1Washed microbiota transplantation vs.manual fecal microbiota transplantation:clinical findings,animal studies and in vitro screening显示文摘Fecal microbiota transplantation(FMT)by manual preparation has been applied to treat diseases for thousands of years.However,this method still endures safety risks and challenges the psychological endurance and acceptance of doctors,patients and donors.Population evidence showed the washed microbiota preparation with microfiltration based on an automatic purification system followed by repeated centrifugation plus suspension for three times significantly reduced FMT-related adverse events.This washing preparation makes delivering a precise dose of the enriched microbiota feasible,instead of using the weight of stool.Intraperitoneal injection in mice with the fecal microbiota supernatant obtained after repeated centrifugation plus suspension for three times induced less toxic reaction than that by the first centrifugation following the microfiltration.The toxic reactions that include death,the change in the level of peripheral white blood cells,and the proliferation of germinal center in secondary lymphoid follicles in spleen were noted.The metagenomic next-generation sequencing(NGS)indicated the increasing types and amount of viruses could be washed out during the washing process.Metabolomics analysis indicated metabolites with pro-inflammatory effects in the fecal microbiota supernatant such as leukotriene B4,corticosterone,and prostaglandin G2 could be removed by repeated washing.Near-infrared absorption spectroscopy could be served as a rapid detection method to control the quality of the washingprocess.In conclusion,this study for the first time provides evidence linking clinical findings and animal experiments to support that washed microbiota transplantation(WMT)is safer,more precise and more quality-controllable than the crude FMT by manual.Ting Zhang Gaochen Lu Zhe Zhao Yafei Liu Quan Shen Pan Li Yaoyao Chen Haoran Yin Huiquan Wang Cicilia Marcella Bota Cui Lei Cheng Guozhong Ji Faming Zhang 2020Protein & Cell2020,11,4:44
2Evolution of dielectric loss-dominated electromagnetic patterns in magnetic absorbers for enhanced microwave absorption performances显示文摘As the growing criterion of electromagnetic wave (EMW) absorption materials, micro/nano-scale magnetic materials are drawing more and more attention for their unique features compared to bulky absorbers. Generally, the complex permeability of micro/nano-scale magnetic absorbers varies in a relatively narrow range, whatever for the storage of magnetic energy or the dissipation of magnetic energy. If so, how the small variation of permeability affects the ultimate performances is still unclear. Here, a strategy of electromagnetic parameters regulation for the magnetic materials is applied to understand the loss contribution in micro/nanoscale magnetic absorbers. After analyzing the evolution of electromagnetic maps of ten ferrosoferric oxide samples, it can be found that the dissipation contribution of permeability for magnetic materials is weaker than that of permittivity, in spite of its significant role in determining the impedance matching characteristics. 3High-efficiency second-order nonlinear processes in an optical microfibre assisted by few-layer GaSe显示文摘The centrosymmetric nature of silica fibre precludes the realisation of second-order nonlinear processes in optical fibre systems.Recently,the integration of 2D materials with optical fibres has opened up a great opportunity to develop allfibre active devices.Here,we demonstrate high-efficiency second-order nonlinear frequency conversions in an optical microfibre assisted with few-layer gallium selenide(GaSe)nanoflakes.Attributed to the strong evanescent field of the microfibre and ultrahigh second-order nonlinearity of the GaSe nanoflakes,second harmonic generation(SHG)and sum-frequency generation(SFG)are effectively achieved with only sub-milliwatt continuous-wave(CW)lasers in the wavelength range of 1500–1620 nm,covering the C and L telecom bands.The SHG intensity from the microfibre is enhanced by more than four orders of magnitude with the assistance of the GaSe nanoflakes on fibre nonlinear processes.Moreover,in the SFG process,the intensity transfer between different frequencies can be effectively manipulated by changing the wavelengths and powers of two pump lasers.The realised strong second-order nonlinearity in the GaSe-integrated microfibre might expand the applications of all-fibre devices in all-optical signal processing and new light source generation at awkward wavelengths.Biqiang Jiang Zhen Hao Yafei Ji Yueguo Hou Ruixuan Yi Dong Mao Xuetao Gan Jianlin Zhao 2020Light(Science & Applications)2020,9,1:4

7The histone methyltransferase SUVR2 promotes DSB repair via chromatin remodeling and liquidliquid phase separation显示文摘Maintaining genomic integrity and stability is particularly important for stem cells,which are at the top of the cell lineage origin.Here,we discovered that the plant-specific histone methyltransferase SUVR2 maintains the genome integrity of the root tip stem cells through chromatin remodeling and liquid-liquid phase separation(LLPS)when facing DNA double-strand breaks(DSBs).The histone methyltransferase SUVR2(MtSUVR2)has histone methyltransferase activity and catalyzes the conversion of histone H3 lysine 9 monomethylation(H3K9me1)to H3K9me2/3 in vitro and in Medicago truncatula.Under DNA damage,the proportion of heterochromatin decreased and the level of DSB damage marker y-H2AX increased in suvr2 mutants,indicating that MtSUVR2 promotes the compaction of the chromatin structure through H3K9 methylation modification to protect DNA from damage.Interestingly,MtSUVR2 was induced by DSBs to phase separate and form droplets to localize at the damage sites,and this was confirmed by immunofluorescence and fluorescence recovery after photobleaching experiments.The IDR1 and lowcomplexity domain regions of MtSUVR2 determined its phase separation in the nucleus,whereas the IDR2 region determined the interaction with the homologous recombinase MtRAD51.Furthermore,we found that MtSUVR2 drove the phase separation of MtRAD51 to form'DNA repair bodies,'which could enhance the stability of MtRAD51 proteins to facilitate error-free homologous recombination repair of stem cells.Taken together,our study reveals that chromatin remodeling-associated proteins participate in DNA repair through LLPS.Qianwen Liu Peng Liu Tuo Ji Lihua Zheng Chen Shen Shasha Ran Jinling Liu Yafei Zhao Yiding Niu Tao Wang Jiangli Dong 2022Molecular Plant2022,15,7:1

11Physical Activity,Sedentary Behavior,and the Risk of Cardiovascular Disease in Type 2 Diabetes Mellitus Patients:The MIDiab Study显示文摘The aim of this study was to explore the associations of moderate-to-vigorous-intensity physical activity(MVPA)time and sedentary(SED)time with a history of cardiovascular disease(CVD)and multifactorial(i.e.,blood pressure(BP),body mass index(BMI),low-density lipoprotein cholesterol(LDL-C),and glycated hemoglobin A1c(HbA1c))control status among type 2 diabetes mellitus(T2DM)patients in China.A cross-sectional analysis of 9152 people with type 2 diabetes from the Multifactorial Intervention on Type 2 Diabetes(MIDiab)study was performed.Patients were grouped according to their self-reported MVPA time(low,<150 min·week−1;moderate,150 to<450 min·week−1;high,≥450 min·week−1)and SED time(low,<4 h·d–1;moderate,4 to<8 h·d–1;high,≥8 h·d–1).Participants who self-reported a history of CVD were identified as having a CVD risk.Odds ratios(ORs)and 95%confidence intervals(CIs)of CVD risk and multifactorial control status associated with MVPA time and SED time were estimated using mixed-effect logistic regression models,adjusting for China’s geographical region characteristics.The participants had a mean±standard deviation(SD)age of(60.87±8.44)years,44.5%were women,and 25.1%had CVD.After adjustment for potential confounding factors,an inverse association between high MVPA time and CVD risk that was independent of SED time was found,whereas this association was not observed in the moderate-MVPA group.A higher MVPA time was more likely to have a positive effect on the control of BMI.Compared with the reference group(i.e.,those with MVPA time≥450 min·week−1 and SED time<4 h·d–1),CVD risk was higher in the low-MVPA group:The OR associated with an SED time<4 h·d–1 was 1.270(95%CI,1.040–1.553)and that associated with an SED time≥8 h·d–1 was 1.499(95%CI,1.149–1.955).We found that a high MVPA time(i.e.,≥450 min·week−1)was associated with lower odds of CVD risk regardless of SED time among patients with T2DM.Yafei Wu Guijun Qin Guixia Wang Libin Liu Bing Chen Qingbo Guan Zhongshang Yuan Xu Hou Ling Gao Chao Xu Haiqing Zhang Xu Zhang Qiu Li Yongfeng Song Fei Jing Shizhan Ma Shanshan Shao Meng Zhao Qingling Guo Nanwei Tong Hongyan Zhao Xiaomin Xie Chao Liu Zhongyan Shan Zhifeng Cheng Xuefeng Yu Shulin Chen Tao Yang Yangang Wang Dongmei Li Zhaoli Yan Lixin Guo Qiuhe Ji Wenjuan Wang Jiajun Zhao 2023Engineering2023,,1:1
12Realizing the potential of exploiting human IPSCs and their derivatives in research of Down syndrome显示文摘Down syndrome(DS)is a genetic condition characterized by intellectual disability,delayed brain development,and early onset Alzheimer’s disease.The use of primary neural cells and tissues is important for understanding this disease,but there are ethical and practical issues,including availability from patients and experimental manipulability.Moreover,there are significant genetic and physiological differences between animal models and humans,which limits the translation of the findings in animal studies to humans.Advancements in induced pluripotent stem cells(iPSC)technology have revolutionized DS research by providing a valuable tool for studying the cellular and molecular pathologies associated with DS.Induced pluripotent stem cells derived from cells obtained from DS patients contain the patient’s entire genome including trisomy 21.Trisomic iPSCs as well as their derived cells or organoids can be useful for disease modeling,investigating the molecular mechanisms,and developing potential strategies for treating or alleviating DS.In this review,we focus on the use of iPSCs and their derivatives obtained from DS individuals and healthy humans for DS research.We summarize the findings from the past decade of DS studies using iPSCs and their derivatives.We also discuss studies using iPSC technology to investigate DS-associated genes(e.g.,APP,OLIG1,OLIG2,RUNX1,and DYRK1A)and abnormal phenotypes(e.g.,dysregulated mitochondria and leukemia risk).Lastly,we review the different strategies for mitigating the limitations of iPSCs and their derivatives,for alleviating the phenotypes,and for developing therapies.YAFEI WANG JIELEI NI YUHAN LIU DINGYING LIAO QIANWEN ZHOU XIAOYANG JI GANG NIU YANXIANG NI 2023BIOCELL2023,47,12:0
13Analytical transient phase change heat transfer model of wearable electronics with a thermal protection substrate显示文摘As thermal protection substrates for wearable electronics,functional soft composites made of polymer materials embedded with phase change materials and metal layers demonstrate unique capabilities for the thermal protection of human skin.Here,we develop an analytical transient phase change heat transfer model to investigate the thermal performance of a wearable electronic device with a thermal protection substrate.The model is validated by experiments and the finite element analysis(FEA).The effects of the substrate structure size and heat source power input on the temperature management efficiency are investigated systematically and comprehensively.The results show that the objective of thermal management for wearable electronics is achieved by the following thermal protection mechanism.The metal thin film helps to dissipate heat along the in-plane direction by reconfiguring the direction of heat flow,while the phase change material assimilates excessive heat.These results will not only promote the fundamental understanding of the thermal properties of wearable electronics incorporating thermal protection substrates,but also facilitate the rational design of thermal protection substrates for wearable electronics.Yingli SHI Junyun JI Yafei YIN Yuhang LI Yufeng XING 2020Applied Mathematics and Mechanics(English Edition)2020,41,11:0

15Metal-free directed C-H borylation of 2-(N-methylanilino)-5-fluoropyridines and 2-benzyl-5-fluoropyridines显示文摘A novel method for metal-free C-H borylation of 2-(N-methylanilino)-5-fluoropyridines and 2-benzyl-5-fluoropyridines has been reported. The 5-fluoropyridine directed borylation reaction exhibited high efficiency and site exclusivity. The useful protocol could be executed on a gram-scale easily and the borylated products showed good derivatization applications.
