic patients. A total of 1,291 type 1 diabetic patients were enrolled in this study. Demographic information was collected. Laboratory tests including mixed-meal tolerance test, human leukocyte antigen alleles, hemoglobin A1 c, lipids, thyroid function and islet autoantibodies were conducted. The frequency of islet-specific autoantibody in newly diagnosed T1 DM patients(duration shorter than half year) was 73% in East China. According to binary logistic regressions, autoantibody positivity, longer duration and lower Body Mass Index were the risk factors of islet failure. As the disease developed, autoantibodies against glutamic acid decarboxylase declined as well as the other two autoantibodies against zinc transporter 8 and islet antigen 2. The decrease of autoantibodies was positively correlated with aggressive beta cell destruction. Autoantibodies can facilitate the identification of classic T1 DM from other subtypes and predict the progression of islet failure. | 4 | The global mean sea surface model WHU2013显示文摘The mean sea surface(MSS) model is an important reference for the study of charting datum and sea level change. A global MSS model named WHU2013, with 20? 20 spatial resolution between 80S and 84N, is established in this paper by combining nearly 20 years of multi-satellite altimetric data that include Topex/Poseidon(T/P), Jason-1, Jason-2, ERS-2,ENVISAT and GFO Exact Repeat Mission(ERM) data, ERS-1/168, Jason-1/C geodetic mission data and Cryosat-2 low resolution mode(LRM) data. All the ERM data are adjusted by the collinear method to achieve the mean along-track sea surface height(SSH), and the combined dataset of T/P, Jason-1 and Jason-2 from 1993 to 2012 after collinear adjustment is used as the reference data. The sea level variations in the non-ERM data(geodetic mission data and LRM data) are mainly investigated, and a combined method is proposed to correct the sea level variations between 66S and 66N by along-track sea level variation time series and beyond 66S or 66N by seasonal sea level variations. In the crossover adjustment between multi-altimetric data, a stepwise method is used to solve the problem of inconsistency in the reference data between the high and low latitude regions. The proposed model is compared with the CNES-CLS2011 and DTU13 MSS models, and the standard derivation(STD) of the differences between the models is about 5 cm between80S and 84N, less than 3 cm between 66S and 66N, and less than 4 cm in the China Sea and its adjacent sea. | 9 | A durable 4-1BB-based CD19 CAR-T cell for treatment of relapsed or refractory non-Hodgkin lymphoma显示文摘Objective:Previous studies reported that 4-1BB-based CD19 chimeric antigen receptor(CAR)-T cells were more beneficial for the clinical outcomes than CD28-based CAR-T cells,especially the lower incidence rate of severe adverse events.However,the median progression-free survival(mPFS)of 4-1BB-based product Kymriah was shorter than that of CD28-based Yescarta(2.9 months vs.5.9 months),suggesting that Kymriah was limited in the long-term efficacy.Thus,a safe and durable 4-1BB-based CD19 CAR-T needs to be developed.Methods:We designed a CD19-targeted CAR-T(named as IM19)which consisted of an FMC63 scFv,4-1BB and CD3ζintracellular domain and was manufactured into a memory T-enriched formulation.A phase I/II clinical trial was launched to evaluate the clinical outcomes of IM19 in relapsed or refractory(r/r)B cell non-Hodgkin lymphoma(B-NHL).Dose-escalation investigation(at a dose of 5×10^(5)/kg,1×10^(6)/kg and 3×106/kg)was performed in 22 r/r B-NHL patients.All patients received a single infusion of IM19 after 3-day conditional regimen.Results:At month 3,the overall response rate(ORR)was 59.1%,the complete response rate(CRR)was 50.0%.The mPFS was 6 months and the 1-year overall survival rate was 77.8%.Cytokine release syndrome(CRS)occurred in 13 patients(59.1%),with 54.5%of grade 1−2 CRS.Only one patient(4.5%)experienced grade 3 CRS and grade 3 neurotoxicity.Conclusions:These results demonstrated the safety and durable efficacy of a 4-1BB-based CD19 CAR-T,IM19,which is promising for further development and clinical investigation. | Zhitao Ying Ting He Shanzhao Jin Xiaopei Wang Wen Zheng Ningjing Lin Meifeng Tu Yan Xie Lingyan Ping Weiping Liu Lijuan Deng Yanping Ding Xuelian Hu Bing Bu Xin’an Lu Yuqin Song Jun Zhu | 2022 | Chinese Journal of Cancer Research2022,34,1: | 2 |

| 19 | AQR is a novel type 2 diabetes-associated gene that regulates signaling pathways critical for glucose metabolism显示文摘Type 2 diabetes mellitus(T2 DM) is a common metabolic disease influenced by both genetic and environmental factors. In this study, we performed an in-house genotyping and meta-analysis study using three independent GWAS datasets of T2 DM and found that rs3743121, located 1 kb downstream of AQR,was a novel susceptibility SNP associated with T2 DM. The risk allele C of rs3743121 was correlated with the increased expression of AQR in white blood cells, similar to that observed in T2 DM models. The knockdown of AQR in HepG2 facilitated the glucose uptake, decreased the expression level of PCK2,increased the phosphorylation of GSK-3β, and restored the insulin sensitivity. Furthermore, the suppression of AQR inhibited the mTOR pathway and the protein ubiquitination process. | 20 | Variability of the Kuroshio extension system in 1992-2013 from satellite altimetry data显示文摘The Kuroshio Extension(KE) plays an important role in climate and environmental change in the North Pacific. In this paper, more than 20 years of merged absolute dynamic topography and merged sea level anomaly products from satellite altimetry are used to analyze the stability of the KE system. By analyzing the annually averaged sea surface topography, the variations of inter-annual path and annually averaged eddy kinetic energy at the KE region, the KE's two dynamic states are given as: the relatively stable state during 1993e1995, 2002e2005, and 2010e2012, and the unstable dynamic state among 1996e2001 and2006e2009. During the stable state, the KE spindle had a shorter path length and smaller time-varying amplitude, as well as a trend to move northward. While during the unstable state, the KE spindle had a longer path length and an integral southward transport trend, and was observed to oscillate significantly over time. The analysis on the KE's upstream and downstream region gives the same variations, indicating that they are significantly affected by the El Nino events. The power spectrum of the mean latitudinal position variation of the KE's upstream and downstream shows significant quasi-decadal oscillation characteristics and strong annual signals. Furthermore, the correlation of the strength variation between the southern RG and the KE's upstream is calculated to be 0.50 after low-pass filtering,and that of the mean latitudinal position variation between the southern RG and the KE's upstream/downstream are 0.75/0.69 after low-pass filtering, respectively.
