| 1 | BLT1 in dendritic cells promotes Th1/Th17 differentiation and its deficiency ameliorates TNBS-induced colitis显示文摘Leukotriene B4(LTB4)synthesis is enhanced in the colonic mucosa in patients with inflammatory bowel disease(IBD).BLT1,a highaffinity receptor for LTB4,exhibits no effect on the progression of dextran sodium sulfate(DSS)-induced colitis,which mostly relies on innate immunity.Here,we reported that BLT1 regulates trinitrobenzene sulfonic acid(TNBS)-induced colitis,which reflects CD4+T-cell-dependent adaptive immune mechanisms of IBD.We found that BLT1 signaling enhanced the progression of colitis through controlling the production of proinflammatory cytokines by dendritic cells(DCs)and modulating the differentiation of Th1 and Th17.BLT1−/−mice displayed an alleviated severity of TNBS-induced colitis with reduced body weight loss and infiltrating cells in the lamina propria.BLT1 deficiency in DCs led to reduced production of proinflammatory cytokines,including IL-6,TNF-α,and IL-12,and these results were further confirmed via treatment with a BLT1 antagonist.The impaired cytokine production by BLT1−/−DCs subsequently led to reduced Th1 and Th17 differentiation both in vitro and in vivo.We further performed a conditional DC reconstitution experiment to assess whether BLT1 in DCs plays a major role in regulating the pathogenesis of TNBS-induced colitis,and the results indicate that BLT1 deficiency in DCs also significantly reduces disease severity.The mechanistic study demonstrated that BLT1-regulated proinflammatory cytokine production through the Gαiβγsubunit-phospholipase Cβ(PLCβ)-PKC pathway.Notably,we found that treatment with the BLT1 antagonist also reduced the production of proinflammatory cytokines by human peripheral blood DCs.Our findings reveal the critical role of BLT1 in regulating adaptive immunity and TNBS-induced colitis,which further supports BLT1 as a potential drug target for adaptive immunity-mediated IBD. | Jinfeng Zhou Weiming Lai Wanjie Yang Juping Pan Hu Shen Yingying Cai Cuixia Yang Ningjia Ma Yue Zhang Ru Zhang Xin Xie Zhongjun Dong Yuan Gao Changsheng Du | 2018 | Cellular & Molecular Immunology2018,15,12: | 9 |
| 2 | Tailoring interphase structure to enable high-rate, durable sodium-ion battery cathode显示文摘Na-based layered transition metal oxides with O_(3)-type structure have been considered to be promising cathodes for Na-ion batteries. However, the intrinsically limited Na-ion conductivity induced by the Otype Na-coordinate environment compromises their rate and cycle capability, hindering their practical application. Here, we report an interphase-structure tailoring strategy that improves the electrochemical properties of O_(3)-type layered cathodes achieved through surface coating and doping processes.Specifically, a Zr-doped interphase structure is designed in the model compound NaNi_(1/3)Mn_(1/3)Fe_(1/3)O_(2) using the ionic conductor Na_(3)Zr_(2)Si_(2)PO_(12) as the surface coating material and Zr-dopant provider. We discover that the modified NaNi_(1/3)Mn_(1/3)Fe_(1/3)O_(2)cathode shows a stable Na-storage structure as well as an enhanced rate/cycle capability. Combined with theoretical calculations, it is suggested that the superior electrochemical performances originate from the Zr-doped interphase structure, which has an enlarged Na layer spacing that forms favorable Na-ion diffusion channels. | 4 | Surface modification of polyolefin separators for lithium ion batteries to reduce thermal shrinkage without thickness increase显示文摘Surface chemical modification of polyolefin separators for lithium ion batteries is attempted to reduce the thermal shrinkage, which is important for the battery energy density. In this study, we grafted organic/inorganic hybrid crosslinked networks on the separators, simply by grafting polymerization and condensation reaction. The considerable silicon-oxygen crosslinked heat-resistance networks are responsible for the reduced thermal shrinkage. The strong chemical bonds between networks and separators promise enough mechanical support even at high temperature. The shrinkage at 150 C for 30 min in the mechanical direction was 38.6% and 4.6% for the pristine and present graft-modified separators, respectively. Meanwhile, the grafting organic-inorganic hybrid crosslink networks mainly occupied part of void in the internal pores of the separators, so the thicknesses of the graft-modified separators were similar with the pristine one. | 7 | Optimal Groundwater Development in Coastal Aquifers Near Beihai, China显示文摘Groundwater resources occur in a multi aquifer system in the alluvial coastal plain near Beihai, China. The aquifers receive recharge from precipitation, canal and reservoir infiltration, and discharge through subterranean drainage into the sea and through artificial pumping. A quasi three dimensional finite element model has been used to simulate the spatial and temporal distribution of groundwater levels in the aquifers. Various input parameters were considered in the simulation model. A linear optimization model has been developed for groundwater development within the coastal aquifers. The objective function of the model is to maximize the total groundwater pumpage from the confined aquifer. The control of sea water intrusion is examined by the restriction of the water levels at points along the coast and of the pumping rates in coastal management cells. The response matrix used in the optimization model was generated from the simulation model by forecasting drawdown produced by pumping at a unit impulse discharge. | 8 | Effect of block length on the self-assembly of end-capping perfluoroalkyl moieties on the polymer surface显示文摘Most research on copolymers with fluorinated monomers has focused on the relationship between fluorinated monomer content and the corresponding surface structure. However,the influence of the non-fluorinated block on the surface structure of the copolymer film is unknown. Various molecular weight poly(butyl methacrylates) (PBMA) end-capped with 2-perfluorooctylethyl methacrylate (FMA) units (PBMA-ec-FMA) have been synthesized by atom transfer radical polymerization (ATRP). The effect of the PBMA block length on the surface structure and properties of the polymers both in the solid state and in solution was investigated using various techniques. X-ray photoelectron spectroscopy (XPS),sum frequency generation (SFG) vibrational spectroscopy and X-ray diffraction (XRD) analyses indicated that longer PBMA blocks enhanced both the enrichment of the fluorinated moieties and the order of the packing orientation of the perfluoroalkyl side chains on the surface. This enhancement was attributed mainly to the molecular aggregate structure of the end-capped polymers with long PBMA blocks in the solution and to the interfacial structure at the air/liquid interface,which favors the -(CF2)7CF3 moieties self-assembling on the polymer surface during film formation. | 10 | Nonpharmaceutical interventions contribute to the control of COVID-19 in China based on a pairwise model显示文摘Nonpharmaceutical interventions(NPIs),particularly contact tracing isolation and household quarantine,play a vital role in effectively bringing the Coronavirus Disease 2019(COVID-19)under control in China.The pairwise model,has an inherent advantage in characterizing those two NPIs than the classical well-mixed models.Therefore,in this paper,we devised a pairwise epidemic model with NPIs to analyze COVID-19 outbreak in China by using confirmed cases during February 3rde22nd,2020.By explicitly incorporating contact tracing isolation and family clusters caused by household quarantine,our model provided a good fit to the trajectory of COVID-19 infections.We calculated the reproduction number R=1.345(95%CI:1.230-1.460)for Hubei province and R=1.217(95%CI:1.207-1.227)for China(except Hubei).We also estimated the peak time of infections,the epidemic duration and the final size,which are basically consistent with real observation.We indicated by simulation that the traced high-risk contacts from incubated to susceptible decrease under NPIs,regardless of infected cases.The sensitivity analysis showed that reducing the exposure of the susceptible and increasing the clustering coefficient bolster COVID-19 control.With the enforcement of household quarantine,the reproduction number R and the epidemic prevalence declined effectively.Furthermore,we obtained the resumption time of work and production in China(except Hubei)on 10th March and in Hubei at the end of April 2020,respectively,which is broadly in line with the actual time.Our results may provide some potential lessons from China on the control of COVID-19 for other parts of the world. | Xiao-Feng Luo Shanshan Feng Junyuan Yang Xiao-Long Peng Xiaochun Cao Juping Zhang Meiping Yao Huaiping Zhu Michael Y.Li Hao Wang Zhen Jin | 2021 | Infectious Disease Modelling2021,6,1: | 2 |
