1CDH13 Genetic Polymorphisms, Adiponectin and Ischemic Stroke: a Chinese Family-based Sib-pair Study显示文摘Objective To understand the relationships between CDH13(T-cadherin) genetic polymorphisms, adiponectin levels and ischemic stroke, and possible interactions between CDH13 polymorphisms and other risk factors. Methods We recruited 342 Chinese ischemic stroke sib pairs. We genotyped rs4783244 and rs7193788 on CDH13 using time-of-flight mass spectrometry genotyping technology and measured total and high-molecular weight(HMW) adiponectin levels. We investigated associations between SNPs and ischemic stroke, and interactions between SNPs and other risk factors using multi-level mixed-effects regression model. Results In individuals without ischemic stroke, CDH13 rs4783244 was associated with total adiponectin levels(per T: Coef =-0.257, P = 0.001). CDH13 rs7193788 was associated with total adiponectin levels(per A: Coef =-0.221, P = 0.001) and HMW adiponectin levels(per A: Coef =-0.163, P = 0.003). rs7193788 was significantly associated with ischemic stroke(GA/AA vs. GG: OR = 1.55, 95% CI: 1.07 to 2.24, P = 0.020) after Bonferroni correction(α = 0.025). There was an interaction between rs7193788 and diabetes(P = 0.036). Compared to diabetes-free individuals with rs7193788 GG genotype, diabetes patients with rs7193788 GA/AA genotypes had higher risks for ischemic stroke(OR = 2.64, 95% CI: 1.58-4.40, P < 0.001). Conclusion CDH13 genetic polymorphisms are associated with adiponectin levels and ischemic stroke. 6Designing specific bacterial 16S primers to sequence and quantitate plant endo-bacteriome显示文摘Plant endophytic bacteria colonize the internal tissues of plants and interact with plants closely.The past two decades have witnessed the increasing application of next-generation 16S r RNA gene sequencing in the investigation of bacterial communities.However,deciphering plant endo-bacterial communities by this method is difficult because of the co-amplification of massive plant organellar DNAs with bacterial 16S.Here,we designed polymerase chain reaction(PCR) primer sets,including799F/1107R,322F/796R,and 322F-Dr/796Rs(primer pair 322F/796R with a penultimate-base substitution in 322F),that can specifically amplify bacterial 16S from plant total DNAs.We computationally and experimentally evaluated the specificity,coverage,and accuracy of the newly designed primer sets.Both 799F/1107R and 322F-Dr/796Rs produced plant DNA-free 16S amplicon libraries or reduced plant DNA contamination to lower than 5% for the plant materials with extremely-low-abundance bacterial communities.The primer set 322F-A/796R was used through absolute quantitative PCR to quantitate the population size of rice leaf or root endo-bacteriome,which revealed 10^(6)–10^(7) and 10^(9)–10^(10) bacteria per gram fresh weight,respectively.These 16S primer sets and amplification methods enable the simple and inexpensive next-generation sequencing and quantification of plant endo-bacteriome,which will significantly advance studies on the plant-related microbiome.Liying Chen Mengting Zhang Da Liu Hongbo Sun Jianxiang Wu Yan Huo Xiaoying Chen Rongxiang Fang Lili Zhang 2022Science China(Life Sciences)2022,65,5:2

12Nucleic acids analysis显示文摘Nucleic acids are natural biopolymers of nucleotides that store, encode, transmit and express genetic information, which play central roles in diverse cellular events and diseases in living things. The analysis of nucleic acids and nucleic acids-based analysis have been widely applied in biological studies, clinical diagnosis, environmental analysis, food safety and forensic analysis.During the past decades, the field of nucleic acids analysis has been rapidly advancing with many technological breakthroughs.In this review, we focus on the methods developed for analyzing nucleic acids, nucleic acids-based analysis, device for nucleic acids analysis, and applications of nucleic acids analysis. The representative strategies for the development of new nucleic acids analysis in this field are summarized, and key advantages and possible limitations are discussed. 16Antagonistic MADS-box transcription factors SEEDSTICK and SEPALLATA3 form a transcriptional regulatory network that regulates seed oil accumulation显示文摘Transcriptional regulation is essential for balancing multiple metabolic pathways that influence oil accumulation in seeds.Thus far,the transcriptional regulatory mechanisms that govern seed oil accumulation remain largely unknown.Here,we identified the transcriptional regulatory network composed of MADS-box transcription factors SEEDSTICK(STK)and SEPALLATA3(SEP3),which bridges several key genes to regulate oil accumulation in seeds.We found that STK,highly expressed in the developing embryo,positively regulates seed oil accumulation in Arabidopsis(Arabidopsis thaliana).Furthermore,we discovered that SEP3 physically interacts with STK in vivo and in vitro.Seed oil content is increased by the SEP3 mutation,while it is decreased by SEP3 overexpression.The chromatin immunoprecipitation,electrophoretic mobility shift assay,and transient dual-luciferase reporter assays showed that STK positively regulates seed oil accumulation by directly repressing the expression of MYB5,SEP3,and SEED FATTY ACID REDUCER 4(SFAR4).Moreover,genetic and molecular analyses demonstrated that STK and SEP3 antagonistically regulate seed oil production and that SEP3 weakens the binding ability of STK to MYB5,SEP3,and SFAR4.Additionally,we demonstrated that TRANSPARENT TESTA 8(TT8)and ACYL-ACYL CARRIER PROTEIN DESATURASE 3(AAD3)are direct targets of MYB5 during seed oil accumulation in Arabidopsis.Together,our findings provide the transcriptional regulatory network antagonistically orchestrated by STK and SEP3,which fine tunes oil accumulation in seeds.Shuangcheng He Yuanchang Min Zijin Liu Fang Zhi Rong Ma Ankang Ge Shixiang Wang Yu Zhao Danshuai Peng Da Zhang Minshan Jin Bo Song Jianjun Wang Yuan Guo Mingxun Chen 2024Journal of Integrative Plant Biology2024,66,1:0
17Development of a pressure coring system for the investigation of deep underground exploration显示文摘To provide a more accurate evaluation of the scale of deep underground resources,a new pressure coring system was carefully developed and its strength and safety of the engineering were verified by theoretical analyses and numerical simulation.The designed pressure coring system can obtain cores with length of 3 m and diameter of 50 mm at 70 MPa.The results of the ball-drop operation experiment demonstrate that differential motion assembly can effectively cut a safety pin by applying a tensile force of 4852 N,and it can lift the core tube through the center pole to complete a series of mechanical actions to seal the pressure.Additionally,by maintaining pressures at 70 MPa for 30 min,the pressure sealing capacity of the system was proven.Furthermore,a core sample with a diameter of 50 mm was obtained through a core drilling experiment and the coring performance of the pressure coring system was verified.This study can not only enrich the existing onshore coring technology but also provide a theoretical guide and design criteria for the development of similar pressure coring systems to meet the demand for deeper underground exploration.Da Guo Ling Chen Zhongya Zhou Dingming Wang Yiwei Zhang Xun Yang Xin Fang Xiaojun Wang Heping Lu Lin Dai Heping Xie 2023International Journal of Mining Science and Technology2023,33,11:0
18Physical and emulsifying properties of pea protein:influence of combined physical modification by flaxseed gum and ultrasonic treatment显示文摘This study characterized and compared the physical and emulsifying properties of pea protein(PP)and its modified proteins(ultrasound treated-PP(PPU),flaxseed gum(FG)treated PP(PPFG)and ultrasound treated-PPFG(PPFGU)).The results showed FG triggered the formation of loosely attached complex with PP via physical modification under gentle magnetic stirring at pH 7.0,while ultrasound played an important role in reducing protein size,increasing surface hydrophobicity and molecular fluidity onto oil-water interface.So ultrasound further enhanced the interaction of PP with FG,and produced the PPFGU complex with smaller droplet size,higherζ-potential and lower turbidity.Further,combination of FG and ultrasound improved the physical properties of PP with higher viscosity,stiffer gels(defined as higher elastic modulus),stronger hydrophobic properties,better thermal stability,and fast protein absorption rate.Therefore,the PPFGU coarse emulsion performed highest emulsifying activity index(EAI)and emulsion stability index(ESI)that the stabilized nanoemulsion obtained smallest droplet size,higherζ-potential,and longest storage stability.The combination of FG and ultrasonic treatment will be an effective approach to improving the emulsifying property and thermal stability of PP,which can be considered as a potential plant-based emulsifier applied in the food industry.Jia Yang Fenghong Huang Qingde Huang Da Ma Yashu Chen Dengfeng Peng Xiao Yu Qianchun Deng Fang Geng 2023Food Science and Human Wellness2023,12,2:0
