| 2 | Pathogenicity of a currently circulating Chinese variant pseudorabies virus in pigs显示文摘AIM:To test the pathogenicity of pseudorabies virus(PRV)variant HN1201 and compare its pathogenicity with a classical PRV Fa strain.METHODS:The pathogenicity of the newly-emerging PRV variant HN1201 was evaluated by different inoculating routes,virus loads,and ages of pigs.The classical PRV Fa strain was then used to compare with HN1201 to determine pathogenicity.Clinical symptoms after virus infection were recorded daily and average daily body weight was used to measure the growth performance of pigs.At necropsy,gross pathology and histopathology were used to evaluate the severity of tissue damage caused by virus infection.RESULTS:The results showed that the efficient infection method of RPV HN1201 was via intranasal inoculation at 107 TCID50,and that the virus has high pathogenicity to 35-to 127-d old pigs.Compared with Fa strain,pigs infected with HN1201 showed more severe clinical symptoms and pathological lesions.Immunochemistry results revealed HN1201 had more abundant antigen distribution in extensive organs.CONCLUSION:All of the above results suggest that PRV variant HN1201 was more pathogenic to pigs than the classical Fa strain. | Qing-Yuan Yang Zhe Sun Fei-Fei Tan Ling-Hua Guo Yu-Zhou Wang Juan Wang Zhi-Yan Wang Li-Lin Wang Xiang-Dong Li Yan Xiao Ke-Gong Tian | 2016 | World Journal of Virology2016,5,1: | 31 |
| 3 | Neuroprotective effects of salidroside on focal cerebral ischemia/reperfusion injury involve the nuclear erythroid 2-related factor 2 pathway显示文摘Salidroside,the main active ingredient extracted from Rhodiola crenulata,has been shown to be neuroprotective in ischemic cerebral injury,but the underlying mechanism for this neuroprotection is poorly understood.In the current study,the neuroprotective effect of salidroside on cerebral ischemia-induced oxidative stress and the role of the nuclear factor erythroid 2-related factor 2(Nrf2)pathway was investigated in a rat model of middle cerebral artery occlusion.Salidroside(30 mg/kg)reduced infarct size,improved neurological function and histological changes,increased activity of superoxide dismutase and glutathione-S-transferase,and reduced malon-dialdehyde levels after cerebral ischemia and reperfusion.Furthermore,salidroside apparently increased Nrf2 and heme oxygenase-1 expression.These results suggest that salidroside exerts its neuroprotective effect against cerebral ischemia through anti-oxidant mechanisms and that activation of the Nrf2 pathway is involved.The Nrf2/antioxidant response element pathway may become a new therapeutic target for the treatment of ischemic stroke. | Jing Han Qing Xiao Yan-hua Lin Zhen-zhu Zheng Zhao-dong He Juan Hu Li-dian Chen | 2015 | Neural Regeneration Research2015,10,12: | 23 |
| 4 | Determination of Seven Major Ginsenosides in Different Parts of Panax quinquefolius L.(American Ginseng) with Different Ages显示文摘Ginsenosides Rg1, Re, Rb1, Rc, Rb2, Rb3, and Rd in different parts of the American ginseng plant were investigated. The extraction process was a pressurized microwave-assisted extraction(PMAE). The seven ginsenosides were separated and determined by high-performance liquid chromatography(HPLC) with a ultraviolet(UV) detector, at 203 nm. The experiment results showed significant variations in the individual ginsenoside contents of the American ginseng in different parts and ages of the plant. The results demonstrated that the leaves, root hairs, and rhizomes of Panax quinquefolius L. contained higher ginsenoside contents, followed by the main roots and stems. The leaves contained dramatically higher levels of ginsenoside Rg1, Rb3, and Rd than the other four parts. Higher contents of Rb1 and Re were present in the main roots, root hairs, and rhizomes. The amount of ginsenoside content in the stems was the lowest. The total content of the seven ginsenosides in main roots, root hairs and rhizomes increased with the age of the plant. | 6 | Hydrogen Sulfide Positively Regulates Abscisic Acid Signaling through Persulf idation of SnRK2.6 in Guard Cells显示文摘The phytohormone abscisic acid(ABA)plays pivotal roles in triggering stomatal closure and facilitating adaptation of plants to drought stress.Hydrogen sulfide(H2S),a small signaling gas molecule,is involved in ABA-dependent stomatal closure.However,how H2S regulates ABA signaling remains largely unclear.Here,we show that ABA induces the production of H2S catalyzed by L-CYSTEINE DESULFHYDRASE1(DES1)in guard cells,and H2S in turn positively regulates ABA signaling through persulfidation of Open Stomata 1(OST1)/SNF1-RELATED PROTEIN KINASE2.6(SnRK2.6).Two cysteine(Cys)sites,Cys131 and Cys137,which are exposed on the surface of SnRK2.6 and close to the activation loop,were identified to be persulfidated,which promotes the activity of SnRK2.6 and its interaction with ABA response element-binding factor 2(ABF2),a transcription factor acting downstream of ABA signaling.When Cys131,Cys137,or both residues in SnRK2.6 were substituted with serine(S),H2S_induced SnRK2_6 activity and SnRK2.6-ABF2 interaction were partially(SnRK2.6c131s and SnRK2.6c137S)or completely(SnRK2.6c131sc137S)compromised.Introduction of SnRK2.6c131s,SnRK2.6c137S,or SnRK2.6c131sc137S into the ost1-3 mutant could not rescue the mutant phenotype:less sensitivity to ABA-and H2S-induced stomatal closure and Ca2+influx as well as increased water loss and decreased drought tolerance.Taken together,our study reveals a novel post-translational regulatory mechanism of ABA signaling whereby H2S persulfidates SnRK2.6 to promote ABA signaling and ABA-induced stomatal closure. | Sisi Chen Honglei Jia Xiaofeng Wang Cong Shi Xiao Wang Peiyun Ma Juan Wang Meijuan Ren Jisheng Li | 2020 | Molecular Plant2020,13,5: | 13 |

| 8 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 10 | The Effects of Demographic Features on Differences in Sensitivity between PCL-C and SCL-90 Scores in a Follow-up Study in Secondary School Students in the Wenchuan Earthquake Region显示文摘Objective To analyze the sensitivity of effect factors between the PCL‐C and the SCL‐90,to provide evidence for social psychological crisis screening and post‐trauma interventions.Methods We administered the PCL‐C and SCL‐90 to screen for PTSD and other psychological problems among students who survived the disaster and continued their school studies.The surveys were carried out 3,6,9,and 12 months after the earthquake.A bivariate 2‐level logistic model was used to explore the different levels of sensitivity among students.The factors influencing the relationships between PTSD and depression,and between PTSD and anxiety were examined.Results We analyzed data from 1 677 students,revealing that female students in higher grades were more likely to exhibit symptoms of depression,rather than PTSD,compared with the control group (males in lower grades),and the difference was significant (P<0.05).In contrast,ethnic minorities were more likely to exhibit PTSD symptoms compared to the others.In addition,female students were more likely to exhibit symptoms of anxiety than PTSD.Other effects that did not reach statistical significance were suggested to have a similar influence on PTSD,depression,and anxiety.Conclusion After a natural disaster,specific aspects of depression and anxiety should be examined,avoiding an overemphasis on PTSD in social psychological crisis interventions. | HOU Feng Su LI Ting LI Juan HU Xiao Qin LIU Zhi Yue YUAN Ping | 2011 | Biomedical and Environmental Sciences2011,24,6: | 11 |
| 11 | Inhibition of bromophenols against PTP1B and anti-hyperglycemic effect of Rhodomela confervoides extract in diabetic rats显示文摘Protein tyrosine phosphatase 1B (PTP1B) plays an important role as a negative regulator in insulin signaling pathways. PTP1B is an effective target for the treatment of type 2 diabetes mellitus. Four bromophenol derivatives from red algae Rhodomela confervoides, 2,2′,3,3′-tetrabromo-4,4′,5,5′-tetra- hydroxydiphenyl methane (1), 3-bormo-4,5-bis(2,3-dibromo-4,5-dihydroxybenzyl) pyrocatechol (2), bis(2,3-dibromo-4,5-dihydroxybenzyl) ether (3) and 2,2′,3-tribromo-3′,4,4′,5-tetrahydroxy-6′-ethyloxy- methyldiphenylmethane (4) showed significant inhibitory activity against PTP1B (IC50 were 2.4, 1.7, 1.5 and 0.84 μmol/L, respectively) as potential therapeutical agents for the treatment of type 2 diabetes mellitus. The anti-hyperglycemic effects of the ethanol extracts from R. confervoides on streptozoto- cin-induced diabetes (STZ-diabetes) in male Wistar rats fed with high fat diet were investigated. The STZ-diabetic rats treated with medium-dose and high-dose alga extracts showed remarkable reductions in fasting blood glucose (FBG) as compared with the STZ-diabetic control. | 12 | Limb remote ischemic postconditioning protects integrity of the blood-brain barrier after stroke显示文摘Integrity of the blood-brain barrier structure is essential for maintaining the internal environment of the brain.Development of cerebral infarction and brain edema is strongly associated with blood-brain barrier leakage.Therefore,studies have suggested that protecting the blood-brain barrier may be an effective method for treating acute stroke.To examine this possibility,stroke model rats were established by middle cerebral artery occlusion and reperfusion.Remote ischemic postconditioning was immediately induced by three cycles of 10-minute ischemia/10-minute reperfusion of bilateral hind limbs at the beginning of middle cerebral artery occlusion reperfusion.Neurological function of rat models was evaluated using Zea Longa's method.Permeability of the blood-brain barrier was assessed by Evans blue leakage.Infarct volume and brain edema were evaluated using 2,3,5-triphenyltetrazolium chloride staining.Expression of matrix metalloproteinase-9 and claudin-5 m RNA was determined by real-time quantitative reverse transcription-polymerase chain reaction.Expression of matrix metalloproteinase-9 and claudin-5 protein was measured by western blot assay.The number of matrix metalloproteinase-9-and claudin-5-positive cells was analyzed using immunohistochemistry.Our results showed that remote ischemic postconditioning alleviated disruption of the blood-brain barrier,reduced infarct volume and edema,decreased expression of matrix metalloproteinase-9 m RNA and protein and the number of positive cells,increased expression of claudin-5 m RNA and protein and the number of positive cells,and remarkably improved neurological function.These findings confirm that by suppressing expression of matrix metalloproteinase-9 and claudin-5 induced by acute ischemia/reperfusion,remote ischemic postconditioning reduces blood-brain barrier injury,mitigates ischemic injury,and exerts protective effects on the brain. | Juan Li Xiao-Song Hu Fang-Fang Zhou Shuai Li You-Sheng Lin Wen-Qian Qi Cun-Fang Qi Xiao Zhang | 2018 | Neural Regeneration Research2018,13,9: | 11 |

| 16 | The Status and Associated Factors of Successful Aging among Older Adults Residing in Longevity Areas in China显示文摘Objective This study aims to assess the status of successful aging(SA) in longevity areas in China and explore multiple factors associated with SA among the young-old and oldest-old. Methods A total of 2296 elderly people aged 65 and older were interviewed in the longevity areas sub-sample of the Chinese Longitudinal Healthy Longevity Survey(CLHLS) in 2012. Baseline assessments included a researcher-administered questionnaire, physical examination, and laboratory testing. A logistic regression model was used to identify factors associated with SA. Results The prevalence of SA was 38.81% in the CLHLS in 2012. There were significant differences between ages groups, with SA compromising 56.85% among ≥65 years group and 20.31% among ≥100 years group(χtrend2=126.73, P<0.01). The prevalence of SA among females was 33.59%, which was significantly lower than that among males(45.58%)(χ2gender=33.65, P<0.05). In the regression analysis, having anemia(OR=0.744, 95% CI: 0.609-0.910), poor lifestyle(OR=0.697, 95% CI: 0.568-0.854), poor sleep quality(OR=0.558, 95% CI: 0.456-0.682), and central obesity(OR=0.684, 95% CI: 0.556-0.841) were the main factors associated with SA. The promoting SA rate decreased as age increased, and the group of 65-79 years had higher odds than the other age group. | 18 | Clinical risk score for invasive fungal diseases in patients with hematological malignancies undergoing chemotherapy:China Assessment of Antifungal Therapy in Hematological Diseases (CAESAR) study显示文摘Invasive fungal disease (IFD) is a major infectious complication in patients with hematological malignancies.In this study,we examined 4889 courses of chemotherapy in patients with hematological diseases to establish a training dataset (n=3500) by simple random sampling to develop a weighted risk score for proven or probable IFD through multivariate regression,which included the following variables: male patients,induction chemotherapy for newly diagnosed or relapsed disease,neutropenia,neutropenia longer than 10 days,hypoalbuminemia,central-venous catheter,and history of IFD.The patients were classified into three groups,which had low (0-10,~1.2%),intermediate (11-15,6.4%),and high risk (> 15,17.5%) of IFD.In the validation set (n=1389),the IFD incidences of the groups were ~1.4%,5.0%,and 21.4%.In addition,we demonstrated that antifungal prophylaxis offered no benefits in low-risk patients,whereas benefits were documented in intermediate (2.1% vs.6.6%,P=0.007) and high-risk patients (8.4% vs.23.3%,P=0.007).To make the risk score applicable for clinical settings,a pre-chemo risk score that deleted all unpredictable factors before chemotherapy was established,and it confirmed that anti-fungal prophylaxis was beneficial in patients with intermediate and high risk of IFD.In conclusion,an objective,weighted risk score for IFD was developed,and it may be useful in guiding antifungal prophylaxis. | Ling Wang Ying Wang Jiong Hu Yuqian Sun He Huang Jing Chen Jianyong Li Jun Ma Juan Li Yingmin Liang Jianmin Wang Yan Li Kang Yu Jianda Hu Jie Jin Chun Wang Depei Wu Yang Xiao Xiaojun Huang | 2019 | Frontiers of Medicine2019,13,3: | 10 |
| 19 | Dairy Consumption and Associations with Nutritional Status of Chinese Children and Adolescents显示文摘Objective This study aimed to describe frequency and quantity of total dairy consumption of Chinese children and adolescents and explore the associations between dairy consumption and nutrition status, including stunting, wasting, overweight, and obesity. Methods Participants included 28,250 children and adolescents aged 6-17 years old. A food frequency questionnaire(FFQ) including 100 kinds of food was used to collect information about frequency and quantity of dairy consumption. Determination of stunting was with a height cutoff value for age and gender, and determination for wasting, overweight, and obesity was with BMI for age and gender. Results Of the total sample, 36.1% of children aged 6-17 reported consuming dairy food more than once per day(≥ 1/day). The average total dairy intake of all the participants was 126.7 g/day. For boys, dairy consumption had an inverse correlation with stunting and wasting after controlling for confounders. For girls, dairy consumption was negatively associated with stunting and obesity after controlling for confounders as above. | 20 | Pathogen-inducible OsMPKKI 0.2-OsMPK6 cascade phosphorylates the Raf-like kinase OsEDRI and inhibits its scaffold function to promote rice disease resistance显示文摘Mitogen-activated protein kinase(MAPK)cascades regulate a myriad of plant biological processes,including disease resistance.Plant genomes encode a large number of MAPK kinase kinases(MAPKKKs)that can be divided into two subfamilies,namely MEKK-like kinases and Raf-like kinases.Thus far,the func-tions of MEKK-like MAPKKKs have been relatively well characterized,but the roles of Raf-like MAPKKKs in.plant MAPK cascades remain less understood.Here,we report the role of OsEDR1,a Raf-like MAPKKK,in the regulation of the MAPK cascade in rice response tp the bacterial pathogen Xanthomonas oryzae pv.oryzicola(Xoc).We found that OsEDR1 inhibits OsMPKK10.2(a MAPK kinase)activity through physical interaction.Upon Xoc infection,OsMPKK10.2 is phosphorylated at S304 to activate OsMPK6(a MAPK).Interestingly,activated OsMPK6 phosphorylates OsEDR1 at S861,which destabilizes OsEDR1 and thus releases the inhibition of OsMPKK10.2,leading to increased OsMPKK10.2 activity and enhanced resistance of rice plants to Xoc.Taken together,these results provide new insights into the functions of Raf-like ki-nases in the regulation of the MAPK cascade in plant immunity. | Haigang Ma Juan Li Ling Ma Peilun Wang Yuan Xue Ping Yin Jinghua Xiao Shiping Wang | 2021 | Molecular Plant2021,14,4: | 9 |
