| 2 | Gene Network Analysis and Functional Studies of Senescence-associated Genes Reveal Novel Regulators of Arabidopsis Leaf Senescence显示文摘Plant leaf senescence has been recognized as the last phase of plant development, a highly ordered process regulated by genes known as senescence associated genes (SAGs). However, the function of most of SAGs in regulating leaf senescence as well as regulators of those functionally known SAGs are still unclear. We have previously developed a curated database of genes potentially associated with leaf senescence, the Leaf Senescence Database (LSD). In this study, we built gene networks to identify common regulators of leaf senescence in Arabidopsis thaliana using promoting or delaying senescence genes in LSD. Our results demonstrated that plant hormones cytokinin, auxin, nitric oxide as well as small molecules, such as Ca2+, delay leaf senescence. By contrast, ethylene, ABA, SA and JA as well as small molecules, such as oxygen, promote leaf senescence, altogether supporting the idea that phytohormones play a critical role in regulating leaf senescence. Functional analysis of candidate SAGs in LSD revealed that a WRKY transcription factor WRKY75 and a Cys2/His2-type transcription factor AZF2 are positive regulators of leaf senescence and loss-of-function of WRKY75 or AZF2 delayed leaf senescence. We also found that silencing of a protein phosphatase, AtMKP2, promoted early senescence. | 3 | Effect of dry density on ^(125)I diffusion in GMZ bentonite显示文摘Gaomiaozi(GMZ) bentonite is regarded as the favorable candidate backfilling material for a potential repository in China.It is important to understand the diffusion behavior of 125 I in GMZ bentonite and compare the diffusion behavior in GMZ and other types of bentonite like MX-80,Avonlea,etc.Therefore,through-and out-diffusion experiments were conducted to obtain the effective diffusion coefficient(D e) and distribution coefficient(K d).A computer code named Fitting for diffusion coefficient(FDP) was used for the experimental data processing and theoretical modeling.At the dry density of GMZ bentonite from 1600-2000 kg/m 3,the D e values of 125 I were(2.4-20.4) × 10-12 m 2 /s and K d values were constants.At dry density above 1800 kg/m 3,the diffusion behaviors were almost the same,indicating that the anion exclusion was ineffective.Out-diffusion results showed that the species of 125 I may be changed during the diffusion processing.It was probably caused by some organic matters or reducing substances in GMZ bentonite.Since the main composition of bentonite is montmorillonite,similar diffusion parameters were obtained in GMZ and other types of bentonite.The relationship of D e and accessible porosity(acc) could be described by Archie's law with exponent n = 1.2-2.8 for 125 I diffusion in bentonite,whereas n = 2.0 in GMZ bentonite.Furthermore,bentonite with the dry density of 1800 kg/m 3 was proposed as the backfilling materials used in the construction of high level radioactivity waste repository. | WU Tao LI JinYing DAI Wei XIAO GuoPing SHU FuJun YAO Jun SU YuLan SHI Lei | 2012 | Science China Chemistry2012,55,9: | 11 |

| 5 | Secondary biogenic coalbed gas in some coal fields of China显示文摘The secondary biogenic coalbed gas, a new genetic and energy source type of coalbed gas in China, has been found in Xinji, Liyazhuang and Enhong areas. The essential characteristics of this type of gas are: (i) the major component of the gas is methane, with C1/C1-5 value higher than 0.99, indicating that the gas is part of dry gas; (ii) theδ13C1 value is in the range of -61.7‰to -47.9‰, mostly lower than -55‰, which is much lower than the estimatedδ13C1 value of thermogenic methane according to the thermal evolution degree of the coal rocks (with R0 value from 0.87% to 1.43%), showing the characteristics of the secondary biogenic gas; (iii) theδ5D value of methane ranges from -244‰to -196‰; (iv)δ13C 2 value ranges from -26.7‰to -15.9‰andδ13C 3 value ranges from -10.8‰to -25.3‰, indicating that the heavier hydrocarbons have a thermogenic origin; (v) the content of CO2 is very low, andδ13CCO2 value changes greatly, reflecting a characteristic of secondary change; (vi)δ15N2 value ranges mainly from -1‰to +1‰, indicating N2 derived significantly from air. The negative linear correlation between the contents of N2 and CH4 reflects the activity of bacteria bearing surface water infiltrating into coal beds. The comprehensive tracing indices show that the coalbed gas in the studied areas is the mixed gas of primarily secondary biogenic gas and a part of remnant thermogenic gas. | 6 | The localization of type 2 diabetes susceptibility gene loci in northern Chinese Han families显示文摘We conducted a genome-wide scan, in which 358 well distributed fluorescent dye-labeled microsatellite marker sets were applied in 32 Chinese Han type 2 diabetes families from Northern China to search for the susceptibility gene loci. The data collected from screening all the chromosomes of genome were genotyped by using genescan and genotyping software, then, parametric and non-parametric multipoint test, and affected sib-pair analysis as well, were used to analyze the data. We identified some susceptibility gene loci residing in chromosomes 1,12,18,20, respectively, or precisely, located around D1S214, D1S207, D1S218, D1S235, D12S336, D18S61 and D20S118. | 7 | MoS2 nanorods with inner caves through synchronous encapsulation of sulfur for high performance Li–S cathodes显示文摘Lithium–sulfur(Li–S)batteries have become one of the most promising candidates for next-generation batteries owing to their high specific capacity,low cost,and environment-friendliness.Many efforts have been made to mitigate the'shuttle effect'through physical adsorption and chemical bonding.MoS2 has been proposed as a cathode material to provide effective anchoring sites for lithium polysulfides(Li PSs),but is still limited by its layer structure.Herein,we designed novel MoS2 nanorods with inner caves based on our previous work,and performed synchronous encapsulation of sulfur during the synthesis process.The outer MoS2 tubular shells physically inhibit the outward diffusion of polysulfide species while the inner particles chemically anchor the polysulfides to prevent shuttling.As the cathode matrix in Li–S batteries,the electrochemical results deliver a high initial discharge capacity of 1213 mAhg^-1 for sulfur at 0.1 C.After cycling at 1 C for 300 cycles,the cells exhibit a capacity decay of only 0.076%per cycle and high average coulombic efficiency over 95%.The tubular MoS2 structure is an innovative and appealing design,which could be regarded as a prospective substrate for the improved performance of Li–S batteries. | Yikun Yi Zechen Liu Pu Yang Tao Wang Xuewen Zhao Hongyang Huang Yonghong Cheng Jinying Zhang Mingtao Li | 2020 | Journal of Energy Chemistry2020,29,6: | 5 |
| 8 | HrcQ is necessary for Xanthomonas oryzae pv. oryzae HR-induction in non-host tobacco and pathogenicity in host rice显示文摘Bacterial blight, caused by Xanthomonas oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice(Oryza sativa L.) worldwide. The type III secretion system(T3SS) of Xoo, encoded by the hrp(hypersensitive response and pathogenicity) genes, plays critical roles in conferring pathogenicity in host rice and triggering a hypersensitive response(HR) in non-host plants. To investigate the major genes conferring the pathogenicity and avirulence of Xoo, we previously constructed a random Tn5-insertion mutant library of Xoo strain PXO99A. We report here the isolation and characterization of a Tn5-insertion mutant PXM69. Tn5-insertion mutants were screened on indica rice JG30, which is highly susceptible to PXO99A, by leaf-cutting inoculation.Four mutants with reduced virulence were obtained after two rounds of screening. Among them, the mutant PXM69 had completely lost virulence to the rice host and ability to elicit HR in non-host tobacco. Southern blotting analysis showed a single copy of a Tn5-insertion in the genome of PXM69. PCR walking and sequencing analysis revealed that the Tn5 transposon was inserted at nucleotide position 70,192–70,201 in the genome of PXO99A, disrupting the type III hrc(hrp-conserved) gene hrcQ, the first gene in the D operon of the hrp cluster in Xoo. To confirm the relationship between the Tn5-insertion and the avirulence phenotype of PXM69, we used the marker exchange mutagenesis to create a PXO99Amutant, ΔhrcQ::KAN, in which the hrcQ was disrupted by a kanamycin-encoding gene cassette at the same site as that of the Tn5-insertion. ΔhrcQ::KAN showed the same phenotype as mutant PXM69. Reintroduction of the wild-type hrcQ gene partially complemented the pathogenic function of PXM69. RT-PCR and cellulase secretion assays showed that the Tn5-disruption of hrcQ did not affect transcription of downstream genes in the D operon and function of the type II secretion system. | 10 | Polarization multiplexing for double images display显示文摘Metasurface provides subwavelength structures for manipulating wavefronts of light. The benefits of subwavelength components offer a continuous modulation of amplitude, phase, and polarization, thus eliminating the production of higher-order images and improving the utilization of light intensity. Despite the rapid progress in this field, multiparameter control of light using single layer metasurface is rarely reported. In fact, multiparameter control of light helps to improve information storage capacity and image fidelity. With simultaneous manipulation of polarization and amplitude at each pixel, it is possible to encode two separate images into one metasurface and reconstruct them under proper conditions. In a proof of concept experiment, we demonstrate an independent display of two binary images at the same position with polarization de-multiplexing from a single metasurface. | 13 | UV light tolerance and reactivation potential of tetracycline-resistant bacteria from secondary effluents of a wastewater treatment plant显示文摘Tetracycline-resistant bacteria(TRB) are of concern as emerging microbial contaminants in reclaimed water.To understand the effects of UV disinfection on TRB,both inactivation and reactivation profiles of TRB,as well as 16 tetracycline-resistant isolates from secondary effluent,were characterized in this study.The inactivation ratio of TRB was significantly lower(3.0-log) than that of heterotrophic bacteria(>4.0-log) in the secondary effluent.Additionally,the proportion of TRB significantly increased from 1.65%to 15.51%under20 mJ/cm^2 ultraviolet(UV) exposure.The inactivation rates of tetracycline-resistant isolates ranged from 0.57/s to 1.04/s,of which tetracycline-resistant Enterobacter-1 was the most tolerant to UV light.The reactivation of TRB,tetracycline-resistant isolated strains,as well as heterotrophic bacteria commonly occurred in the secondary effluent even after20 mJ/cm^2 UV exposure.The colony forming ability of TRB and heterotrophic bacteria reached 3.2-log and 3.0-log under 20 mJ/cm^2 UV exposure after 22 hr incubation.The final inactivation ratio of tetracycline-resistant Enterobacter-1 was 1.18-log under 20 mJ/cm^2 UV exposure after 22 hr incubation,which is similar to those of TRB(1.18-log) and heterotrophic bacteria(1.19-log).The increased proportion of TRB and the reactivation of tetracycline-resistant enterobacteria in reclaimed water could induce a microbial health risk during wastewater reuse. | Jing-Jing Huang Jinying Xi Hong-Ying Hu Yi Li Sun-Qin Lu Fang Tang Yu-Chen Pang | 2016 | Journal of Environmental Sciences2016,28,3: | 2 |

| 18 | 2,3-Dihydroxybiphenyl dioxygenase gene was first discovered in Arthrobacter sp. strain PJ3显示文摘Bacterium strain PJ3,isolated from wastewater and identified as Arthrobacter sp. bacterium based on its 16S rDNA gene,could use carbazole as the sole carbon,nitrogen and energy source. The genomic library of strain PJ3 was constructed and a positive clone JM109(pUCW402) was screened out for the expression of dioxygenase by the ability to form yellow ring-fission product. A 2,3-dihydroxybiphenyl dioxygenase(23DHBD) gene of 933 bp was found in the 3360 bp exogenous fragment of pUCW402 by GenSCAN software and BLAST analysis. The phylogenetic analysis showed that 23DHBD from strain PJ3 formed a deep branch separate from a cluster containing most known 23DHBD in GenBank. Southern hybridization confirmed for the first time that the 23DHBD gene was from the genomic DNA of Arthrobacter sp. PJ3. In order to test the gene function,recombinant bacterium BL21(pETW-8) was constructed to express 23DHBD. The expression level in BL21(pETW-8) was highest compared with the recombinant bacteria JM109(pUCW402) and strain PJ3. We observed that 23DHBD was not absolute specific. The enzyme activity was higher with 2,3-dihydroxybiphenyl as a substrate than with catechol.
