| 3 | From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. | 5 | Predictors of healthcare-seeking behavior among Chinese patients with irritable bowel syndrome显示文摘AIM To analyze predictors of healthcare-seeking behavior among Chinese patients with irritable bowel syndrome(IBS) and their satisfaction with medical care.METHODS Participating patients met IBS Rome Ⅲ criteria(excluding those with organic diseases) and were enrolled in an IBS database in a tertiary university hospital. Participants completed IBS questionnaires in face-to-face interviews. The questionnaires covered intestinal and extra-intestinal symptoms, medical consultations, colonoscopy,medications, and self-reported response to medications during the whole disease course and in the past year. Univariate associations and multivariate logistic regression were used to identify predictors for frequent healthcareseeking behavior(≥ 3 times/year), frequent colonoscopies(≥ 2 times/year), long-term medications, and poor satisfaction with medical care.RESULTS In total, 516 patients(293 males, 223 females) were included. Participants' average age was 43.2 ± 11.8 years. Before study enrollment, 55.2% had received medical consultations for IBS symptoms. Ordinary abdominal pain/discomfort(non-defecation) was an independent predictor for healthcare-seeking behavior(OR = 2.07, 95%CI: 1.31-3.27). Frequent colonoscopies were reported by 14.7% of patients(3.1 ± 1.4 times per year). Sensation of incomplete evacuation was an independent predictor for frequent colonoscopies(OR = 2.76, 95%CI: 1.35-5.67). During the whole disease course, 89% of patients took medications for IBS symptoms, and 14.7% reported they were satisfied with medical care. Patients with anxiety were more likely to report dissatisfaction with medical care(OR = 2.08, 95%CI: 1.20-3.59). In the past year, patients with severe(OR = 1.74, 95%CI: 1.06-2.82) and persistent(OR = 1.66, 95%CI: 1.01-2.72) IBS symptoms sought medical care more frequently.CONCLUSION Chinese patients with IBS present high rates of frequent healthcare-seeking behavior, colonoscopies, and medications, and low satisfaction with medical care. | 14 | Ectopic expression of a male fertility gene,LOGL8,represses LOG and hinders panicle and ovule development显示文摘Grain number and seed-setting rate are components of crop yield.Cytokinin influences grain yield.However,emerging studies suggest that high cytokinin signals often lead to reduced branching or seed-setting rate,leading to reduced grain yield,although the mechanisms remain unclear.In this study,we identified and characterized the rice(Oryza sativa L.)gene LONELY GUY-LIKE 8(LOGL8),based on analysis of the LOGL8-pm(promoter mutant of LOGL8)mutant,which harbors a T-DNA insertion in the promoter of this gene.The mutation in LOGL8-pm causes ectopic hyperexpression of LOGL8 in inflorescence organs,resulting in plants with smaller panicles and defective ovules lacking archesporial cells and integuments.Knockout of LOGL8 caused pollen abortion,leading to a reduced seed-setting rate.LOGL8 encodes a putative cytokinin-activating enzyme.Our results showed that LOGL8 directly catalyzes the biosynthesis of bioactive cytokinins.Therefore,we propose that the ectopic expression of LOGL8 disrupts cytokinin spatiotemporal distribution and causes inhibition of LONELY GUY(LOG),which affects panicle branching and female organ development.These findings reveal the important role of LOGL8 in male development,and highlight the delicate balance of local cytokinin levels during panicle branching and female organ development. | Shuifu Chen Sulin Lou Xiucai Zhao Shijuan Zhang Letian Chen Ping Huang Guande Li Yingying Li Yao-Guang Liu Yuanling Chen | 2022 | The Crop Journal2022,10,6: | 0 |

| 16 | An Early Paleozoic Ultramafic Complex in the North Wulan Metamorphic Complex,North Qaidam:Contraints on the Nature of the Alaskan-type Continental Arc Root显示文摘Orogenic peridotite is an important component of orogenic belts and retains crucial information on mantle magmatic activity,slab subduction,and melt or fluid metasomatism.To determine the source of the mantle-derived parental magma of the peridotite and to investigate the metasomatism that it experienced,we undertook an integrated study of the petrography,whole-rock major-and trace-element compositions,in situ zircon U-Pb geochronology,and mineral majorand trace-element compositions of an early Paleozoic ultramafic complex in the North Wulan area of North Qaidam.The Halihatu ultramafic-mafic complex is composed of dunite,pyroxene peridotite,and gabbro,which are characteristic of Alaskan-type complexes.The dunite yields a weighted mean^(206)Pb/^(238)U age of 479±5 Ma(MSWD=0.7),which reflects the age of the metasomatism rather than the crystallization age of the ultramafic magma.The peridotites have high Mg^(#)(89.8-91.8)and Cr contents(2419-5190 ppm),low Al_(2)O_(3)(0.20-1.68 wt%)and Ni(289-1012 ppm)contents,and high olivine Fo contents(87-91),suggesting a large degree(~15%-22%)of partial melting of lithospheric ultramafic rocks followed by variable degrees of fractional crystallization of olivine and pyroxene.This is consistent with estimates of 15%-22.3%partial melting calculated using the Cr#of spinel crystals and with the low Yb(0.04-0.33 ppm)and Y(0.72-1.29 ppm)contents of clinopyroxene crystals.Whole-rock trace-element patterns show enrichment in large ion lithophile elements and depletion in high field strength elements,along with high Al_(2)O_(3)(2.10-6.47 wt%)and low TiO_(2)(0.01-0.21 wt%)contents of clinopyroxene crystals,suggesting an arc magma cumulate trend.These features,along with the high olivine Fo contents(87-91 ppm),imply that the Halihatu peridotite is an Alaskan-type crustal cumulates derived from Mgrich hydrous basaltic melts.The high estimated f O_(2)(FMQ+1.97 to FMQ+3.81)further supports the idea that they formed in an arc setting.The Ni/Co and Ni/Mn ratios and cumulate textures of the olivine,quenched boundaries between mafic and felsic melts,and the occurrence of tremolite and phlogopite reflect interactions between the Halihatu peridotite and injected silicate and carbonatitic melts in the lower crust.Therefore,we propose a new cumulate-infiltration model for the petrogenesis of Alaskan-type ultramafic complexes,which improves our understanding of the nature of Alaskan-type continental arc root. | SHAN Jinming NIU Manlan LI Xiucai LI Chen WANG Lei ZHANG Shuai | 2023 | Acta Geologica Sinica(English Edition)2023,97,5: | 0 |
