1T cells expressing CD5/CD7 bispecific chimeric antigen receptors with fully human heavy-chain-only domains mitigate tumor antigen escape显示文摘Bispecific chimeric antigen receptor T-cell(CAR-T)therapies have shown promising results in clinical trials for advanced B-cell malignancies.However,it is challenging to broaden the success of bispecific CAR-T therapies to treat refractory/relapse(r/r)T-cell leukemia/lymphoma because targeting multiple T-cell-expressing antigens leads to exacerbated CAR-T cell fratricide and potential safety concerns.Fully human heavy chain variable(FHVH)antibodies that specifically target CD5 or CD7 were screened and constructed to CD5/CD7 bispecific CARs.A truncated Epidermal growth factor receptor were integrated into CAR constructs to address safety concerns.To tackle the fratricidal issue of CAR-T cells targeting T-cell-pan marker(s),CRISPR/Cas9-based CD5 and CD7 genes knockout were performed before lentiviral transduction of bispecific CARs.Functional comparison between different bispecific CAR structures:tandem CARs and dual CAR were performed in vitro and in vivo to determine the optimal construct suitable for addressing T-cell malignancy antigen escape in clinical setting.Knockout of CD5 and CD7 prevents fratricide of CD5/CD7 bispecific CAR-T cells,and FHVH-derived CD5/CD7 bispecific CAR-T cells demonstrate potent antitumor activity in vitro and in vivo.The fratricide-resistant FHVH-derived CD5/CD7 bispecific CAR-T cells have potent antitumor activity against T-cell malignancies,and tandem CARs are more effective than dual CAR in preventing tumor escape in heterogeneous leukemic cells.The meaningful clinical efficacy and safety of tandem CD5/CD7 CAR-T cells deserve to be explored urgently.Zhenyu Dai Wei Mu Ya Zhao Jiali Cheng Haolong Lin Kedong Ouyang Xiangyin Jia Jianwei Liu Qiaoe Wei Meng Wang Chaohong Liu Taochao Tan Jianfeng Zhou 2022Signal Transduction and Targeted Therapy2022,7,4:3

5Targeted core-shell nanoparticles for precise CTCF gene insert in treatment of metastatic breast cancer显示文摘Clustered regularly interspaced short palindromic repeats(CRISPR)technology emerges a remarkable potential for cure of refractory cancer like metastatic breast cancer.However,how to efficiently deliver the CRISPR system with non-viral carrier remains a major issue to be solved.Here,we report a kind of targeted core-shell nanoparticles(NPs)carrying dual plasmids(pHR-pCas9)for precise CCCTC-binding factor(CTCF)gene insert to circumvent metastatic breast cancer.The targeted core-shell NPs carrying pHR-pCas9 can accomplishγGTP-mediated cellular uptake and endosomal escape,facilitate the precise insert and stable expression of CTCF gene,inhibit the migration,metastasis,and colonization of metastatic breast cancer cells.Besides,the finding further reveals that the inhibitory mechanism of metastasis could be associated with up-regulating CTCF protein,followed by down-regulating stomatin(STOM)protein.The study offers a universal nanostrategy enabling the robust non-viral delivery of gene-editing system for treatment of severe illness.Jialun Duan Chunjie Bao Ying Xie Haitao Guo Yixuan Liu Jianwei Li Rui Liu Peishan Li Jing Bai Yan Yan Limin Mu Xueqi Li Guiling Wang Wanliang Lu 2022Bioactive Materials2022,7,5:1

7Sensitivity assessment of Phytophthora nicotianae to fungicides of fluazinam and its mixture with carboxylic acid amide显示文摘Tobacco black shank caused by Phytophthora parasitica var.nicotianae(Breda de Hean)is one of the most serious diseases in tobacco growing,and few effective fungicides are available currently.The purpose of this study was to assess the feasibility of using fluazinam to treat tobacco black shank disease.In vitro toxicities of fluazinam to 62 isolates of P.nicotianae were determined by mycelial growth rate method.The EC50 values ranged from 0.294 6 to 2.389 9 μg/mL with a mean of 1.084 0 μg/mL.The sensitivity distribution was unimodal,and there was no resistant isolate in P.nicotianae population.The carboxylic acid amide(CAA)fungicides were single-site in action and activity to oomycetes.The synergistic interactions were tested by Wadley method between fluazinam and dimethomorph,flumorph,mandipropamid,respectively.The mixture containing dimethomoph:Huazinam at 1:2 ratio showed additive actions,with the synergy ratio of 1.2.The mixture containing flumorph and fluazinam at 1:5 ratio had the highest synergy ratio(SR 2.07).Mandipropamid and fluazinam had an additive interaction and the proportion of 2:1 had the lowest EC50 value(0.020 7 μg/mL).This study demonstrated the sensitivity of P.nicotianae to fluazinam,and provided the experimental foundation to use fluazinam for tobacco black shank management.MU Wenjun LI Qi CHEN Xiuhua LI Haijiang DONG Kunle WANG Jianwei ZHANG Youwu FENG Xiaohu ZHANG Zhigao WANG Libing SONG Jizhen 2018烟草科技2018,51,A01:1

10Wide measurement range and high sensitivity spongy MWCNT/polydimethylsiloxane pressure sensor based on a single-electrode enhanced triboelectric nanogenerator显示文摘Flexible pressure sensors have broad application prospects,such as human motion monitoring and personalized recognition.However,their applicability is limited by complex structures,low output performance,low sensitivity,and narrow measurement range.In this study,we report a single-electrode spongy triboelectric sensor(SSTS)mainly composed of spongy composite multi-walled carbon nanotubes/polydimethylsiloxane(MWCNT/PDMS)film and conductive fabric,which can simultaneously generate contact electrification and electrostatic induction coupling in a single-electrode contact-separation mode.The SSTS combines the triboelectric effect,properties of doping material,and spongy porous structure(soft sugar as a sacrificial template).An SSTS with an MWCNT content of 10 wt%and a porosity of 64%exhibits high sensitivity,a wide measurement range,and excellent linearity.It also displays two sensitivity regions(slopes):1.324 V/kPa from 1.5 to 28 kPa in the low-pressure range and 0.096 V/kPa from 28 to 316.5 kPa in the high-pressure range,with linearities of 0.980 and 0.979,respectively.Furthermore,the SSTS delivers a high-performance output and high stability,thus enhancing the monitoring of hand pressure changes,human movement,personalized spatial recognition,and other detection tasks.This new strategy for human motion monitoring shows great potential in the healthcare fields,sports rehabilitation,and human-computer interactions.XIE Xin HOU XiaoJuan QIAN Shuo HOU JianWei ZHANG Jie BI XiaoXue GENG WenPing MU JiLiang HE Jian CHOU XiuJian 2023Science China(Technological Sciences)2023,66,6:0
11Further ideal multipartite access structures from integer polymatroids显示文摘Ideal access structures admit ideal secret sharing schemes where the shares have the minimal size.As multipartite access structures can well mirror the real social organizations, of which the participants are partitioned into disjoint groups according to their properties, it is desirable to find expressive ideal multipartite access structures. Integer polymatroids, due to their close relationship with ideal multipartite access structures,have been shown as a powerful tool to study the ideality of some multipartite access structures. In this paper, to cater for flexible applications, we consider several ideal multipartite access structures that further extend some known results. We first explore a type of compartmented access structures with strictly lower bounds, which provide fairness among all the participant groups when recovering the secret. Then, we investigate ideal bench access structures where the participant set is divided into two parts, that is, line-up section and bench section.The participants in line-up section can delegate their capabilities to the participants in bench section in such a way that the participants in bench section can take over the role of their delegators in line-up section, which is applicable to emergency situations when there are no enough participants in line-up section for recovering the secret. 12Performance Limit Evaluation Strategy for Automated Driving Systems显示文摘Efficient detection of performance limits is critical to autonomous driving.As autonomous driving is difficult to be realized under complicated scenarios,an improved genetic algorithm-based evolution test is proposed to accelerate the evaluation of performance limits.It conducts crossover operation at all positions and mutation several times to make the high-quality chromosome exist in candidate offspring easily.Then the normal offspring is selected statistically based on the scenario com-plexity,which is designed to measure the difficulty of realizing autonomous driving through the Analytic Hierarchy Process.The benefits of modified cross/mutation operators on the improvement of scenario complexity are analyzed theoretically.Finally,the effectiveness of improved genetic algorithm-based evolution test is validated after being applied to evaluate the collision avoidance performance of an automatic parallel parking system.Feng Gao Jianwei Mu Xiangyu Han Yiheng Yang Junwu Zhou 2022Automotive Innovation2022,5,1:0
