2Biodegradable nanocarriers for small interfering ribonucleic acid (siRNA) co-delivery strategy increase the chemosensitivity of pancreatic cancer cells to gemcitabine显示文摘Ribonucleic acid (RNA) interference (RNAi) therapies are promising cancertreatment modalities that can specifically target abnormal proto-oncogenes, thusimproving the therapeutic effect. For the treatment of pancreatic cancer, targetingone mutant proto-oncogene by RNAi usually does not yield the desired therapeuticefficiency. Both K-ras gene mutations and Notch1 overexpression are commonsymptoms in pancreatic cancer patients, and play a crucial role in pancreaticcancer cell drug resistance. In this study, biodegradable charged polyester-basedvectors (BCPVs) were synthesized for the co-delivery of K-ras and Notch1 smallinterfering ribonucleic acid (siRNA) into MiaPaCa-2 cells (pancreatic cancercell line) to overcome drug resistance to gemcitabine (GEM), a first-line chem-otherapeutic drug used in the clinic. BCPVs could effectively absorb negative siRNAto form a capsule-like structure, prevent siRNA from nuclease digestion in theserum, and promote effective siRNA cell internalization and endosomal escape.Through K-ras and Notch1 gene silencing in MiaPaCa-2 cells, BCPV-siRNAKsiRNAN^tcm nanocomplexes effectively reversed the epithelia-mesenchymaltransition (EMT) in MiaPaCa-2 cells, thereby greatly enhancing the sensitivity ofMiaPaCa-2 cells to GEM. MiaPaCa-2 cell proliferation, migration, and invasionwere effectively inhibited, and cell apoptosis was also significantly enhanced bythe synergistic antitumor effect of BCPV-siRNAK siRNAN^t~l nanocomplexesand GEM. These results suggest that this combination RNAi therapy can be usedto improve cancer cell sensitivity to chemotherapeutic drugs. 3Total Ginsenoside Extract from Panax ginseng Enhances Neural Stem Cell Proliferation and Neuronal Differentiation by Inactivating GSK-3β显示文摘Objective:To study the effects of total ginsenosides(TG)extract from Panax ginseng on neural stem cell(NSC)proliferation and differentiation and their underlying mechanisms.Methods:The migration of NSCs after treatment with various concentrations of TG extract(50,100,or 200μg/mL)were monitored.The proliferation of NSCs was examined by a combination of cell counting kit-8 and neurosphere assays.NSC differentiation mediated by TG extract was evaluated by Western blotting and immunofluorescence staining to monitor the expression of nestin and microtubule associated protein 2(MAP2).The GSK-3β/β-catenin pathway in TG-treated NSCs was examined by Western blot assay.The NSCs with constitutively active GSK-3βmutant were made by adenovirus-mediated gene transfection,then the proliferation and differentiation of NSCs mediated by TG were further verified.Results:TG treatment significantly enhanced NSC migration(P<0.01 or P<0.05)and increased the proliferation of NSCs(P<0.01 or P<0.05).TG mediation also significantly upregulated MAP2 expression but downregulated nestin expression(P<0.01 or P<0.05).TG extract also significantly induced GSK-3βphosphorylation at Ser9,leading to GSK-3βinactivation and,consequently,the activation of the GSK-3β/β-catenin pathway(P<0.01 or P<0.05).In addition,constitutive activation of GSK-3βin NSCs by the transfection of GSK-3βS9 A mutant was found to significantly suppress TG-mediated NSC proliferation and differentiation(P<0.01 or P<0.05).Conclusion:TG promoted NSC proliferation and neuronal differentiation by inactivating GSK-3β.LIN Kai-li ZHANG Ji CHUNG Hau-lam WU Xin-yi LIU Bin ZHAO Bo-xin SZE Stephen Cho-wing ZHOU Ping-zheng YUNG Ken Kin-lam ZHANG Shi-qing 2022Chinese Journal of Integrative Medicine2022,28,3:2

5ABO blood type, long-standing diabetes, and the risk of pancreatic cancer显示文摘AIM: To retrospectively study pancreatic cancer patients with respect to their ABO blood type and diabetes. METHODS: Our analysis included a cohort of 1017 patients with pancreatic ductal cancer diagnosed at our hospital in Tokyo. They were divided into two groups: 114 patients with long-standing type 2 diabetes (DM group, defined as diabetes lasting for at least three years before the diagnosis of pancreatic cancer) and 903 patients without diabetes (non-DM group). Multivariate analysis was performed to identify factors that are associated with long-standing diabetes. The DM group was further divided into three subgroups according to the duration of diabetes (3-5 years, 5.1-14.9 years, and 15 years or more) and univariate analyses were performed. RESULTS: Of the 883 pancreatic cancer patients with serologically assessed ABO blood type, 217 (24.6%) had blood type O. Compared with the non-DM group, the DM group had a higher frequency of blood type B [odds ratio (OR) = 2.61, 95%CI: 1.24-5.47; reference group: blood type A]. Moreover, male (OR = 3.17, 95%CI: 1.67-6.06), older than 70 years of age (OR = 2.19, 95%CI: 1.20-3.98) and presence of a family history of diabetes (OR = 6.21, 95%CI: 3.38-11.36) were associated with long-standing type 2 diabetes. The mean ages were 64.8 ± 9.2 years, 67.1 ± 9.8 years, and 71.7 ± 7.0 years in the subgroups with the duration of diabetes, 3-5 years, 5.1-14.9 years, and 15 years or more, respectively (P = 0.007). A comparison of ABO blood type distribution among the subgroups also showed a significant difference (P = 0.03).
