| 1 | 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). | 2 | Osteoblastic and anti-osteoclastic activities of strontium-substituted silicocarnotite ceramics: In vitro and in vivo studies显示文摘Osteoporosis bone defect is a refractory orthopaedic disease which characterized by impaired bone quality and bone regeneration capacity.Current therapies,including antiosteoporosis drugs and artificial bone grafts,are not always satisfactory.Herein,a strontium-substituted calcium phosphate silicate bioactive ceramic(Sr-CPS)was fabricated.In the present study,the extracts of Sr-CPS were prepared for in vitro study and Sr-CPS scaffolds were used for in vivo study.The cytocompatibility,osteogenic and osteoclastogenic properties of Sr-CPS extracts were characterized in comparison to CPS.Molecular mechanisms were also evaluated by Western blot.Sr-CPS extracts were found to promote osteogenesis by upregulating Wnt/β-catenin signal pathways and inhibit osteoclastogenesis through downregulating NF-κB signal pathway.In vivo,micro-CT,histological and histomorphometric observation were conducted after 8 weeks of implantation to evaluate the bone formation using calvarial defects model in ovariectomized rats.Compared with CPS,Sr-CPS significantly promoted critical sized ovariectomy(OVX)calvarial defects healing.Among all the samples,Sr-10 showed the best performance due to a perfect match of bone formation and scaffold degradation rates.Overall,the present study demonstrated that Sr-CPS ceramic can dually modulate both bone formation and resorption,which might be a promising candidate for the reconstruction of osteoporotic bone defect. | Junkai Zeng Jingshu Guo Zhenyu Sun Fanyan Deng Congqin Ning Youzhuan Xie | 2020 | Bioactive Materials2020,5,3: | 9 |
| 3 | Genome re-sequencing analysis uncovers pathogenecity-related genes undergoing positive selection in Magnaporthe oryzae显示文摘Rice blast caused by Magnaporthe oryzae(M.oryzae) is one of the most destructive diseases,which causes significant rice yield losses and affects global food security.To better understand genetic variations among different isolates of M.oryzae in nature,we re-sequenced the genomes of two field isolates,CH43 and Zhong-10-8-14,which showed distinct pathogenecity on most of the rice cultivars.Genome-wide genetic variation analysis reveals that ZHONG-10-8-14 exhibits higher sequence variations than CH43.Structural variations(SVs) detection shows that the sequence variations primarily occur in exons and intergenic regions.Bioinformatics analysis for gene variations reveals that many pathogenecity-related pathways are enriched.In addition,193 candidate effectors with various DNA polymorphisms were identified,including two known effectors AVR-Pik and AVR-Pital.Comparative polymorphism analysis of thirteen randomly selected effectors suggests that the genetic variations of effectors are under positive selection.The expression pattern analysis of several pathogenecity-related variant genes indicates that these genes are differentially regulated in two isolates,with much higher expression levels in Zhong-10-8-14 than CH43.Our data demonstrate that the genetic variations of effectors and pathogenecity-related genes are under positive selection,resulting in the distinct pathogenicities of CH43 and Zhong-10-8-14 on rice. | Jidong Cao Yongqi Yu Junkai Huang Rui Liu Yan Chen Suiyan Li Jun Liu | 2017 | Science China(Life Sciences)2017,60,8: | 6 |
| 4 | Isolation and characterization of rl_(t), a gene that controls leaf rolling in rice显示文摘Moderate leaf rolling is one of the most important morphological traits in rice breeding for plant ideotype.Previous studies have shown that the rl(t)gene has a high breeding potential for developing hybrid-rice varieties with an ideal ideotype,because it leads to an appropriate leaf rolling index(LRI)of about 30%in the heterozygous state,and had a positive effect on grain yield.In this study,we isolated rl(t)and performed a preliminary investigation of its function in regulating leaf rolling in rice.DNA sequencing identified a single base change(G to T)in the finely mapped region(11 kb)containing rl(t),and this is located in 30-untranslated region(30-UTR)of the only predicted gene,Roc5(Rice outermost cell-specific).The expression level of Roc5 is significantly higher in the rl(t)mutant than in the wild-type.Using RNAi and overexpression analysis,we found that the expression level of Roc5 correlated with LRI and leaf bulliform area,and wasalso associated with leaf abaxial or adaxial rolling.These results confirmed that Roc5 controls leaf rolling in a dosage-dependent manner.Bioinformatics analysis revealed a conserved 17-nt sequence(called the GU-rich element)in the 30-UTR of HD-GL2(Homeodomain-Glabra2)family genes including Roc5.Based on the model of this element in regulating mRNA stability in mammals,we speculate that the single nucleotide change in this element accounts for the higher expression level of Roc5 in the rl(t)mutant compared to the wild-type,which ultimately leads to adaxial rolling of the leaf.This discovery further enhances our knowledge of the molecular mechanisms underlying leaf rolling in rice. | Lei Li Xiang Xue Zongxiang Chen Yafang Zhang Yuyin Ma Cunhong Pan Junkai Zhu Xuebiao Pan Shimin Zuo | 2014 | Chinese Science Bulletin2014,59,25: | 4 |

| 12 | Distinct Roles of Dopamine Receptors in the Lateral Thalamus in a Rat Model of Decisional Impulsivity显示文摘The thalamus and central dopamine signaling have been shown to play important roles in high-level cognitive processes including impulsivity. However, little is known about the role of dopamine receptors in the thalamus in decisional impulsivity. In the present study,rats were tested using a delay discounting task and divided into three groups: high impulsivity(HI), medium impulsivity(MI), and low impulsivity(LI). Subsequent in vivo voxel-based magnetic resonance imaging revealed that the HI rats displayed a markedly reduced density of gray matter in the lateral thalamus compared with the LI rats. In the MI rats, the dopamine D1 receptor antagonist SCH23390 or the D2 receptor antagonist eticlopride was microinjected into the lateral thalamus. SCH23390 significantly decreased their choice of a large, delayed reward and increased their omission of lever presses. In contrast,eticlopride increased the choice of a large, delayed reward but had no effect on the omissions. Together, our results indicate that the lateral thalamus is involved in decisional impulsivity, and dopamine D1 and D2 receptors in the lateral thalamus have distinct effects on decisional impulsive behaviors in rats. | 15 | OsMLH1 interacts with OsMLH3 to regulate synapsis and interference-sensitive crossover formation during meiosis in rice显示文摘Meiotic recombination is essential for reciprocal exchange of genetic information between homologous chromosomes and their subsequent proper segregation in sexually reproducing organisms. MLH1 and MLH3 belong to meiosis-specific members of the Mut L-homolog family, which are required for normal level of crossovers(COs) in some eukaryotes. However, their functions in plants need to be further elucidated.Here, we report the identification of Os MLH1 and reveal its functions during meiosis in rice. Using CRISPRCas9 approach, two independent mutants, Osmlh1-1 and Osmlh1-2, are generated and exhibited significantly reduced male fertility. In Osmlh1-1, the clearance of PAIR2 is delayed and partial ZEP1 proteins are not loaded into the chromosomes, which might be due to the deficient in resolution of interlocks at late zygotene. Thus, Os MLH1 is required for the assembly of synapsis complex. In Osmlh1-1, CO number is dropped by ~53% and the distribution of residual COs is consistent with predicted Poisson distribution,indicating that Os MLH1 is essential for the formation of interference-sensitive COs(class I COs). Os MLH1 interacts with Os MLH3 through their C-terminal domains. Mutation in Os MLH3 also affects the pollen fertility.
