| 1 | Mechanically induced Ca^(2+) oscillations in osteocytes release extracellular vesicles and enhance bone formation显示文摘The vast osteocytic network is believed to orchestrate bone metabolic activity in response to mechanical stimuli through production of sclerostin, RANKL, and osteoprotegerin(OPG). However, the mechanisms of osteocyte mechanotransduction remain poorly understood. We've previously shown that osteocyte mechanosensitivity is encoded through unique intracellular calcium (Ca^(2+) ) dynamics. Here, by simultaneously monitoring Ca^(2+) and actin dynamics in single cells exposed to fluid shear flow, we detected actin network contractions immediately upon onset of flow-induced Ca^(2+) transients, which were facilitated by smooth muscle myosin and further confirmed in native osteocytes ex vivo. Actomyosin contractions have been linked to the secretion of extracellular vesicles(EVs), and our studies demonstrate that mechanical stimulation upregulates EV production in osteocytes through immunostaining for the secretory vesicle marker Lysosomal-associated membrane protein 1(LAMP1) and quantifying EV release in conditioned medium, both of which are blunted when Ca^(2+) signaling was inhibited by neomycin. Axial tibia compression was used to induce anabolic bone formation responses in mice, revealing upregulated LAMP1 and expected downregulation of sclerostin in vivo. This load-related increase in LAMP1 expression was inhibited in neomycin-injected mice compared to vehicle.Micro-computed tomography revealed significant load-related increases in both trabecular bone volume fraction and cortical thickness after two weeks of loading, which were blunted by neomycin treatment. In summary, we found mechanical stimulation of osteocytes activates Ca^(2+) -dependent contractions and enhances the production and release of EVs containing bone regulatory proteins. | 2 | Current Status and Future Challenges of Weather Radar Polarimetry: Bridging the Gap between Radar Meteorology/Hydrology/Engineering and Numerical Weather Prediction显示文摘After decades of research and development, the WSR-88 D(NEXRAD) network in the United States was upgraded with dual-polarization capability, providing polarimetric radar data(PRD) that have the potential to improve weather observations,quantification, forecasting, and warnings. The weather radar networks in China and other countries are also being upgraded with dual-polarization capability. Now, with radar polarimetry technology having matured, and PRD available both nationally and globally, it is important to understand the current status and future challenges and opportunities. The potential impact of PRD has been limited by their oftentimes subjective and empirical use. More importantly, the community has not begun to regularly derive from PRD the state parameters, such as water mixing ratios and number concentrations, used in numerical weather prediction(NWP) models.In this review, we summarize the current status of weather radar polarimetry, discuss the issues and limitations of PRD usage, and explore potential approaches to more efficiently use PRD for quantitative precipitation estimation and forecasting based on statistical retrieval with physical constraints where prior information is used and observation error is included. This approach aligns the observation-based retrievals favored by the radar meteorology community with the model-based analysis of the NWP community. | 3 | Initiation of Parkinson’s disease from gut to brain byδ-secretase显示文摘Lewy pathology,composed ofα-Synuclein(α-Syn)inclusions,a hallmark of Parkinson’s disease(PD),progressively spreads from the enteric nervous system(ENS)to the central nervous system(CNS).However,it remains unclear how this process is regulated at a molecular level.Here we show thatδ-secretase(asparagine endopeptidase,AEP)cleaves bothα-Syn at N103 and Tau at N368,and mediates their fibrillization and retrograde propagation from the gut to the brain,triggering nigra dopaminergic neuronal loss associated with Lewy bodies and motor dysfunction.α-Syn N103 and Tau N368 robustly interact with each other and are highly elevated in PD patients’gut and brain.Chronic oral administration of the neurotoxin rotenone induces AEP activation andα-Syn N103/Tau N368 complex formation in the gut,eliciting constipation and dopaminergic neuronal death in an AEP-dependent manner.Preformed fibrils(PFFs)ofα-Syn N103/Tau N368 are more neurotoxic and compact,and aggregate more quickly along the vagus nerve than their FL/FL counterparts or the individual fragments’fibrils.Colonic injection of PFFs induces PD pathologies,motor dysfunctions,and cognitive impairments.Thus,δ-secretase plays a crucial role in initiating PD pathology progression from the ENS to the CNS. | Eun Hee Ahn Seong Su Kang Xia Liu Guiqin Chen Zhentao Zhang Bindu Chandrasekharan Ashfaqul M.Alam Andrew S.Neish Xuebing Cao Keqiang Ye | 2020 | Cell Research2020,30,1: | 9 |

| 6 | Mobility in China,2020:a tale of four phases显示文摘2020 was an unprecedented year;with rapid and drastic changes in human mobility due to the COVID-19 pandemic.To understand the variation in commuting patterns among the Chinese population across stable and unstable periods,we used nationwide mobility data from 318 million mobile phone users in China to examine the extreme fluctuations of population movements in 2020,ranging from the Lunar New Year travel season(chunyun),to the exceptional calm of COVID-19 lockdown,and then to the recovery period.We observed that cross-city movements;which increased substantially in chunyun and then dropped sharply during the lockdown,are primarily dependent on travel distance and the socio-economic development of cities.Following the Lunar New Year holiday,national mobility remained low until mid-February,and COVID-19 interventions delayed more than 72.89 million people returning to large cities.Mobility network analysis revealed clusters of highly connected cities,conforming to the social-economic division of urban agglomerations in China.While the mass migration back to large cities was delayed,smaller cities connected more densely to form new clusters.During the recovery period after travel restrictions were lifted,the netflows of over 55%city pairs reversed in direction compared to before the lockdown.These findings offer the most comprehensive picture of Chinese mobility at fine resolution across various scenarios in China and are of critical importance for decision making regarding future public-health-emergency response,transportation planning and regional economic development,among others. | Suoyi Tan Shengjie Lai Fan Fang Ziqiang Cao Bin Sai Bing Song Bitao Dai Shuhui Guo Chuchu Liu Mengsi Cai Tong Wang Mengning Wang Jiaxu Li Saran Chen Shuo Qin Jessica R.Floyd Zhidong Cao Jing Tan Xin Sun Tao Zhou Wei Zhang Andrew J.Tatem Petter Holme Xiaohong Chen Xin Lu | 2021 | National Science Review2021,8,11: | 3 |
