| 1 | Assessing the effects of surface-bound humic acid on the phototoxicity of anatase and rutile TiO_2 nanoparticles in vitro显示文摘In this study,the cytotoxicity of two different crystal phases of TiO_2 nanoparticles,with surface modification by humic acid(HA),to Escherichia coli,was assessed.The physicochemical properties of TiO_2 nanoparticles were thoroughly characterized.Three different initial concentrations,namely 50,100,and 200 ppm,of HA were used for synthesis of HA coated TiO_2 nanoparticles(denoted as A/RHA50,A/RHA100,and A/RHA200,respectively).Results indicate that rutile(LC50(concentration that causes 50%mortality compared the control group)=6.5)was more toxic than anatase(LC_50=278.8)under simulated sunlight(SSL)irradiation,possibly due to an extremely narrow band gap.It is noted that HA coating increased the toxicity of anatase,but decreased that of rutile.Additionally,AHA50 and RHA50had the biggest differences compared to uncoated anatase and rutile with LC_50of 201.9 and21.6,respectively.We then investigated the formation of reactive oxygen species(ROS)by TiO_2 nanoparticles in terms of hydroxyl radicals(OH)and superoxide anions(O_2^-).Data suggested that O_2^- was the main ROS that accounted for the higher toxicity of rutile upon SSL irradiation.We also observed that HA coating decreased the generation of OH and O_2^- on rutile,but increased O_2^- formation on anatase.Results from TEM analysis also indicated that HA coated rutile tended to be attached to the surface of E.coli more than anatase. | Xiaojia He Sabrieon Sanders Winfred G.Aker Yunfeng Lin Jessica Douglas Huey-min Hwang | 2016 | Journal of Environmental Sciences2016,28,4: | 6 |
| 2 | A Zebrafish Model of 5q-Syndrome Using CRISPR/Cas9 Targeting RPS14 Reveals a p53-Independent and p53-Dependent Mechanism of Erythroid Failure显示文摘5q-syndrome is a distinct form of myelodysplastic syndrome(MDS) where a deletion on chromosome 5 is the underlying cause.MDS is characterized by bone marrow failures,including macrocytic anemia.Genetic mapping and studies using various models support the notion that ribosomal protein S14(RPS14) is the candidate gene for the erythroid failure.Targeted disruption of RPS14 causes an increase in p53 activity and p53-mediated apoptosis,similar to what is observed with other ribosomal proteins.However,due to the higher risk for cancer development in patients with ribosome deficiency,targeting the p53 pathway is not a viable treatment option.To better understand the pathology of RPS14 deficiency in 5q-deletion,we generated a zebrafish model harboring a mutation in the RPS14 gene.This model mirrors the anemic phenotype seen in 5q-syndrome.Moreover,the anemia is due to a late-stage erythropoietic defect,where the erythropoietic defect is initially p53-independent and then becomes p53-dependent.Finally,we demonstrate the versatility of this model to test various pharmacological agents,such as RAP-011,L-leucine,and dexamethasone in order to identify molecules that can reverse the anemic phenotype. | Jason Ear Jessica Hsueh Melinda Nguyen QingHua Zhang Victoria Sung Rajesh Chopra Kathleen M.Sakamoto Shuo Lin | 2016 | Journal of Genetics and Genomics2016,43,5: | 5 |

| 5 | Procedural complexity independent of P2Y12 reaction unit(PRU)values is associated with acute in situ thrombosis in Pipeline flow diversion of cerebral aneurysms显示文摘background Acute in situ thrombosis is an ischaemic phenomenon during Pipeline embolisation device(PED)procedures with potentially high morbidity and mortality.There is controversy regarding the role of platelet function testing with P2Y12 assay as a predictor of intraprocedural thromboembolic events.There is limited knowledge on whether procedural complexity influences these events.Methods Data were collected retrospectively on 742 consecutive PED cases at a single institution.Patients with intraprocedural acute thrombosis were compared with patients without these events.results A cohort of 37 PED cases with acute in situ thrombosis(mean age 53.8 years,mean aneurysm size 8.4 mm)was matched with a cohort of 705 PED cases without intraprocedural thromboembolic events(mean age 56.4 years,mean aneurysm size 6.9 mm).All patients with in situ thrombosis received intra-arterial and/or intravenous abciximab.The two groups were evenly matched in patient demographics,previous treatment/subarachnoid hemorrhage(SAH)and aneurysm location.There was no statistical difference in postprocedural P2Y12 reaction unit(PRU)values between the two groups,with a mean of 156 in the in situ thrombosis group vs 148 in the control group(p=0.5894).Presence of cervical carotid tortuosity,high cavernous internal carotid artery grade,need for multiple PED and vasospasm were not significantly different between the two groups.The in situ thrombosis group had statistically significant longer fluoroscopy time(60.4 vs 38.4 min,p<0.0001),higher radiation exposure(3476 vs 2160 mGy,p<0.0001),higher rates of adjunctive coiling(24.3% vs 8.37%,p=0.0010)and higher utilisation of balloon angioplasty(37.8% vs 12.2%,p<0.0001).Clinically,the in situ thrombosis cohort had higher incidence of major and minor stroke,intracerebral haemorrhage and length of stay.Conclusions Predictors of procedural complexity(higher radiation exposure,longer fluoroscopy time,adjunctive coiling and need for balloon angioplasty)are associated with acute thrombotic events during PED placement,independent of PRU values. | Bowen Jiang Matthew T Bender Erick M Westbroek Jessica K Campos Li-Mei Lin Risheng Xu Rafael J Tamargo Judy Huang Geoffrey P Colby Alexander L Coon | 2018 | Stroke & Vascular Neurology2018,3,3: | 2 |

| 18 | The function of floral orientation in bluebells:interactions with pollinators and rain in two species of Mertensia(Boraginaceae)显示文摘Aims Pollinators are traditionally considered to be the primary agent of selection on floral traits.However,floral traits may also be under selection from abiotic agents(e.g.rain),which makes considering the relative importance of pollinators and abiotic selective agents on floral traits essential.The functional significance of floral orien-tation is often ascribed to pollinator attraction,but orientation can also protect reproductive structures from rain.Therefore,a study that incorporates both factors will enhance our understanding of the ecological roles of floral orientation in plant fitness.Mertensia brevistyla and M.fusiformis are herbaceous species that differ in their floral orientations.A series of field and laboratory experi-ments was used to investigate the adaptive function of floral orien-tation in these species,particularly with respect to pollinators and rain.Methods We measured and compared floral orientation and visitor assem-blages between M.brevistyla and M.fusiformis populations in west-ern Colorado,USA.We manipulated floral stems and conducted a choice experiment with floral visitors,and also compared orienta-tions of pollinator-visited stems with those of unvisited stems in a natural setting.We examined pollinator-and rain-mediated selec-tion on floral orientation by manipulating orientation,conducting supplemental pollinations,applying watering treatments and meas-uring subsequent seed set.We also experimentally tested the like-lihood of rain contact with anthers,and the effect of rainwater on pollen germinability.Important Findings Mertensia brevistyla had a significantly more upright floral orienta-tion than M.fusiformis,and seed set was highest in upright M.bre-vistyla and in horizontal/pendant M.fusiformis stems,supporting an adaptive function(via female fitness)of the interspecific difference in orientation.However,floral visitor assemblages did not differ significantly between the two species;visitors did not exhibit sig-nificant preference for either orientation;and pollinator-mediated selection on orientation was undetectable.Similarly,there was lit-tle effect of water on seed set in either species,regardless of floral orientation.However,pollen germinability was reduced in both species by immersion in water;and water was more likely to contact anthers in M.fusiformis than in M.brevistyla,due to interspecific differences in floral morphology.We conclude that pollinators are likely not the primary selective agent driving differences in orienta-tion in these Mertensia species.Instead,the negative effect of rain on pollen germinability helps explain the more pendant orientation of M.fusiformis,while short anthers in more upright M.brevistyla provide an alternative adaptation to rain.The selective agent driving effects of orientation on seed set remains unclear.This study illus-trates the necessity of considering male fitness and abiotic agents in interpreting the functional significance of inflorescence traits. | Shang-Yao Lin Jessica R.K.Forrest | 2019 | Journal of Plant Ecology2019,12,1: | 1 |
