| 4 | DOCK2 regulates antifungal immunity by regulating RAC GTPase activity显示文摘Fungal infections cause~1.5 million deaths each year worldwide,and the mortality rate of disseminated candidiasis currently exceeds that of breast cancer and malaria.The major reasons for the high mortality of candidiasis are the limited number of antifungal drugs and the emergence of drug-resistant species.Therefore,a better understanding of antifungal host defense mechanisms is crucial for the development of effective preventive and therapeutic strategies.Here,we report that DOCK2(dedicator of cytokinesis 2)promotes indispensable antifungal innate immune signaling and proinflammatory gene expression in macrophages.DOCK2-deficient macrophages exhibit decreased RAC GTPase(Rac family small GTPase)activation and ROS(reactive oxygen species)production,which in turn attenuates the killing of intracellular fungi and the activation of downstream signaling pathways.Mechanistically,after fungal stimulation,activated SYK(spleen-associated tyrosine kinase)phosphorylates DOCK2 at tyrosine 985 and 1405,which promotes the recruitment and activation of RAC GTPases and then increases ROS production and downstream signaling activation.Importantly,nanoparticle-mediated delivery of in vitro transcribed(IVT)Rac1 mRNA promotes the activity of Rac1 and helps to eliminate fungal infection in vivo.Taken together,this study not only identifies a critical role of DOCK2 in antifungal immunity via regulation of RAC GTPase activity but also provides proof of concept for the treatment of invasive fungal infections by using IVT mRNA. | Xiaojian Ma Xi Tan Bingbing Yu Wanwei Sun Heping Wang Huijun Hu Yanyun Du Ruirui He Ru Gao Qianwen Peng Zhihui Cui Ting Pan Xiong Feng Junhan Wang Chengqi Xu Bin Zhu Wei Liu Chenhui Wang | 2022 | Cellular & Molecular Immunology2022,19,5: | 1 |
| 5 | Training needs in intravenous infusion care for children in China:A cross-sectional study显示文摘Aim:This study aims to evaluate demands for general and pharmacological knowledge and training related to intravenous(IV)therapy among pediatric nurses.Materials and Methods:This multicentric cross‑sectional study involved 12,707 pediatric nurses from 100 hospitals in China.A questionnaire was used to assess information about IV drug administration training received,and the demands for pharmacology‑related knowledge,and corresponding methods of acquisition.A generalized linear model using Logit link function was employed to assess relationships between factors and multivariate analysis was conducted.Results:More than 99%of participants showed their training demands for IV therapy training.Pediatric nurses’demands and methods for acquiring IV therapy knowledge and the knowledge related to IV therapy pharmacology have significant differences in social demographic factors,previous knowledge,whether they have received training or not,and other factors(all P<0.05).Received a needlestick injury in the past month(P=0.007)and knowledge acquired through in‑hospital or out‑of‑hospital training(P=0.039)were factors that reduced the demands for methods to acquire further pharmacology‑related knowledge of IV therapy.Working in internal medicine(P=0.025)and not having experienced a needlestick injury in the past year(P=0.007)reduced the demands for IV therapy knowledge.Attended hospital(P=0.007)or departmental meetings(P=0.009),being pediatric primary nurses(P=0.044),and studied special IV guidelines(P=0.006)reduced the desire for methods to acquire IV therapy knowledge.Conclusion:There was a high demand for greater general and pharmacological knowledge related to IV therapy among pediatric nurses.Resources should be coordinated to provide ongoing training to nurses to improve the quality of IV care. | Jingjing LI Jiaxin FANG Liu HAN Junhan YANG Xueyan FAN Dahua ZHANG | 2023 | Journal of Integrative Nursing2023,5,2: | 0 |
| 6 | Biomimetic“Gemini nanoimmunoregulators”orchestrated for boosted photoimmunotherapy by spatiotemporally modulating PD-L1 and tumor-associated macrophages显示文摘A novel strategy of not only stimulating the immune cycle but also modulating the immunosuppressive tumor microenvironment is of vital importance to efficient cancer immunotherapy.Here,a new type of spatiotemporal biomimetic“Gemini nanoimmunoregulators”was engineered to activate robust systemic photoimmunotherapy by integrating the triple-punch of amplified immunogenic cell death(ICD),tumor-associated macrophages(TAMs)phenotype reprogramming and programmed cell death ligand 1(PD-L1)degradation.The“Gemini nanoimmunoregulators”PM@RM-T7 and PR@RM-M2 were constructed by taking the biocompatible mesoporous polydopamine(mPDA)as nanovectors to deliver metformin(Met)and toll-like receptor 7/8 agonist resiquimod(R848)to cancer cells and TAMs by specific biorecognition via wrapping of red blood cell membrane(RM)inlaid with T7or M2 peptides.mPDA/Met@RM-T7(abbreviated as PM@RM-T7)was constructed to elicit an amplified in situ ICD effect through the targeted PTT and effectively stimulated the anticancer immunity.Meanwhile,PD-L1 on the remaining cancer cells was degraded by the burst metformin to prevent immune evasion.Subsequently,mPDA/R848@RM-M2(abbreviated as PR@RM-M2)specifically recognized TAMs and reset the phenotype from M2 to M1 state,thus disrupting the immunosuppressive microenvironment and further boosting the function of cytotoxic T lymphocytes.This pair of sister nanoimmunoregulators cooperatively orchestrated the comprehensive anticancer activity,which remarkably inhibited the growth of primary and distant 4T1 tumors and prevented malignant metastasis.This study highlights the spatiotemporal cooperative modalities using multiple nanomedicines and provides a new paradigm for efficient cancer immunotherapy against metastatic-prone tumors. | Honglin Huang Ningxi Li Xiaodan Wei Qingzhi Li Junhan Guo Geng Yang Hong Yang Lulu Cai Yiyao Liu Chunhui Wu | 2024 | Acta Pharmaceutica Sinica B2024,14,3: | 0 |
