| 4 | High-throughput fat quantifications of hematoxylin-eosin stained liver histopathological images based on pixel-wise clustering显示文摘Besides diagnosis of fatty liver disease(FLD) using multiple medical imaging techniques in clinic,accurate fat quantification of liver tissue slice, especially the fat droplets measurement, is still a critical indicator in related pathological researches. Stained by hematoxylin-eosin(HE), different tissue components with different colors need to be identified and measured manually in conventional approaches. Automated liver fat quantification of HE stained images remains challenging because forms and distributions of fat are extremely irregular with no clear boundaries, especially in conducting high-throughput analysis which demands quick processing and higher accuracy for the reference of pathologists. To solve this problem, we propose an automated liver fat quantifications pipeline of HE stained images based on pixel-wise clustering, which firstly extracts high-relevant pixel-level features with color mode transformation, then locates boundaries between nuclei, fat and other components by clustering image pixels in an unsupervised mode, and finally identifies indicative fat droplets based on a set of morphological criteria. The pipeline was verified in analysis of multifold fatty liver treatment assays, with experimental results showing high accuracy and adaptability in fat droplets quantification despite data variance. Quantitative indicators provide a reliable evidence for relevant pathological researches or therapy selection, in which number and average area of indicative fat droplets increased sharply in severe and moderate-grade FLD respectively. | 5 | Identification of two migratory colon ILC2 populations differentially expressing IL-17A and IL-5/IL-13显示文摘Group 2 innate lymphoid cells(ILC2s)play important tissue resident roles in anti-parasite immunity,allergic immune response,tissue homeostasis,and tumor immunity.ILC2s are considered tissue resident cells with little proliferation at steady state.Recent studies have shown that a subset of small intestinal ILC2s could leave their residing tissues,circulate and migrate to different organs,including lung,liver,mesenteric LN and spleen,upon activation.However,it remains unknown whether other ILC populations with migratory behavior exist.In this study,we find two major colon ILC2 populations with potential to migrate to the lung in response to IL-25 stimulation.One subset expresses IL-17A and resembles inflammatory ILC2s(iILC2s)but lacks CD27 expression,whereas the other expresses CD27 but not IL-17A.In addition,the IL-17A^(+)ILC2s express lower levels of CD127,CD25,and ST2 than CD27^(+)ILC2s,which express higher levels of IL-5 and IL-13.Surprisingly,we found that both colon ILC2 populations still maintained their colonic features of preferential expression of IL-17A and CD27,IL-5/IL-13,respectively.Together,our study identifies two migratory colon ILC2 subsets with unique surface markers and cytokine profiles which are critical in regulating lung and colon immunity and homeostasis. | Hongzhi Liu Liang Li Yanyun Hao Jialu Li Zhaoyuan Liu Jingjing Qi Jingjing Zhang Ningbo Wu Dandan Wu Caixia Gao Lei Chen Lei Shen Jinke Cheng Bing Su | 2023 | Science China(Life Sciences)2023,66,1: | 1 |

| 19 | Towards Efficient Blue DelayedFluorescence Molecules by Modulating Torsion Angle Between Electron Donor and Acceptor显示文摘Constructing blue thermally activated delayedfluorescence materials for high-performance organic light-emitting diodes(OLEDs)remains challenging due to the intrinsically strong intramolecular charge transfer nature of the nearly orthogonal connection of electron donor(D)and acceptor(A),which results in long-wavelength emission.Herein,an effective delayed-fluorescence design strategy of modulating D–A torsion angles is proposed and efficient sky-blue,pure-blue,and deep-blue delayed-fluorescence molecules consisting of a xanthenone acceptor and carbazole-based donors are created by decreasing the torsion angles.They exhibit strong delayed fluorescence with high photoluminescence quantum yields of 85–94%in doped films,and their delayed-fluorescence lifetimes are elongated from 1.0 to 27.6μs as the torsion angles decrease.These molecules can function as excellent emitters in OLEDs,providing efficient electroluminescence peaking at 442 nm(CIEx,y=0.15,0.08),462 nm(CIEx,y=0.15,0.18),and 482 nm(CIEx,y=0.17,0.30)with state-of-the-art external quantum efficiencies of up to 22.2%,33.7%,and 32.1%,respectively,demonstrating the proposed molecular design for efficient blue delayed-fluorescence molecules is successful and promising. | Jinke Chen Xing Wu Hao Liu Nuoling Qiu Zhangshan Liu Dezhi Yang Dongge Ma Ben Zhong Tang Zujin Zhao | 2023 | CCS Chemistry2023,5,3: | 0 |
