1Identification and characterization of the cellular subclones that contribute to the pathogenesis of mantle cell lymphoma显示文摘Mantle cell lymphoma(MCL)is a B-cell malignancy with poor clinical outcome and undefined pathogenesis.Development of clinically relevant cellular models for MCL research is an urgent need.Our preliminary observations lead the development of two novel hypotheses that we tested in this study:1.multicellular spheroid might be a unique growth mode of earlystage cells in MCL;2.MCL might be a polyclonal tumor.We made the following original observations that have not been reported:First,we have provided a new experiment method for enriching MCL early-stage cells and characterized the spheroid mode of growth as a unique feature of early-stage MCL cells in cell line as well as in clinical samples.Second,we have established a clinically relevant cellular model of MCL,the JeKo-1-spheroid cell line,that was highly enriched in early-stage sub-clones.JeKo-1-spheroid cells and the spheroid growing cells enriched from MCL patients exhibited comparably enhanced tumorigenic abilities and similar biological features.Third,Immunophenotypic analysis has revealed that MCL may be derived from precursor-B(pre-B),immature-B and mature-B cells,not only the mature-B cells as WHO classified in 2016.Fourth,MCL may be a polyclonal disease composed of CD19e/IgMe,CD19 e/IgMt,CD19t/IgMt three sub-clones,of which the CD19e/IgMt sub-clone might be the dominant sub-clone with the strongest tumorigenic ability.Fifth,CD19t/IgMe that differentiates MCL and normal B cells may represent a new marker for MCL early detection,minor residual disease monitoring after therapies and prognosis.Junling Tang Li Zhang Tiejun Zhou Zhiwei Sun Liangsheng Kong Li Jing Hongyun Xing Hongyan Wu Yongli Liu Shixia Zhou Jingyuan Li Mei Chen Fang Xu Jirui Tang Tao Ma Min Hu Dan Liu Jing Guo Xiaofeng Zhu Yan Chen Ting Ye Jianyu Wang Xiaoming Li H.Rosie Xing 2019Genes & Diseases2019,6,4:2
2Expression of MxA Protein in Triple Negative Breast Cancer and its Relationship with Prognosis显示文摘Objective:To explore the expression of human myxovirus resistance protein A(MxA)in triple negative breast cancer(TNBC)and its relationship with prognosis.Methods:144 cases of TNBC confirmed by pathology before or after surgery from January 2014 to January 2017 in the First Central Hospital of Baoding City were retrospectively collected.Western blotting was used to detect the expression of MxA protein in TNBC and adjacent breast tissues.According to the expression of MxA protein,144 TNBC patients were divided into low MxA protein expression group(n=91)and MxA protein high expression group(n=53)for subsequent comparison of prognosis of patients in between these two groups.Results:The expression of MxA protein in TNBC tissue was lower than that of adjacent breast tissue,and the difference was statistically significant(P<0.05).The patients in high MxA expression group had higher loco-regional recurrence-free rate,disease-free survival(DFS)rate,and overall survival(OS)rate than those in low MxA expression group for 3 years.On the other hand,the distant metastasis rate was lower in the high expression group compared to that in the low MxA expression group,and the difference was statistically significant(P<0.05).Conclusion:In triple-negative breast cancer,high MxA expression is a potential predictor of TNBC prognosis.Jinmei Li Jirui Sun Hong Chen Qiushuang Ma Jinku Zhang 2020Proceedings of Anticancer Research2020,4,3:0

5Macrophage CCL22 Secretion Under Hypoxic Conditions Promotes the Metastasis of Triple-Negative Breast Cancer显示文摘Objective:To explore the mechanism by which macrophages secrete CCL22 to promote the metastasis of triple-negative breast cancer under hypoxic conditions.Methods:20 ng/mL mass concentration of phorbol 12-myristate 13-acetate(PMA)cell culture medium,4′,6-diamidino-2-phenylindole(DAPI),dimethyl sulfoxide(DMSO),trypsin digestion solution,CCL18 Kit,Interleukin(IL)-10 Kit,CCL17 Kit,CCL22 Kit,TRIzol^(TM)Reagent Reverse Transcriptase Polymerase Chain Reaction(RT-PCR)Kit,triple-negative breast cancer cells MDA-MB-231 and BT-549,as well as other reagents were used to culture triple-negative breast cancer(TNBC)cells MDA-MB-231 and BT-549 as well as human mononuclear cells THP-1,analyze and observe the metastasis of triple-negative breast cancer cells to the lungs,the secretion of CCL22,the migration of triple-negative breast cancer,and the situation of CCR4.Results:Compared with normal tumor-associated macrophage(TAM),hypoxic TAM further promoted the migration of tumor cells.The number of tumor metastases in the lungs,induced by hypoxic TAM,was significantly higher than that of normoxic TAM.Hypoxia can significantly stimulate the expression of CCL22.CCL22 can significantly promote the migration of MDA-MB-231 and BT-549 cells.The expressions of CCR3,CCR4,and CCR5 in tumor tissues were significantly increased compared with normal tissues,in which CCR4 showed the most significant increase.Conclusion:TAM cultured under hypoxia significantly enhanced the migration ability of TNBC cells and promoted the metastasis of cancer cells to the lungs in vivo.The hypoxic condition induced TAM to secrete CCL22;the expression of CCL22 receptor,CCR4,in breast cancer tissues was significantly higher than that in normal tissues.Wenming Zhao Bingjuan Zhou Jirui Sun Haizhi Qiao Jinmei Li Qiushuang Ma Xu Wang Jinku Zhang 2022Proceedings of Anticancer Research2022,6,6:0

7Mechanism of Ginsenoside Rh1 in Regulating Breast Cancer Cell Extravasation Based on CCL20- CCR6显示文摘Objective:To explore the regulatory mechanism of ginsenoside Rh1 on breast cancer cell extravasation based on CCL20-CCR6.Methods:In 2021,a total of 34 patients with breast cancer were treated in Baoding First Central Hospital.During hospitalization,pathological examinations were performed,and all the patients were diagnosed with invasive ductal carcinoma.Out of the 34 cases,16 cases were found to have CCR6 expression in breast cancer tissues,in which they were recorded as the CCR6 expression group,whereas 18 cases did not have CCR6 expression;these cases were recorded as the CCR6 non-expression group.During the same period,21 normal patients were selected as the control group.The peripheral blood CCL20 level and the expression of CCR6 on the surface of CD3+T lymphocytes were analyzed.The extracts of cancer cells were collected,purified,and cultured,and the effect of ginsenoside Rh1 on the invasion and metastasis of breast cancer cells was analyzed.Results:The peripheral blood CCL20 level in the CCR6 expression group was significantly higher than that in the CCR6 non-expression group and the control group,in which p<0.05,indicating that the difference was statistically significant;at 12,24,and 48 hours,the cell survival rate of each dose group was significantly higher than that of the blank control group and the dimethyl sulfoxide(DMSO)group(p<0.05).At 48 hours,comparing the low-dose group with the high-dose group,the cell survival rate significantly decreased(p<0.05).Compared with the blank control group and DMSO group,the invasion ability of breast cancer cells could be reduced in both,high-and medium-dose groups,where p<0.05,indicating that the difference was statistically significant.Conclusion:CCL20 may play a role in the pathogenesis of certain breast cancers,and ginsenoside Rh1 can effectively regulate the invasion and migration of breast cancer cells.Wenming Zhao Binjuan Zhou Jirui Sun Haizhi Qiao Jinmei Li Shuangqiu Ma Xu Wang Jinku Zhang 2022Proceedings of Anticancer Research2022,6,5:0
8A telomere-to-telomere genome assembly of Zhonghuang 13,a widely-grown soybean variety from the original center of Glycine max显示文摘Soybean(Glycine max)stands as a globally significant agricultural crop,and the comprehensive assembly of its genome is of paramount importance for unraveling its biological characteristics and evolutionary history.Nevertheless,previous soybean genome assemblies have harbored gaps and incompleteness,which have constrained in-depth investigations into soybean.Here,we present Telomere-to-Telomere(T2T)assembly of the Chinese soybean cultivar Zhonghuang 13(ZH13)genome,termed ZH13-T2T,utilizing PacBio Hifi and ONT ultralong reads.We employed a multi-assembler approach,integrating Hifiasm,NextDenovo,and Canu,to minimize biases and enhance assembly accuracy.The assembly spans 1,015,024,879 bp,effectively resolving all 393 gaps that previously plagued the reference genome.Our annotation efforts identified 50,564 high-confidence protein-coding genes,707 of which are novel.ZH13-T2T revealed longer chromosomes,421 not-aligned regions(NARs),112 structure variations(SVs),and a substantial expansion of repetitive element compared to earlier assemblies.Specifically,we identified 25.67 Mb of tandem repeats,an enrichment of 5S and 48S rDNAs,and characterized their genotypic diversity.In summary,we deliver the first complete Chinese soybean cultivar T2T genome.The comprehensive annotation,along with precise centromere and telomere characterization,as well as insights into structural variations,further enhance our understanding of soybean genetics and evolution.Anqi Zhang Tangchao Kong Baiquan Sun Shizheng Qiu Jiahe Guo Shuyong Ruan Yu Guo Jirui Guo Zhishuai Zhang Yue Liu Zheng Hu Tao Jiang Yadong Liu Shuqi Cao Shi Sun Tingting Wu Huilong Hong Bingjun Jiang Maoxiang Yang Xiangyu Yao Yang Hu Bo Liu Tianfu Han Yadong Wang 2024The Crop Journal2024,12,1:0
9Clinical Value of cfDNA Content in Peripheral Blood of Patients with Triple-Negative Breast Cancer显示文摘Objective:To explore the value of circulating free(cfDNA)content in the clinical diagnosis and treatment of triple-negative breast cancer(TNBC).Methods:A total of 39 TNBC patients,45 non-TNBC patients,and 50 healthy individuals admitted to the Baoding First Central Hospital during 2019-2022 were recruited.The clinical data,peripheral blood cfDNA concentration,and clinicopathological indicators of the patients were observed and analyzed.Results:The difference in clinical indicators such as age,age range,tumor size,clinical stage,and lymph node metastasis between patients with TNBC and non-TNBC was insignificant(P>0.05).The cfDNA concentrations(ng/mL)of the TNBC group,non-TNBC group,and healthy group were 24.12±4.98,15.36±4.12,and 3.12±1.02,respectively,and they are statistically different(P<0.05).The difference in cfDNA concentration was insignificant between TNBC patients with tumors≤2 cm and>2 cm(P>0.05)but was significant between TNBC patients with clinical stages I+II and III+IV(P<0.05).The cfDNA concentration in TNBC patients with lymph node metastasis was significantly higher than those without lymph node metastasis(P<0.05).Conclusion:cfDNA has an important application value in the diagnosis and treatment of breast cancer.By detecting the cfDNA level and its gene variation,valuable information about the progress and treatment effects of breast cancer can be obtained.This non-invasive detection method has a wide range of applications and can be used for early screening,auxiliary diagnosis,efficacy evaluation,and recurrence monitoring of breast cancer.Jirui Sun Qiushuang Ma Hong Chen Xing Zhou Bingjuan Zhou Jinku Zhang 2023Proceedings of Anticancer Research2023,7,4:0
10The Expression and Significance of CyclinD2 and Bcl-2 in Diffuse Large B-cell Lymphoma显示文摘Objective:To study the expression and significance of cyclinD2 and Bcl-2 in diffuse large B-cell lymphoma.Methods:This project used immunohistochemical methods to detect the expression of cyclinD2 and Bcl-2 in 120 cases of diffuse large B-cell lymphoma and 80 cases of reactive hyperplasia of lymphoid tissue.The materials were collected from the hospital from January 2018 to 2020.In March 2015,120 patients had lymphoma tissue removed during the month of surgery.The postoperative pathological diagnosis was DLBCL.Another 80 cases of lymphoid hyperplasia(RLH)tissues were selected as controls.Results:CyclinD2 and BCL-2 expression were not statistically different in patients with diffuse large B-cell lymphoma in different ages,genders,locations,tissue types,and degree of differentiation;but statistically significant in different Ann Arbor stages,immunotypes,IPI index and first treatment efficacy.Conclusion:This research not only has important theoretical value,but also important economic value and social significance.Qiushuang Ma Huijuan Geng Yafei Zhao Jirui Sun Xing Zhou Haiying Yao Jinku Zhang 2020Proceedings of Anticancer Research2020,4,6:0

13Detection and Clinical Significance of Peripheral Blood CTC, CA125, CA153, and CEA Levels in Patients with Ductal Carcinoma in situ显示文摘Objective:To explore the detection and clinical significance of peripheral blood CTC,CA125,CA153,and CEA levels in patients with DCIS.Methods:210 patients who received surgical treatment for breast cancer in our hospital from January 2019 to December 2022 were selected as the research subjects.According to the postoperative pathology,100 cases were divided into breast cancer and 110 benign breast tumor groups.One hundred ten healthy patients undergoing physical examination during the same period were selected as the control group.CA153,CA125,and CEA levels were observed in the three groups.Results:The levels of CA153,CA125,and CEA in the breast cancer group were significantly higher than those in the benign breast tumor group and the control group(P<0.05);the levels of CA153,CA125,and CEA in the benign breast tumor group were all higher than those in the control group,but the differences were not statistically significant;among the three tumor markers,CA153 has the highest sensitivity at 39.00%,CA125 has the second highest sensitivity at 18.00%,and CEA has the lowest sensitivity at 17.00%;The sensitivity of the two joint tests of CA153+CA125,CA153+CEA,and CA125+CEA for the diagnosis of breast cancer were 50.00%,48.00%,and 26.00%respectively;the sensitivity of the three joint tests is the highest,reaching 53.00%,while the specificity of the joint tests was lower than the individual tests.Conclusion:Detection of peripheral blood CTC,CA125,CA153,and CEA levels has specific reference significance for the treatment and prognosis of DCIS patients.Jirui Sun Qiushuang Ma Hong Chen Xing Zhou Jinku Zhang 2023Proceedings of Anticancer Research2023,7,5:0

17The Expression and Significance of CyclinD2,MPGES-1,Bcl2 in Diffuse Large B-cell Lymphoma显示文摘Objective:To study the expression and significance of cell cycle proteins CyclinD2,mPGES-1,Bcl2 in diffuse large B-cell lymphoma.Methods:Choose lymphoma and sexually hyper-plastic lymphoid tissues as control.Immunohistoc-hemical methods were used to detect the expression of CyclinD2,mPGES-1,and Bcl2,and to compare the positive expression rates of CyclinD2,MPGES-1 and Bcl2 in diffuse large B-cell lymphoma and reactive proliferative lymphoid tissues to compare their diffusion formation.B-cell lymphoma was analyzed for its clinicopathological features.Results:The positive expression rate of CyclinD2,mPGES-1 and Bcl2 in diffuse large B-cell lymphoma is higher than that in reactive proliferative lymphoid tissue,and the difference between the two is statistically significant.There was no statistical difference in CyclinD2,mPGES-1 and Bcl2 in diffuse large B-cell lymphoma between patients according to the age,sex,location,tissue type and degree of differentiation.Conclusion:CyclinD2,mPGES-1 and Bcl2 are highly expressed in patients with diffuse large B-cell lymphoma,and can be used as reference indicators for evaluating the malignant degree and efficacy of dysplasia.Qiushuang Ma Peng Lin Bingjuan Zhou Yafei Zhao Ling Li Jinku Zhang Jirui Sun 2021Proceedings of Anticancer Research2021,5,2:0
18Clinical Value of Peripheral Blood Circulating Tumor Cells and Cell-Free DNA Combined Detection in Triple-Negative Breast Cancer显示文摘Objective:To determine the clinical value of combined detection of circulating tumor cells(CTCs)and cell-free DNA(cfDNA)in peripheral blood of patients with triple-negative breast cancer.Method:41 patients with breast cancer admitted to the First Central Hospital of Baoding from January 2020 to December 2021 were selected and recruited into the experimental group,42 patients with benign breast cancer admitted during the same period were recruited into the conditional control group,and 41 healthy patients admitted during the same period were recruited into the blank control group.The positive rate of peripheral blood CTCs,the level of cfDNA,and the diagnostic efficacy of peripheral blood CTCs,cfDNA alone and the combination thereof for breast cancer were analyzed.Result:The positive rates of peripheral blood CTCs in the experimental group,the conditional control group,and the blank control group were 43.90%,11.90%,and 9.74%,respectively,and there was significant difference among the groups.The levels of cfDNA in peripheral blood of the experimental group,the conditional control group,and the blank control group were 0.26±0.08 bp,0.17±0.03 bp,and 0.15±0.04 bp,respectively,which were statistically significant.The detection levels of 100 bp hTERT/ng mT1 and 241 bp hTERT/ng-mT1 in the experimental group were significantly higher than those in the conditional control group and the blank control group.The accuracy of peripheral blood CTCs detection in the three groups was 66.21%,the accuracy of cfDA241 bp/100 hp hTERT detection was 80.41%,and the accuracy of combined detection of peripheral blood CTCs and cfDNA was 94.03%.Conclusion:The clinical application of peripheral blood CTCs combined with cfDNA level detection can increase detection accuracy,provide data support for clinicians,and improve the clinical diagnostic effect of triple-negative breast cancer.Jirui Sun Shuangqiu Ma Hong Chen Huijuan Geng Jinku Zhang 2022Proceedings of Anticancer Research2022,6,3:0
19Evaluation of Clinical Application of Chemiluminescence and Real-time,Fluorescence-based Quantitative PCR in Diagnosis of Epstein-Barr Virus lnfection显示文摘Objective:To compare the effects of clinical application of chemiluminescence and real-time,fluorescence-based quantitative PCR in the detection Epstein-Barr virus(EBV).Methods:The data of chemiluminescence and real-time fluorescent quantitative PCR.fromipaEsfwo were suspected of being infectea w1tn rito1 roro January 2016 to January 2019 in our hospital were analyzed.The specific stage of EBV infection was analyzed,and the differences in results of the two detection methods were compared.Results:Chemiluminescence method was used to detect EBV infection during the active phase.The sensitivity of the chemiluminescence method was 76.7%(56/73)and the real-time quantitative PCRmethod was 90.4%(66/73).There was a statistical difference between the two detection methods(P<0.05).Conclusion:There was no statistical difference in positive predictive values between the chemiluminescence method and the real-time,fluorescence-based quantitative PCR method in the detection of EBV infection,but the sensitivity of chemiluminescence method is slightly lower than the real-time quantitative PCRmethod.It is noteworthy that chemiluminescence method is convenient and fast while the real-time,fluorescence-based quantitative PCR method is more accurate,which can provide a more accurate reference for clinical treatment.Huijuan Geng Yan Wang Hao Wang Jirui Sun Hui Tang 2020Journal of Clinical and Nursing Research2020,4,4:0
