1Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically.Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang 2020Cell Research2020,30,1:11
2Advances in Cadaverine Bacterial Production and Its Applications显示文摘本文探讨了微波加热在工业锡粉熔化中的应用。对离心雾化法制备的金属锡粉的形貌和粒径进行了表征。锡粉颗粒均匀且呈球形,90%的颗粒粒径为38~75μm。通过介电性能测试,研究了锡粉的微波吸收特性。微波的穿透作用对锡粉具有良好的整体加热性能。温度高于150℃时,微波加热锡粉的主要机制是电导损耗。设计开发了20 k W微波熔炼锡合金装备,并将其应用于产业化。该装备的加热速率与常规方法相比能提高10倍以上,缩短了熔炼工艺。研究结果表明,微波加热加快了升温速率,缩短了熔炼时间,锡回收率为97.79%,渣量仅为1.65%,其他损失低于0.56%;单位能耗仅为0.17 k W·h·kg^(–1),远低于传统方法所需要的能量。因此微波冶炼提高了加热效率,降低了能耗。Weichao Ma Kequan Chen Yan Li Ning Hao Xin Wang Pingkai Ouyang 2017Engineering2017,3,3:7
3Chemical genomics reveals inhibition of breast cancer lung metastasis by Ponatinib via c-Jun显示文摘Metastasis is the leading cause of human cancer deaths.Unfortunately,no approved drugs are available for antimetastatic treatment.In our study,high-throughput sequencing-based high-throughput screening(HTS^2)and a breast cancer lung metastasis(BCLM)-associated gene signature were combined to discover anti-metastatic drugs.After screening of thousands of compounds,we identified Ponatinib as a BCLM inhibitor.Ponatinib significantly inhibited the migration and mammosphere formation of breast cancer cells in vitro and blocked BCLM in multiple mouse models.Mechanistically,Ponatinib represses the expression of BCLM-associated genes mainly through the ERK/c-Jun signaling pathway by inhibiting the transcription of JUN and accelerating the degradation of c-Jun protein.Notably,JUN expression levels were positively correlated with BCLM-associated gene expression and lung metastases in breast cancer patients.Collectively,we established a novel approach for the discovery of anti-metastatic drugs,identified Ponatinib as a new drug to inhibit BCLM and revealed c-Jun as a crucial factor and potential drug target for BCLM.Our study may facilitate the therapeutic treatment of BCLM as well as other metastases.Wei Shao Shasha Li Lu Li Kequan Lin Xinhong Liu Haiyan Wang Huili Wang Dong Wang 2019Protein & Cell2019,10,3:7
4Diversity of arbuscular mycorrhizal fungal spore communities and its relations to plants under increased temperature and precipitation in a natural grassland显示文摘Arbuscular mycorrhizal fungi(AMF)form mutualisms with most plant species,and therefore,understanding how AMF communities will respond to climate change is essential for predictions of changes in plant communities.To evaluate the impact of global climate change on AMFs and plant-AMF interactions in a natural grassland in Inner Mongolia,both artificial warming and watering treatments were assigned to experimental plots.Our results indicate that(1)warming and precipitation significantly affected the relative spore abundance of abundant sporulating AMF species;(2)the relative abundance of weak sporulating AMF species and AMF diversity decreased under experimental warming;(3)evidence was found that the composition of the AMF community in a given year might be correlated with plant community composition in the following year;and(4)grasses and forbs showing different preferences to Claroideoglomus etunicatum or Ambispora gerdemannii dominated plots.Our results imply that climate change appears to induce changes in AMF assemblages with knock-on effects on grassland plant communities.AMF communities may play a much more important role than we have thought in the responses of ecosystem to global climate changes.SUN XiuFeng SU YuanYing ZHANG Ying WU MingYu ZHANG Zhe PEI KeQuan SUN LiFu WAN ShiQiang LIANG Yu 2013Chinese Science Bulletin2013,58,33:6
5Diversity of root-associated fungi of Vaccinium mandarinorum along a human disturbance gradient in subtropical forests,China显示文摘Aims Ericaceous plant species can host diverse fungi in their roots,including ericoid mycorrhizal fungi(ERMF),endophytes,pathogens and some species with unknown functions.However,how this diversity of fungi responds to different human disturbances is not well understood.Methods In this study,we examined the effects of different human disturbance on fungal diversity in hair roots of Vaccinium mandarinorum,an ericaceous plant.Fungal DNA was extracted from hair roots of V.mandarinorum and high-throughput sequencing was applied to detect the diversity of root-associated fungi along a human disturbance gradient in subtropical forests in Gutianshan National Nature Reserve(GNNR)in East China.The four forest types with different disturbance regime were:old growth forest(OGF),secondary forest with once cut(SEC I),secondary forest with twice cut(SEC II)and Cunninghamia lanceolata plantation(PLF).Important Findings The results showed that:(i)diverse fungal operational units(OTUs)were detected in hair roots of V.mandarinorum in the four types of forests,covering fungal phyla of Ascomycota,Basidiomycota,Chytridiomycota,Glomeromycota and Zygomycota;(ii)Community composition of root-associated fungi of V.mandarinorum in PLF was distinct from those in the other three forest types,and two types of secondary forests had similar fungal community composition;(iii)Different fungal families respond differently to human disturbances:fungal families with significant preference to OGF were ectomycorrhizal or saprophytic fungi while fungal families with higher relative abundance in PLF were plant pathogenic or saprophytic fungi;(iv)The first principal component(PC1)of plant community had a significant effect on composition of root-associated fungal community,while edaphic parameters showed no significant effect on fungal community composition in roots of V.mandarinorum.Our results help to better understand the responses of both ericaceous plants and their fungal partners to human disturbances and forest managements.Yanhua Zhang Jian Ni Fangping Tang Lifen Jiang Tianrong Guo Kequan Pei Lifu Sun Yu Liang 2017Journal of Plant Ecology2017,10,1:6

8Ameliorating end-product inhibition to improve cadaverine production in engineered Escherichia coli and its application in the synthesis of bio-based diisocyanates显示文摘Cadaverine is an important C5 platform chemical with a wide range of industrial applications.However,the cadaverine inhibition on the fermenting strain limited its industrial efficiency of the strain.In this study,we report an engineered Escherichia coli strain with high cadaverine productivity that was generated by developing a robust host coupled with metabolic engineering to mitigate cadaverine inhibition.First,a lysine producing E.coli was treated with a combination of radiation(ultraviolet and visible spectrum)and ARTP(atmospheric and room temperature plasma)mutagenesis to obtain a robust host with high cadaverine tolerance.Three mutant targets including HokD,PhnI and PuuR are identified for improved cadaverine tolerance.Further transcriptome analysis suggested that cadaverine suppressed the synthesis of ATP and lysine precursor.Accordingly,the related genes involved in glycolysis and lysine precursor,as well as cadaverine exporter was engineered to release the cadaverine inhibition.The final engineered strain was fed-batch cultured and a titer of 58.7 g/L cadaverine was achieved with a yield of 0.396 g/g,both of which were the highest level reported to date in E.coli.The bio-based cadaverine was purified to>99.6%purity,and successfully used for the synthesis of polyurethane precursor 1,5-pentamethylene diisocyanate(PDI)through the approach of carbamate decomposition.Xin Wang Xing Guo Jing Wang Hui Li Feng He Sheng Xu Kequan Chen Pingkai Ouyang 2021Synthetic and Systems Biotechnology2021,6,4:3
9METTL5 stabilizes c-Myc by facilitating USP5 translation to reprogram glucose metabolism and promote hepatocellular carcinoma progression显示文摘Background:Hepatocellular carcinoma(HCC)is one of the most prevalent cancers in the world,with a high likelihood of metastasis and a dismal prognosis.The reprogramming of glucosemetabolism is critical in the development ofHCC.TheWarburg effect has recently been confirmed to occur in a variety of cancers,including HCC.However,little is known about the molecular biological mechanisms underlying the Warburg effect in HCC cells.In this study,we sought to better understand how methyltransferase 5,N6-adenosine(METTL5)controls the development of HCC and theWarburg effect.Methods:In the current study,quantitative real-time polymerase chain reaction and Western blotting were used to detect the expression of METTL5 in HCC tissues and cell lines.Several different cell models and animal models were established to determine the role of METTL5 in glucose metabolism reprogramming and the underlying molecularmechanism of HCC.Glutathione-S-transferase pulldown,coimmunoprecipitation,RNA sequencing,non-targeted metabolomics,polysome profiling,and luciferase reporter assays were performed to investigate the molecular mechanisms of METTL5 in HCC cells.Results:We discovered that METTL5 drove glucose metabolic reprogramming to promote the proliferation and metastasis of HCC.Mechanistically,upregulation of METTL5 promoted c-Myc stability and thus activated its downstream glycolytic genes lactate dehydrogenase A(LDHA),enolase 1(ENO1),triosephosphate isomerase 1(TPI1),solute carrier family 2 member 1(SLC2A1),and pyruvate kinase M2(PKM2).The c-Box and ubiquitin binding domain(UBA)regions of ubiquitin specific peptidase 5(USP5)binded to c-Myc protein and inhibited K48-linked polyubiquitination of c-Myc.Further study revealed that METTL5 controled the USP5 translation process,which in turn regulated the ubiquitination of c-Myc.Furthermore,we identified cAMP responsive element binding protein 1(CREB1)/P300 as a critical transcriptional regulator ofMETTL5 that promoted the transcription of METTL5 in HCC.In patient-derived tumor xenograft(PDX)models,adenovirus-mediated knockout of METTL5 had a good antitumor effect and prolonged the survival of PDX-bearing mice.Conclusions:These findings point to a novel mechanism by which CREB1/P300-METTL5-USP5-c-Myc controls abnormal glucose metabolism and promotes tumor growth,suggesting that METTL5 is a potential therapeutic target and prognostic biomarker for HCC.Peng Xia Hao Zhang Haofeng Lu Kequan Xu Xiang Jiang Yuke Jiang Xiangdong Gongye Zhang Chen Jie Liu Xi Chen Weijie Ma Zhonglin Zhang Yufeng Yuan 2023Cancer Communications2023,43,3:3
10The influence of the NCO/OH ratio and the 1,6-hexanediol/ dimethylol propionic acid molar ratio on the properties of waterborne polyurethane dispersions based on 1,5-pentamethylene diisocyanate显示文摘1,5-Pentamethylene diisocyanate, a novel aliphatic diisocyanate formed from bio-based 1,5-pentamethylenediamine, has been used as a hard segmented material to synthesize polyurethane. In this study, several waterborne polyurethane (WPU) dispersions have been successfully prepared by a prepolymer process from 1,5-pentamethylene diisocyanate poly(polyether) with different NCO/OH ratios and 1,6-hexanediol (HDO)/dimethylol propionic acid (DMPA) molar ratios. The Fourier transfonn infrared (FTIR) spectra, thermogravimetric analysis, differential scanning calorimetry, X-ray diffiraction, and a mechanical tensile test were used to investigate the structures, thermal stability, phase separation, crystallinity, mechanical properties, and adhesive performance of the WPU dispersions. The FTIR results indicate that the degree of hydrogen bonding and the numbers of urea groups increase as the NCO/OH ratio and HDO/DMPA molar ratio increase. Furthermore, the phase separation increases and the thermal stability decreases as the NCO/OH ratio increases or the HDO/DMPA molar ratio decreases. Finally, WPU3.0-2.4 (NCO/OH = 3, HDO/DMPA = 2.4) exhibits a maximum tensile strength and shear strength, pointing to its possible use as an adhesive. 11Optical Properties and Source Analysis of Aerosols over a Desert Area in Dunhuang,Northwest China显示文摘Aerosol observational data for 2012 obtained from Dunhuang Station of CARE-China(Campaign on Atmospheric Aerosol Research Network of China) were analyzed to achieve in-depth knowledge of aerosol optical properties over Dunhuang region. The results showed that the annual average aerosol optical depth(AOD) at 500 nm was 0.32 ± 0.06, and the ?ngstr?m exponent(α) was 0.73 ± 0.27. Aerosol optical properties revealed significant seasonal characteristics. Frequent sandstorms in MAM(March–April–May) resulted in the seasonal maximum AOD, 0.41 ± 0.04, and a relatively smaller αvalue, 0.44 ± 0.04. The tourism seasons, JJA(June–July–August) and SON(September–October–November) coincide with serious emissions of small anthropogenic aerosols. While in DJF(December–January–February), the composition of the atmosphere was a mixture of dust particles and polluted aerosols released by domestic heating; the average AOD and αwere 0.29 ± 0.02 and 0.66 ± 0.17, respectively. Different air masses exhibited different degrees of influence on the aerosol concentration over Dunhuang in different seasons. During MAM, ranges of AOD(0.11–1.18) and α(0.06–0.82) were the largest under the dust influence of northwest-short-distance air mass in the four trajectories. Urban aerosols transported by northwest-short-distance air mass accounted for a very large proportion in JJA and the mixed aerosols observed in SON were mainly conveyed by air masses from the west. 12Combination of ARTP mutagenesis and color-mediated highthroughput screening to enhance 1-naphthol yield from microbial oxidation of naphthalene in aqueous system显示文摘Strain QCG of the aerobic bacteria Bacillus cereus is capable of producing l-naphthol from naphtha-lene,this strain was first isolated and characterized in this study.Strain QCG was mutagenized to enhance l-naphthol production,using atmospheric and room temperature plasma(ARTP)technology.Then,a microbial clone screening system was used to accelerate the operation.Meanwhile,a novel color-mediated high-throughput screening using 4-aminoantipyrine was performed to screen mutants.The optimal mutant strain QCG4 produced 19.58土0.34 mg·L-1-naphthol from naphthalene that was 47.32%higher than that of the original strain(13.29+0.28 mg·L-1).In addition,the optimal conditions for l-naphthol production via whole-cell catalysis of strain QCG4 were determined to be an OD600 of40,150 mg.L I naphthalene,and 7.5%dimethyl formamide as a co-solvent at pH 7.5 and 26℃ for 3 h,resulting in 41.18士0.12 mg·L-l-naphthol,i.e.,the mutant strain produces a 2.1-fold higher yield compared to the original strain.Chenggang Qiu Alei Zhang Sha Tao Kang Li Kequan Chen Pingkai Ouyang 2020Frontiers of Chemical Science and Engineering2020,14,5:2

14Efficient production of cyclic adenosine monophosphate from adenosine triphosphate by the N-terminal half of adenylate cyclase from Escherichia coli显示文摘In this study,we aimed at developing an efficient biocatalytic process for bio-production of cyclic adenosine monophosphate(c AMP)from adenosine triphosphate(ATP).First,adenylate cyclase from Escherichia coli MG1655(EAC)and Bordetella Pertussis(BAC)were expressed in E.coli BL21(DE3)and comparatively analyzed for their activities.As a result,EAC from E.coli MG1655 exhibited a higher activity.However,amount of EAC were obtained in an insoluble form.Therefore,we expressed the first 446 amino acids of EAC(EAC446)to avoid the inclusion body.The effects of induction temperature,incubation time,and incubation p H were further evaluated to improve the expression of EAC446.Subsequently,the reaction process for the production of c AMP with ATP as a starting material was investigated.As none of c AMP was detected in the whole-cell based biocatalytic process,the reaction catalyzed by the crude enzyme was determined for c AMP production.What's more,the reaction temperature,reaction p H,metal ion additives and substrate concentration was optimized,and the maximum c AMP production of 18.45 g·L^-1was achieved with a yield of 95.4%after bioconversion of 6 h.Chen Ma Jing Wang Xuelin Wang Dandan Mai Yuqi Jin Kequan Chen Xin Wang Pingkai Ouyang 2020Chinese Journal of Chemical Engineering2020,28,8:1

18Mass Transfer, Gas Holdup, and Kinetic Models of Batch and Continuous Fermentation in a Novel Rectangular Dynamic Membrane Airlift Bioreactor显示文摘Compared with conventional cylinder airlift bioreactors(CCABs)that produce coarse bubbles,a novel rectangular dynamic membrane airlift bioreactor(RDMAB)developed in our lab produces fine bubbles to enhance the volumetric oxygen mass transfer coefficient(k_(L)a)and gas holdup,as well as improve the bioprocess in a bioreactor.In this study,we compared mass transfer,gas holdup,and batch and con-tinuous fermentation for RNA production in CCAB and RDMAB.In addition,unstructured kinetic models for microbial growth,substrate utilization,and RNA formation were established.In batch fermentation,biomass,RNA yield,and substrate utilization in the RDMAB were higher than those in the CCAB,which indicates that dynamic membrane aeration produced a high k_(L)a by fine bubbles;a higher k_(L)a is more bene-ficial to aerobic fermentation.The starting time of continuous fermentation in the RDMAB was 20 h ear-lier than that in the CCAB,which greatly improved the biological process.During continuous fermentation,maintaining the same dissolved oxygen level and a constant dilution rate,the biomass accumulation and RNA concentration in the RDMAB were 9.71% and 11.15% higher than those in the CCAB,respectively.Finally,the dilution rate of RDMAB was 16.7% higher than that of CCAB during con-tinuous fermentation while maintaining the same air aeration.In summary,RDMAB is more suitable for continuous fermentation processes.Developing new aeration and structural geometry in airlift bioreac-tors to enhance k_(L)a and gas holdup is becoming increasingly important to improve bioprocesses in a bioreactor.Ganlu Li Kequan Chen Yanpeng Wei Jinlei Zeng Yue Yang Feng He Hui Li Pingkai Ouyang 2022Engineering2022,8,6:1
