| 6 | Different n-6/n-3 PUFA diets with fish oil attenuated osteoarthritis in ovariectomized mice via targeting the NLRP3 inflammasome显示文摘Osteoporosis(OP)and osteoarthritis(OA)co-exist with increased inflammatory cytokines as a common feature.Modern diet containing high n-6/n-3 PUFA was suggested contributing to these chronic inflammatory diseases.Although fish oil(FO)rich in n-3 PUFAs has been confirmed beneficial for OA and bone loss in previous studies,whether small amounts of FO could alleviate osteoporotic OA under a high n-6/n-3 PUFA condition has not been reported.In this study,the osteoporotic OA mice were fed with 20:1 or 6:1 n-6/n-3 PUFA diets with or without FO for 12 weeks.The results showed that 20:1 diet with FO ameliorated systemic inflammation and cartilage degradation in model mice,compared to the non-FO 20:1 diet.Specifically,diets with FO altered the fatty acids composition with elevating n-3/n-6 PUFA in articular cartilage,thus restoring the cartilage homeostasis by inhibiting the catabolic process.Mechanistically,20:1 non-FO diet promoted the expression of NLRP3 via PGE2/EP4/NFκB.However,20:1 FO-H diet significantly inhibited the expression of NLRP3 and further exhibited a greater affinity for GPR120 than 20:1 non-FO diet to inactivate caspase-1-dependent NLRP3 inflammasome.As a consequence,20:1 FO-H diet inhibited chondrocyte pyroptosis and followed by the reduction of the two catabolic cytokines:IL-1β and IL-18.Notably,the 20:1 FO-H diet performed as well as the 6:1 non-FO diet,and 6:1 diet with FO could further improve osteoporotic OA in a dose-dependent manner.This study demonstrated that FO could attenuate the adverse effect of 20:1 n-6/n-3 PUFA diet on osteoporotic OA,providing skeletal evidence for healthy diet. | Tianqi Zhang Xiaolin Bai Yufeng Dai Ping Dong Jingfeng Wang | 2022 | Food Bioscience2022,45,1: | 1 |

| 10 | A new method for uniform local heating deep in body using ultrasound phased-array system显示文摘A new method for targeted heating of deep tissue was developed by using an ultrasound phased-arraysystem which can generate various multiple foci patterns by electronically changing its amplitude or phasepattern.This method involves using a technique of combining switching and rotating of multiple foci pat-terns to create a uniform temperature over tissue volumes in various size.Using this method,the targettissue deep in the body can be heated to a specified temperature,which gives conditions for thermo-sensi-tire liposomes release.A simulation study for a 108-element,spherically sectioned array was performed todetermine an optimal heating scheme from a set of multiple focus fields which were produced by inputtingdifferent combinations of phases and amplitudes.Comparisons of a static multiple foci field,the switchedfields and the switched-rotated fields indicated that the technique of combining switching and rotating ofmultiple foci patterns has advantages of both lowering the peak temperature and evening the temperaturedistribution.The simulation results also show that the therapeutic heating zones in various size(φ5mm~φ40mm)with uniform temperature distributions can be obtained employing the combined method.Theseresults offer significant data for designing thermotherapy equipment for tumor-specific drug release withthermo-sensitive hposomes. | 章琛曦 Bai Jingfeng Chen Yazhu | 2008 | High Technology Letters2008,14,1: | 0 |
| 11 | Epidemiological characteristics and clinical manifestations of pediatric patients with COVID-19 in China: A multicenter retrospective study显示文摘Importance:The Coronavirus disease 2019(COVID-19)global pandemic poses a considerable challenge for pediatricians.Objective:This study aimed to identify the epidemiological characteristics and clinical features of pediatric patients with COVID-19 in China.Methods:This multicenter retrospective study included pediatric patients from 46 hospitals in China,covering 12 provinces and two municipalities.Epidemiological,demographic,clinical,laboratory,treatment,and outcome data were analyzed.Results:In total,211 pediatric patients with COVID-19 were included in this study.The median age was 7.0 years(range:22 days to 18 years).Approximately 16.3%of the patients exhibited asymptomatic infections,23.0%had upper respiratory tract infections,and 60.7%had pneumonia,including two with severe pneumonia and one with critical illness.Approximately 78.7%of the pediatric patients occurred in familial clusters.The most three common symptoms or signs at onset in children with COVID-19 were fever(54.5%),cough(49.3%),and pharyngeal congestion(20.8%).Only 17.6%of the patients presented with decreased lymphocyte count,whereas 13.6%had increased lymphocyte count.Among the patients with pneumonia who exhibited abnormal chest computed tomography findings,18.2%(23/127)of the patients had no other symptoms.Generally,the chest radiographs showed abnormalities that affected both lungs(49.6%);ground-glass opacity(47.2%)was the most common manifestation.The cure and improvement rates were 86.7%(183/211)and 13.3%(28/211),respectively.Only one patient with an underlying condition received invasive mechanical ventilation;none of the patients died.Interpretation:Similar to adults,children of all age groups are susceptible to COVID-19.Fortunately,most pediatric patients have mild symptoms or remain asymptomatic,despite the high incidence of pneumonia.Decreased proportions of white blood cells and lymphocytes are less frequent in children than in adults. | Yan Bai Liwei Gao Xianfeng Wang Lili Zhong Jingfeng Li Shenggang Ding Yuejie Zheng Jun Liu Yuxia Cui Lina Wang Mingfeng Han Jun Liu Xinping Jiang Min Jiang Junhua Li Ning Chen Yunxiao Shang Hourong Zhou Yi Xu Gen Lu Xing Chen Jizhi Xu Qihong Fan Yu Tang Jiang Wu Cen Li Xiaoxiang Yang Chunxi Chen Yonghong Yang Gary Wing-Kin Wong Adong Shen Tianyou Wang Runming Jin Baoping Xu Kunling Shen | 2021 | Pediatric Investigation2021,5,3: | 0 |
| 12 | Efficient production of chimeric mice from embryonic stem cells injected into 4-to 8-cell and blastocyst embryos显示文摘Background: Production of chimeric mice is a useful tool for the elucidation of gene function. After successful isolation of embryonic stem(ES) cell lines, there are many methods for producing chimeras, including co-culture with the embryos, microinjection of the ES cells into pre-implantation embryos, and use of tetraploid embryos to generate the full ES-derived transgenic mice. Here, we aimed to generate the transgenic ES cell line, compare the production efficiency of chimeric mice and its proportion to yield the male chimeric mice by microinjected ES cells into 4-to 8-cell and blastocysts embryos with the application of Piezo-Micromanipulator(PMM), and trace the fate of the injected ES cells. Results: We successfully generated a transgenic ES cell line and proved that this cell line still maintained pluripotency. Although we achieved a satisfactory chimeric mice rate, there was no significant difference in the production of chimeric mice using the two different methods, but the proportion of the male chimeric mice in the 4-to 8-cell group was higher than in the blastocyst group. We also found that there was no tendency for ES cells to aggregate into the inner cell mass using in vitro culture of the chimeric embryos, indicating that they aggregated randomly. Conclusions: These results showed that the PMM method is a convenient way to generate chimeric mice and microinjection of ES cells into 4-to 8-cell embryos can increase the chance of yielding male chimeras compared to the blastocyst injection.
