| 6 | Association of Helicobacter pylori infection with colorectal polyps and malignancy in China显示文摘BACKGROUND Gastric Helicobacter pylori(H.pylori)infection is related to chronic gastritis,gastroduodenal ulcer,and gastric malignancies;whether this infection is related to colorectal polyps and colorectal cancer(CRC),remains debatable.AIM To investigate the relationship between gastric H.pylori infection and the risk of colorectal polyps and CRC.METHODS We retrospectively analyzed 3872 patients with colorectal polyps who underwent colonoscopy and pathological diagnosis.We also analyzed 304 patients with primary CRC.The characteristics of these patients were compared with those of the control group,which included 2362 patients with the normal intestinal mucosa.All subjects completed a 14C-urea breath test,bidirectional gastrointestinal endoscopy,and a biopsy on the same day.Data on the number,size,location,and pathology of the polyps,the location,and pathology of the CRC,the detection of H.pylori,and the incidence of H.pylori-associated atrophic gastritis or intestinal metaplasia were obtained.A logistic regression model was used to analyze the relationship between gastric infection due to H.pylori,and the incidence of colorectal polyps and CRC.RESULTS The prevalence of H.pylori infection was higher in the multiple polyps group than in the solitary polyp group and the control group[95%confidence interval(CI)=1.02-1.31,P=0.03;95%CI:2.12-2.74,P<0.001].The patients with adenomatous polyps had a higher incidence of H.pylori infection than patients with non-adenomatous polyps[59.95%vs 51.75%,adjusted odds ratio(OR)=1.41,95%CI:1.24-1.60,P<0.01].Patients with H.pylori-associated atrophic gastritis or intestinal metaplasia were at high risk of CRC(adjusted OR=3.46,95%CI:2.63-4.55,P<0.01;adjusted OR=4.86,95%CI:3.22-7.34,P<0.01,respectively).The size and location of the polyps,the histopathological characteristics and the location of CRC were not related to H.pylori infection.CONCLUSION Our study demonstrates that the incidence of gastric H.pylori infection and H.pylori-associated atrophic gastritis or intestinal metaplasia elevates the risk of colorectal polyps and CRC. | Man Wang Wen-Jie Kong Jing-Zhan Zhang Jia-Jie Lu Wen-Jia Hui Wei-Dong Liu Xiao-Jing Kang Feng Gao | 2020 | World Journal of Gastrointestinal Oncology2020,12,5: | 15 |

| 8 | Binding Activity Difference of Anti-CD20 scFv-Fc Fusion Protein Derived from Variable Domain Exchange显示文摘Two novel engineered antibody fragments binding to antigen CD20 were generated by fusing a murine IgM-type anti-CD20 single-chain Fv fragment (scFv) to the human IgG1 CH2 (i.e., Cγ2) and CH3 (i.e., Cγ3) domains with the human IgG1 hinge (i.e. Hγ). Given the relationship between structure and function of protein, the 3-D structures of the two engineered antibody fragments were modeled using computer-aided homology modeling method. Furthermore, the relationship between 3-D conformation and their binding activity was evaluated theoretically. Due to the change of active pocket formed by CDRs, the HL23 (VH-Linker-VL-Hγ-Cγ2-Cγ3) remained its activity because of its preserved conformation, while the binding activity of the LH23 (VL-Linker-VH-Hγ-Cγ2-Cγ3) was impaired severely. Experimental studies by flow cytometry and fluorescence microscopy showed that HL23 possessed significantly superior binding activity to CD20-expressing target cells than LH23. That is to say, the order of variable regions could influence the binding activity of the fusion protein to CD20+ cell lines, which was in accordance with the theoretical results. | 14 | Esophagojejunostomy after laparoscopic total gastrectomy by Or VilTM or hemi-double stapling technique显示文摘AIM: To investigate the feasibility, advantages and disadvantages of two types of anvil insertion techniques for esophagojejunostomy after laparoscopic total gastrectomy.METHODS: This was an open-label prospective cohort study. Laparoscopy-assisted radical total gastrectomy with D2 lymph node dissection was performed in 84 patients with primary non-metastatic gastric cancer confirmed by pre-operative histological examination. Overweight patients were excluded, as well as patients with peritoneal dissemination and invasion of adjacent organs. After total gastrectomy, all patients were randomized into two groups. Patients in Group Ⅰ underwent esophagojejunostomy using a transorally-inserted anvil(Or VilTM), while patients in Group Ⅱ underwent esophagojejunostomy using the hemi-double stapling technique(HDST). Both types of esophagojejunostomy were performed under laparoscopy. Patients' baseline characteristics, preoperative characteristics, perioperative characteristics, short-term postoperative outcomes and operation cost were comparedbetween the two groups. The primary endpoint was evaluation of the surgical outcome(operating time, time of digestive tract reconstruction and time of anvil insertion) and the medical cost of each operation(operation cost and total cost of hospitalization). The secondary endpoints were time to solid diet, post-surgical hospitalization time, time to defecation, time to ambulation and intra-operative blood loss. In addition, complications were assessed and compared. RESULTS: Laparoscopic total gastrectomy and esophagojejunostomy were successfully performed in all 84 patients, without conversion to laparotomy. There were no significant differences in the operative time and time for total gastrectomy between the two groups(287.8 ± 38.4 min vs 271.8 ± 46.1 min, P = 0.09, and 147.7 ± 31.6 min vs 159.8 ± 33.8 min, P = 0.09, respectively). The time for digestive tract reconstruction and for anvil insertion were significantly decreased in Group Ⅱ compared with Group I(47.8 ± 12.1 min vs 55.4 ± 15.7 min, P = 0.01, and 12.6 ± 4.7 min vs 18.7 ± 7.5 min, P = 0.001, respectively). Intraoperative blood loss(96.4 ± 32.7 m L vs 88.2 ± 36.9 m L, P = 0.28), time to defecation(3.5 ± 0.9 d vs 3.2 ± 1.1 d, P = 0.12), time to ambulation(3.9 ± 0.7 d vs 3.6 ± 1.1 d, P = 0.12), time to solid diet(7.6 ± 1.4 d vs 8.0 ± 2.7 d, P = 0.31) and total hospitalization(10.6 ± 2.6 d vs 10.8 ± 3.5 d, P = 0.80) were similar between the two groups. | 15 | Plant Polyploidy: Origin, Evolution, and Its Influence on Crop Domestication显示文摘The prevalence and recurrence of polyploidization in plant species make it one of the most important evolutionary events in plants, and as a result, polyploidization is an extensively investigated research field. Due to the rapid development of sequencing technologies, there is increased evidence to support that polyploidization plays an important role in the diversification of plant species, evolution of genes, and the domestication of crops. Here, we reviewed the influence of polyploidization on various aspects of plant evolution, mainly focused on polyploid origin, characteristics, subsequent genome divergence, and its impact on gene function innovation and crop domestication. The occurrence of many independent polyploidization events in plants was found to be tightly associated with the timing of extreme climate events or natural disasters on earth, leading to mass extinction while possibly facilitating increased polyploidization. Following allo-polyploidization, a distinct phenomenon known as sub-genome dominance occurred during sub-genome evolution, which was found to be associated with the methylation of transposons. Extensive gene fractionations(lost) following polyploidization were reported in almost all paleo-polyploids, and the evolutionary fates of multi-copy genes, such as sub-/neo-functionalization, were further proposed to illustrate their underlying mechanisms. Moreover,polyploidization was found to significantly impact species diversification, with subsequent effects on crop domestication and the development of traits with agronomic importance. | 17 | Characteristics of the crustal magnetic anomaly and regional tectonics in the Qinghai-Tibet Plateau and the adjacent areas显示文摘Study of the characteristics of the crustal magnetic anomaly in the Qinghai-Tibet Plateau and the adjacent areas helps better understand the lithospheric structure and evolution, as well as the regional geodynamic processes. Here we analyze the distribution laws of the crustal magnetic anomaly and its vertical gradient, the decay characteristics of the anomaly, the contributions from different wavelength bands to the anomaly, and the relationship between the anomaly and the crustal regional tectonics in the plateau and nearby, based on a new and higher degree geomagnetic model NGDC-EMM-720-V3 constructed from the surface, aeromagnetic, marine and satellite survey data. The results reveal that the positive and negative anomalies in the Qinghai-Tibet Plateau are weak, while those of the surrounding areas are strong. The boundary agrees well with the border of the plateau regional tectonics. The anomaly is nearly east-west in the central and western plateau, arc-shaped in the southwestern and eastern, and nearly north-south in the southeastern, consistent with the tectonic trends. There are strong negative anomaly foci in the east and west syntaxis, whereas no significant differences exist among the Cenozoic blocks in the plateau interior. No direct correspondence exists between the anomaly and the crustal depth. On the background of a weak magnetic anomaly in the plateau, relatively stronger short wavelength fields from the shallower crust are overlapped in the Lhasa, Qilian, Qaidam, and Sichuan-Yunnan rhombic blocks. A strong negative anomaly in the east-west direction is distributed along the Himalayas, mainly caused by the middle and long wavelength bands in the deep and central crust. The magnetic structural layers are stable in the Sichuan and Tarim basins. | 20 | Spatial and temporal variation of haze in China from 1961 to 2012显示文摘The purpose of this study is to analyze the climatic characteristics and long-term spatial and temporal variations of haze occurrence in China. The impact factors of haze trends are also discussed. Meteorological data from 1961 to 2012 and daily PM10 concentrations from2003 to 2012 were employed in this study. The results indicate that the annual-average hazy days at all stations have been increasing rapidly from 4 days in 1961 to 18 days in 2012. The maximum number of haze days occur in winter(41.1%) while the minimum occur in summer(10.4%). During 1961-2012, the high occurrence areas of haze shifted from central to south and east regions of China. The Beijing-Tianjin-Hebei(Jing-Jin-Ji) region, Shanxi,Shaanxi, and Henan Province are the high occurrence areas for haze, while the Yangtze River Delta(YRD) and the Pearl River Delta(PRD) have become regions with high haze occurrences in the last 25 years. Temperature and pressure are positively correlated with the number of haze days. However, wind, relative humidity, precipitation, and sunshine duration are negatively correlated with the number of haze days. The key meteorological factors affecting the formation and dissipation of haze vary for high and low altitudes, and are closely related to anthropogenic activities.
