1Transforming growth factor-β in stem cells and tissue homeostasis显示文摘TGF-β 1–3 are unique multi-functional growth factors that are only expressed in mammals, and mainly secreted and stored as a latent complex in the extracellular matrix(ECM). The biological functions of TGF-β in adults can only be delivered after ligand activation, mostly in response to environmental perturbations. Although involved in multiple biological and pathological processes of the human body, the exact roles of TGF-β in maintaining stem cells and tissue homeostasis have not been well-documented until recent advances, which delineate their functions in a given context. Our recent findings, along with data reported by others, have clearly shown that temporal and spatial activation of TGF-β is involved in the recruitment of stem/progenitor cell participation in tissue regeneration/remodeling process, whereas sustained abnormalities in TGF-β ligand activation, regardless of genetic or environmental origin, will inevitably disrupt the normal physiology and lead to pathobiology of major diseases. Modulation of TGF-β signaling with different approaches has proven effective pre-clinically in the treatment of multiple pathologies such as sclerosis/fibrosis, tumor metastasis, osteoarthritis, and immune disorders. 2Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 3Aberrant activation of latent transforming growth factor-β initiates the onset of temporomandibular joint osteoarthritis显示文摘There is currently no effective medical treatment for temporomandibular joint osteoarthritis(TMJ-OA) due to a limited understanding of its pathogenesis. This study was undertaken to investigate the key role of transforming growth factor-β(TGF-β)signalling in the cartilage and subchondral bone of the TMJ using a temporomandibular joint disorder(TMD) rat model, an ageing mouse model and a Camurati–Engelmann disease(CED) mouse model. In the three animal models, the subchondral bone phenotypes in the mandibular condyles were evaluated by μCT, and changes in TMJ condyles were examined by TRAP staining and immunohistochemical analysis of Osterix and p-Smad2/3. Condyle degradation was confirmed by Safranin O staining, the Mankin and OARSI scoring systems and type X collagen(Col X), p-Smad2/3 a and Osterix immunohistochemical analyses. We found apparent histological phenotypes of TMJ-OA in the TMD, ageing and CED animal models, with abnormal activation of TGF-βsignalling in the condylar cartilage and subchondral bone. Moreover, inhibition of TGF-β receptor I attenuated TMJ-OA progression in the TMD models. 5Up-regulation of miR-139-5p protects diabetic mice from liver tissue damage and oxidative stress through inhibiting Notch signaling pathway显示文摘The occurrence and development of diabetes seriously threaten the health of patients.Therefore,the mechanism exploration of diabetes is of great significance for more effective control of this disease.In this study,we aimed to investigate the regulatory mechanism of miR-139-5p and Notch signaling pathway on liver damage and oxidative stress in diabetic mice.The mouse model of diabetes was established,and the mice were divided into normal group,model group,negative control(NC)group,miR-139-5p mimic group,miR-139-5p inhibitor group,DAPT group,and miR-139-5p inhibitor+DAPT group.The mRNA expressions of miR-139-5p,Notchl,Jaggedl,and NICD1,and the protein expressions of Notchl,Jaggedl,and NICD1 were detected.In addition,HepG2 cells were cultured for high glucose induction,and cell cycle distribution and apoptosis were detected by flow cytometry.The results showed that the body weights of mice in the model,NC,miR-139-5p mimic,miR-139-5p inhibitor,DAPT,and miR-139-5p inhibitor+DAPT groups were all lower than that in the normal group.Co-localization of miR-139-5p and Notchl was observed in the fluorescence in situ hybridization assay,and miR-139-5p was found to negatively regulate Notch!Furthermore,reduced blood glucose level and inhibited liver oxidative stress were observed in mice with miR-139-5p overexpression or DAPT treatment.DAPT treatment reversed the increase of blood glucose level and oxidative stress injury caused by miR-139-5p silencing.In conclusion,upregulation of miR-139-5p expression can protect liver tissue from oxidative stress injury in diabetic mice,and its mechanism may be related to the inhibition of Notch signaling pathway.Hua Wei Liwei Huang Fenghua Wei Guangzhi Li Bin Huang Jun Li Cong Cao 2020Acta Biochimica et Biophysica Sinica2020,52,4:3

8Fluid simulation of internal leakage for continuous rotary electro-hydraulic servo motor显示文摘The internal leakage has important influence on low speed performance of continuous rotary electro-hydraulic motors,and research on internal leakage is of important significance to improve the motor low speed performance. UG software is adopted to establish a flow field inside a motor,which ignores the gap between the blade and the blade groove,and a motor fluid model in different working conditions is established. Using ICEM-CFD to generate high-quality O-Net mesh for meeting the computing requirements,the flow field inside the motor is simulated using fluid simulation software CFX. Applying reasonable boundary conditions and then solving it,pressure field distribution and the variation rule of internal leakage with different pressure difference in the flow field are obtained when the clearance value of the motor key components is 0.01 mm. 11Tetrahedral framework nucleic acids promote the biological functions and related mechanism of synovium-derived mesenchymal stem cells and show improved articular cartilage regeneration activity in situ显示文摘Many recent studies have shown that joint-resident mesenchymal stem cells(MSCs)play a vital role in articular cartilage(AC)in situ regeneration.Specifically,synovium-derived MSCs(SMSCs),which have strong chondrogenic differentiation potential,may be the main driver of cartilage repair.However,both the insufficient number of MSCs and the lack of an ideal regenerative microenvironment in the defect area will seriously affect the regeneration of AC.Tetrahedral framework nucleic acids(tFNAs),notable novel nanomaterials,are considered prospective biological regulators in biomedical engineering.Here,we aimed to explore whether tFNAs have positive effects on AC in situ regeneration and to investigate the related mechanism.The results of in vitro experiments showed that the proliferation and migration of SMSCs were significantly enhanced by tFNAs.In addition,tFNAs,which were added to chondrogenic induction medium,were shown to promote the chondrogenic capacity of SMSCs by increasing the phosphorylation of Smad2/3.In animal models,the injection of tFNAs improved the therapeutic outcome of cartilage defects compared with that of the control treatments without tFNAs.In conclusion,this is the first report to demonstrate that tFNAs can promote the chondrogenic differentiation of SMSCs in vitro and enhance AC regeneration in vivo,indicating that tFNAs may become a promising therapeutic for AC regeneration.Liwei Fu Pinxue Li Junyao Zhu Zhiyao Liao Cangjian Gao Hao Li Zhen Yang Tianyuan Zhao Wei Chen Yu Peng Fuyang Cao Chao Ning Xiang Sui Quanyi Guo Yunfeng Lin Shuyun Liu 2022Bioactive Materials2022,7,3:1

17Fault-Tolerant Routing Algorithm for Network-on-Chip Based on Dynamic XY Routing显示文摘In order to ensure the reliability of network-on-chip(NoC) under faulty circumstance,a dynamic fault tolerant routing algorithm is proposed.This algorithm can implement detour routing when there are both static and dynamic permanent faults in the network.That means the packet is able to move around the faults to the destination with a non-minimum path.In addition,the multilevel congestion control mechanism gives the algorithm the ability to distribute the load over the whole network and to avoid hotspots around the faults.Simulation results demonstrate the advantage of the proposed routing algorithm in terms of average packet latency and packet loss rate compared with negative-first routing algorithm and DyAD routing algorithm in the presence of permanent faults.For the proposed algorithm,it can get much less average packet latency and lead to less than 20% packet loss rate.LI Xiaohui CAO Yang WANG Liwei CAI Tian 2009Wuhan University Journal of Natural Sciences2009,14,4:1

19Deep learning model improves radiologists'performance in detection and classification of breast lesions显示文摘Objective:Computer-aided diagnosis using deep learning algorithms has been initially applied in the field of mammography,but there is no large-scale clinical application.Methods:This study proposed to develop and verify an artificial intelligence model based on mammography.Firstly,mammograms retrospectively collected from six centers were randomized to a training dataset and a validation dataset for establishing the model.Secondly,the model was tested by comparing 12 radiologists’performance with and without it.Finally,prospectively enrolled women with mammograms from six centers were diagnosed by radiologists with the model.The detection and diagnostic capabilities were evaluated using the freeresponse receiver operating characteristic(FROC)curve and ROC curve.Results:The sensitivity of model for detecting lesions after matching was 0.908 for false positive rate of 0.25 in unilateral images.The area under ROC curve(AUC)to distinguish the benign lesions from malignant lesions was0.855[95%confidence interval(95%CI):0.830,0.880].The performance of 12 radiologists with the model was higher than that of radiologists alone(AUC:0.852 vs.0.805,P=0.005).The mean reading time of with the model was shorter than that of reading alone(80.18 s vs.62.28 s,P=0.032).In prospective application,the sensitivity of detection reached 0.887 at false positive rate of 0.25;the AUC of radiologists with the model was 0.983(95%CI:0.978,0.988),with sensitivity,specificity,positive predictive value(PPV),and negative predictive value(NPV)of94.36%,98.07%,87.76%,and 99.09%,respectively.Conclusions:The artificial intelligence model exhibits high accuracy for detecting and diagnosing breast lesions,improves diagnostic accuracy and saves time.Yingshi Sun Yuhong Qu Dong Wang Yi Li Lin Ye Jingbo Du Bing Xu Baoqing Li Xiaoting Li Kexin Zhang Yanjie Shi Ruijia Sun Yichuan Wang Rong Long Dengbo Chen Haijiao Li Liwei Wang Min Cao 2021Chinese Journal of Cancer Research2021,33,6:1
20Suppression of LjBAK1-mediated immunity by SymRK promotes rhizobial infection in Lotus japonicus.显示文摘An important question in biology is how organisms can associate with different microbes that pose no threat (commensals), pose a severe threat (pathogens) and those that are beneficial (symbionts). The root nodule symbiosis serves as important model system to address such questions in the context of plant-microbe interactions. It is now generally accepted that rhizobia have the abilities to actively suppress host immune responses during the infection process, analogous to the way in which plant pathogens can evade immune recognition. However, much remains to be elucidated with regard to the mechanisms by which the host recognizes the rhizobia as pathogens and how, subsequently, these pathways are suppressed to allow establishment of the nitrogen fixing symbiosis. In this study, we found that SymRK (Symbiosis Receptor-like Kinase) is required for rhizobial suppression of plant innate immunity in Lotus japonicus. SymRK associates with LjBAK1 (BRASSINOSTEROID INSENSITIVE 1-Associated receptor Kinase 1), a well characterized, positive regulator of plant innate immunity, and directly inhibits LjBAK1 kinase activity. Rhizobial inoculation enhances the association between SymRK and LjBAK1 in planta. LjBAK1 is required to regulate plant innate immunity and plays a negative role in mediating rhizobial infection in L. japonicus.
