| 2 | Danggui Buxue Tang ameliorates bleomycin-induced pulmonary fibrosis in rats through inhibiting transforming growth factor-β1/Smad3/plasminogen activator inhibitor-1 signaling pathway显示文摘OBJECTIVE: To investigate the effect of Danggui Buxue Tang(DBT), a decoction from Traditional Chinese Medicine, on bleomycin-induced pulmonary fibrosis in rats, and to propose the possible underlying mechanism.METHODS: Forty male Sprague-Dawley rats were randomly divided into sham group, model group,prednisone group and DBT group. Pulmonary fibrosis rat model was established by intratracheal injection with bleomycin. Body weight and lung index were monitored. Histopathologic examination and collagen deposition were determined using Hematoxylin and eosin(HE) and Masson's trichrome staining. Immunohistochemistry staining was applied to observe the expression of alpha-smooth muscle actin(α-SMA). m RNA expression of α-SMA,collagen Ⅰ and collagen Ⅲ were measured by realtime fluorescence quantitative PCR(RT-q PCR). Inflammatory cytokines, including tumor necrosis factor alpha(TNF-α), interleukin-6(IL-6) and IL-1β in serum were detected by Enzyme-linked immunosorbent assay. Alkali hydrolysis method was conducted to investigate the content of hydroxyproline(HYP). Transforming growth factor-β1(TGF-β1),Smad3 and plasminogen activator inhibitor-1(PAI-1) protein level were examined by Western blot assay.RESULTS: DBT significantly reduced the severity of bleomycin-induced pulmonary fibrosis and inflammation as indicated by minimizing the lost of weight, and by lowering the levels of lung index, inflammation score, Ashcroft score, collagen volume fraction(%), HYP, α-SMA, collagen Ⅰ, collagen Ⅲ,TNF-α, IL-6, IL-1β, TGF-β1, Smad3 and PAI-1, consistent with the effect of prednisone.CONCLUSION: Our findings suggest that DBT is able to ameliorate the pulmonary fibrosis, the possible mechanism may involve inhibition of pulmonary inflammation and collagen deposition, possibly via suppressing TGF-β1/Smad3/PAI-1 signaling pathway. | Wang Jiepeng Fang Chaoyi Wang Shaoxian Fang Fang Chu Xinqiao Liu Na Lu Chenxi Wang Shuo Li Wei | 2020 | Journal of Traditional Chinese Medicine2020,40,2: | 5 |
| 3 | Seismic behavior of thin-walled circular and stiffened square steel tubed-reinforced-concrete columns显示文摘Steel tubed-reinforced-concrete(TRC) columns have been gradually used in the construction of high-rise buildings recently because of their high axial load-carrying capacities and excellent seismic behavior. Existing studies about their seismic behavior were focused on columns with relatively thick tubes, i.e., diameter-to-thickness/width-to-thickness(D/t) ratios were below 100,while little is known about thin-walled TRC columns, especially for square TRC columns. Considering the infilled concrete of square TRC columns is non-uniformly and non-effectively confined, accordingly, stiffened square TRC columns are usually adopted in practice. Thus, two thin-walled circular TRC columns(D/t=120) and two stiffened square ones with diagonal stiffeners in plastic hinge regions(D/t=106) were tested under a constant axial compression combined with cyclic lateral loading.Both the circular and stiffened square TRC columns had the same cross sectional area, tube thickness, reinforcing bar ratio and column height. Flexural failure occurred for all the four specimens. Test results showed the strengths of the stiffened square TRC columns were a little higher in comparison to their circular counterparts; the ductility and energy dissipation capacities were excellent for both the stiffened and circular TRC columns, indicating very good confinement was gained from the yielded steel tubes of the plastic hinge regions at the peak loads. And shear stresses(35–90 MPa) in the sheared plates showed their moderate contribution of carrying lateral loads. | 4 | Numerical study on effects of the cofferdam area in liquefied natural gas storage tank on the leakage and diffusion characteristics of natural gas显示文摘The leakage and diffusion characteristics of natural gas were investigated in the condition of the leakage of liquefied natural gas(LNG) in the storage tank.Fluent was adopted to simulate the process in a series of three-dimension unsteady state calculations.The effects of different heights of the cofferdam(1.0 m, 2.0 m and 3.0 m),wind directions,ambient temperature,leakage location,leakage volume on the diffusion process of natural gas were investigated.The diffusion characteristics of the natural gas clouds over cofferdam were found.Under windless condition,when the gas clouds met,the gas clouds rose due to the collision,which made them easier to cross the cofferdam and spread out.The higher the ambient temperature was,the higher the gas concentration around the cofferdam was,and the smaller the gas concentration difference was.When the leakage occurred,the higher coffe rdam was more beneficial to delay the outward diffusion of gas clouds.However,when the leaka ge stopped,the higher cofferdam went against the dissipation of gas clouds.Under windy condition,the time to form stable leakage flow field was faster than that of windless,and the lower cofferdam further reduced this time.Therefore,considering the effect of barrier and dissipation,it was suggested that the rational height of cofferdam should be designed in the range of 1.0 m to 2.0 m.In case of emergency,the leakage of gas should be deduced reasonably by combining the measurement of gas concentration with the rolling of gas clouds.When windless,the leakage area should be entered between the overflows of gas clouds. | Zirong Lin Shuangfeng Wang Shuxun Fu Jiepeng Huo | 2021 | Chinese Journal of Chemical Engineering2021,34,1: | 2 |
| 5 | Preparation and toxicity evaluation of a novel nattokinase-tauroursodeoxycholate complex显示文摘Nattokinase(NK), which has been identified as a potent fibrinolytic protease, has remarkable potential in treatment of thrombolysis, and even has the ability to ameliorate chronic vein thrombosis. To reduce the hemorrhagic risk from an intravenous injection of NK,nattokinase-tauroursodeoxycholate(NK-TUDCA) complex was prepared at different pH values and with different ratios of NK and TUDCA. When assessing survival time, survival state,tail injury, and the body weight of mice, it was found that the NK-TUDCA complex(NK: 10 k IU/ml; TUDCA: 10 mg/ml; pH 5.0) had a lower toxicity when administered at an NK dosage of 130 kIU/kg in the acute toxicity test and 13 kIU/kg in the repeated low-dose challenge. From the results of the in vitro thrombolytic test and characterization of NKTUDCA, we speculated that the delayed release of NK-TUDCA might be the main cause of toxicity reduction by the complex. | 7 | Thermal expansion behaviors of Mn(Ⅱ)-pyridylbenzoate frameworks based on metal-carboxylate chains显示文摘Solvothermal reactions of MnCl2with pyridylbenzoic acids gave three-dimensional metal-carboxylate frameworks(MCFs),named MCF-34,MCF-43,and MCF-44,based on one-dimensional Mn-carboxylate chains.The crystal structure,stability,porosity,and framework flexibility of the new compound MCF-44 were studied in detail and compared with its analogs.Depending on their shapes and the bridging angles of the ligands,these compounds possess different network connectivities and porosities.Considering the pyridylbenzote ligands and Mn(II)ions as,respectively,3-and 6-connected nodes,they resemble either the anatase(ant)or rutile(rtl)polymorph of TiO2.Variable-temperature single-crystal X-ray diffraction studies revealed large thermal expansion coefficients for these compounds,which are probably related to the relatively flexible edge-sharing polyhedral structure of their Mn-carboxylate chains.Interestingly,the new compound MCF-44,with its highly porous rtl structure exhibits the largest thermal expansion coefficienct among the coordination polymers reported so far. | ZHOU HaoLong Li Mian LI Dan ZHANG JiePeng CHEN XiaoMing | 2014 | Science China Chemistry2014,57,3: | 2 |
