| 2 | Three-dimensional finite element analysis of weakened roots restored with different cements in combination with titanium alloy posts显示文摘Background It is very difficult and relatively unpredictable to preserve and restore severely weakened pulpless roots. To provide much needed benefit basis for clinical practice, this study was carried out to analyze the stress distribution in weakened roots restored with different cements in combination with titanium alloy posts. Finite element analysis (FEA) was employed in the study. Methods A pseudo three-dimensional model of a maxillary central incisor with flared root canal, theoretically restored with titanium alloy posts in combination with different cements, was established. The analysis was performed by use of ANSYS software. The tooth was assumed to be isotropic, homogenous and elastic. A load of 100 N at an angle of 45°to the longitudinal axis was applied at the palatal surface of the crown. The distributions of stresses in weakened roots filled with cements of different elastic modulus were analyzed by the three-dimensional FEA model. Results Several stress trends were observed when the stress cloud atlas obtained in the study was analyzed. With the increase of the elastic modulus of cements from 1.8 GPa to 22.4 GPa, the stress values in dentin decreased from 39.58 MPa to 31.43 MPa and from 24.51 MPa to 20.76 MPa (respectively, for maximum principle stress values and Von Mises stress values). When Panavia F and zinc phosphate cement were used, the stress peak values in dentin were very small with no significant difference observed, and the Von Mises stress values were 20.87 MPa and 20.76 MPa respectively. On the other hand, maximum principle stress value and Von Mises stress value in cement layer increased with the increase of the elastic modulus of cements. Conclusions The result of this study demonstrated that elastic modulus was indeed one of the important parameters to evaluate property of the cements. Our three-dimensional FEA model study also found that the cement with elastic modulus similar to that of dentin could reinforce weakened root and reduce the stress in dentin. | 3 | High-resolution records of Holocene from the Shuangchi Maar Lake in Hainan Island显示文摘The results of sedimentary characteristics, grain-size, clay mineral, C/N and pollen analysis reveal the evolution history of the Shuangchi Maar Lake of Hainan Island as well as the short-term climate variations in tropical areas. It is testified that the Maar sediment system was sensitive in recording the rapid climate changes in the Holocene. These multidisciplinary results demonstrate that the thermal maximum in the Holocene climate lasted from 7200 to 2700 aBP, with several abrupt ephemeral vibrations occurring between 7250-6120 aBP and at around 4460, 3850 and 2700 aBP. In the late Holocene, particularly at ~2700 aBP, a distinct environmental change indicating an important climate transition in the tropical eastern Asia was recorded, probably responding to the enhancement of ENSO phenomena in the Pacific Ocean. | 4 | Retrospective and perspective of rice breeding in China显示文摘Breeding is the art and science of selecting and changing crop traits for the benefit of human beings. For several decades, tremendous efforts have been made by Chinese scientists in rice breeding in improving grain yield, nutrition quality, and environmental performance, achieving substantial progress for global food security. Several generations of crop breeding technologies have been developed, for example,selection of better performance in the field among variants(conventional breeding), application of molecular markers for precise selection(molecular marker assisted breeding), and development of molecular design(molecular breeding by rational design). In this review, we briefly summarize the advances in conventional breeding, functional genomics for genes and networks in rice that regulate important agronomic traits, and molecular breeding in China with focuses on high yield, good quality,stress tolerance, and high nutrient-use efficiency. | 5 | Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. | 6 | Tracing the first hematopoietic stem cell generation in human embryo by single-cell RNA sequencing显示文摘Tracing the emergence of the first hematopoietic stem cells(HSCs)in human embryos,particularly the scarce and transient precursors thereof,is so far challenging,largely due to the technical limitations and the material rarity.Here,using single-cell RNA sequencing,we constructed the first genome-scale gene expression landscape covering the entire course of endothelial-to-HSC transition during human embryogenesis.The transcriptomically defined HSC-primed hemogenic endothelial cells(HECs)were captured at Carnegie stage(CS)12-14 in an unbiased way,showing an unambiguous feature of arterial endothelial cells(ECs)with the up-regulation of RUNX1,MYB and ANGPTL Importantly,subcategorizing CD34+CD45-ECs into a CD44+population strikingly enriched HECs by over 10-fold.We further mapped the developmental path from arterial ECs via HSC-primed HECs to hematopoietic stem progenitor cells,and revealed a distinct expression pattern of genes that were transiently over-represented upon the hemogenic fate choice of arterial ECs#including EMCN,PROCR and RUNX1T1.We also uncovered another temporally and molecularly distinct intra-embryonic HEC population,which was detected mainly at earlier CS 10 and lacked the arterial feature.Finally,we revealed the cellular components of the putative aortic niche and potential cellular interactions acting on the HSCprimed HECs.The cellular and molecular programs that underlie the generation of the first HSCs from HECs in human embryos,together with the ability to distinguish the HSC-primed HECs from others,will shed light on the strategies for the production of clinically useful HSCs from pluripotent stem cells. | Yang Zeng Jian He Zhijie Bai Zongcheng Li Yandong Gong Chen Liu Yanli Ni Junjie Du Chunyu Ma Lihong Bian Yu Lan Bing Liu | 2019 | Cell Research2019,29,11: | 14 |
| 7 | Choice of approach for hepatectomy for hepatocellular carcinoma located in the caudate lobe:Isolated or combined lobectomy?显示文摘AIM:To investigate the significance of the surgical approaches in the prognosis of hepatocellular carcinoma(HCC) located in the caudate lobe with a multivariate regression analysis using a Cox proportional hazard model.METHODS:Thirty-six patients with HCC underwent caudate lobectomy at a single tertiary referral center between January 1995 and June 2010.In this series,left-sided,right-sided and bilateral approaches were used.The outcomes of patients who underwent isolated caudate lobectomy or caudate lobectomy combined with an additional partial hepatectomy were compared.The survival curves of the isolated and combined resection groups were generated by the Kaplan-Meier method and compared by a log-rank test.RESULTS:Sixteen(44.4%) of 36 patients underwent isolated total or partial caudate lobectomy whereas 20(55.6%) received a total or partial caudate lobectomy combined with an additional partial hepatectomy.The median diameter of the tumor was 6.7 cm(range,2.1-15.8 cm).Patients who underwent an isolated caudate lobectomy had significantly longer operative time(240 min vs 170 min),longer length of hospital stay(18 d vs 13 d) and more blood loss(780 mL vs 270 mL) than patients who underwent a combined caudate lobectomy(P < 0.05).There were no perioperative deaths in both groups of patients.The complication rate was higher in the patients who underwent an isolated caudate lobectomy than in those who underwent combined caudate lobectomy(31.3% vs 10.0%,P < 0.05).The 1-,3-and 5-year disease-free survival rates for the isolated caudate lobectomy and the combined caudate lobectomy groups were 54.5%,6.5% and 0% and 85.8%,37.6% and 0%,respectively(P < 0.05).The corresponding overall survival rates were 73.8%,18.5% and 0% and 93.1%,43.6% and 6.7%(P < 0.05).CONCLUSION:The caudate lobectomy combined with an additional partial hepatectomy is preferred because this approach is technically less demanding and offers an adequate surgical margin. | Peng Liu Bao-An Qiu Gang Bai Hong-Wei Bai Nian-Xin Xia Ying-Xiang Yang Jian-Yong Zhu Yang An Bing Hu | 2012 | World Journal of Gastroenterology2012,18,29: | 13 |
| 8 | Estimating Aboveground Fresh Biomass of Different Cotton Canopy Types with Homogeneity Models Based on Hyper Spectrum Parameters显示文摘AGB(aboveground fresh biomass)is one of the most important parameters of the crop condition monitored with remote sensing.Hyper spectrum remote sensing with the fine spectrum information becomes the efficient method estimating the vegetation AGB.The research was conducted in Xinjiang,the largest cotton planting region of China.The paper analyzed the correlation between the cotton AGB and reflective spectrum and the first derivative spectrum,and the variation coefficient of the waveband reflectance.According to the analysis above,all of 23 parameters,including the hyper spectrum reflectance,the first derivative spectrum parameters and normalization vegetation indexes,were established. | 9 | Inhibition of pancreatic carcinoma cell growth in vitro by DPC4 gene transfection显示文摘AIM: To detect the expression of DPC4 in malignant and non-malignant specimens of human pancreas,and observe the inhibition of retroviral pLXSN containing DPC4 on pancreatic carcinoma cells in vitro.METHODS: The expression of DPC4 was determined in 40 pancreatic adenocarcinoma and 36 non-malignant pancreatic specimens by reverse-transcriptase polymerase chain reaction (RT-PCR) and immunohisto-chemistry.Furthermore,we constructed retroviral vectors containing DPC4,which then infected the pancreatic carcinoma cell line BxPC-3.Cell growth in vitro after being infected was observed,and the vascular endothelial growth factor (VEGF) mRNA level in the daughter cells was determined by semi-quantitative PCR assay.RESULTS: The RT-PCR assay showed a positive rate of DPC4 mRNA in 100% (36/36) of normal specimens,compared to 40% (16/40) in adenocarcinoma specimens.The regional and intense positive cases of DPC4 expression in adenocarcinoma detected by immunohistochemistry were 10 and four,whereas it was all positive expression in normal tissues.There was a significant difference of DPC4 expression between them.The stable expression of DPC4 in the pancreatic carcinoma cells BxPC-3 could be resumed by retroviral vector pLXSN transfection,and could inhibit cell growth in vitro.Rather,DPC4 could decrease VEGF mRNA transcription levels.CONCLUSION: The deletion of DPC4 expression in pancreatic carcinoma suggests that loss of DPC4 may be involved in the development of pancreatic carcinoma.The retroviral vector pLXSN containing DPC4 can inhibit the proliferation of pancreatic carcinoma cells,and down-regulate the level of VEGF. | Wei Shen Guo-Qing Tao Bing Cai De-Chun Li Xing-Guo Zhu Xia Bai | 2008 | World Journal of Gastroenterology2008,14,40: | 11 |

| 14 | GCH1 plays a role in the high-altitude adaptation of Tibetans显示文摘Tibetans are well adapted to high-altitude hypoxia. Previous genome-wide scans have reported many candidate genes for this adaptation, but only a few have been studied. Here we report on a hypoxia gene(GCH1, GTP-cyclohydrolase I), involved in maintaining nitric oxide synthetase(NOS) function and normal blood pressure, that harbors many potentially adaptive variants in Tibetans. We resequenced an 80.8 kb fragment covering the entire gene region of GCH1 in 50 unrelated Tibetans. Combined with previously published data, we demonstrated many GCH1 variants showing deep divergence between highlander Tibetans and lowlander Han Chinese. Neutrality tests confirmed a signal of positive Darwinian selection on GCH1 in Tibetans. Moreover, association analysis indicated that the Tibetan version of GCH1 was significantly associated with multiple physiological traits in Tibetans, including blood nitric oxide concentration, blood oxygen saturation, and hemoglobin concentration. | 15 | BRIF and CARIF progress显示文摘China Institute of Atomic Energy (CIAE) is currently constructing Beijing rare ion beam facility (BRIF) and is proposing China advanced rare ion beam facility (CARIF). This paper is aiming at introducing the progress of BRIF project and the con ceptual design CARIF. The ISOL type facility BRIF under construction is composed of a 100 MeV 300 ?A proton cyclotron, an ISOL with mass resolution of 20000, and a super-conducting LINAC of 2 MeV/q, and will be commissioned in 2013. CARIF facility proposed is planned to use both ISOL and PF techniques. It is based on a China advanced research reactor CARR that was critical, with ISOL separation of fission fragment, post acceleration to 150 MeV/u, and fragmentation of neu tron-rich fission fragment beam like 132Sn. | 16 | The geomechanics of Shenhua carbon dioxide capture and storage(CCS) demonstration project in Ordos Basin,China显示文摘Carbon dioxide(CO_2) capture and storage(CCS) is considered widely as one of promising options for CO_2emissions reduction,especially for those countries with coal-dominant energy mix like China.Injecting and storing a huge volume of CO_2 in deep formations are likely to cause a series of geomechanical issues,including ground surface uplift,damage of caprock integrity,and fault reactivation.The Shenhua CCS demonstration project in Ordos Basin,China,is the first and the largest full-chain saline aquifer storage project of CO_2 in Asia.The injection started in 2010 and ended in 2015.during which totally 0.3 million tonnes(Mt) CO_2 was injected.The project is unique in which CO_2 was injected into 18 sandstone formations simultaneously and the overlying coal seams will be mined after the injection stopped in 2015.Hence,intense geomechanical studies and monitoring works have been conducted in recent years,including possible damage resulting from the temperature difference between injected CO_2 and formations,injection induced stress and deformation change,potential failure mode and safety factor,interaction between coal mining and CO_2 geological storage,determination of injection pressure limit,and surface monitoring by the interferometric synthetic aperture radar(InSAR) technology.In this paper,we first described the background and its geological conditions of the Shenhua CCS demonstration project.Then,we gave an introduction to the coupled thermo-hydro-mechano-chemical(THMC) processes in CO_2 geological storage,and mapped the key geomechanical issues into the THMC processes accordingly.Next,we proposed a generalized geomechanical research flowchart for CO_2 geological storage projects.After that,we addressed and discussed some typical geomechanical issues,including design of injection pressure limit.CO_2 injection induced near-field damage,and interaction between CO_2geological storage and coal mining,in the Shenhua CCS demonstration project.Finally,we concluded some insights to this CCS project. | Xiaochun Li Qi Li Bing Bai Ning Wei Wei Yuan | 2016 | Journal of Rock Mechanics and Geotechnical Engineering2016,8,6: | 8 |
| 17 | Behavioral Abnormality along with NMDAR-related CREB Suppression in Rat Hippocampus after Shortwave Exposure显示文摘Objective To estimate the detrimental effects of shortwave exposure on rat hippocampal structure and function and explore the underlying mechanisms. Methods One hundred Wistar rats were randomly divided into four groups(25 rats per group) and exposed to 27 MHz continuous shortwave at a power density of 5, 10, or 30 m W/cm^2 for 6 min once only or underwent sham exposure for the control. The spatial learning and memory, electroencephalogram(EEG), hippocampal structure and Nissl bodies were analysed. Furthermore, the expressions of N-methyl-D-aspartate receptor(NMDAR) subunits(NR1, NR2 A, and NR2 B), c AMP responsive element-binding protein(CREB) and phosphorylated CREB(p-CREB) in hippocampal tissue were analysed on 1, 7, and 14 days after exposure. Results The rats in the 10 and 30 m W/cm^2 groups had poor learning and memory, disrupted EEG oscillations, and injured hippocampal structures, including hippocampal neurons degeneration, mitochondria cavitation and blood capillaries swelling. The Nissl body content was also reduced in the exposure groups. Moreover, the hippocampal tissue in the 30 m W/cm^2 group had increased expressions of NR2 A and NR2 B and decreased levels of CREB and p-CREB. Conclusion Shortwave exposure(27 MHz, with an average power density of 10 and 30 m W/cm^2) impaired rats' spatial learning and memory and caused a series of dose-dependent pathophysiological changes. | 19 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. | 20 | Mn-containing bioceramics inhibit osteoclastogenesis and promote osteoporotic bone regeneration via scavenging ROS显示文摘Osteoporosis is caused by an osteoclast activation mechanism.People suffering from osteoporosis are prone to bone defects.Increasing evidence indicates that scavenging reactive oxygen species(ROS)can inhibit receptor activator of nuclear factorκB ligand(RANKL)-induced osteoclastogenesis and suppress ovariectomy-induced osteoporosis.It is critical to develop biomaterials with antioxidant properties to modulate osteoclast activity for treating osteoporotic bone defects.Previous studies have shown that manganese(Mn)can improve bone regeneration,and Mn supplementation may treat osteoporosis.However,the effect of Mn on osteoclasts and the role of Mn in osteoporotic bone defects remain unclear.In present research,a model bioceramic,Mn-containedβ-tricalcium phosphate(Mn-TCP)was prepared by introducing Mn intoβ-TCP.The introduction of Mn intoβ-TCP significantly improved the scavenging of oxygen radicals and nitrogen radicals,demonstrating that Mn-TCP bioceramics might have antioxidant properties.The in vitro and in vivo findings revealed that Mn^(2+)ions released from Mn-TCP bioceramics could distinctly inhibit the formation and function of osteoclasts,promote the differentiation of osteoblasts,and accelerate bone regeneration under osteoporotic conditions in vivo.Mechanistically,Mn-TCP bioceramics inhibited osteoclastogenesis and promoted the regeneration of osteoporotic bone defects by scavenging ROS via Nrf2 activation.These results suggest that Mn-containing bioceramics with osteoconductivity,ROS scavenging and bone resorption inhibition abilities may be an ideal biomaterial for the treatment of osteoporotic bone defect. | Jianmei Li Cuijun Deng Wanyuan Liang Fei Kang Yun Bai Bing Ma Chengtie Wu Shiwu Dong | 2021 | Bioactive Materials2021,6,11: | 7 |
