1Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum显示文摘1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. 2The characterization of auditory brainstem response(ABR) waveforms: A study in tree shrews(Tupaia belangeri)显示文摘To characterize the patterns of ABR waves in tree shrews, we must understand the hearing sensitivity and auditory function of healthy adult tree shrews. Fifteen tree shrews(30 ears) were stimulated with clicks and tone-pips at 11 different frequencies from 1 to 60 kHz. The ABR waves were recorded and analyzed. The ABR consisted of five to seven positive waves in the first 10 ms after a click stimulus, and the average hearing threshold of component III was 27.86 ± 3.78 dB SPL. Wave III was the largest and most clear. The ABR threshold was related to the tone-pip sitmulus by a 'U' shaped curve. The sensitive frequency was approximately 8 kHz in tree shrews. The latencies systematically decreased with increasing stimulus frequencies. The ABR amplitudes of wave III increased as the sound pressure level increased. 3Marsdenia tenacissima injection induces the apoptosis of prostate cancer by regulating the AKT/GSK3β/STAT3 signaling axis显示文摘Marsdenia tenacissima injection,a standard Marsdenia tenacissima extract(MTE),has been approved as an adjuvant therapeutic agent for various cancers.Our previous study showed that MTE inhibited the proliferation and metastasis of prostate cancer(PCa)cells.However,the underlying mechanisms and active ingredients of MTE against PCa were not completely understood.This study revealed that MTE induced significant decreases in cell viability and clonal growth in PCa cells.In addition,MTE induced the apoptosis of DU145 cells by reducing the mitochondrial membrane potential and increasing the expression of Cleaved Caspase 3/7,Cyt c,and Bax.In vivo,DU145 xenografted NOD-SCID mice treated with MTE showed significantly decreased tumor size.TUNEL staining and Western blot confirmed the pro-apoptotic effects of MTE.Network pharmacology analysis collected 196 ingredients of MTE linked to 655 potential targets,and 709 PCa-associated targets were retrieved,from which 149 overlapped targets were screened out.Pathway enrichment analysis showed that the HIF-1,PI3K-AKT,and ErbB signaling pathways were closely related to tumor apoptosis.Western blot results confirmed that MTE increased the expression of p-AKT^(Ser473) and p-GSK3β^(Ser9),and decreased the expression of p-STAT3^(Tyr705) in vitro and in vivo.A total of 13 compounds in MTE were identified by HPLC-CAD-QTOF-MS/MS and UPLCQTOF-MS/MS.Molecular docking analysis indicated that six compounds may interact with AKT,GSK3β,and STAT3.In conclusion,MTE induces the endogenous mitochondrial apoptosis of PCa by regulating the AKT/GSK3β/STAT3 signaling axis,resulting in inhibition of PCa growth in vitro and in vivo.LI Xiaolan HE Songhua LIANG Wei ZHANG Weiquan CHEN Xin LI Qiaofeng YANG Xin LIU Yanying ZHU Dan LI Li LIU Buming SU Zhiheng CHEN Jie GUO Hongwei 2023Chinese Journal of Natural Medicines2023,21,2:2
4Active anticorrosion and self-healing coatings:A review with focus on multi-action smart coating strategies显示文摘The development of smart coatings with potential for active anticorrosion and self-healing protection of metals is essential for long-term performance of metallic structures in aggressive chemical environments.Presently,emphasis has been placed on the development of advanced smart coatings for corrosion protection in different applications.Innovative multifunctional coatings with fascinating stimuliresponsive functionalities are considered“smart”.The stimuli-responsive functionalities of these smart coatings when properly harnessed result in a class of coatings with inherent autonomous control of corrosion.Fundamentally,when metals are exposed to aggressive environments,occurrences at the metalsolution interface cause environmental changes.These changes can be controlled when triggers from external environment set off active components of smart coating,thereby enhancing coating’s life and functionality.Common triggers include the availability of moisture,concentration of chloride ion,p H gradient,mechanical damage,impact,fatigue,light,redox activity and temperature.In this review,recent technological trends in active anticorrosion and self-healing coatings as functional routes for metal protection are summarized,stimuli responsiveness and mechanisms of inhibition are discussed,and recent multi-action protective systems are particularly focused on.Inime Ime Udoh Hongwei Shi Enobong Felix Daniel Jianyang Li Songhua Gu Fuchun Liu En-Hou Han 2022Journal of Materials Science & Technology2022,,21:2

20Spatial Distribution Pattern of the Steppe Toad-headed Lizard(Phrynocephalus frontalis) and Its Influencing Factors显示文摘Spatial distribution patterns are associated with life history and behavioral adaptations of animals. For studying the spatial distribution pattern of the steppe toad-headed lizard(Phrynocephalus frontalis) and its influencing factors,we conducted experiments in Hunshandake Sandy Land in Inner Mongolia,China in July 2009. By calculating the clustered indices,we found that the lizard was aggregately distributed when the sampling quadrat was smaller than 10 m × 10 m,and uniformly distributed when it was greater than 10 m × 10 m. The Nearest Neighbor Rule showed a clustering distribution pattern for P. frontalis and the distribution pattern was quadrat-sampling dependent. Furthermore,the cluster was determined by environmental factors when the sampling quadrat was smaller than 20 m × 20 m,but it was determined by both environmental factors and characteristics of the lizard when it was larger than 20 m × 20 m. Our results suggested that the steppe toad-headed lizards tended to aggregate into suitable habitat patches in desert areas.
