1Myeloid-specific targeting of Notch ameliorates murine renal fibrosis via reduced infiltration and activation of bone marrowderived macrophage显示文摘Macrophages play critical roles in renal fibrosis.However,macrophages exhibit ontogenic and functional heterogeneities,and which population of macrophages contributes to renal fibrosis and the underlying mechanisms remain unclear.In this study,we genetically targeted Notch signaling by disrupting the transcription factor recombination signal binding protein-JK(RBP-J),to reveal its role in regulation of macrophages during the unilateral ureteral obstruction(UUO)-induced murine renal fibrosis.Myeloid-specific disruption of RBP-J attenuated renal fibrosis with reduced extracellular matrix deposition and myofibroblast activation,as well as attenuated epithelial-mesenchymal transition,likely owing to the reduced expression of TGF-β.Mean while,RBP-J deletion significantly hampered macrophage infiltration and activation in fibrotic kidney,although their proliferation appeared unaltered.By using macrophage clearance experiment,we found that kidney resident macrophages made negligible contribution,but bone marrow(BM)-derived macrophages played a major role in renal fibrogenesis.Further mechanistic analyses showed that Notch blockade reduced monocyte emigration from BM by down-regulating CCR2 expression.Finally,we found that myeloid-specific Notch activation aggravated renal fibrosis,which was mediated by CCR2^+macrophages infiltration.In summary,our data have unveiled that myeloid-specific targeting of Notch could ameliorate renal fibrosis by regulating BM-derived macrophages recruitment and activation,providing a novel strategy for intervention of this disease.Yali Jiang Yuanyuan Wang Pengfei Ma Dongjie An Junlong Zhao Shiqian Liang Yuchen Ye Yingying Lu Peng Zhang Xiaowei Liu Hua Han Hongyan Qin 2019Protein & Cell2019,10,3:10

3Terrestrial input and nutrient change reflected by sediment records of the Changjiang River Estuary in recent 80 years显示文摘A variety of environmental problems have been observed in the Changjiang River Estuary and adjacent coastal area, including eutrophication, harmful algal blooms(HABs), and hypoxia in recent decades. Application of sedimentary biogenic element indicators on the study of paleoenvironment can reconstruct environmental evolution history of waters. Two 210Pb-dated cores were collected from the Changjiang River Estuary(S3) and adjacent coastal area(Z13), and total organic carbon(TOC), total nitrogen(TN), biogenic silicon(BSi), total phosphorus(TP) and phosphorus(P) species were analyzed. Three stages of environmental changes are deduced by the nutrient sedimentary records. First, nutrient concentration increased rapidly since the 1950 s, which attributed to agriculture development and overused chemical fertilizers. Second, nutrient concentration kept high and primary production began to promote during the 1960 s to 1980 s, while diatom abundance and proportion began to decline since the 1970 s, accompanied by reduced 23 Si O- concentration and flux from the river. Third, due to several dams and bridges constructed, river runoff and coastal hydrodynamic conditions reduced to a certain extent since the 1990 s, which aggravated the unbalance in nutrient structure. 4Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. 5Mission-oriented dynamic optimization model of carrying spares for warship redundant system显示文摘Redundancy is a common structure for warship system,and it is an effective way to improve the reliability of the system.In this paper, warship system is taken as the object of study,based on the system reliability equivalence principle, a spares demand rate calculation method for redundant system is proposed through structure transformation. According to the system analysis method, the general modeling data structure of spares support echelon and system indenture is established, and the mission success probability is taken as the optimization target to build the dynamic optimization model of carrying spares during the process of multi-phase. By introducing the Lagrange multiplier, the spares weight, volume and cost are transformed to the single target of the spares total scale, and the initial Lagrange factors and its dynamic adjustment policy is proposed. 6Structural variation during dog domestication: insights from gray wolf and dhole genomes显示文摘Several processes like phenotypic evolution, disease susceptibility and environmental adaptations, which fashion the domestication of animals, are largely attributable to structural variations(SVs) in the genome.Here, we present high-quality draft genomes of the gray wolf(Canis lupus) and dhole(Cuon alpinus) with scaffold N50 of 6.04 Mb and 3.96 Mb, respectively. Sequence alignment comprising genomes of three canid species reveals SVs specific to the dog, particularly 16 315 insertions, 2565 deletions, 443 repeats, 16 inversions and 15 translocations. Functional annotation of the dog SVs associated with genes indicates their enrichments in energy metabolisms, neurological processes and immune systems. Interestingly, we identify and verify at population level an insertion fully covering a copy of the AKR1 B1(Aldo-Keto Reductase Family 1 Member B) transcript. Transcriptome analysis reveals a high level of expression of the new AKR1 B1 copy in the small intestine and liver, implying an increase in de novo fatty acid synthesis and antioxidant ability in dog compared to gray wolf, likely in response to dietary shifts during the agricultural revolution. For the first time, we report a comprehensive analysis of the evolutionary dynamics of SVs during the domestication step of dogs.
