1The adult human testis transcriptional cell atlas显示文摘Human adult spermatogenesis balances spermatogonial stem cell (SSC) self-renewal and differentiation,alongside complex germ cell-niche interactions,to ensure long-term fertility and faithful genome propagation. Here,we performed single-cell RNA sequencing of -6500 testicular cells from young adults. We found five niche/somatic cell types (Leydig,myoid,Sertoli,endothelial,macrophage),and observed germline-niche interactions and key human-mouse differences. Spermatogenesis,including meiosis,was reconstructed computationally,revealing sequential coding,non-coding,and repeat-element transcriptional signatures. Interestingly,we identified five discrete transcriptional/developmental spermatogonial states,including a novel early SSC state,termed State 0. Epigenetic features and nascent transcription analyses suggested developmental plasticity within spermatogonial States. To understand the or丨gin of State 0,we profiled testicular cells from infants,and identified distinct similarities between adult State 0 and infant SSCs. Overall,our datasets describe key transcriptional and epigenetic signatures of the normal adult human testis,and provide new insights into germ cell developmental transitions and plasticity.Jingtao Guo Edward J. 2Synthesis and electrochemical performance of YF_3-coated LiMn_2O_4 cathode materials for Li-ion batteries显示文摘Spinel LiMn2O4 cathodes were coated with 1 mol% YF3.X-ray diffraction(XRD) analyses showed that Y and/or F did not enter the lattice of the LiMn2O4 crystal.Transmission electron microscopy(TEM) showed that a compact YF3 layer of 5-20 nm in thickness was coated onto the surface of LiMn2O4 particles.Scanning electron microscopy(SEM) observation showed that the YF3 coating caused the agglomeration of LiMn2O4 particles.The cycling test demonstrated that the YF3 coating can improve the electrochemical performance of LiMn2O4 at both 20 and 55°C.Moreover,YF3-coated LiMn2O4 exhibited an improved rate capability compared with the uncoated one at high rates over 5C.The immersion test in electrolytes showed that YF3-coated LiMn2O4 is more erosion resistant than the uncoated one.CAO Jina CAO Gaoshao YU Hongming XIE Jian ZHAO Xinbing 2011Rare Metals2011,30,1:7
3Properties of sodium silicate bonded sand hardened by microwave heating显示文摘The sodium silicate bonded sand hardened by microwave heating has many advantages,such as low sodium silicate adding quantity,fast hardening speed,high room temperature strength,good collapsibility and certain surface stability. However,it has big moisture absorbability in the air,which would lead to the compression strength and the surface stability of the sand molds being sharply reduced. In this study,the moisture absorbability of the sodium silicate bonded sand hardened by microwave heating in different humidity conditions and the effect factors were investigated. Meanwhile,the reasons for the big moisture absorbability of the sand were analyzed. Some measures to overcome the problems of high moisture absorbability,bad surface stability and sharply reducing strength in the air were discussed. 5Inflammation-induced inhibition of chaperone-mediated autophagy maintains the immunosuppressive function of murine mesenchymal stromal cells显示文摘Macroautophagy has been implicated in modulating the therapeutic function of mesenchymal stromal cells(MSCs).However,the biological function of chaperone-mediated autophagy(CMA)in MSCs remains elusive.Here,we found that CMA was inhibited in MSCs in response to the proinflammatory cytokines interferon-γ(IFN-γ)and tumor necrosis factor-α(TNF-α).In addition,suppression of CMA by knocking down the CMA-related lysosomal receptor lysosomal-associated membrane protein 2(LAMP-2A)in MSCs significantly enhanced the immunosuppressive effect of MSCs on T cell proliferation,and as expected,LAMP-2A overexpression in MSCs exerted the opposite effect on T cell proliferation.This effect of CMA on the immunosuppressive function of MSCs was attributed to its negative regulation of the expression of chemokine C-X-C motif ligand 10(CXCL10),which recruits inflammatory cells,especially T cells,to MSCs,and inducible nitric oxide synthase(iNOS),which leads to the subsequent inhibition of T cell proliferation via nitric oxide(NO).Mechanistically,CMA inhibition dramatically promoted IFN-γplus TNF-α-induced activation of NF-κB and STAT1,leading to the enhanced expression of CXCL10 and iNOS in MSCs.Furthermore,we found that IFN-γplus TNF-α-induced AKT activation contributed to CMA inhibition in MSCs.More interestingly,CMA-deficient MSCs exhibited improved therapeutic efficacy in inflammatory liver injury.Taken together,our findings established CMA inhibition as a critical contributor to the immunosuppressive function of MSCs induced by inflammatory cytokines nd highlighted a previously unknown function of CMA.Jie Zhang Jiefang Huang Yuting Gu Mingxing Xue Fengtao Qian Bei Wang Wanlin Yang Hongshuang Yu Qiwei Wang Xin Guo Xinyuan Ding Jina Wang Min Jin Yanyun Zhang 2021Cellular & Molecular Immunology2021,18,6:3
6Dual-peptide-modified alginate hydrogels for the promotion of angiogenesis显示文摘The ability to supply suitable blood vessel system is a major challenge for artificial thick tissue engineering. Angiogenesis is a key point during the process of microvascular formation. Many bioactive molecules such as extra cellular matrix(ECM) proteins and adhesion peptides derived from the ECM are applied to promote angiogenesis. In this work, two adhesion peptides, YIGSR and REDV, were selected to modify sodium alginate(ALG) to obtain YIGSR- and REDV-alginate conjugates(ALG-YIGSR, and ALG-REDV, respectively). We mixed the two peptide-conjugates together in a series of concentration ratios to prepare bioactive surfaces for in vitro studies and hydrogel scaffolds for in vivo studies. In vitro studies showed that surfaces modified with 1.09 pmol/mm2 peptide had the best affinity to human umbilical vein endothelial cells(HUVECs) than that with high or low concentrations of peptides. In addition, surfaces modified with dual peptides could significantly promote HUVECs proliferation, where ALG-YIGSR:ALG-REDV at a mole ratio of 5:1 exhibited the best enhancement ability. Furthermore, the in vivo angiogenesis results demonstrated that hydrogel scaffolds composed of mixed ALG-YIGSR and ALG-REDV at the 5:1 ratio had angiogenic induction potential by stimulating new blood vessel formation, and showed higher blood vessel density than scaffolds composed of a single peptide. 7An experimental study of premixed laminar methane/oxygen/argon flames doped with hydrogen at low pressure with synchrotron photoionization显示文摘Laminar premixed stoichiometric methane/hydrogen/oxygen/argon flames were investigated with tun- able synchrotron vacuum ultraviolet (VUV) photoionization and molecular-beam sampling mass spec- trometry techniques. The methane/hydrogen fuel blends with hydrogen volumetric fraction of 0, 20%, 40%, 60% and 80% were studied. All observed flame species, including stable intermediates and radi- cals in the flames, were detected by measuring photoionization mass spectra and photoionization effi- ciency (PIE) spectra. Mole fraction profiles of major species and intermediates were derived by scan- ning burner at some selected photon energies near ionization thresholds. The influence of hydrogen addition on mole fraction of major species and intermediates was analyzed. The results show that the major species mole fraction of CO, CO2 and CH4 decreases with the increase of hydrogen fraction. The mole fraction of intermediates measured in this experiment decreases remarkably with the increase of hydrogen fraction. This would be due to the increase of H and OH radicals by hydrogen addition and the high diffusivity and activity of H radical promoting the chemical reaction. 9Engineering a cell-penetrating hyperstable antibody scFv(Ras)-An extraordinary approach to cancer therapeutics显示文摘In the modern pharmaceutical industry,monoclonal antibodies are often used as therapeutic agents.However,they are restricted to cell surface antigens due to their inability to penetrate the outer cell membrane and maintain normal function in the reducing environment.Additionally,it can lead to cytotoxicity since it attacks cancerous cells by mimicking the human immune system.As an alternative,this study modifies the hyperstable single-chain fragment variable(scFv)antibody to eliminate cancer using its linear shape.The scFv(F8)antibody model was modified to recognize human Ras protein by altering residues in the antigen-binding site.Furthermore,a cell-penetrating peptide(CPP)was attached to the scFv(Ras)antibody model to allow entrance to the cell,creating CPP-scFv(Ras).Sodium dodecyl sulfate polyacrylamide gel electrophoresis(SDS-PAGE),western blotting,and the binding assay were performed to prove its effectiveness.As a result,CPP-scFv(Ras)was successfully engineered and bound to the antigen,HRas(G12V).Jina Bae Yoonyee Song 2021Synthetic and Systems Biotechnology2021,6,4:2

12Effect of K-doping on the Electrochemical Performance of Ca_3Co_4O_9 Anode for Li-ion Batteries显示文摘Ca3Co4O9 (CCO) and Ca2.95K0.05Co4O9 (CKCO) powders have been prepared by the polyacrylamide gel method. CKCO shows increased capacity and better cycling stability compared with CCO. After cycled for 50 cycles at 0.5 C, CKCO retains a capacity of 223 mAh·g-1, almost twice than CCO. 13Advances of supercritical fluid chromatography in lipid profiling显示文摘Supercritical fluid chromatography(SFC) meets with great favor due to its high efficiency, low organic solvent consumption, and the specialty for the identification of the isomeric species. This review describes the advances of SFC in targeted and untargeted lipid profiling. The advancement of the SFC instruments and the stationary phases are summarized. Typical applications of SFC to the targeted and untargeted lipid profiling are discussed in detail. Moreover, the perspectives of SFC in the lipid profiling are also proposed. 20Tracking the reactivity of ozonation towards effluent organic matters from WWTP using two-dimensional correlation spectra显示文摘The characteristics of effluent organic matter(EfOM) from a wastewater treatment plant(WWTP) during ozonation were investigated using excitation and emission matrix(EEM)spectra, Fourier transform infrared spectroscopy(FT-IR) and high-performance size exclusion chromatography(HPSEC) at different ozone dosages. The selectivity of ozonation towards different constituents and functional groups was analysed using two-dimensional correlation spectra(2D-COS) probed by FT-IR, synchronous fluorescence spectra and HPSEC.The results indicated that ozonation can destroy aromatic structures of EfOM and change its molecular weight distribution(MWD). According to 2D-COS analysis, microbial humiclike substances were preferentially removed, and then the protein-like fractions. Terrestrial humic-like components exhibited inactivity towards ozonation compared with the above two fractions. Protein-like substances with small molecular weight were preferentially reacted during ozonation based on 2D-COS probed by HPSEC. In addition, the selectivity of ozone towards different functional groups of EfOM exhibited the following sequence:phenolic and alcoholic C\O groups > aromatic structures containing C_C double bonds >aliphatic C\H.
