| 1 | Myeloid-specific targeting of Notch ameliorates murine renal fibrosis via reduced infiltration and activation of bone marrowderived macrophage显示文摘Macrophages play critical roles in renal fibrosis.However,macrophages exhibit ontogenic and functional heterogeneities,and which population of macrophages contributes to renal fibrosis and the underlying mechanisms remain unclear.In this study,we genetically targeted Notch signaling by disrupting the transcription factor recombination signal binding protein-JK(RBP-J),to reveal its role in regulation of macrophages during the unilateral ureteral obstruction(UUO)-induced murine renal fibrosis.Myeloid-specific disruption of RBP-J attenuated renal fibrosis with reduced extracellular matrix deposition and myofibroblast activation,as well as attenuated epithelial-mesenchymal transition,likely owing to the reduced expression of TGF-β.Mean while,RBP-J deletion significantly hampered macrophage infiltration and activation in fibrotic kidney,although their proliferation appeared unaltered.By using macrophage clearance experiment,we found that kidney resident macrophages made negligible contribution,but bone marrow(BM)-derived macrophages played a major role in renal fibrogenesis.Further mechanistic analyses showed that Notch blockade reduced monocyte emigration from BM by down-regulating CCR2 expression.Finally,we found that myeloid-specific Notch activation aggravated renal fibrosis,which was mediated by CCR2^+macrophages infiltration.In summary,our data have unveiled that myeloid-specific targeting of Notch could ameliorate renal fibrosis by regulating BM-derived macrophages recruitment and activation,providing a novel strategy for intervention of this disease. | Yali Jiang Yuanyuan Wang Pengfei Ma Dongjie An Junlong Zhao Shiqian Liang Yuchen Ye Yingying Lu Peng Zhang Xiaowei Liu Hua Han Hongyan Qin | 2019 | Protein & Cell2019,10,3: | 10 |
| 2 | The radioactive abnormality characteristics of typical regions in Ordos Basin and its geological implications显示文摘There are many results of single mineral enrichment characteristic, such as oil, gas, coal and uranium, but little is known about the synergistic research of these important minerals and the study of uranium enrichment features in the deep basin. So, the study on the paragenesis regularity and coexisting relation of many minerals in the basin will promote the integrated forecast and cooperative exploitation of the basin. Based on the plentiful logging data and geological data, this paper studies the distributing feature of higher Gamma abnormality. The analysis on 33 core samples' test results indicates that the increasing of Gamma abnormality is due to the increasing of the uranium element, and the enrichment of uranium is a result of the activation and conglomeration of uranium. | 4 | Structural variation during dog domestication: insights from gray wolf and dhole genomes显示文摘Several processes like phenotypic evolution, disease susceptibility and environmental adaptations, which fashion the domestication of animals, are largely attributable to structural variations(SVs) in the genome.Here, we present high-quality draft genomes of the gray wolf(Canis lupus) and dhole(Cuon alpinus) with scaffold N50 of 6.04 Mb and 3.96 Mb, respectively. Sequence alignment comprising genomes of three canid species reveals SVs specific to the dog, particularly 16 315 insertions, 2565 deletions, 443 repeats, 16 inversions and 15 translocations. Functional annotation of the dog SVs associated with genes indicates their enrichments in energy metabolisms, neurological processes and immune systems. Interestingly, we identify and verify at population level an insertion fully covering a copy of the AKR1 B1(Aldo-Keto Reductase Family 1 Member B) transcript. Transcriptome analysis reveals a high level of expression of the new AKR1 B1 copy in the small intestine and liver, implying an increase in de novo fatty acid synthesis and antioxidant ability in dog compared to gray wolf, likely in response to dietary shifts during the agricultural revolution. For the first time, we report a comprehensive analysis of the evolutionary dynamics of SVs during the domestication step of dogs.
