2Potassium Channels: A Potential Therapeutic Target for Parkinson's Disease显示文摘The pathogenesis of the second major neurodegenerative disorder, Parkinson's disease(PD), is closely associated with the dysfunction of potassium(K^+ ) channels. Therefore, PD is also considered to be an ion channel disease or neuronal channelopathy. Mounting evidence has shown that K^+ channels play crucial roles in the regulations of neurotransmitter release, neuronal excitability, and cell volume. Inhibition of K^+ channels enhances the spontaneous firing frequency of nigral dopamine(DA)neurons, induces a transition from tonic firing to burst discharge, and promotes the release of DA in the striatum.Recently, three K^+ channels have been identified to protect DA neurons and to improve the motor and non-motor symptoms in PD animal models: small conductance(SK)channels, A-type K^+ channels, and KV7/KCNQ channels.In this review, we summarize the physiological and pharmacological effects of the three K^+ channels. 4Neuroprotective Effects of Brain-Gut Peptides: A Potential Therapy for Parkinson’s Disease显示文摘Parkinson's disease(PD) is the second most common neurodegenerative disease and is typically associated with progressive motor and non-motor dysfunctions.Currently, dopamine replacement therapy is mainly used to relieve the motor symptoms, while its long-term application can lead to various complications and does not cure the disease. Numerous studies have demonstrated that many brain-gut peptides have neuroprotective effects in vivo and in vitro, and may be a promising treatment for PD. In recent years, some progress has been made in studies on the neuroprotective effects of some newly-discovered braingut peptides, such as glucagon-like peptide 1, pituitary adenylate cyclase activating polypeptide, nesfatin-1, and ghrelin. However, there is still no systematic review on the neuroprotective effects common to these peptides. 6The zinc transporter Slc39a5 controls glucose sensing and insulin secretion in pancreatic β-cells via Sirt1- and Pgc-1α-mediated regulation of Glut2显示文摘Zinc levels are high in pancreatic β-cells, and zinc is involved in the synthesis, processing and secretion of insulin in these cells. However, precisely how cellular zinc homeostasis is regulated in pancreatic β-cells is poorly understood. By screening the expression of 14 Slc39a metal importer family member genes, we found that the zinc transporter Slc39a5 is significantly downregulated in pancreatic β-cells in diabetic db/db mice, obese ob/ob mice and high-fat diet-fed mice. Moreover,β-cell-specific Slc39a5 knockout mice have impaired insulin secretion. In addition, Slc39a5-deficient pancreatic islets have reduced glucose tolerance accompanied by reduced expression of Pgc-1α and its downstream target gene Glut2. The down-regulation of Glut2 in Slc39a5-deficient islets was rescued using agonists of Sirt1, Pgc-1α and Ppar-γ. At the mechanistic level, we found that Slc39a5-mediated zinc influx induces Glut2 expression via Sirt1-mediated Pgc-1α activation. 10Preparation and characterization of some surface negatively charged residue mutants of cytochrome b_5显示文摘Site-directed mutagenesis was used to obtain seven variants of tryptic fragment of bovine liver cytochrome bs (cyt bs), in which the negatively charged residues around the heme exposed edge of cyt bs were replaced by hydropho-bic amino acid alanine. Double-site mutants, triple-site mutants and even quadruple-site mutants were obtained. DNA sequencing and molecular weight measurements of the mutant proteins both confirmed that these site-directed muta-genesises were successfully performed. Spectroelectrochem-istry of these mutant proteins revealed that the apparent redox potentials of these mutant proteins caused a positive shift of 2-10 mV. 11Proteomics Study of COI1-regulated Proteins in Arabidopsis Flower显示文摘Jasmonates(JAs) are a new class of plant hormone that regulate expression of diverse genes to mediate various plant responses.The Arabidopsis F-box protein COI1 is required for plant defense and male fertility in JA signal pathway.To further investigate the regulatory role of COI1 in male fertility,we compared the proteomics profiles of Arabidopsis wild type(WT) flowers with coi1-1 mutan male-sterile flowers using two-dimensional difference gel electrophoresis coupled with matrix-assisted laser desoption/ionization-time-of-flight mass spectrometry.Sixteen proteins with potential function in specific biological processes such as metabolism processes and defense/stress responses were differentially expressed in WT and coi1-1 mutant flowers.Verification on a phi class glutathione transferase AtGSTF9,one out of these 16 identified proteins,revealed that the expression of AtGSTF9 was severely downregulated in flowers of coi1-1 mutant compared with that of WT.Further function analyses of these genes would provide new insights into the molecular basis of COI1-regulated male fertility.Lingling Chua Xiaoyi Shan Junxia Wang Wen Peng Guiyou Zhang Daoxin Xie 2010Journal of Integrative Plant Biology2010,52,4:1
12NAC1 regulates root ground tissue maturation by coordinating with the SCR/SHR-CYCD6;1 module in Arabidopsis显示文摘Precise spatiotemporal control of the timing and extent of asymmetric cell divisions(AcDs)is essential for plant development.In the Arabidopsis root,ground tissue maturation involves an additional AcD of the endo-dermis that maintains the inner cell layer as the endodermis and generates the middle cortex to the outside.Through regulation of the cell cycle regulator CYCLIND6;1(CYCD6;1),the transcription factors SCARECROW(SCR)and SHORT-ROOT(SHR)play critical roles in this process.In the present study,we found that loss of function of NAC1,a NAC transcription factor family gene,causes markedly increased periclinal cell divisions in the root endodermis.Importantly,NAC1 directly represses the transcription of CYCD6;1 by recruiting the co-repressor TOPLESS(TPL),creating a fine-tuned mechanism to maintain proper root ground tissue patterning by limiting production of middle cortex cells.Biochemical and genetic analyses further showed that NAC1 physically interacts with SCR and SHR to restrict excessive periclinal cell divisions in the endo-dermis during root middle cortex formation.Although NAC1-TPL is recruited to the CYCD6;1 promoter and represses its transcription in an SCR-dependent manner,NAC1 and SHR antagonize each other to regu-late the expression of CYCD6;1.Collectively,our study provides mechanistic insights into how the NAC1-TPL module integrates with the master transcriptional regulators SCR and SHR to control root ground tissue patterning by fine-tuning spatiotemporal expression of CYCD6;1 in Arabidopsis.Chuantian Xie Cuiling Li Fengxia Wang Feng Zhang Jiajia Liu Junxia Wang Xiansheng Zhang Xiangpei Kong Zhaojun Ding 2023Molecular Plant2023,16,4:1

17Astrocyte dysfunction in Parkinson's Check for updates disease:from the perspectives of transmittedα-synuclein and genetic modulation显示文摘Parkinson's disease(PD)is a common neurodegenerative disorder that primarily affects the elderly.While the etiology of PD is likely multifactorial with the involvement of genetic,environmental,aging and other factors,α-synuclein(α-syn)pathology is a pivotal mechanism underlying the development of PD.In recent years,astrocytes have attracted considerable attention in the field.Although astrocytes perform a variety of physiological functions in the brain,they are pivotal mediators ofα-syn toxicity since they internalizeα-syn released from damaged neurons,and this triggers an inflammatory response,protein degradation dysfunction,mitochondrial dysfunction and endoplasmic reticulum stress.Astrocytes are indispensable coordinators in the background of several genetic mutations,including PARK7,GBA1,LRRK2,ATP13A2,PINK1,PRKN and PLA2G6.As the most abundant glial cells in the brain,functional astrocytes can be replenished and even converted to functional neurons.In this review,we discuss astrocyte dysfunction in PD with an emphasis onα-syn toxicity and genetic modulation and conclude that astrocyte replenishment is a valuable therapeutic approach in PD.Changjing Wang Tongtong Yang Meiyu Liang Junxia Xie Ning Song 2021Translational Neurodegeneration2021,10,4:1
