Tumour invasion and metastasis is the biggest impediment to gastric carcinoma cure. Active migration of tumour cells is now considered as the pivotal step in cancer invasion and metastasis. RhoC is a member of the Ras-superfamily of small guanosine triphosphatases that can regulate many cellular functions, especially cytoskeletal organization and cell locomotion. | 4 | Expression of tumor related genes NGX6,NAG-7,BRD7 in gastric and colorectal cancer显示文摘AIM: NGX6, NAG-7 and BRD7 genes are tumor related genes, which have been newly cloned by positional candidate cloning strategy. This study was designed to investigate the expression levels of NGX6, NAG-7 and BRD7 genes in human gastric and colorectal cancer tissues, and their corresponding normal tissues, and to investigate whether these genes play a role in the pathogenesis of gastric and colorectal cancers.METHODS: Reverse transcription-polymerase chain reaction (RT-PCR), dot hybridization and Northern blot analysis were used to compare the expression levels of NGX6, NAG-7 and BRD7 genes in 34 gastric cancer tissues and 34 colorectal cancer tissues with their corresponding normal tissues of the same patients, respectively.RESULTS: Among the 34 colorectal cancer specimens and the 34 gastric cancer specimens, the expression of NGX6 in 25 colorectal cancer tissues was absent or very weak (73.5 %) by RT-PCR analysis. The down-regulation rate of NGX6 in colorectal cancer tissues was significantly higher than that in corresponding normal tissues (26.5 %,9/34)(P<0.005). Moreover, the down-regulation of NGX6 was significantly correlated with lymph node and/or distance metastases. Patients with lymph node and/or distance metastasis had much higher down-regulation rate of NGX6 than patients without metastases (93.8 % vs 55.6 %, P<0.05).However no correlation was found between the expression of NGX6 and pathologic type of colorectal cancer in this study, and also the expression of NGX6 did not display any difference between gastric cancer and corresponding normal tissues (58.8 % vs 70.6 %, P>0.25). Dot hybridization and Northern blot analysis confirmed the results of RT-PCR.Furthermore, NAG-7 and BRD7 mRNA was not up- or downregulated in gastric and colorectal cancers compared with their corresponding normal tissues in our study.CONCLUSION: The down-regulation of NGX6 may be closely associated with tumorigenesis and metastasis of colorectal carcinoma. However, it may not contribute to the development and progression of gastric carcinoma. In addition, the expression levels of NAG-7, and BRD7 did not alter in gastric and colorectal cancers. | 5 | Sensitivity-enhanced surface plasmon resonance sensor utilizing a tungsten disulfide (WS_2) nanosheets overlayer显示文摘Tungsten disulfide(WS_2), as a representative layered transition metal dichalcogenide(TMDC) material, possesses important potential for applications in highly sensitive sensors. Here, a sensitivity-enhanced surface plasmon resonance(SPR) sensor with a metal film modified by an overlayer of WS_2 nanosheets is proposed and demonstrated. The SPR sensitivity is related to the thickness of the WS_2 overlayer, which can be tailored by coating a WS_2 ethanol suspension with different concentrations or by the number of times of repeated post-coating.Benefitting from its large surface area, high refractive index, and unique optoelectronic properties, the WS_2 nanosheet overlayer coated on the gold film significantly improves the sensing sensitivity. The highest sensitivity(up to 2459.3 nm∕RIU) in the experiment is achieved by coating the WS_2 suspension once. Compared to the case without a WS_2 overlayer, this result shows a sensitivity enhancement of 26.6%. The influence of the WS_2 nanosheet overlayer on the sensing performance improvement is analyzed and discussed. Moreover, the proposed WS_2 SPR sensor has a linear correlation coefficient of 99.76% in refractive index range of 1.333 to 1.360. | 8 | Construction of phospholamban antisense RNA recombinant adenoassociated virus vector and its effects in rat cardiomyocytes显示文摘Aim: To construct a recombinant adeno-associated virus (rAAV) vector containing gene encoding phospholamban antisense RNA (asPLB), and analyse its effect on expression of PLB, expression and activity of sarco-endoplasmic reticulum Ca^2+-ATPase (SERCA), and the change of intracellular free Ca^2+ concentration ([Ca^2+]i) in rat cardiomyocytes. Methods: The target gene encoding PLB antisense RNA was inserted inversely into the adeno-associated virus plasmid pAAV-MCS digested by corresponding restricted endonuclease enzyme. The recombinant plasmid and pAAV-RC and pHelper were co-transfected into 293 cell. At the same time, a viral production positive control (rAAV-LacZ) and negative control were performed. The recombinant viruses were used to transfect the cultured rat cardiomyocytes. Site β-Galactosidase staining were performed to observe the transfer efficiency. Reverse transcription-PCR and Western blot were used to determine the mRNA and protein expression of PLB and SERCA. The activity of SERCA and the [Ca^2+]i were measured. Results: The rAAV vectors were constructed successfully and were transfected into rat cardiomyocytes effectively.The PLB mRNA and protein expression were reduced in rat cardiomyocytes transfected by rAAV-asPLB compared with controls. The activity of SERCA was increased. In rest state, the level of [Ca^2+]i in the rAAV-asPLB transfected groupdecreased. Its optical properties were analyzed and it was introduced into a dye-sensitized solar cell (DSSC). As a luminescence medium, Gd_2O_3 :Eu^(3+) improved light harvesting via conversion luminescence and increased the photocurrent of the DSSC. As a p-type dopant insulating rare earth oxides form an energy barrier, and the Gd_2O_3 :Eu^(3+) elevated the energy level of the oxide film and increased the photovoltage. | 10 | Reconstructing the extraordinary palaeoflood events during 3200-2800 a BP in the upper reaches of Hanjiang River Valley, China显示文摘Palaeoflood hydrology study is a leading subject in global change study. Through field investigation in the upper reaches of Hanjiang River, palaeoflood slackwater deposits(SWD) were found in the bedrock gorges of the Yunxi reach. The results of field observation, laboratory analysis including particle-size distribution and magnetic susceptibility, and comparison with modern flood deposits, the SWD were identified as the most typical Holocene palaeoflood deposits of the Hanjiang River. By using stratigraphic correlation and OSL dating method, the palaeoflood event was dated to be 3200–2800 a BP at the turn from the middle to late Holocene. According to the palaeoflood peak stage and hydraulic parameters, the peak discharges of the palaeoflood were reconstructed as 48,830–51,710 m3/s by using the slope-area method. At same time, the flood peak discharges of 1983, 2005 and 2010 severe floods were reconstructed with the same method and hydraulic parameters in the same cross section. The error between the reconstructed and gauged discharges was 1.99%–4.21%. This showed that the reconstructed palaeoflood peak discharges were reliable. The flood peak discharge-frequency relationship at 10,000-year time scale was established by a combination of the gauged flood, historical flood and palaeoflood hydrological data. All the ceramic samples showed single perovskite phase except the sam- ple with x = 0.1. The effects of gallium doping on microstructure, ferromagnetic, ferroelectric, dielectric properties and conductivity were investigated. The results indicated that Ga-dopant could improve the sintering ability of the solid solution when the Ga content x was below 0.05. When x was over 0.05, the sintering ability of the solid solution was weakened, and the phase structure of 0.7BiFeO3-0.3BaTiO3 solid solution changed from rhombohedral phase into tetragonal ferroelectric phase. The electrical resistivity increased with the Ga content increasing. Both ferroelectricity and ferromagnetism were observed in all the ceramic samples. Based on the experience of HIT Hand and DLR Hand II, a smaller and easier manufactured dexterous robot hand with multisensory function and high integration is jointly developed. The prototype of the hand is successfully built. It has 4 fingers in total 13-DOFs (degree of freedom). Each finger has 3-DOFs and 4 joints, the last 2 joints are mechanically coupled by means of four-bar linkage mechanism. It also has an additional DOF to realize motion of the thumb relative to the palm. The fingertip force can reach up to 10 N. Full integration of mechanical body, actuation system, multisensory system and electronics is a significant feature. | 17 | Geochemical characterization of loess-paleosol sequences:Comparison between the upper reaches of the Hanjiang and Weihe river valleys,China显示文摘This paper aims to compare the geochemical characteristics of loess-paleosol sequences in the upper reaches of the Hanjiang and Weihe river valleyswhich are located in the semi-humid temperate zone and humid subtropical zonerespectively. The Mituosi(MTS) profile in the upper reaches of the Hanjiang River valley and the Yaohecun(YHC) profile in the Weihe River valley were selected for this comparative research. The stratigraphic characteristicscompositionchemical weathering intensityleaching rates of Ca and Namobility of major elementsand transport features of Na and Fe were analyzed with respect to depth and compared between the two profiles. This study reached the following conclusions.(1) The composition of the loess-paleosol sequences in two regions are quite similar to the average composition of the upper continental crust(UCC)indicating that the loess in the two regions came from multiple sources and was mixed well. Thereforethe loess in the two regions is considered aeolian loess.(2) Compared with the loess-paleosol sequence in the Weihe River valleythe loess-paleosol sequence in the upper reaches of the Hanjiang River valley features a darker color; a higher chemical index of alteration(CIA) value; higher leaching rates of Na and Ca; higher migration ratio(relative to K) of AlSiMgand Na; and lower migration ratio of Fe and Ca. This evidence indicates that the loess-paleosol sequence in the humid subtropical environment experienced stronger chemical weathering intensity than the loess-paleosol sequence in the semi-humid temperate zone.(3) Both the YHC profile and MTS profile record a period of climate deterioration at 6000–5000 a BP.
