1Wnt and BMP signaling crosstalk in regulating dental stem cells:Implications in dental tissue engineering显示文摘Tooth is a complex hard tissue organ and consists of multiple cell types that are regulated by important signaling pathways such as Wnt and BMP signaling.Serious injuries and/or loss of tooth or periodontal tissues may significantly impact aesthetic appearance,essential oral functions and the quality of life.Regenerative dentistry holds great promise in treating oral/dental disorders.The past decade has witnessed a rapid expansion of our understanding of the biological features of dental stem cells,along with the signaling mechanisms governing stem cell self-renewal and differentiation.In this review,we first summarize the biological characteristics of seven types of dental stem cells,including dental pulp stem cells,stem cells from apical papilla,stem cells from human exfoliated deciduous teeth,dental follicle precursor cells,periodontal ligament stem cells,alveolar bone-derived mesenchymal stem cells(MSCs),and MSCs from gingiva.We then focus on how these stem cells are regulated by bone morphogenetic protein(BMP)and/or Wnt signaling by examining the interplays between these pathways.Lastly,we analyze the current status of dental tissue engineering strategies that utilize oral/dental stem cells by harnessing the interplays between BMP and Wnt pathways.We also highlight the challenges that must be addressed before the dental stem cells may reach any clinical applications.Thus,we can expect to witness significant progresses to be made in regenerative dentistry in the coming decade.Fugui Zhang Jinlin Song Hongmei Zhang Enyi Huang Dongzhe Song Viktor Tollemar Jing Wang Jinhua Wang Maryam Mohammed Qiang Wei Jiaming Fan Junyi Liao Yulong Zou Feng Liu Xue Hu Xiangyang Qu Liqun Chen Xinyi Yu Hue H.Luu Michael J.Lee Tong-Chuan He Ping Ji 2016Genes & Diseases2016,3,4:16
2Joint contractures in severe burn patients with early rehabilitation intervention in one of the largest burn intensive care unit in China:a descriptive analysis显示文摘Background:Joint contracture is the major clinical complication in burn patients,especially,the severe burn patients.This study aimed to investigate the number and severity of joint contractures in patients with burns affecting greater than or equal to 50%of the total body surface area(TBSA)undergoing early rehabilitation in a burn intensive care unit(BICU).Methods:We analyzed burn patients with burns affecting greater than or equal to 50%of the TBSA admitted to a BICU who received early rehabilitation within 7 days post-injury from January 2011 to December 2015.Demographic and medical information was collected.The range of motion(ROM)of different joints was measured 1 month post-admission.Spearman’s correlation coefficient and logistic regression analysis was used to determine predictors of the presence and severity of contractures.Result:The average affected TBSA of the included burn patients was 67.4%,and the average length of stay in the BICU was 46.2±28.8 days.One hundred and one of 108 burn patients(93.5%)developed at least one joint contracture.The ROM in 67.9%of the affected joints was mildly limited.The majority of contractures in severe burn patients were mild(37.7%)or moderate(33.2%).The wrist was the most commonly affected joint(18.2%),followed by the shoulder,ankle,hip,knee,and elbow.A predictor of the presence of contractures was the length of hospital stay(p=0.049).The severe contracture was related to the area of full-thickness burns,the strict bed rest time,and the duration of rehabilitation in BICU.The length of rehabilitation stay(days)in patients with moderate contracture is 54.5%longer than that in severe contracture(p=0.024)Conclusion:During the long stay in BICU,the length of rehabilitation stay in a BICU could decrease the severity of contractures from severe to moderate in the patients with equal to 50%of the TBSA.Hence,this research reveals the important role of early rehabilitation interventions in severe burn patients.Jianglin Tan Jian Chen Junyi Zhou Huapei Song Huan Deng Ming Ao Gaoxing Luo Jun Wu 2019Burns & Trauma2019,7,1:9
3Allogeneic Vγ9Vδ2 T-cell immunotherapy exhibits promising clinical safety and prolongs the survival of patients with late-stage lung or liver cancer显示文摘Vγ9Vδ2 T cells are promising candidates for cellular tumor immunotherapy.Due to their HLA-independent mode of action,allogeneic Vγ9Vδ2 T cells can be considered for clinical application.To apply allogeneic Vγ9Vδ2 T cells in adoptive immunotherapy,the methodology used to obtain adequate cell numbers with optimal effector function in vitro needs to be optimized,and clinical safety and efficacy also need to be proven.Therefore,we developed a novel formula to improve the expansion of peripheralγδT cells from healthy donors.Then,we used a humanized mouse model to validate the therapeutic efficacy of expandedγδT cells in vivo;furthermore,the expandedγδT cells were adoptively transferred into late-stage liver and lung cancer patients.We found that the expanded cells possessed significantly improved immune effector functions,including proliferation,differentiation,and cancer cell killing,both in vitro and in the humanized mouse model.Furthermore,a phase I clinical trial in 132 late-stage cancer patients with a total of 414 cell infusions unequivocally validated the clinical safety of allogeneic Vγ9Vδ2 T cells.Among these 132 patients,8 liver cancer patients and 10 lung cancer patients who received≥5 cell infusions showed greatly prolonged survival,which preliminarily verified the efficacy of allogeneic Vγ9Vδ2 T-cell therapy.Our clinical studies underscore the safety and efficacy of allogeneic Vγ9Vδ2 T-cell immunotherapy,which will inspire further clinical investigations and eventually benefit cancer patients.Yan Xu Zheng Xiang Mohammed Alnaggar Léonce Kouakanou Jiawei Li Junyi He Jiashuang Yang Yi Hu Yan Chen Li Lin Jianlei Hao Jingxia Li Jibing Chen Man Li Qingling Wu Christian Peters Qinghua Zhou Jianshuang Li Yingqing Liang Xiaohua Wang Baohui Han Meili Ma Dieter Kabelitz Kecheng Xu Wenwei Tu Yangzhe Wu Zhinan Yin 2021Cellular & Molecular Immunology2021,18,2:8
4Sox9 augments BMP2-induced chondrogenic differentiation by downregulating Smad7 in mesenchymal stem cells(MSCs)显示文摘Cartilage injuries caused by arthritis or trauma pose formidable challenges for effective clinical management due to the limited intrinsic proliferative capability of chondrocytes.Autologous stem cell-based therapies and transgene-enhanced cartilage tissue engineering may open new avenues for the treatment of cartilage injuries.Bone morphogenetic protein 2(BMP2)induces effective chondrogenesis of mesenchymal stem cells(MSCs)and can thus be explored as a potential therapeutic agent for cartilage defect repair.However,BMP2 also induces robust endochondral ossification.Although the precise mechanisms through which BMP2 governs the divergence of chondrogenesis and osteogenesis remain to be fully understood,blocking endochondral ossification during BMP2-induced cartilage formation may have practical significance for cartilage tissue engineering.Here,we investigate the role of Sox9-donwregulated Smad7 in BMP2-induced chondrogenic differentiation of MSCs.We find that overexpression of Sox9 leads to a decrease in BMP2-induced Smad7 expression in MSCs.Sox9 inhibits BMP2-induced expression of osteopontin while enhancing the expression of chondrogenic marker Col2a1 in MSCs.Forced expression of Sox9 in MSCs promotes BMP2-induced chondrogenesis and suppresses BMP2-induced endochondral ossification.Constitutive Smad7 expression inhibits BMP2-induced chondrogenesis in stem cell implantation assay.Mouse limb explant assay reveals that Sox9 expands BMP2-stimulated chondrocyte proliferating zone while Smad7 promotes BMP2-intitated hypertrophic zone of the growth plate.Cell cycle analysis indicates that Smad7 induces significant early apoptosis in BMP2-stimulated MSCs.Taken together,our results strongly suggest that Sox9 may facilitate BMP2-induced chondrogenesis by downregulating Smad7,which can be exploited for effective cartilage tissue engineering.Chen Zhao Wei Jiang Nian Zhou Junyi Liao Mingming Yang Ning Hu Xi Liang Wei Xu Hong Chen Wei Liu Lewis L.Shi Leonardo Oliveira Jennifer Moriatis Wolf Sherwin Ho Aravind Athiviraham H.M.Tsai Tong-Chuan He Wei Huang 2017Genes & Diseases2017,4,4:7
5A genome-wide analysis of SWEET gene family in cotton and their expressions under different stresses显示文摘Background: The SWEET(Sugars will eventually be exported transporters) gene family plays multiple roles in plant physiological activities and development process. It participates in reproductive development and in the process of sugar transport and absorption, plant senescence and stress responses and plant-pathogen interaction. However,thecomprehensive analysis of SWEET genes has not been reported in cotton.Results: In this study, we identified 22, 31, 55 and 60 SWEET genes from the sequenced genomes of Gossypium arboreum, G. raimondii, G. hirsutum and G. barbadense, respectively. Phylogenetic tree analysis showed that the SWEET genes could be divided into four groups, which were further classified into 14 sub-clades. Further analysis of chromosomal location, synteny analysis and gene duplication suggested that the orthologs showed a good collinearity and segmental duplication events played a crucial role in the expansion of the family in cotton. Specific MtN3_slv domains were highly conserved between Arobidopsis and cotton by exon-intron organization and motif analysis. In addition, the expression pattern in different tissues indicated that the duplicated genes in cotton might have acquired new functions as a result of sub-functionalization or neo-funtionalization. The expression pattern of SWEET genes showed that the different genes were induced by diverse stresses. The identification and functional analysis of SWEET genes in cotton may provide more candidate genes for genetic modification.Conclusion: SWEET genes were classified into four clades in cotton. The expression patterns suggested that the duplicated genes might have experienced a functional divergence. 8Whole-genome resequencing of Osmanthus fragrans provides insights into flower color evolution显示文摘Osmanthus fragrans is a well-known ornamental plant that has been domesticated in China for 2500 years.More than 160 cultivars have been found during this long period of domestication,and they have subsequently been divided into four cultivar groups,including the Yingui,Jingui,Dangui,and Sijigui groups.These groups provide a set of materials to study genetic evolution and variability.Here,we constructed a reference genome of O.fragrans‘Liuyejingui’in the Jingui group and investigated its floral color traits and domestication history by resequencing a total of 122 samples,including 119 O.fragrans accessions and three other Osmanthus species,at an average sequencing depth of 15×.The population structure analysis showed that these 119 accessions formed an apparent regional cluster.The results of linkage disequilibrium(LD)decay analysis suggested that varieties with orange/red flower color in the Dangui group had undergone more artificial directional selection;these varieties had the highest LD values among the four groups,followed by the Sijigui,Jingui,and Yingui groups.Through a genome-wide association study,we further identified significant quantitative trait loci and genomic regions containing several genes,such as ethylene-responsive transcription factor 2 and Arabidopsis pseudoresponse regulator 2,that are positively associated with petal color.Moreover,we found a frameshift mutation with a 34-bp deletion in the first coding region of the carotenoid cleavage dioxygenase 4 gene.This frameshift mutation existed in at least one site on both alleles in all varieties of the Dangui group.The results from this study shed light on the genetic basis of domestication in woody plants,such as O.fragrans.Hongguo Chen Xiangling Zeng Jie Yang Xuan Cai Yumin Shi Riru Zheng Zhenqi Wang Junyi Liu Xinxin Yi Siwei Xiao Qiang Fu Jingjing Zou Caiyun Wang 2021Horticulture Research2021,8,1:5

11Exploration of presence/absence variation and corresponding polymorphic markers in soybean genome显示文摘This study was designed to reveal the genome‐wide distribution of presence/absence variation(PAV) and to establish a database of polymorphic PAV markers in soybean. The 33 soybean whole‐genome sequences were compared to each other with that of Williams 82 as a reference genome. A total of 33,127 PAVs were detected and 28,912 PAV markers with their primer sequences were designed as the database NJAUSoyPAV_1.0. The PAVs scattered on whole genome while only 518(1.8%) overlapped with simple sequence repeats(SSRs) in BARCSOYSSR_1.0database. In a random sample of 800 PAVs, 713(89.13%) showed polymorphism among the 12 differential genotypes. Using 126 PAVs and 108 SSRs to test a Chinese soybean germplasm collection composed of 828 Glycine soja Sieb. et Zucc. and Glycine max(L.) Merr. accessions, the per locus allele number and its variation appeared less in PAVs than in SSRs. The distinctness among alleles/bands of PCR(polymerase chain reaction) products showed better in PAVs than in SSRs, potential in accurate marker‐assisted allele selection. 12Wavelet analysis of coastal-trapped waves along the China coast generated by winter storms in 2008显示文摘This study applies the wavelet analysis to the tidal gauge records, alongshore winds, atmospheric temperature and pressure along the China coast in winter 2008. The analysis results show three events of sea level oscillations(SLOs) on the shelf induced by winter storms. The first event occurred from January 9 to 21. The SLO periods were double-peaked at 1.6–5.3 and 7.0–16.0 d with the power densities of 0.04–0.05 and 0.10–0.15 m2·d, respectively.The second event occurred from February 5 to 18. The SLO period was single-peaked at 2.3–3.5 d with power density of 0.03–0.04 m2·d. The third event occurred from February 20 to March 8. The SLO periods were doublepeaked at 1.5–4.3 and 6.1–8.2 d with the power densities of 0.08–0.11 and 0.02–0.08 m2·d, respectively. The SLOs propagated along the coast from Zhejiang in north to Guangdong in south. The phase speeds ranged about 9–29m/s from Kanmen to Pingtan, 5–11 m/s from Xiamen to Huizhou and 11–22 m/s from Huizhou to Shuidong. 13Distr ibution, risk and bioavailability of metals in sediments of Lake Yamdrok Basin on the Tibetan Plateau, China显示文摘Total contents of metals in soil and sediments on the Tibetan Plateau of China have been widely analyzed,but existing information is insufficient to effectively evaluate metal ecological risk because of a lack of metal bioavailability data.In this study,distribution,potential risk,mobility and bioavailability of metals in sediments of Lake Yamdrok Basin in Tibet of China were explored by combined use of total digestion,sequential extraction and the diffusive gradient in thin-films(DGT).Average concentrations of Cr,Ni,Cu,Zn,As,Cd and Pb in surface sediments were 31.25,30.31,22.00,45.04,31.32,0.13 and 13.39 mg/kg,respectively.Higher levels of metals were found near the inflowing rivers.Residual form was dominant in Cr,Ni,Zn,Cd and Pb,and reducible form was dominant in As and Cd.Metals in surface sediments showed a low enrichment degree overall,but Cd and As had higher ecological risk levels than the other metals.Furthermore,there was a larger average proportion of exchangeable form of As(20.4%)and Cd(9.0%)than the other metals(1.7%-3.3%),implying their higher mobility and release risk.Average DGT-labile concentrations of Cr,Ni,Cu,Zn,As,Cd and Pb were 0.5,4.5,0.7,25.1,60.0,0.22 and 1.0μg/L,respectively.The DGT-labile As was significantly correlated with extractable As forms(p<0.01),suggesting that extractable As in sediments acts as a'mobile pool'for bioavailable As.These results suggest potential risks of As and Cd,especially As,deserve further attention in Lake Yamdrok Basin.Feifei Che Junyi Chen Bo Zhang Xia Jiang Shuhang Wang 2020Journal of Environmental Sciences2020,32,11:4

16Mapping Quantitative Trait Loci Associated with Aluminum Toxin Tolerance in NJRIKY Recombinant Inbred Line Population of Soybean (Glycine max)显示文摘To investigate the genetic mechanism of Al-tolerance in soybean, a recombinant inbred line population (RIL) with 184 F2:7:11 lines derived from the cross of Kefeng No.1 × Nannong 1138-2 (Al-tolerant × Al-sensitive) were tested in pot experiment with sand culture medium in net room in Nanjing. Four traits, i.e. plant height, number of leaves, shoot dry weight and root dry weight at seedling stage, were evaluated and used to calculate the average membership index (FAi) as the indicator of Al-tolerance. The composite interval mapping (CIM) under WinQTL Cartographer v. 2.5 detected five QTLs (i.e. qFAi-1, qFAi- 2, qFAi-3, qFAi-4 and qFAi-5), explaining 5.20%-9.07% of the total phenotypic variation individually. While with the multiple interval mapping (MIM) of the same software, five QTLs (qFAi-1, qFAi-5, qFAi-6, qFAi-7, and qFAi-8) explaining 5.7%-24.60% of the total phenotypic variation individually were mapped. Here qFAi-1 and qFAi-5 were detected by both CIM and MIM with the locations in a same ?anking marker region, GMKF046-GMKF080 on B1 and satt278-sat_95 on L, respectively. While qFAi-2 under CIM and qFAi-6 under MIM both on D1b2 were located in neighboring regions with their confidence intervals overlapped and might be the same locus. Segregation analysis under major gene plus polygene inheritance model showed that Al-tolerance was controlled by two major genes (h2mg = 33.05%) plus polygenes (h2pg = 52.73%). 17Prevalence and clinical significance of pathogenic germline BRCA1/2 mutations in Chinese non-small cell lung cancer patients显示文摘Objective: Germline alterations in the breast cancer susceptibility genes type 1 and 2, BRCA1 and BRCA2, predispose individuals to hereditary cancers, including breast, ovarian, prostate, pancreatic, and stomach cancers.Accumulating evidence suggests inherited genetic susceptibility to lung cancer.The present study aimed to survey the prevalence of pathogenic germline BRCA mutations(gBRCAm) and explore the potential association between gBRCAm and disease onset in Chinese advanced non-small cell lung cancer(NSCLC) patients.Methods: A total of 6,220 NSCLC patients were screened using capture-based ultra-deep targeted sequencing to identify patients harboring germline BRCA1/2 mutations.Results: Out of the 6,220 patients screened, 1.03%(64/6,220) of the patients harbored the pathogenic gB RCAm, with BRCA2 mutations being the most predominant mutations(49/64, 76.5%).Patients who developed NSCLC before 50 years of age were more likely to carry gBRCAm(P = 0.036).Among the patients harboring classic lung cancer driver mutations, those with concurrent gBRCAm were significantly younger than those harboring the wild-type gBRCA(P = 0.029).By contrast, the age of patients with or without concurrent gBRCAm was comparable to those of patients without the driver mutations(P = 0.972).In addition, we identified EGFR-mutant patients with concurrent gBRCAm who showed comparable progression-free survival but significantly longer overall survival(P = 0.002) compared to EGFR-mutant patients with wild-type germline BRCA.Conclusions: Overall, our study is the largest survey of the prevalence of pathogenic gBRCAm in advanced Chinese NSCLC patients.Results suggested a lack of association between germline BRCA status and treatment outcome of EGFR-TKI.In addition,results showed a positive correlation between pathogenic gB RCAm and an early onset of NSCLC.Xingsheng Hu Dongyong Yang Yalun Li Li Li Yan Wang Peng Chen Song Xu Xingxiang Pu Wei Zhu Pengbo Deng Junyi Ye Hanhan Zhang Analyn Lizaso Hao Liu Xinru Mao Hai Huang Qian Chu Chengping Hu 2019Cancer Biology & Medicine2019,16,3:4
18A modified HLA-A^(*)0201-restricted CTL epitope from human oncoprotein (hPEBP4) induces more efficient antitumor responses显示文摘We previously identified human phosphatidylethanolamine-binding protein 4(hPEBP4)as an antiapoptotic protein with increased expression levels in breast,ovarian and prostate cancer cells,but low expression levels in normal tissues,which makes hPEBP4 an attractive target for immunotherapy.Here,we developed hPEBP4-derived immunogenic peptides for inducing antigen-specific cytotoxic T lymphocytes(CTLs)targeting breast cancer.A panel of hPEBP4-derived peptides predicted by peptide-MHC-binding algorithms was evaluated to characterize their HLAA2.1 affinity and immunogenicity.We identified a novel immunogenic peptide,P40–48(TLFCQGLEV),that was capable of eliciting specific CTL responses in HLA-A2.1/K^(b) transgenic mice,as well as in peripheral blood lymphocytes from breast cancer patients.Furthermore,amino-acid substitutions in the P40–48 sequence improved its immunogenicity against hPEBP4,a self-antigen,thus circumventing tolerance.We designed peptide analogs by preferred auxiliary HLA-A^(*)0201 anchor residue replacement,which induced CTLs that were crossreactive to the native peptide.Several analogs were able to stably bind to HLA-A^(*)0201 and elicit specific CTL responses better than the native sequence.Importantly,adoptive transfer of CTLs induced by vaccination with two analogs more effectively inhibited tumor growth than the native peptide.These data indicate that peptide analogs with high immunogenicity represent promising candidates for peptide-mediated therapeutic cancer vaccines.Weihong Sun Junyi Shi Jian Wu Junchu Zhang Huabiao Chen Yuanyuan Li Shuxun Liu Yanfeng Wu Zhigang Tian Xuetao Cao Nan Li 2018Cellular & Molecular Immunology2018,15,8:4
19Precipitation behaviors and mechanical properties of a solution-treated Mg-Gd-Nd-Zn-Zr alloy during equal-channel angular pressing process显示文摘A magnesium alloy processed by equal-channel angular pressing (ECAP) exhibited excellent microstructure refinement and improved strength and hardness.The comprehensive mechanical properties of magnesium alloys have supported the expansion of their applications in the automotive,aerospace,and biomedical industries.Herein,pre-treatment of a solution-treated Mg-2.9Gd-1.5Nd-0.3Zn-0.3Zr alloy was conducted to investigate the precipitate behavior and microstructure evolution during the ECAP process.β;phase grains quickly precipitated from the solution-treated alloy,which accelerated grain refinement and enhanced the ductility after the ECAP process,as compared to the as-cast alloy reported in our previous study.Moreover,spherical precipitates (~200 nm) and fine phases (~100 nm) precipitated along the stripe-like Zn;Zr;phase,which formed a kabap-like structure dispersing homogeneously in the solution-treated alloy during the ECAP process.Owing to grain refinement,dislocations,sphericalβ;precipitates,and texture evolution,the solution-treated alloy after eight passes of ECAP exhibited good comprehensive mechanical properties,with the ultimate tensile strength,yield strength,and elongation values reaching210.9 MPa,263.9 MPa,and 27.9%,respectively.Zhenzhen Gui Fen Wang Junyi Zhang Dexin Chen Zhixin Kang 2022Journal of Magnesium and Alloys2022,10,1:3
20Molecular markers linked to Rsa resistant to soybean mosaic virus显示文摘Soybean mosaic virus (SMV) is a severe disease in worldwide soybean production. A cross was made between Kefeng No.1 with broad spectrum resistance to SMV and Nannong 1138-2, a susceptible cultivar. The inheritance of resistance to SMV strain Sa prevailing in southern China was analyzed. Results of x2 test from inoculation experiment on parents F1, F2 and F3 lines showed that the resistance to strain Sa was controlled by a single dominant gene Rsa. BSA method was adopted and 900 random 10-mer primers were used to amplify total DNA from resistant pool and susceptible pool in order to obtain polymorphic bands in two bulks.16 primers could generate polymorphic bands, of which OPW-05 and OPAS-06 could generate the most stable RAPD patterns. RAPD markers OPW-05660 and OPAS-061800 were found to be linked to Rsa.Their order and genetic distance were OPAS-06(1800)22.2 cM Rsa 10.1 cM OPW-05660.
