1A feasible,economical,and accurate analytical method for simultaneous determination of six alkaloid markers in Aconiti Lateralis Radix Praeparata from different manufacturing sources and processing ways显示文摘Aconiti Lateralis Radix Praeparata(Fuzi) is a commonly used traditional Chinese medicine in clinic for its potency in restoring yang and rescuing from collapse. Aconiti alkaloids, mainly including monoester-diterpenoidaconitines(MDAs) and diester-diterpenoidaconitines(DDAs), are considered to act as both bioactive and toxic constituents. In the present study, a feasible, economical, and accurate HPLC method for simultaneous determination of six alkaloid markers using the Single Standard for Determination of Multi-Components(SSDMC) method was developed and fully validated. Benzoylmesaconine was used as the unique reference standard. This method was proven as accurate(recovery varying between 97.5%-101.8%, RSD < 3%), precise(RSD 0.63%-2.05%), and linear(R > 0.999 9) over the concentration ranges, and subsequently applied to quantitative evaluation of 62 batches of samples, among which 45 batches were from good manufacturing practice(GMP) facilities and 17 batches from the drug market. The contents were then analyzed by principal component analysis(PCA) and homogeneity test. The present study provided valuable information for improving the quality standard of Aconiti Lateralis Radix Praeparata. 3CDH13 Genetic Polymorphisms, Adiponectin and Ischemic Stroke: a Chinese Family-based Sib-pair Study显示文摘Objective To understand the relationships between CDH13(T-cadherin) genetic polymorphisms, adiponectin levels and ischemic stroke, and possible interactions between CDH13 polymorphisms and other risk factors. Methods We recruited 342 Chinese ischemic stroke sib pairs. We genotyped rs4783244 and rs7193788 on CDH13 using time-of-flight mass spectrometry genotyping technology and measured total and high-molecular weight(HMW) adiponectin levels. We investigated associations between SNPs and ischemic stroke, and interactions between SNPs and other risk factors using multi-level mixed-effects regression model. Results In individuals without ischemic stroke, CDH13 rs4783244 was associated with total adiponectin levels(per T: Coef =-0.257, P = 0.001). CDH13 rs7193788 was associated with total adiponectin levels(per A: Coef =-0.221, P = 0.001) and HMW adiponectin levels(per A: Coef =-0.163, P = 0.003). rs7193788 was significantly associated with ischemic stroke(GA/AA vs. GG: OR = 1.55, 95% CI: 1.07 to 2.24, P = 0.020) after Bonferroni correction(α = 0.025). There was an interaction between rs7193788 and diabetes(P = 0.036). Compared to diabetes-free individuals with rs7193788 GG genotype, diabetes patients with rs7193788 GA/AA genotypes had higher risks for ischemic stroke(OR = 2.64, 95% CI: 1.58-4.40, P < 0.001). Conclusion CDH13 genetic polymorphisms are associated with adiponectin levels and ischemic stroke. 4Human podocyte injury in the early course of hypertensive renal injury显示文摘BACKGROUND Hypertension is prevalent in the general population and is regarded as the second leading cause of renal damage and dysfunction,outnumbered only by diabetes.However,the mechanisms remain unclear.AIM To investigate podocyte injury induced by hypertension in the early course without massive proteinuria or renal dysfunction.METHODS The hypertension group comprised 18 patients with hypertension accompanied by microalbuminuria,diagnosed with hypertensive renal injury according to biopsy results.For a comparison of pathological changes in renal tissue,control group 1 comprised 10 healthy volunteers,and control group 2 comprised 16 patients who underwent surgery for renal trauma.RESULTS The hypertension group had significantly higher blood pressure(P=0.000)and microalbuminuria(P=0.000)compared with control group 1.In the hypertension group,urinary podocytes were detected following positive staining of podocytespecific nephrin and/or CD2-associated protein(CD2AP)in urine sediment.Podocyte foot process fusion and a significant decrease in nephrin and/or CD2AP expression in glomeruli were observed in the hypertension group compared with control group 2.This indicated that hypertension caused podocyte injury and detachment from the glomerular basement membrane,which was consistent with urinary detection of podocytes.CONCLUSION Our results suggest that podocyturia appears early in the course of hypertensive renal injury,and may be a sensitive marker for early prediction of hypertensive renal injury.Da Sun Jiao-Jiao Wang Wei Wang Juan Wang Li-Ning Wang Li Yao Ying-Hui Sun Zi-Long Li 2019World Journal of Clinical Cases2019,7,22:3
5Diameter-controlled growth of aligned single-walled carbon nanotubes on quartz using molecular nanoclusters as catalyst precursors显示文摘Molecular nanoclusters containing Fe and Mo atoms have been used as catalyst precursors for the growth of single-walled carbon nanotubes (SWNTs) on stable temperature (ST)-cut quartz substrates by chemical vapor deposition. Attribute to the uniform catalyst nanoparticles and the confinement effect of the crystalline substrates, well-aligned SWNTs with narrow diameter distribution have been synthesized. Atomic force microscopy measurements show that the mean diameter of the nanotubes obtained by thermal decomposition of ethanol at 900°C is 0.76 ± 0.16 nm, which is the smallest among all reported results for aligned SWNTs. The mean diameter of the nanotubes increases with growth temperature. In addition to using identical nanoclusters as the catalyst precursors, the avoidance of annealing treatment of catalyst precursors is also a key point for obtaining SWNTs with controlled diameters. 6Quantitative Analysis of Eight Ginsenosides in Red Ginseng Using Ginsenoside Rg1 as Single Reference Standard显示文摘Objective:To develop a reversed-phase high-performance liquid chromatography method for the quantification of major ginsenosides in red ginseng(RG,the steamed and dried root of the cultivar of Panax ginseng C.A.Mey).Methods:A feasible method was developed in strict accordance with chromatographic properties of eight ginsenosides.Their contents could be unveiled with conventional external standard method,or as an alternative,using ginsenoside Rg1 as the single reference standard by means of seven conversion factors.Those parameters had been validated on different chromatographic columns and instruments.Results:Twenty-one batches of RG samples were determined.In addition,the chromatograms of RG and confusing species,including Panax ginseng,Panax quinquefolium,and Panax notoginseng,were apparently different.Conclusions:The method was proved to be efficient for the quality control of RG.Juan Da Qiu-Rong Wang Yan Wang Shuai Yao Yong Huang Wen-Long Wei Jian Liang Yao Shen Gerhard Franz De-An Guo 2021World Journal of Traditional Chinese Medicine2021,7,1:2

12Simple and robust differentiation of Ganoderma species by high performance thin-layer chromatography coupled with single quadrupole mass spectrometry QDa显示文摘In this study,a high performance thin-layer chromatography/single quadrupole mass spectrometry QDa(HPTLC-QDa)method for robust authentication of Ganoderma lucidum,a popular and valuable herbal medicine,has been developed.This method is simple and practical,which allows direct generation of characteristic mass spectra from the HPTLC plates automatically with the application of in situ solvent desorption interface.The HPTLC silica gel plates were developed with toluene-ethyl formate-formic acid(5:5:0.2,V/V)and all bands were transferred to QDa system directly in situ using 80%methanol with 0.1%formic acid as desorption solvent.The acquired HPTLC-QDa spectra showed that luminous yellow band b3,containing ganoderic acid B/G/H and ganodeneric acid B,the major active components of Ganoderma,could be found only in G.lucidum and G.lucidum(Antler-shaped),but not in G.sinense and G.applanatum.Moreover,bands b13 and b14 with m/z 475/477 and m/z 475/491/495,respectively,could be detected in G.lucidum(Antler-shaped),but not in G.lucidum,thus allowing simple and robust authentication of G.lucidum with confused species.This method is proved to be simple,practical and reproducible,which can be extended to analyze other herbal medicines.YAO Shuai ZHANG Jian-Qing HOU Jin-Jun HU Xiao-Su WANG Ling DA Juan RAO Wei WANG Dan-Dan HUANG Yong WU Wan-Ying GUO De-An 2021Chinese Journal of Natural Medicines2021,19,4:1

17Comparative proteomics analysis of pomegranate seeds on fruit maturation period(Punica granatum L.)显示文摘Seeds play a central role in the life cycle of plants. Seed hardness in pomegranates is of economic relevance, yet scarcely studied and poorly understood in China. In this study, we compared the proteomic differences between Zhongnonghong(soft-seeded) and Sanbai(hard-seeded) pomegranates. A total of 892 protein spots from both varieties were detected on two-dimensional electrophoresis gels(2-DE); 76 spots showed greater than a 1.5-fold or less than a 0.66-fold difference(P<0.05) in Zhongnonghong compared to Sanbai, of which 24 exhibited greater than a 2-fold change. Compared with Sanbai, Zhongnonghong possessed 14 up-regulated, and 10 down-regulated proteins. We identified and annotated 5 of these by using MALDI-TOF-TOF MS: pyruvate dehydrogenase(PDH) E1-β family protein(spot 4 609); alanine aminotransferase 2-like(ALT2L); mitochondrial glycine decarboxylase complex P-protein(spot 5 803); phosphofructokinase B(Pfk B)-type family of carbohydrate kinase(spot 8 411); and putative dna K-type molecular chaperone heat shock cognate protein 70(Hsc70)(spot 9 006). Of these, 3 proteins(spots 4 609, 5 608, 5 803) were hypothesized to play a role in the formation of seed hardness. The other two proteins(spots 8 411, 9 006) were theorized to play a role in protecting the seeds from adverse stress during periods of fruit maturation.
