Neuroprotective effect of baicalin on focal cerebral ischemia in rats显示文摘Baicalin, a flavonoid compound from the root of the herb Scutellaria baicalensis Georgi, has been widely used to treat patients with inflammatory disease. The aim of this study was to assess the efficacy of baicalin in a rat model of focal cerebral ischemia. Adult male Sprague-Dawley rat models of cerebral artery occlusion were established and then randomly and equally divided into three groups: ischemia(cerebral ischemia and reperfusion), valproic acid(cerebral ischemia and reperfusion + three intraperitoneal injections of valproic acid; positive control), and baicalin(cerebral ischemia and reperfusion + intraperitoneal injection of baicalin for 21 days). Neurological deficits were assessed using the postural reflex test and forelimb placing test at 3, 7, 14, and 21 days after ischemia. Rat cerebral infarct volume was measured using 2,3,5-triphenyltetrazolium chloride(TTC) staining method. Pathological change of ischemic brain tissue was assessed using hematoxylin-eosin staining. In the baicalin group, rat neurological function was obviously improved, cerebral infarct volume was obviously reduced, and the pathological impairment of ischemic brain tissue was obviously alleviated compared to the ischemia group. Cerebral infarct volume was similar in the valproic acid and baicalin groups. These findings suggest that baicalin has a neuroprotective effect on cerebral ischemia. | Jiong Dai Yong-Ming Qiu Zheng-Wen Ma Guo-Feng Yan Jing Zhou Shan-Quan Li Hui Wu Yi-Chao Jin Xiao-Hua Zhang | 2018 | Neural Regeneration Research2018,13,12: | 7
On Weyl’s Theorem for Functions of Operators显示文摘Let H be a complex separable infinite dimensional Hilbert space. In this paper, a variant of the Weyl spectrum is discussed. Using the new spectrum, we characterize the necessary and sufficient conditions for both T and f(T) satisfying Weyl’s theorem, where f ∈ Hol(σ(T)) and Hol(σ(T)) is defined by the set of all functions f which are analytic on a neighbourhood of σ(T) and are not constant on any component of σ(T). Also we consider the perturbations of Weyl’s theorem for f(T). | Jiong DONG Xiao Hong CAO Lei DAI | 2019 | Acta Mathematica Sinica,English Series2019,35,8: | 5
Prior exposure to ciprofloxacin disrupts intestinal homeostasis and predisposes ayu(Plecoglossus altivelis)to subsequent Pseudomonas plecoglossicida-induced infection显示文摘With the rapid development of intensive farming, the aquaculture industry uses a great many antibiotics for the prevention and treatment of bacterial diseases.Despite their therapeutic functions, the overuse and accumulation of antibiotics also pose a threat to aquaculture organisms. In the present study, ayu(Plecoglossus altivelis)was used as a fish model to study the impacts of ciprofloxacin(CIP)overuse on intestinal homeostasis and immune response during subsequent Pseudomonas plecoglossicida infection.Based on 16S rRNA gene amplification and Illumina sequencing,we found that CIP pre-exposure caused significant variation in intestinal microbiota, including increased species richness, altered microbiota composition and interaction networks, and increased metabolic dysfunction.Furthermore, immunohistochemical analysis indicated that CIP pre-exposure resulted in severe mucosal layer damage, goblet cell reduction, and epithelial cell necrosis of the intestinal barrier in infected ayu.Quantitative real-time polymerase chain reaction(qRT-PCR)showed that disruption of intestinal homeostasis impaired systemic antiinfection immune responses in the intestine, gill, spleen, and head kidney, while inhibiting IL-1β,TNF-α,and IL-10 expression and promoting TGF-βexpression.Our findings indicated that CIP administration can directly affect intestinal microbiota composition and intestinal integrity in ayu fish. This perturbation of intestinal homeostasis is likely responsible for the lower survival rate of hosts following subsequent infection as the capacity to mount an effective immune response is compromised. This study also provides preliminary clues for understanding the effects of antibiotic overuse on higher vertebrates through trophic transfer. | Xiang-Yu Wu Jin-Bo Xiong Chen-Jie Fei Ting Dai Ting-Fang Zhu Zi-Yue Zhao Jing Pan Li Nie Jiong Chen | 2022 | Zoological Research2022,43,4: | 2
XAFS study of Cu^(2+) in aqueous solution of CuBr_2显示文摘Copper ion is the essential microelement to many organisms. In this paper, the local structure of Cu2+ in CuBr2 aqueous solutions with different concentrations are investigated by using X-ray absorption fine structure (XAFS) technique. XANES (X-ray Absorption Near Edge Structure) spectra indicate that charge transfer from Br- to Cu2+ decreases with the solution concentration, which lead to a shift of the absorption edge. The shoulder appearing at the rising edge proves to be characteristic of a tetragonal distortion. The Fourier transform magnitudes of EXAFS (Extended X-ray absorption fine structure) data of Cu species suggest that more Cu-Br bonds may exist in high concentrations. A fivefold coordination configuration like a pyramid is used as the fitting parameters. From the analysis of the coordination numbers, the proportion of Cu-O and Cu-Br is 4:1 in the saturated solution. The Br atom is on the equatorial plane of the model. The fitting results agree well with the experiment data. | DAI Binbin WANG Qian MA Jingyuan LI Jiong ZHANG Shuo HUANG Yuying HUANG Wei WU Guozhong ZOU Yang JIANG Zheng XU Hongjie | 2012 | Nuclear Science and Techniques2012,23,3: | 2
Leukocyte cell-derived chemotaxin 2 inhibits development of atherosclerosis in mice显示文摘Leukocyte cell-derived chemotaxin 2 (LECT2), a multifunctional hepatokine, is involved in many pathological conditi ons. However, its role in atherosclerosis remains undefined. In this study, we admimistered vehicle or LECT2 to male Apoe^-/- mice fed a Western diet for 15 weeks. Atherosclerotic lesions were visualized and quantified with Oil-red O and hematoxylin staining. The mRNA expression levels of MCP-1, MMP-1, IL-8 IL-1β, and TNF-a were analyzed by quantitative real-time polymerase chain reaction. Serum TNF-a, IL-1β, IL-8, MCP-1, and MMP-1 concentrations were measured by en zyme-li nked immuno sorbent assay. CD68, CD31, and a-SMA, markers of macrophages, endothelial cells, and smooth muscle cells, respectively, were detected by immuno staining. Results showed that LECT2 reduced total cholesterol and low-density lipoprotein concentrations in serum and inhibited the development of atherosclerotic lesions, accompanied by reductions in inflammatory cytokines and lower MCP-1, MMP-1, TNF-a, IL-8, and IL-1β mRNA abundanee. Furthermore, LECT2 decreased CD68, but in creased cr SMA in atherosclerotic lesi ons, suggesting an in crease in smooth muscle cells and reduction in macrophages. In summary, LECT2 inhibited the development of atherosclerosis in mice, accompanied by reduced serum total cholesterol concentration and lower inflammatory responses. | Wen-Ming He Ting Dai Jiong Chen Jian-An Wang | 2019 | Zoological Research2019,40,4: | 2
Histones released by NETosis enhance the infectivity of SARS-CoV-2 by bridging the spike protein subunit 2 and sialic acid on host cells显示文摘Neutrophil extracellular traps(NETs)can capture and kill viruses, such as influenza viruses, human immunodeficiency virus(HIV),and respiratory syncytial virus(RSV),thus contributing to host defense.Contrary to our expectation,we show here that the histones released by NETosis enhance the infectivity of SARS-CoV-2,as found by using live SARS-CoV-2 and two pseudovirus systems as well as a mouse model. The histone H3 or H4 selectively binds to subunit 2 of the spike(S)protein,as shown by a biochemical binding assay, surface plasmon resonance and binding energy calculation as well as the construction of a mutant S protein by replacing four acidic amino acids.Sialic acid on the host cell surface is the key molecule to which histones bridge subunit 2 of the S protein. Moreover, histones enhance cell-cell fusion. Finally, treatment with an inhibitor of NETosis, histone H3 or H4,or sialic acid notably affected the levels of sgRNA copies and the number of apoptotic cells in a mouse model. These findings suggest that SARS-CoV-2 could hijack histones from neutrophil NETosis to promote its host cell attachment and entry process and may be important in exploring pathogenesis and possible strategies to develop new effective therapies for COVID-19. | Weiqi Hong Jingyun Yang Jun Zou Zhenfei Bi Cai He Hong Lei Xuemei He Xue Li Aqu Alu Wenyan Ren Zeng Wang Xiaohua Jiang Kunhong Zhong Guowen Jia Yun Yang Wenhai Yu Qing Huang Mengli Yang Yanan Zhou Yuan Zhao Dexuan Kuang Junbin Wang Haixuan Wang Siyuan Chen Min Luo Ziqi Zhang Tiangi Lu Li Chen Haiying Que Zhiyao He Qiu Sun Wei Wang Guobo Shen Guangwen Lu Zhiwei Zhao Li Yang Jinliang Yang Zhenling Wang Jiong Li Xiangrong Song Lunzhi Dai Chong Chen Jia Geng Maling Gou Lu Chen Haohao Dong Yong Peng Canhua Huang Zhiyong Qian Wei Cheng Changfa Fan Yuquan Wei Zhaoming Su Aiping Tong Shuaiyao Lu Xiaozhong Peng Xiawei Wei | 2022 | Cellular & Molecular Immunology2022,19,5: | 2

The application of intraoperative neurophysiological monitoring in selective dorsal neurotomy for primary premature ejaculation:a prospective single-center study显示文摘Selective dorsal neurotomy(SDN)is a surgical treatment for primary premature ejaculation(PE),but there is still no standard surgical procedure for selecting the branches of the dorsal penile nerves to be removed. We performed this study to explore the value of intraoperative neurophysiological monitoring(IONM)of the penile sensory-evoked potential(PSEP)for standard surgical procedures in SDN.One hundred and twenty primary PE patients undergoing SDN were selected as the PE group and 120 non-PE patients were selected as the normal group. The PSEP was monitored and compared between the two groups under both natural and general anesthesia(GA)states. In addition, patients in the PE group were randomly divided into the IONM group and the non-IONM group.During SDN surgery,PSEP parameters of the IONM group were recorded and analyzed. The differences in PE-related outcome measurements between the perioperative period and 3 months'postoperation were compared for the PE patients, and the differences in effectiveness and complications between the IONM group and the non-IONM group were compared. The results showed that the average latency of the PSEP in the PE group was shorter than that in the normal group under both natural and GA states(P<0.001).Three months after surgery, the significant effective rates in the IONM and non-IONM groups were 63.6%and 34.0%,respectively(P<0.01),and the difference in complications between the two groups was significant(P<0.05).IONM might be useful in improving the short-term therapeutic effectiveness and reducing the complications of SDN. | Qing-Lai Tang Tao Song You-Feng Han Bai-Bing Yang Jian-Huai Chen Zhi-Peng Xu Chun-Lu Xu Yang Xu Wen Yu Wei Qiu Jiong Shi En-Si Zhang Yu-Tian Dai | 2023 | Asian Journal of Andrology2023,25,1: | 0
S19W,T27W,and N33OY mutations in ACE2 enhance SARS-CoV-2 S-RBD binding toward both wild-type and antibody-resistant viruses and its molecular basis显示文摘SARS-CoV-2 recognizes, via its spike receptor-binding domain(S-RBD),human angiotensin-converting enzyme 2(ACE2)to initiate infection.Ecto-domain protein of ACE2 can therefore function as a decoy. Here we show that mutations of S19W,T27W,and N330Y in ACE2 could individually enhance SARS-CoV-2 S-RBD binding.Y330 could be synergistically combined with either W19 or W27,whereas W19 and W27 are mutually unbeneficial. The structures of SARS-CoV-2S-RBD bound to the ACE2 mutants reveal that the enhan ced binding is mainly con tributed by the van der Waals interactio ns mediated by the aromatic side-chai ns from W19,W27,and Y330.While Y330 and W19/W27 are distantly located and devoid of any steric interference,W19 and W27 are shown to orient their side-chains toward each other and to cause steric conflicts, explai ning their in compatibility. Finally, using pseudotyped SARS-CoV-2 viruses,we dem on strate that these residue substitutions are associated with dramatically improved entry-inhibition efficacy toward both wild-type and antibody-resistant viruses.Taken together, our biochemical and structural data have delineated the basis for the elevated S-RBD binding associated with S19W,T27W,and N330Y mutations in ACE2,paving the way for potential application of these mutants in dinical treatment of COVID-19. | Fei Ye Xi Lin Zimin Che Fanli Yang Sheng Lin Jing Yang Hua Chen Honglu Sun Lingling Wang Ao Wen Xindan Zhang Yushan Dai Yu Cao Jingyun Yang Guobo Shen Li Yang Jiong Li Zhenling Wang Wei Wang Xiawei Wei Guangwen Lu | 2021 | Signal Transduction and Targeted Therapy2021,6,10: | 0
A 3D microseismic data-driven damage model for jointed rock mass under hydro-mechanical coupling conditions and its application显示文摘Rock mass is a fractured porous medium usually subjected to complex geostress and fluid pressure simultaneously. Moreover, the properties of rock mass change in time and space due to mining-induced fractures. Therefore,it is always challenging to accurately measure rock mass properties. In this study,a three-dimensional(3D)microseismic(MS)data-driven damage model for jointed rock mass under hydro-mechanical coupling conditions is proposed. It is a 3D finite element model that takes seepage, damage and stress field effects into account jointly.Multiple factors(i.e.joints,water and microseismicity)are used to optimize the rock mass mechanical parameters at different scales. The model is applied in Shirengou iron mine to study the damage evolution of rock mass and assess the crown pillar stability during the transition from open-pit to underground mining. It is found that the damage pattern is mostly controlled by the structure, water and rock mass parameters. The damage pattern is evidently different from the two-dimensional result and is more consistent with the field observations. This difference is caused by the MS-derived damage acting on the rock mass.MS data are responsible for gradually correcting the damage zone, changing the direction in which it expands, and promoting it to evolve close to reality. For the crown pillar, the proposed model yields a more trustworthy safety factor. In order to guarantee the stability of the pillar,it is suggested to take waterproof and reinforcement measures in areas with a high degree of damage. | Jingren Zhou Jinfu Lou Jiong Wei Feng Dai Jiankang Chen Minsi Zhang | 2023 | Journal of Rock Mechanics and Geotechnical Engineering2023,15,4: | 0
