1An Analysis of Historical and Future Temperature Fluctuations over China Based on CMIP5 Simulations显示文摘The trends and fluctuations of observed and CMIP5-simulated yearly mean surface air temperature over China were analyzed. In general, the historical simulations replicate the observed increase of temperature, but the multi-model ensemble(MME) mean does not accurately reproduce the drastic interannual fluctuations. The correlation coefficient of the MME mean with the observations over all runs and all models was 0.77, which was larger than the largest value(0.65) from any single model ensemble. The results showed that winter temperatures are increasing at a higher rate than summer temperatures, and that winter temperatures exhibit stronger interannual variations. It was also found that the models underestimate the differences between winter and summer rates. The ensemble empirical mode decomposition technique was used to obtain six intrinsic mode functions(IMFs) for the modeled temperature and observations. The periods of the first two IMFs of the MME mean were 3.2 and 7.2, which represented the cycle of 2–7-yr oscillations. The periods of the third and fourth IMFs were 14.7 and 35.2, which reflected a multi-decadal oscillation of climate change. The corresponding periods of the first four IMFs were 2.69, 7.24, 16.15 and 52.5 in the observed data. The models overestimate the period of low frequency oscillation of temperature, but underestimate the period of high frequency variation. The warming rates from different representative concentration pathways(RCPs) were calculated, and the results showed that the temperature will increase by approximately 0.9℃, 2.4℃, 3.2℃ and 6.1℃ in the next century under the RCP2.6, RCP4.5, RCP6.0 and RCP8.5 scenarios, respectively.LIU Yonghe FENG Jinming MA Zhuguo 2014Advances in Atmospheric Sciences2014,31,2:15
2Noise and delay sustained chimera state in small world neuronal network显示文摘Chimera state in neuronal network means the coexistence of synchronized and desynchronized firing patterns. It attracts much attention recently due to its possible relevance to the phenomenon of unihemispheric sleep in mammals. In this paper, we search for chimera state in a noisy small-world neuronal network, in which the neurons are delayed coupled. We found both transient and permanent chimera state when time delay is close to a critical value. The chimera state occurs due to the competition between the synchronized and desynchronized patterns in the neuronal network. On the other hand, intermediate intensity of noise facilitates the occurrence of delay-sustained chimera states. 3Anti-PD-L1/TGF-BR fusion protein (SHR-1701) overcomes disrupted lymphocyte recovery-induced resistance to PD-1/PD-L1 inhibitors in lung cancer显示文摘Background:Second-generation programmed cell death-protein 1/pro-grammed death-ligand 1(PD-1/PD-L1)inhibitors,such as bintrafusp alfa(M7824),SHR-1701,and YM101,have been developed to simultaneously block PD-1/PD-L1 and transforming growth factor-beta/transforming growth factor-beta receptor(TGF-P/TGF-βR).Consequently,it is necessary to identify predictive factors of lung cancer patients who are not only resistant to PD-1/PD-LI inhibitors but also sensitive to bifunctional drugs.The purpose of this study was to search for such predictors.Methods:Multivariable Cox regression was used to study the association between the clinical outcome of treatment with PD-1/PD-L1 inhibitors and lym-phocyte recovery after lymphopenia in lung cancer patients.Murine CMT167 lung cancer cells were engineered to express the firefly luciferase gene and implanted orthotopically in the lung of syngeneic mice.Bioluminescence imag-ing,flow cytometry,and immunohistochemistry were employed to detrmine response to immunotherapy and function of tumor-infiltrating immune cells.Results:For lung cancer patients treated with anti-PD-1/PD-LI antibodies,poor lymphocyte recovery was associated with a shorter progression-free survival(PFS;P<0.001),an accumulation of regulatory T cells(Tregs),and an elimi-nation of CD8+T cells in the peripheral blood.Levels of CD8+T cells and Treg cells were also imbalanced in the tumors and peripheral immune organs of mice with poor lymphocyte recovery after chemotherapy.Moreover,these mice failed to respond to anti-PD-1 antibodies but remained sensitive to the anti-PD-LI/TGF-βR fusion protein(SHR-1701).Consistently,SHR-1701 but not anti-PD-1 antibod-ies,markedly enhanced IFN-γproduction and Ki-67 expression in peripheral CD8+T cells from patients with impaired lymphocyte recovery.Conclusions:Lung cancer patients with poor lymphocyte recovery and suffer-ing from persistent lymphopenia after previous chemotherapy are resistant to anti-PD-1/PD-L1 antibodies but might be sensitive to second-generation agents such as SHR-1701.Bo Cheng Kaikai Ding Pengxiang Chen Jianxiong Ji Tao Luo Xiaofan Guo Wei Qiu Chunhong Ma Xue Meng Jian Wang Jinming Yu Yuan Liu 2022Cancer Communications2022,42,1:5

5Ticagrelor inhibits the NLRP3 inflammasome to protect against inflammatory disease independent of the P2Y_(12 ) signaling pathway显示文摘Ticagrelor is the first reversibly binding oralP2Y_(12) receptor antagonist to inhibit platelet activation and has been approved by the Food and Drug Administration for the treatment of coronary artery disease. At present, the other pharmacological functions of ticagrelor remain poorly understood. The NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome plays a critical role in the innate immune system, but its excessive activation also contributes to the pathogenesis of complex diseases. In this study, we systematically examined the effects of ticagrelor on the NLRP3 inflammasome and found that ticagrelor inhibits NLRP3 inflammasome activation in macrophages independent of its classic inhibitory effect on theP2Y_(12) signaling pathway. Further mechanistic studies demonstrate that ticagrelor attenuates the oligomerization of apoptosis-associated speck-like protein containing a CARD (ASC) by blocking chloride efflux, an effect achieved through the degradation of chloride intracellular channel proteins (CLICs) and blockade of the translocation of CLICs to the plasma membrane. Moreover, experiments on lipopolysaccharide-induced sepsis and alum-induced peritonitis in mice confirmed that ticagrelor mitigates the severity of systemic inflammation independent ofP2Y_(12) receptor antagonism. Importantly, oral administration of ticagrelor rapidly and strongly inhibited NLRP3 inflammasome activation in peripheral blood mononuclear cells from patients with acute coronary syndrome. 8Lipid profiling of mammalian cells with in situ matrix-assisted laser desorption ionization-mass spectrometry显示文摘The analysis of cellular lipids and phospholipids has been of continuously increasing research interest due to the importance of these molecules in psychological process.In this work,a mass spectrometry-based method for direct,in situ analysis of lipids in cells was reported.Mammalian cells were directly cultured on ITO-coated glass and then analyzed by matrix-assisted laser desorption ionization-mass spectrometry(MALDI-MS).The matrix application process was achieved by electrospray coating,which produced a homogenous layer of matrix crystal on the sample.The detection results and reproducibility are satisfactory.With this method,a profile of abundant membrane lipids is generated,which is characteristic of cell type.In conclusion,this in situ MALDI-MS cellular lipid analysis method provides a platform for sensitive and robust molecular profiling of mammalian cells.ZHANG YanDong LI HaiFang MA Yuan LIN JinMing 2014Science China Chemistry2014,57,3:1

10Control factors of DIC in the Y3 seamount waters of the Western Pacific Ocean显示文摘An investigation was carried out in the Y3 seamount area of the Western Pacific Ocean in December 2014,and the distribution of dissolved inorganic carbon(DIC)and its relationship with environmental factors in this area were explored.The results show that DIC concentration was higher in the adjacent waters of the Y3 seamount area,and the uplift of DIC isolines at the stations was close to the seamount.Meanwhile,interaction between the North Equatorial Current(NEC)and the Y3 seamount affected the DIC distribution,i.e.,the upwelling in the same direction of the NEC was obvious,resulting in a decreasing trend of average concentration of DIC in the 200 m water column from the top to the two sides in this direction but in the cross direction.The DIC concentration increased with the water depth increase,and its distribution was affected by various environmental factors.In the surface water,high temperature was a decisive factor for the decrease of the DIC concentration,but the photosynthesis of phytoplankton showing only a weak influence.In the North Pacific Tropic Water(NPTW),DIC production rate from organic matter decomposition was higher than that of DIC consumption by phytoplankton photosynthesis,leading to a continual increase of DIC.In the North Pacific Intermediate Water(NPIW),organic matter decomposition played a leading role in the increase of DIC.In the deep water,decomposition of organic matter weakened,and the dissolution of CaCO3 controlled the carbonate system,and DIC had the smallest variation range.MA Jun SONG Jinming LI Xuegang YUAN Huamao LI Ning DUAN Liqin WANG Qidong 2020Journal of Oceanology and Limnology2020,38,4:1
11Identification and characterization of loop7 motif and its role in regulating biological function of human APOBEC3G through molecular modeling and biological assay显示文摘Human APOBEC3G(h A3G) is a cytidine deaminase which inhibits HIV-1 replication. The HIV-1 accessory protein viral infectivity factor(Vif) counteracts with hA3G by targeting it for proteasomal degradation. In this work, we constructed and optimized molecular models of the hA3G dimer and the h A3G–Vif complex. The molecular modeling study revealed that the loop7 motif of hA3G appears on the interfaces of both the h A3G–Vif complex and the hA3G dimer. Biochemical analysis provided evidence suggesting that binding of Vif to hA3G results in steric blocking of hA3G dimerization, implying that monomeric hA3G serves as a substrate for Vif-mediated degradation.Furthermore, we presented evidence for the important roles of the loop7 motif, especially the central residues within the region, in hA3G dimerization, h A3G–Vif interaction, Vif-mediated hA3G degradation as well as subcellular localization of hA3G. 13Preliminary validation of GLASS-DSSR products using surface measurements collected in arid and semi-arid regions of China显示文摘Global Land Surface Satellite-downward surface shortwave radiation(GLASSDSSR)products have been routinely produced from 20082010 based on an improved look-up table algorithm,which explicitly accounts for the variations of cloud optical depth,water vapor content,and elevation.In this study,we validated and assessed the accuracy of these products in arid and semiarid regions of China.Toward this goal,observation data-sets provided by the Arid and Semiarid Region Collaborative Observation Project as well as four other metrological sites were collected,chosen,and preprocessed for the final validation.Due to the possible effect of spatial collocation and the strong adjacency pixel effect in instantaneous products,we used a more sophisticated validating scheme in order to reduce the impacts from these effects as much as possible.Evidences indicate that the GLASS-DSSR products are considerably accurate over most parts of arid and semiarid regions in China,but in complex terrain areas the products might need further refinements.The R2 at all sites(except Naqu)was larger than 0.8 with a root mean square error(RMSE)range of about in 90130 W/m2.Linear regression analyses suggest that GLASS-DSSR products tend to overestimate DSSR in the interval of low surface-measured values and symmetrically underestimate DSSR in the interval of high values.This systematic error may result from inappropriate assumptions about clouds and aerosol loadings over the regions in the operational algorithm.Guanghui Huang Weizhen Wang Xiaotong Zhang Shunlin Liang Shaomin Liu Tianbao Zhao Jinming Feng Zhuguo Ma 2013International Journal of Digital Earth2013,6,S01:1
14Modeling the Warming Impact of Urban Land Expansion on Hot Weather Using the Weather Research and Forecasting Model: A Case Study of Beijing, China显示文摘The impacts of three periods of urban land expansion during 1990–2010 on near-surface air temperature in summer in Beijing were simulated in this study, and then the interrelation between heat waves and urban warming was assessed. We ran the sensitivity tests using the mesoscale Weather Research and Forecasting model coupled with a single urban canopy model,as well as high-resolution land cover data. The warming area expanded approximately at the same scale as the urban land expansion. The average regional warming induced by urban expansion increased but the warming speed declined slightly during 2000–2010. The smallest warming occurred at noon and then increased gradually in the afternoon before peaking at around 2000 LST—the time of sunset. In the daytime, urban warming was primarily caused by the decrease in latent heat flux at the urban surface. Urbanization led to more ground heat flux during the day and then more release at night, which resulted in nocturnal warming. Urban warming at night was higher than that in the day, although the nighttime increment in sensible heat flux was smaller. This was because the shallower planetary boundary layer at night reduced the release efficiency of near-surface heat. The simulated results also suggested that heat waves or high temperature weather enhanced urban warming intensity at night. Heat waves caused more heat to be stored in the surface during the day, greater heat released at night, and thus higher nighttime warming. 15Treatment‐related adverse events of antibody‐drug conjugates in clinical trials:A systematic review and meta‐analysis显示文摘Background:The wide use of antibody‐drug conjugates(ADCs)is transforming the cancer‐treatment landscape.Understanding the treatment‐related adverse events(AEs)of ADCs is crucial for their clinical application.We conducted a meta‐analysis to analyze the profile and incidence of AEs related to ADC use in the treatment of solid tumors and hematological malignancies.Methods:We searched the PubMed,Embase,and Cochrane Library databases for articles published from January 2001 to October 2022.The overall profile and incidence of all‐grade and grade≥3 treatment‐related AEs were the primary outcomes of the analysis.Results:A total of 138 trials involving 15,473 patients were included in this study.The overall incidence of any‐grade treatment‐related AEs was 100.0%(95%confidence interval[CI]:99.9%–100.0%;I2=89%)and the incidence of grade≥3 treatment‐related AEs was 6.2%(95%CI:3.0%–12.4%;I2=99%).Conclusions:This study provides a comprehensive overview of AEs related to ADCs used for cancer treatment.ADC use resulted in a high incidence of any‐grade AEs but a low incidence of grade≥3 AEs.The AE profiles and incidence differed according to cancer type,ADC type,and ADC components.Jinming Li Guoshuang Shen Zhen Liu Yaobang Liu Miaozhou Wang Fuxing Zhao Dengfeng Ren Qiqi Xie Zitao Li Zhilin Liu Yi Zhao Fei Ma Xinlan Liu Zhengbo Xu Jiuda Zhao 2023Cancer Innovation2023,2,5:1

18Pivotal roles of tumor-draining lymph nodes in the abscopal effects from combined immunotherapy and radiotherapy显示文摘Background:Currently,due to synergy enhancement of anti-tumor effects and potent stimulation of abscopal effects,combination therapy with irradiation and programmed cell death protein 1/programmed death-ligand 1(PD-1/PD-L1)immune checkpoint inhibition(immuno-radiotherapy,iRT)has revolutionized the therapeutic guidelines.It has been demonstrated that tumor-draining lymph nodes(TDLN)are essential for effective antitumor immunity induced by radiotherapy,immunotherapy,or iRT.Given that the function of TDLN in iRT remains unclear,this study aimed to investigate the function and mechanism of TDLN in iRT-induced abscopal effects.Methods:The function of TDLN was evaluated using unilateral or bilateral MC38 and B16F10 subcutaneous tumor models with or without indicated TDLN.The flow cytometry,multiple immunofluorescence analysis,and NanoString analysis were utilized to detect the composition and function of the immune cells in the primary and abscopal tumor microenvironment.Additionally,we tempted to interrogate the possible mechanisms via RNA-sequencing of tumor-infiltrating lymphocytes and TDLN.Results:TDLN deficiency impaired the control of tumor growth by monotherapy.Bilateral TDLN removal rather than unilateral TDLN removal substantially curtailed iRT-stimulated anti-tumor and abscopal effects.Furthermore,in the absence of TDLN,the infiltration of CD45+and CD8+T cells was substantially reduced in both primary and abscopal tumors,and the anti-tumor function of CD8+T cells was attenuated as well.Additionally,the polarization of tumor-associated macrophages in primary and abscopal tumors were found to be dependent on intact bilateral TDLN.RNA-sequencing data indicated that impaired infiltration and anti-tumor effects of immune cells partially attributed to the altered secretion of components from the tumor microenvironment.Conclusions:TDLN play a critical role in iRT by promoting the infiltration of CD8+T cells and maintaining the M1/M2 macrophage ratio.Zhaoyun Liu Zhiyong Yu Dawei Chen Vivek Verma Chenxi Yuan Minglei Wang Fei Wang Qing Fan Xingwu Wang Yang Li Yuequn Ma Meng Wu Jinming Yu 2022Cancer Communications2022,42,10:1

20Experiments and evidences: jellyfish (Nemopilema nomurai) decomposing and nutrients (nitrogen and phosphorus) released显示文摘The aim of this study was to investigate nitrogen and phosphorus released in the process of the decomposition of giant jellyfish in the laboratory and found the evidence to verify the influence of nutrients released by the decomposition of jellyfish on the ecosystem in the field. The release of nitrogen and phosphorus from the decomposition of Nemopilema nomurai was examined in a series of experiments under different incubation conditions such as different p H values, salinity values, temperatures and nitrogen and phosphorus concentrations. The results showed that the complete decomposition of Nemopilema nomurai generally took about 4–8 d. The release of nitrogen and phosphorus from the decomposition of Nemopilema nomurai could be divided into two stages: the early stage and the later stage, although the efflux rate of nitrogen was one order more than phosphorus. In the early stage of the decomposition of Nemopilema nomurai, the concentrations of dissolved nitrogen, dissolved phosphorus, total nitrogen and total phosphorus in seawater increased rapidly, and the concentration of nitrogen could reach the highest level in the whole degradation process. In the later stage of the decomposition, the concentrations of dissolved nitrogen and total nitrogen declined slowly, while the concentration of phosphorus in water could reach a maximum in the degradation process. High p H, low salinity,high temperature and N/P will promote the release of nitrogen; low p H is unfavorable to the release of nitrogen but favorable to the release of phosphorus. In addition, we found the concentrations of ammonium and phosphate in the bottom water were higher than those in the surface water during the period of jellyfish bloom in the Jiaozhou Bay, proving that nutrients released by the decomposition of jellyfish have significant influence on nitrogen and phosphorus in the field. For the whole Yellow Sea, nutrients released by jellyfish carcasses may reach up to(2.63±2.98)×107 mol/d of dissolved nitrogen(DN) and(0.74±0.84)×106 mol/d of dissolved phosphorus(DP) during the period of jellyfish bloom. The values are comparable to riverine inputs in a day, but much higher than sediment–water exchange flux in the Yellow Sea. The great amounts of nutrients must have significant influence on the nutrients balance of the Yellow Sea during the period of jellyfish dead and decomposition.
