Multicenter study of the clinicopathological features and recurrence risk prediction model of early-stage breast cancer with low-positive human epidermal growth factor receptor 2 expression in China (Chinese Society of Breast Surgery 021)显示文摘Background:Breast cancer with low-positive human epidermal growth factor receptor 2(HER2)expression has triggered further refinement of evaluation criteria for HER2 expression. We studied the clinicopathological features of early-stage breast cancer with low-positive HER2 expression in China and analyzed prognostic factors. Methods:Clinical and pathological data and prognostic information of patients with early-stage breast cancer with low-positive HER2 expression treated by the member units of the Chinese Society of Breast Surgery and Chinese Society of Surgery of Chinese Medical Association, from January 2015 to December 2016 were collected. The prognostic factors of these patients were analyzed.Results:Twenty-nine hospitals provided valid cases. From 2015 to 2016,a total of 25,096 cases of early-stage breast cancer were treated,7642(30.5%)of which had low-positive HER2 expression and were included in the study. After ineligible cases were excluded,6486 patients were included in the study. The median follow-up time was 57 months(4-76 months). The disease-free survival rate was 92.1%at 5 years, and the overall survival rate was 97.4%at 5 years. At the follow-up,506(7.8%)cases of metastasis and 167(2.6%)deaths were noted.Multivariate Cox regression analysis showed that tumor stage, lymphvascular invasion, and the Ki67 index were related to recurrence and metastasis(P<0.05).The recurrence risk prediction model was established using a machine learning model and showed that the area under the receiving operator characteristic curve was 0.815(95%confidence interval:0.750-0.880).Conclusions:Early-stage breast cancer patients with low-positive HER2 expression account for 30.5%of all patients.Tumor stage, lymphvascular invasion, and the Ki67 index are factors affecting prognosis. The recurrence prediction model for breast cancer with low-positive HER2 expression based on a machine learning model had a good clinical reference value for predicting the recurrence risk at 5 years.Trial registration:ChiCTR.org.cn,ChiCTR2100046766.Ling Xin Qian Wu Chongming Zhan Hongyan Qin Hongyu Xiang Ling Xu Jingming Ye Xuening Duan Yinhua Liu Chinese Society of Breast Surgery(CSBrS) Chinese Society of Surgery of Chinese Medical Association 2022Chinese Medical Journal2022,,6:9
A retrospective prognostic evaluation analysis using the 8th edition of American Joint Committee on Cancer(AJCC)cancer staging system for luminal A breast cancer显示文摘Objective: We retrospectively analyzed the clinical prognostic value of the 8th edition of the American Joint Committee on Cancer(AJCC) staging system for luminal A breast cancer. Methods: Using both the anatomic and prognostic staging in the 8th edition of AJCC cancer staging system, we restaged patients with luminal A breast cancer treated at the Breast Disease Center, Peking University First Hospital from 2008 to 2014. Follow-up data including 5-year disease free survival(DFS), overall survival(OS) and other clinic-pathological data were collected to analyze the differences between the two staging subgroups. Results: This study included 421 patients with luminal A breast cancer(median follow-up, 61 months). The 5-year DFS and OS rates were 98.3% and 99.3%, respectively. Significant differences in 5-year DFS but not OS were observed between different anatomic disease stages. Significant differences were observed in both 5-year DFS and OS between different prognostic stages. Application of the prognostic staging system resulted in assignment of 175 of 421 patients(41.6%) to a different group compared to their original anatomic stages. In total, 102 of 103 patients with anatomic stage IIA changed to prognostic stage IB, and 24 of 52 patients with anatomic stage IIB changed to prognostic stage IB, while 1 changed to prognostic stage IIIB. Twenty-two of 33 patients with anatomic stage IIIA were down-staged to IIA when staged by prognostic staging system, and the other 11 patients were down-staged to IIB. Two patients with anatomic stage IIIB were down-staged to IIIA. Among seven patients with anatomic stage IIIC cancer, two were down-staged to IIIA and four were down-staged to stage IIIB.Conclusions: The 8th edition of AJCC prognostic staging system is an important supplement to the breast cancer staging system. More clinical trials are needed to prove its ability to guide selection of proper systemic therapy and predict prognosis of breast cancer.jingming ye wenjun wang ling xu xuening duan yuanjia cheng ling xin hong zhang shuang zhang ting li yinhua liu 2017Chinese Journal of Cancer Research2017,29,4:8
A novel STING agonist-adjuvanted pan-sarbecovirus vaccine elicits potent and durable neutralizing antibody and T cell responses in mice, rabbits and NHPs显示文摘The emergence of SARS-CoV-2 variants and potentially other highly pathogenic sarbecoviruses in the future highlights the need for pan-sarbecovirus vaccines. Here,we discovered a new STING agonist,CF501,and found that CF501-adjuvanted RBD-FC vaccine(CF501/RBD-FC)elicited significantly stronger neutralizing antibody(nAb)and T cell responses than Alum-and cGAMP-adjuvanted RBD-FC in mice.Vaccination of rabbits and rhesus macaques(nonhuman primates,NHPs)with CF501/RBD-FC elicited exceptionally potent nAb responses against SARS-CoV-2 and its nine variants and 41 S-mutants,SARS-CoV and bat SARSr-CoVs.CF501/RBD-FC-immunized hACE2-transgenic mice were almost completely protected against SARS-CoV-2 challenge, even 6 months after the initial immunization.NHPs immunized with a single dose of CF501/RBD-FC produced high titers of nAbs.The immunized macaques also exhibited durable humoral and cellular immune responses and showed remarkably reduced viral load in the upper and lower airways upon SARS-CoV-2 challenge even at 108 days post the final immunization.Thus,CF501/RBD-Fc can be further developed as a novel pan-sarbecovirus vaccine to combat current and future outbreaks of sarbecovirus diseases.Zezhong Liu Jie Zhou Wei Xu Wei Deng Yanqun Wang Meiyu Wang Qian Wang Ming Hsieh Jingming Dong Xinling Wang Weijin Huang Lixiao Xing Miaoling He Chunlin Tao Youhua Xie Yilong Zhang Youchun Wang Jincun Zhao Zhenghong Yuan Chuan Qin Shibo Jiang Lu Lu 2022Cell Research2022,32,3:5

Gap-free genome assembly and comparative analysis reveal the evolution and anthocyanin accumulation mechanism of Rhodomyrtus tomentosa显示文摘Rhodomyrtus tomentosa is an important f leshy-fruited tree and a well-known medicinal plant of the Myrtaceae family that is widely cultivated in tropical and subtropical areas of the world. However, studies on the evolution and genomic breeding of R.tomentosa were hindered by the lack of a reference genome. Here,we presented a chromosome-level gap-free T2T genome assembly of R.tomentosa using PacBio and ONT long read sequencing. We assembled the genome with size of 470.35 Mb and contig N50 of∼43.80 Mb with 11 pseudochromosomes.A total of 33382 genes and 239.31 Mb of repetitive sequences were annotated in this genome.Phylogenetic analysis elucidated the independent evolution of R.tomentosa starting from 14.37MYA and shared a recent WGD event with other Myrtaceae species. We identified four major compounds of anthocyanins and their synthetic pathways in R.tomentosa.Comparative genomic and gene expression analysis suggested the coloring and high anthocyanin accumulation in R.tomentosa tends to be determined by the activation of anthocyanin synthesis pathway. The positive selection and up-regulation of MYB transcription factors were the implicit factors in this process. The copy number increase of downstream anthocyanin transport-related OMT and GST gene were also detected in R.tomentosa.Expression analysis and pathway identification enriched the importance of starch degradation, response to stimuli, effect of hormones, and cell wall metabolism during the f leshy fruit development in Myrtaceae. Our genome assembly provided a foundation for investigating the origins and differentiation of Myrtaceae species and accelerated the genetic improvement of R.tomentosa.angping Li Shiqiang Xu Zitong Xiao Jingming Wang Yu Mei Haifei Hu Jingyu Li Jieying Liu Zhuangwei Hou Junliang Zhao Shaohai Yang Jihua Wang 2023Horticulture Research2023,10,3:3
Magnetic resonance imaging for non-invasive clinical evaluation of normal and regenerated cartilage显示文摘With the development of tissue engineering and regenerative medicine,it is much desired to establish bioimaging techniques to monitor the real-time regeneration efficacy in vivo in a non-invasive way.Herein,we tried magnetic resonance imaging(MRI)to evaluate knee cartilage regeneration after implanting a biomaterial scaffold seeded with chondrocytes,namely,matrix-induced autologous chondrocyte implantation(MACI).After summary of the T2 mapping and the T1-related delayed gadolinium-enhanced MRI imaging of cartilage(dGEMRIC)in vitro and in vivo in the literature, these two MRI techniques were tried clinically. In this study,18 patients were followed up for 1 year.It was found that there was a significant difference between the regeneration site and the neighboring normal site(control),and the difference gradually diminished with regeneration time up to 1 year according to both the quantitative T1 and T2 MRI methods. We further established the correlation between the quantitative evaluation of MRI and the clinical Lysholm scores for the first time.Hence, the MRI technique was confirmed to be a feasible semiquantitative yet non-invasive way to evaluate the in vivo regeneration of knee articular cartilage.Xian Xu Jingming Gao Shuyun Liu Liang Chen Min Chen Xiaoye Yu Ning Ma Jun Zhang Xiaobin Chen Lisen Zhong Lin Yu Liming Xu Quanyi Guo Jiandong Ding 2021Regenerative Biomaterials2021,8,5:2

Studies on the preparation of active oxygen-deficient copper ferrite and its application for hydrogen production through thermal chemical water splitting显示文摘Hydrogen generation through thermal chemical water splitting technology has recently received in- creasingly international interest in the nuclear hydrogen production field. Besides the main known sulfur-iodine (S-I) cycle developed by the General Atomics Company and the UT3 cycle (iron, calcium, and bromine) developed at the University of Tokyo, the thermal cycle based on metal oxide two-step water splitting methods is also receiving research and development attention worldwide. In this work, copper ferrite was prepared by the co-precipitation method and oxygen-deficient copper ferrite was synthesized through first and second calcination steps for the application of hydrogen production by a two-step water splitting process. The crystal structure, properties, chemical composition and δ were investigated in detail by utilizing X-ray diffraction (XRD), thermogravimetry (TG) and differential thermal analysis (DTA), atomic absorption spectrometer (AAS), ultraviolet spectrophotometry (UV), gas chro- matography (GC), and so on. The experimental two-step thermal chemical cycle reactor for hydrogen generation was designed and developed in this lab. The hydrogen generation process of water splitting through CuFe2O4-δ and the cycle performance of copper ferrite regeneration were firstly studied and discussed.YU Bo ZHANG Ping ZHANG Lei CHEN Jing XU JingMing 2008Science China Chemistry2008,51,9:1
